    Gene expression studies in Arabidopsis in response to South African Cassava Mosaic Virus infection utilizing microarrays

    Student Number : 9610284H - MSc dissertation - School of Molecular and Cell Biology - Faculty of ScienceCassava Mosaic Disease is the most devastating disease affecting cassava (Manihot esculenta Crantz) crops worldwide. This disease is associated with eight species of geminiviruses, all belonging to the genus Begomovirus of the family Geminiviridae. In South Africa, in particular, CMD is caused by South African cassava mosaic virus (SACMV). Currently, there are no adequate methods for control of this disease as mechanisms within virus-host interactions are poorly understood. This brings about the need for development of virus-disease control strategies. This study was therefore conducted to identify the host’s response to an invading virus. The model plant, Arabidopsis was chosen as it is a well-characterized plant system, with expression databases readily available as its entire genome has been sequenced. This study was conducted, firstly, to phenotypically determine if Arabidopsis was resistant or susceptible to SACMV infection, and secondly, to identify the host’s response to pathogen infection on a molecular level through gene expression studies utilizing microarrays. Results from the symptomatology study revealed that Arabidopsis plants were fully symptomatic 28 days post-inoculation, displaying characteristic disease symptoms such as stunting, yellowing, and leaf deformation. This indicated that Arabidopsis was susceptible to SACMV infection. Microarray analyses revealed 86 differentially expressed genes, of which 48 showed up-regulation and 38 down-regulation. Relative quantification real-time PCR was performed on selected genes to confirm these results. Many up-regulated genes were shown to be primarily involved in a general stress response induced by the host, whereas those genes that were downregulated seemed to be involved in more specific responses to viral invasion, probably a consequence of suppression of host genes by SACMV to enhance its own replication. The majority of genes identified fell under the predominant functional categories involved in metabolism, transcription, and transport.     Professionals Who Care: A Nonprofit Design for Inclusivity of Caregivers in the Workplace

    Informal caregivers provide essential care to the disabled, ill, aging, or injured, services that are valued over $470 billion annually in the United States. Research has shown that nearly half of these caregivers had no choice in taking on their role, and that this population experiences significant financial, physical, and mental health struggles. 32.3 million of these caregivers must balance both care and job responsibilities, and these employed caregivers face widespread discrimination in the workplace. The bias against care providers is based on historic expectations of what makes an ideal worker: traditional hours, uninterrupted, and in person. These cultural standards have limited relevancy in today’s society with high rates of caregiving in the home, advanced technology, and the proven success of remote and flexible work during the covid-19 pandemic. While the workplace has made strides in recognizing the benefits and becoming more thoughtful of diversity-equity-inclusion issues and employment well-being, employed caregivers often encounter experiences as an invisible and marginalized class. This report will explore the struggles of employed caregivers, describe workplace advocacy gaps, and detail the design, benefits, and incorporation for Professionals Who Care, a nonprofit dedicated to inclusivity for employed caregivers in the workplace

    Do GPs use electronic mental health resources? A qualitative study

    BACKGROUNDThe Better Outcomes in Mental Health Care (BOMHC) initiative encourages general practitioners to use electronic mental health resources (EMHRs) during consultation with patients requiring psychological assistance. However, there is little data on GPs&rsquo; acceptance and use of EMHRs.METHODSemistructured interviews were conducted with 27 GPs to determine their attitude toward EMHRs, and their use during consultation with patients.RESULTSFew GPs reported frequently using EMHRs in consultation. Identified barriers to use included lack of familiarity with information technology, and insufficient knowledge of available resources. Identified advantages of electronic resources included high patient acceptance, time efficiency, and improved quality of information.DISCUSSIONGeneral practitioners recognise several advantages of utilising electronic resources for managing patients with mental illness. However, GPs are not sufficiently familiar with electronic resources to use them effectively. This could be overcome by education.<br /

    Urban Living Labs: how to enable inclusive transdisciplinary research?

    The Urban Living Lab (ULL) approach has the potential to create enabling environments for social learning and to be a successful arena for innovative local collaboration in knowledge co-creation and experimentation in the context of research and practice in sustainability transitions. Nevertheless, complex issues such as the urban Food-Water-Energy (FWE) Nexus present a challenge to the realization of such ULL, especially regarding their inclusiveness. We present ULL as a frame for a local knowledge co-creation and participation approach based on the project "Creating Interfaces - Building capacity for integrated governance at the Food-Water-Energy-nexus in cities on the water". This project aims at making FWE Nexus linkages better understandable to the stakeholders (citizens and associations, city government, science, businesses), and to facilitate cooperation and knowledge exchange among them. This paper focuses on and discusses inclusiveness as a key aspect and challenge of ULLs and on what literature and our experiences in this regard suggest for the advancement of the concept of ULL towards ULL 2.0. These findings often also relate to framing transdisciplinary research in a wider sense

    A case study in leveraging strategic partnerships through trust-based philanthropy

    This practice note highlights a case study of leveraging strategic partnerships through trust-based philanthropy, a set of practices rooted in values, relationship building, mutual learning, and equity. It describes the motivations, planning, and execution of a symposium organized by, and held for, a Foundation and four of its grantees. The symposium led to the development of sustained pathways between and among the partners, resulting in productive collaborations and shared projects. This case study is shared to illustrate the argument that it is the responsibility of funders, and certainly in their self-interest, to eliminate competition between organizations to whom they provide financial resources and support. By facilitating trust and collaboration, funders are uniquely positioned to foster collective, higher-impact work. © 2024 The Authors.     Digital tools for knowledge exchange and sustainable public food procurement in community kindergartens: A case study in SƂupsk, Poland

    This article presents a case study on the experimental co-creation process of a digital platform supporting Sustainable Public Food Procurement (SPFP) in public kindergartens in a medium-sized city in Poland. The organisation of SPFP requires a dedicated technological infrastructure to ensure the information flow among food producers, kindergarten employees, children and parents. To this end, a digital platform was designed to enable contact, assessment of food quality and food procurement environmental impact, and the communication of needs and problems among all the actors involved in the food procurement system for kindergartens. The article also discusses the results of the field research and the method of Urban Living Labs, highlighting the key challenges faced by those seeking to combine knowledge about food and the natural environment with public food procurement. The principal difficulties include the availability, accessibility and possible application of data on the environmental costs of food production, the individualisation of needs and motivations related to public catering in educational facilities, and the specific nature of the public sector responsible for public food procurement

    A SMART decade: outcomes of an integrated, inclusive, first-year college-level STEM curricular innovation

    In the early 2000s, our primarily undergraduate, white institution (PUI/PWI), began recruiting and enrolling higher numbers of students of color and first-generation college students. However, like many of our peer institutions, our established pedagogies and mindsets did not provide these students an educational experience to enable them to persist and thrive in STEM. Realizing the need to systematically address our lack of inclusivity in science majors, in 2012 faculty from multiple disciplines developed the Science, Math, and Research Training (SMART) program. Here, we describe an educational innovation, originally funded by a grant from the Howard Hughes Medical Institute, designed to support and retain students of color, first generation college students, and other students with marginalized identities in the sciences through a cohort-based, integrated, and inclusive first-year experience focused on community and sense of belonging. The SMART program engages first-year students with semester-long themed courses around “real world” problems of antibiotic resistance and viral infections while integrating the fields of Biology, Chemistry, Mathematics, and an optional Computer Science component. In the decade since its inception, 97% of SMART students have graduated or are on track to graduate, with 80.9% of these students earning a major in a STEM discipline. Here, we present additional student outcomes since the initiation of this program, results of the student self-evaluative surveys SALG and CURE, and lessons we have learned from a decade of this educational experience

    Validation of a breast cancer assay for radiotherapy omission: an individual participant data meta-analysis

    Background: There are currently no molecular tests to identify individual breast cancers where radiotherapy (RT) offers no benefit. Profile for the Omission of Local Adjuvant Radiotherapy (POLAR) is a 16-gene molecular signature developed to identify low risk cancers where RT will not further reduce recurrence rates. Methods: An individual participant data meta-analysis was performed in 623 cases of node-negative ER+/HER2-negative early breast cancer enrolled in three RT randomized trials for whom primary tumor material was available for analysis. A Cox proportional hazards model on time to locoregional recurrence (LRR) was used to test the interaction between POLAR score and RT.Results: 429 (69%) patients’ tumors had a high POLAR score and 194 (31%) had a low score. Patients with high POLAR score had, in the absence of RT, a 10-year cumulative incidence of LRR: 20% (15%-26%) vs 5% (2%-11%) for those with a low score. Patients with a high POLAR score had a large benefit from RT (hazard ratio [HR] for RT vs no RT: 0.37 [0.23-0.60], p&lt;0.001). In contrast, there was no evidence of benefit from RT for patients with a low POLAR score (HR: 0.92 [0.42-2.02], p = 0.832). The test for interaction between RT and POLAR was statistically significant (p = 0.022).Conclusions: POLAR is not only prognostic for locoregional recurrence but also predictive of benefit from radiotherapy in selected patients. Patients ≄ 50 years with ER+/HER2-negative disease and a low POLAR score could consider omitting adjuvant RT. Further validation in contemporary clinical cohorts is required.<br/
