    Increased Circulating VAP-1 Levels Are Associated with Liver Fibrosis in Chronic Hepatitis C Infection

    Vascular adhesion protein-1 (VAP-1) is a multifunction protein. While membrane-bound VAP-1 is an adhesion protein, soluble VAP-1 catalyzes the deamination of primary amines through its semicarbazide-sensitive amino oxidase (SSAO) activity. VAP-1 supports the transmigration of leukocytes and increases oxidative stress. In chronic liver diseases, it plays a role in leukocyte infiltration and fibrogenesis. Here, we measured VAP-1 plasma concentration and its SSAO activity in 322 patients with chronic hepatitis C infection and evaluated the association of VAP-1 with fibrosis stages. VAP-1 concentration strongly correlated with liver stiffness and was the second strongest influencing variable after gamma-glutamytransferase (GGT) for liver stiffness in regression analysis. The VAP-1 concentration increased with advancing fibrosis stages and the highest concentrations were found in patients with cirrhosis. According to the receiver operating characteristic (ROC) analysis, a VAP-1 cut-off value of 541 ng/mL predicted histologically confirmed cirrhosis (sensitivity 74%; specificity 72%). SSAO activity correlated only moderately with liver stiffness, showing a relatively small increase in advanced fibrosis. To our knowledge, this is the first study on VAP-1 in chronic hepatitis C infection showing its association with progressive fibrosis. In conclusion, VAP-1 plasma concentration, rather than its SSAO activity, may represent a non-invasive biomarker for monitoring fibrogenesis in patients with chronic hepatitis C infection

    Prevalence and determinants of osteoporosis in patients with type 1 and type 2 diabetes mellitus

    Background: Increased risk of osteoporosis and its clinical significance in patients with diabetes is controversial. We analyze osteoporosis prevalence and determinants of bone mineral density (BMD) in patients with type 1 and 2 diabetes. Methods: Three hundred and ninety-eight consecutive diabetic patients from a single outpatient clinic received a standardized questionnaire on osteoporosis risk factors, and were evaluated for diabetes-related complications, HbA1c levels, and lumbar spine (LS) and femoral neck (FN) BMD. Of these, 139 (71 men, 68 women) type 1 and 243 (115 men, 128 women) type 2 diabetes patients were included in the study. BMD (T-scores and values adjusted for age, BMI and duration of disease) was compared between patient groups and between patients with type 2 diabetes and population-based controls (255 men, 249 women).     First data set of H<sub>2</sub>O/HDO columns from the Tropospheric Monitoring Instrument (TROPOMI)

    Global measurements of atmospheric water vapour isotopologues aid to better understand the hydrological cycle and improve global circulation models. This paper presents a new data set of vertical column densities of H2O and HDO retrieved from short-wave infrared (2.3 µm) reflectance measurements by the Tropospheric Monitoring Instrument (TROPOMI) onboard the Sentinel-5 Precursor satellite. TROPOMI features daily global coverage with a spatial resolution of up to 7 km×7 km. The retrieval utilises a profile-scaling approach. The forward model neglects scattering, and strict cloud filtering is therefore necessary. For validation, recent ground-based water vapour isotopologue measurements by the Total Carbon Column Observing Network (TCCON) are employed. A comparison of TCCON δD with ground-based measurements by the Multi-platform remote Sensing of Isotopologues for investigating the Cycle of Atmospheric water (MUSICA) project for data prior to 2014 (where MUSICA data are available) shows a bias in TCCON δD estimates. As TCCON HDO is currently not validated, an overall correction of recent TCCON HDO data is derived based on this finding. The agreement between the corrected TCCON measurements and co-located TROPOMI observations is good with an average bias of (−0.2±3)×1021^{21} molec cm2^{-2} ((1.1±7.2) %) in H2_{2}O and (−2±7)×1017^{17} molec cm2^{-2} ((−1.1±7.3) %) in HDO, which corresponds to a mean bias of (−14±17) ‰ in a posteriori δD. The bias is lower at low- and mid-latitude stations and higher at high-latitude stations.     Comprehensive Analyses of Coagulation Parameters in Patients with Vascular Anomalies.

    BACKGROUND Vascular anomalies comprise a diverse group of rare diseases with altered blood flow and are often associated with coagulation disorders. The most common example is a localized intravascular coagulopathy in venous malformations leading to elevated D-dimers. In severe cases, this may progress to a disseminated intravascular coagulopathy with subsequent consumption of fibrinogen and thrombocytes predisposing to serious bleeding. A separate coagulopathy is the Kasabach-Merritt phenomenon in kaposiform hemangioendothelioma characterized by platelet trapping leading to thrombocytopenia and eventually consumptive coagulopathy. Our previous work showed impaired von Willebrand factor and platelet aggregometry due to abnormal blood flow, i.e., in ventricular assist devices or extracorporeal membrane oxygenation. With altered blood flow also present in vascular anomalies, we hypothesized that, in particular, the von Willebrand factor parameters and the platelet function may be similarly impacted. METHODS We prospectively recruited 73 patients with different vascular anomaly entities and analyzed their coagulation parameters. RESULTS Acquired von Willebrand syndrome was observed in both of our patients with Kasabach-Merritt phenomenon. In six out of nine patients with complex lymphatic anomalies, both the vWF antigen and activity were upregulated. Platelet aggregometry was impaired in both patients with Kasabach-Merritt phenomenon and in seven out of eight patients with an arteriovenous malformation. CONCLUSIONS The analysis of coagulation parameters in our patients with vascular anomalies advanced our understanding of the underlying pathophysiologies of the observed coagulopathies. This may lead to new treatment options for the, in part, life-threatening bleeding risks in these patients in the future

    Flux Periodicities in Loops of Nodal Superconductors

    Supercurrents in superconducting flux threaded loops are expected to oscillate with the magnetic flux with a period of hc/2e. This is indeed true for s-wave superconductors larger than the coherence length xi_0. Here we show that for superconductors with gap nodes, there is no such strict condition for the supercurrent to be hc/2e rather than hc/e periodic. For nodal superconductors, the flux induced Doppler shift of the near nodal states leads to a flux dependent occupation probability of quasi-particles circulating clockwise and counter clockwise around the loop, which leads to an hc/e periodic component of the supercurrent, even at zero temperature. We analyze this phenomenon on a cylinder in an approximative analytic approach and also numerically within the framework of the BCS theory. Specifically for d-wave pairing, we show that the hc/e periodic current component decreases with the inverse radius of the loop and investigate its temperature dependence

    Osteocalcin Is Independently Associated with C-Reactive Protein during Lifestyle-Induced Weight Loss in Metabolic Syndrome

    Bone-derived osteocalcin has been suggested to be a metabolic regulator. To scrutinize the relation between osteocalcin and peripheral insulin sensitivity, we analyzed changes in serum osteocalcin relative to changes in insulin sensitivity, low-grade inflammation, and bone mineral density following lifestyle-induced weight loss in individuals with metabolic syndrome (MetS). Participants with MetS were randomized to a weight loss program or to a control group. Before and after the 6-month intervention period, clinical and laboratory parameters and serum osteocalcin levels were determined. Changes in body composition were analyzed by dual-energy X-ray absorptiometry (DXA). In participants of the intervention group, weight loss resulted in improved insulin sensitivity and amelioration of inflammation. Increased serum levels of osteocalcin correlated inversely with BMI (r = −0.63; p < 0.001), total fat mass (r = −0.58, p < 0.001), total lean mass (r = −0.45, p < 0.001), C-reactive protein (CRP) (r = −0.37; p < 0.01), insulin (r = −0.4; p < 0.001), leptin (r = −0.53; p < 0.001), triglycerides (r = −0.42; p < 0.001), and alanine aminotransferase (ALAT) (r = −0.52; p < 0.001). Regression analysis revealed that osteocalcin was independently associated with changes in CRP but not with changes in insulin concentration, fat mass, or bone mineral density, suggesting that weight loss-induced higher serum osteocalcin is primarily associated with reduced inflammation

    Monitoring System for Laboratory Mice Transportation: A Novel Concept for the Measurement of Physiological and Environmental Parameters

    Laboratory mice are used in biomedical research as “models” for studying human disease. These mice may be subject to significant levels of stress during transportation that can cause alterations that could negatively affect the results of the performed investigation. Here, we present the design and realization of a prototypical transportation container for laboratory mice, which may contribute to improved laboratory animal welfare. This prototype incorporates electric potential integrated circuit (EPIC) sensors, which have been shown to allow the recording of physiological parameters (heart rate and breathing rate) and other sensors for recording environmental parameters during mouse transportation. This allows for the estimation of the stress levels suffered by mice. First experimental results for capturing physiological and environmental parameters are shown and discussed
