Data were analyzed with the following steps: (1) identify the data and classifies data based on type, recalled strategy, context, and tells of the effects of linguistic politeness strategies in the directive speech act, (2) connecting the data with the data of the other, (3 ) conducted a study of data inference.The findings of this study are as follows. First, there are 5 types of directive speech acts, that is telling, asking, advising, challenging, and suggest. Second, there are 3 strategies recalled, that tells directly without further ado, instantly recalled with niceties positive politeness, recalled instantly with niceties negative politeness. Evidence for conserved DNA and histone H3 methylation reprogramming in mouse, bovine and rabbit zygotes
<p>Abstract</p> <p>Background</p> <p>In mammals the parental genomes are epigenetically reprogrammed after fertilization. This reprogramming includes a rapid demethylation of the paternal (sperm-derived) chromosomes prior to DNA replication in zygotes. Such active DNA demethylation in the zygote has been documented for several mammalian species, including mouse, rat, pig, human and cow, but questioned to occur in rabbit.</p> <p>Results</p> <p>When comparing immunohistochemical patterns of antibodies against 5-methyl-cytosine, H3K4me3 and H3K9me2 modifications we observe similar pronuclear distribution and dynamics in mouse, bovine and rabbit zygotes. In rabbit DNA demethylation of the paternal chromosomes occurs at slightly advanced pronuclear stages. We also show that the rabbit oocyte rapidly demethylates DNA of donor fibroblast after nuclear transfer.</p> <p>Conclusion</p> <p>Our data reveal that major events of epigenetic reprogramming during pronuclear maturation, including mechanisms of active DNA demethylation, are apparently conserved among mammalian species.</p
Spectral and Timing Analysis of the accretion-powered pulsar 4U 1626-67 observed with Suzaku and NuSTAR
We present an analysis of the spectral shape and pulse profile of the
accretion-powered pulsar 4U 1626-67 observed with Suzaku and NuSTAR during a
spin-up state. The pulsar, which experienced a torque reversal to spin-up in
2008, has a spin period of 7.7 s. Comparing the phase-averaged spectra obtained
with Suzaku in 2010 and with NuSTAR in 2015, we find that the spectral shape
changed between the two observations: the 3-10 keV flux increased by 5% while
the 30-60 keV flux decreased significantly by 35%. Phase-averaged and
phase-resolved spectral analysis shows that the continuum spectrum observed by
NuSTAR is well described by an empirical NPEX continuum with an added broad
Gaussian emission component around the spectral peak at 20 keV. Taken together
with the observed Pdot value obtained from Fermi/GBM, we conclude that the
spectral change between the Suzaku and NuSTAR observations was likely caused by
an increase of the accretion rate. We also report the possible detection of
asymmetry in the profile of the fundamental cyclotron line. Furthermore, we
present a study of the energy-resolved pulse profiles using a new relativistic
ray tracing code, where we perform a simultaneous fit to the pulse profiles
assuming a two-column geometry with a mixed pencil- and fan-beam emission
pattern. The resulting pulse profile decompositions enable us to obtain
geometrical parameters of accretion columns (inclination, azimuthal and polar
angles) and a fiducial set of beam patterns. This information is important to
validate the theoretical predictions from radiation transfer in a strong

reChIP-seq reveals widespread bivalency of H3K4me3 and H3K27me3 in CD4+ memory T cells
The combinatorial action of co-localizing chromatin modifications and
regulators determines chromatin structure and function. However, identifying
co-localizing chromatin features in a high-throughput manner remains a
technical challenge. Here we describe a novel reChIP-seq approach and tailored
bioinformatic analysis tool, normR that allows for the sequential enrichment
and detection of co-localizing DNA-associated proteins in an unbiased and
genome-wide manner. We illustrate the utility of the reChIP-seq method and
normR by identifying H3K4me3 or H3K27me3 bivalently modified nucleosomes in
primary human CD4+ memory T cells. We unravel widespread bivalency at
hypomethylated CpG-islands coinciding with inactive promoters of developmental
regulators. reChIP-seq additionally uncovered heterogeneous bivalency in the
population, which was undetectable by intersecting H3K4me3 and H3K27me3 ChIP-
seq tracks. Finally, we provide evidence that bivalency is established and
stabilized by an interplay between the genome and epigenome. Our reChIP-seq
approach augments conventional ChIP-seq and is broadly applicable to unravel
combinatorial modes of action
Acceptance of Enhanced Robotic Assistance Systems in People With Amyotrophic Lateral Sclerosis–Associated Motor Impairment: Observational Online Study
Background:
Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by a progressive paresis of the extremities and the loss of manual functioning. Due to the severe functional impairment that the disease entails, ALS requires the provision of comprehensive nursing care and a complex set of assistive technology devices. To relieve caregivers and promote autonomy of people with ALS, robotic assistance systems are being developed. This trial aims to evaluate the acceptance of technology, in general, and of robotic arm assistance among people with ALS in order to lay the groundwork for the development of a semiautomatic robotic arm that can be controlled by humans via a multimodal user interface and that will allow users to handle objects and attend to their own bodies.
Objective:
The aim of this study was to perform a systematic analysis of technology commitment and acceptance of robotic assistance systems from the perspective of physically limited people living with ALS.
Methods:
The investigation was conducted as a study of a prospective cohort. Participants were only included if they had received a medical diagnosis of ALS. Data collection took place via an online questionnaire on the Ambulanzpartner Soziotechnologie internet platform. Technological commitment was measured using the Neyer short scale. Furthermore, a multidimensional questionnaire was specially developed to analyze participant acceptance of robotic arm assistance: the Acceptance Measure of Robotic Arm Assistance (AMRAA). This questionnaire was accompanied by a video introducing the robot arm. ALS severity was ascertained using the ALS Functional Rating Scale–Extended (ALSFRS-EX).
Results:
A total of 268 people with ALS participated in the survey. Two-thirds of the participants were male. The overall mean ALS severity score was 42.9 (SD 11.7) points out of 60 on the ALSFRS-EX, with the most relevant restrictions on arms and legs (<60% of normal functioning). Technological commitment ranked high, with the top third scoring 47.2 points out of 60. Younger participants and males showed significantly higher values. The AMRAA score was, again, significantly higher among younger participants. However, the gender difference within the overall cohort was not significant. The more limited the arm functioning of participants according to the ALSFRS-EX subscale, the higher the acceptance rate of robotic assistance. This relationship proved significant.
Conclusions:
People with ALS display high technological commitment and feel positive about using technological assistance systems. In our study, younger participants were more open to technology use, in general, and robotic assistance, in particular. Self-appraisal of technology acceptance, competence, and control conviction were generally higher among men. However, any presumed gender difference vanished when users were asked to rate the anticipated usefulness of the technology, in particular the robotic arm. The acceptance was also reflected in users’ increased willingness to use a robotic arm as the functionality of their own arms decreased. From the perspective of people with ALS, robotic assistance systems are critical to promoting individual autonomy. Another key consideration in the development of future assistive technologies should be the reduction of caregiver burden.

Zurich Consensus: German Expert Opinion on the St. Gallen Votes on 15 March 2009 (11th International Conference at St. Gallen: Primary Therapy of Early Breast Cancer)
A German working group of 23 breast cancer experts discussed the results from the vote at this year's St. Gallen Consensus Conference on Primary Therapy for Early Breast Cancer ( March 11-14, 2009) and came up with some concrete recommendations for day-to-day therapeutic decisions in Germany. Due the fact that the concept of the St. Gallen Consensus Conference merely allows for a minimal consensus, the objective of the working group was to provide practice-related recommendations for day-to-day clinical decisions in Germany. One area of emphasis at St. Gallen was tumor biology as a starting point for reaching individual therapeutic decisions. Intensive discussion was necessary with respect to the clinical relevance of predictive and prognostic factors. A new addition to the area of systemic therapy was a first-ever discussion of the adjuvant administration of bisphosponates and the fact that therapy with trastuzumab in HER2 overexpressing breast cancer has been defined as the standard for neoadjuvant therapy. Glutamatergic and Resting-State Functional Connectivity Correlates of Severity in Major Depression – The Role of Pregenual Anterior Cingulate Cortex and Anterior Insula
Glutamatergic mechanisms and resting-state functional connectivity alterations have been recently described as factors contributing to major depressive disorder (MDD). Furthermore, the pregenual anterior cingulate cortex (pgACC) seems to play an important role for major depressive symptoms such as anhedonia and impaired emotion processing. We investigated 22 MDD patients and 22 healthy subjects using a combined magnetic resonance spectroscopy (MRS) and resting-state functional magnetic resonance imaging (fMRI) approach. Severity of depression was rated using the 21-item Hamilton depression scale (HAMD) and patients were divided into severely and mildly depressed subgroups according to HAMD scores. Because of their hypothesized role in depression we investigated the functional connectivity between pgACC and left anterior insular cortex (AI). The sum of Glutamate and Glutamine (Glx) in the pgACC, but not in left AI, predicted the resting-state functional connectivity between the two regions exclusively in depressed patients. Furthermore, functional connectivity between these regions was significantly altered in the subgroup of severely depressed patients (HAMD > 15) compared to healthy subjects and mildly depressed patients. Similarly the Glx ratios, relative to Creatine, in the pgACC were lowest in severely depressed patients. These findings support the involvement of glutamatergic mechanisms in severe MDD which are related to the functional connectivity between pgACC and AI and depression severity
The Venus Emissivity Mapper Concept
Based on experience gained from using the VIRTIS instrument on Venus Express to observe the surface of Venus and the new high temperature laboratory experiments, we have developed the multispectral Venus Emissivity Mapper (VEM) to study the surface of Venus. VEM imposes minimal requirements on the spacecraft and mission design and can therefore be added to any future Venus mission. Ideally, the VEM instrument will be combined with a high-resolution radar mapper to provide accurate topographic information, as it will be the case for the NASA Discovery VERITAS mission or the ESA EnVision M5 proposal
