    Hearing-Related Health Among Adult American Indians From a Pacific Northwest Tribe

    INTRODUCTION: Hearing loss and tinnitus are common in most populations, although few data have addressed hearing-related health among tribal members and the need for public health interventions. METHODS: This cross-sectional study examined prevalence and risk factors for hearing loss and tinnitus among 217 adults in a Pacific Northwest tribe. Frequency measures were conducted for difficulty hearing certain sounds and hearing aid use. In 2006, risk factors were examined for two outcomes-hearing loss and tinnitus-with analysis conducted in the same year. RESULTS: Although self-reported hearing loss was more common in men (24%) than women (13%), a larger percentage of women compared with men reported difficulty hearing certain sounds. Only 8% of study participants reported hearing aid use. After age adjustment, significant noise exposure was associated with hearing loss (OR=8.30, 95% CI=1.84, 37.52). The overall prevalence of tinnitus was 33% (similar in men and women). After adjusting for age, the odds of tinnitus in individuals with more than four ear infections was 4.77 (95% CI=1.89, 12.02) times the odds in those who never had an ear infection. Tinnitus was also associated with significant noise exposure (OR=2.24, 95% CI=1.28, 6.73) even after age adjustment. CONCLUSIONS: Increasing age and significant noise exposure were associated with hearing loss in this tribe. Tinnitus was associated with significant noise exposure and history of otitis media, even after age adjustment. Public health efforts are needed to improve hearing-related health in this tribe through messages about noise exposure and use of hearing protection

    Do we miss the hot spots? ? The use of very high resolution aerial photographs to quantify carbon fluxes in peatlands

    International audienceAccurate determination of carbon balances in heterogeneous ecosystems often requires the extrapolation of point based measurements. The ground resolution (pixel size) of the extrapolation base, e.g. a land-cover map, might thus influence the calculated carbon balance, in particular if biogeochemical hot spots are small in size. In this paper, we test the effects of varying ground resolution on the calculated carbon balance of a boreal peatland consisting of hummocks (dry), lawns (intermediate) and flarks (wet surfaces). The generalizations in lower resolution imagery led to biased area estimates for individual micro-site types. While areas of lawns and hummocks were stable below a threshold resolution of ~60 cm, the maximum of the flark area was located at resolutions below 25 cm and was then decreasing with coarsening resolution. Using a resolution of 100 cm instead of 6 cm led to an overestimation of total CO2 uptake of the studied peatland area (approximately 14 600 m2) of ~6% and an underestimation of total CH4 emission of ~11%. To accurately determine the surface area of scattered and small-sized micro-site types in heterogeneous ecosystems (e.g. flarks in peatlands), a minimum ground resolution appears necessary. In our case this leads to a recommended resolution of 25 cm, which can be derived by conventional airborne imagery. The usage of high resolution imagery from commercial satellites, e.g. Quickbird, however, is likely to underestimate the surface area of biogeochemical hot spots. It is important to note that the observed resolution effect on the carbon balance estimates can be much stronger for other ecosystems than for the investigated peatland where the relative hot spot area of the flarks is very small and their hot spot characteristics with respect to CH4 and CO2 fluxes is rather modest

    IL-15 trans-presentation by pulmonary dendritic cells promotes effector CD8 T cell survival during influenza virus infection

    We have recently demonstrated that peripheral CD8 T cells require two separate activation hits to accumulate to high numbers in the lungs after influenza virus infection: a primary interaction with mature, antigen-bearing dendritic cells (DCs) in the lymph node, and a second, previously unrecognized interaction with MHC I–viral antigen–bearing pulmonary DCs in the lungs. We demonstrate that in the absence of lung-resident DC subsets, virus-specific CD8 T cells undergo significantly increased levels of apoptosis in the lungs; however, reconstitution with pulmonary plasmacytoid DCs and CD8α+ DCs promotes increased T cell survival and accumulation in the lungs. Further, our results show that the absence of DCs after influenza virus infection results in significantly reduced levels of IL-15 in the lungs and that pulmonary DC–mediated rescue of virus-specific CD8 T cell responses in the lungs requires trans-presentation of IL-15 via DC-expressed IL-15Rα. This study demonstrates a key, novel requirement for DC trans-presented IL-15 in promoting effector CD8 T cell survival in the respiratory tract after virus infection, and suggests that this trans-presentation could be an important target for the development of unique antiviral therapies and more effective vaccine strategies

    Dancers experience daily pain that impacts their health, but the pain is mediated by coping with pain. We elaborated the Scale of Coping with Pain for Dancers (COPAIN-Dancer) based on the Motivational Theory of Coping with 12 categories of coping. This study presents the validity of the COPAIN-Dancer. The content analysis conducted judges reduced 118 items to 35, that were then administered to 386 Brazilian dancers between 10 and 19 years old (M=15.35; SD=3.36). Applying the Exploratory Factor Analysis, a low correlation resulted among the items that represent the 12 coping categories. However, after conducting Item Response Theory and Network Analysis, we obtained two factors: Adaptive Coping and Maladaptive Coping. The COPAIN-DANCER shows reliability and can be used in adolescent dancers.Los bailarines viven diariamente con dolores que afectan su salud, pero que son mediados por el proceso de afrontamiento. Este estudio presenta la Escala de afrontamiento del dolor para bailarines (COPAIN-Dancer), basado en la Motivational Theory of Coping, y sus evidencias de validez. Jueces evaluaron 118 ítems a su validez de contenido. Los ítems se redujeron a 35 y se administraron a 386 bailarines entre 10-19 años (M=15.35; DE=3.36). Aplicando análisis factorial exploratorio (EPT), se dio una baja correlación entre los ítems que representan a las 12 familias de afrontamiento teorizadas. Sin embargo, utilizando la Teoría de Respuesta al Ítem y Análisis de Redes, se obtuvo dos factores: Afrontamiento Adaptativo y Afrontamiento Maladaptivo. El COPAIN-Dancer muestra fiabilidad, y puede ser utilizado en bailarines adolescentes.Les danseurs vivent chaque jour dans une douleur qui a un impact sur leur santé, mais ils sont médiés par le processus d’adaptation. Pour accéder à ce processus, nous avons élaboré l’Échelle d’Adaptation à la Douleur pour les Danseurs (COPAIN-DANCER), basée sur la Motivational Theory of Coping, avec 12 familles de coping.     Pan-Cancer Analysis of lncRNA Regulation Supports Their Targeting of Cancer Genes in Each Tumor Context

    Long noncoding RNAs (lncRNAs) are commonly dys-regulated in tumors, but only a handful are known toplay pathophysiological roles in cancer. We inferredlncRNAs that dysregulate cancer pathways, onco-genes, and tumor suppressors (cancer genes) bymodeling their effects on the activity of transcriptionfactors, RNA-binding proteins, and microRNAs in5,185 TCGA tumors and 1,019 ENCODE assays.Our predictions included hundreds of candidateonco- and tumor-suppressor lncRNAs (cancerlncRNAs) whose somatic alterations account for thedysregulation of dozens of cancer genes and path-ways in each of 14 tumor contexts. To demonstrateproof of concept, we showed that perturbations tar-geting OIP5-AS1 (an inferred tumor suppressor) andTUG1 and WT1-AS (inferred onco-lncRNAs) dysre-gulated cancer genes and altered proliferation ofbreast and gynecologic cancer cells. Our analysis in-dicates that, although most lncRNAs are dysregu-lated in a tumor-specific manner, some, includingOIP5-AS1, TUG1, NEAT1, MEG3, and TSIX, synergis-tically dysregulate cancer pathways in multiple tumorcontexts

    Pan-cancer Alterations of the MYC Oncogene and Its Proximal Network across the Cancer Genome Atlas

    Although theMYConcogene has been implicated incancer, a systematic assessment of alterations ofMYC, related transcription factors, and co-regulatoryproteins, forming the proximal MYC network (PMN),across human cancers is lacking. Using computa-tional approaches, we define genomic and proteo-mic features associated with MYC and the PMNacross the 33 cancers of The Cancer Genome Atlas.Pan-cancer, 28% of all samples had at least one ofthe MYC paralogs amplified. In contrast, the MYCantagonists MGA and MNT were the most frequentlymutated or deleted members, proposing a roleas tumor suppressors.MYCalterations were mutu-ally exclusive withPIK3CA,PTEN,APC,orBRAFalterations, suggesting that MYC is a distinct onco-genic driver. Expression analysis revealed MYC-associated pathways in tumor subtypes, such asimmune response and growth factor signaling; chro-matin, translation, and DNA replication/repair wereconserved pan-cancer. This analysis reveals insightsinto MYC biology and is a reference for biomarkersand therapeutics for cancers with alterations ofMYC or the PMN

    Spatial Organization and Molecular Correlation of Tumor-Infiltrating Lymphocytes Using Deep Learning on Pathology Images

    Beyond sample curation and basic pathologic characterization, the digitized H&E-stained images of TCGA samples remain underutilized. To highlight this resource, we present mappings of tumorinfiltrating lymphocytes (TILs) based on H&E images from 13 TCGA tumor types. These TIL maps are derived through computational staining using a convolutional neural network trained to classify patches of images. Affinity propagation revealed local spatial structure in TIL patterns and correlation with overall survival. TIL map structural patterns were grouped using standard histopathological parameters. These patterns are enriched in particular T cell subpopulations derived from molecular measures. TIL densities and spatial structure were differentially enriched among tumor types, immune subtypes, and tumor molecular subtypes, implying that spatial infiltrate state could reflect particular tumor cell aberration states. Obtaining spatial lymphocytic patterns linked to the rich genomic characterization of TCGA samples demonstrates one use for the TCGA image archives with insights into the tumor-immune microenvironment

    Dystrophin Is Required for the Normal Function of the Cardio-Protective KATP Channel in Cardiomyocytes

    Duchenne and Becker muscular dystrophy patients often develop a cardiomyopathy for which the pathogenesis is still unknown. We have employed the murine animal model of Duchenne muscular dystrophy (mdx), which develops a cardiomyopathy that includes some characteristics of the human disease, to study the molecular basis of this pathology. Here we show that the mdx mouse heart has defects consistent with alteration in compounds that regulate energy homeostasis including a marked decrease in creatine-phosphate (PC). In addition, the mdx heart is more susceptible to anoxia than controls. Since the cardio-protective ATP sensitive potassium channel (KATP) complex and PC have been shown to interact we investigated whether deficits in PC levels correlate with other molecular events including KATP ion channel complex presence, its functionality and interaction with dystrophin. We found that this channel complex is present in the dystrophic cardiac cell membrane but its ability to sense a drop in the intracellular ATP concentration and consequently open is compromised by the absence of dystrophin. We further demonstrate that the creatine kinase muscle isoform (CKm) is displaced from the plasma membrane of the mdx cardiac cells. Considering that CKm is a determinant of KATP channel complex function we hypothesize that dystrophin acts as a scaffolding protein organizing the KATP channel complex and the enzymes necessary for its correct functioning. Therefore, the lack of proper functioning of the cardio-protective KATP system in the mdx cardiomyocytes may be part of the mechanism contributing to development of cardiac disease in dystrophic patients
