    SYNTHESIS AND IN-VITRO ANTIMICROBIAL STUDIES OF SOME NEW PYRAZOLONES

    In the present scenario antibacterial agents are the greatest contribution of chemotherapy. They have great importance in the developing countries where infectious diseases predominate. Pyrazolone derivatives are an important class of heterocyclic compounds that occur in many drugs and synthetic products. It has a particular value due to their broad spectrum of biological activities and their utility as synthetic tools in the design of various bioactive molecules. It is also exhibited analgesic, antipyretic and anti-inflammatory activity. In the present study, a series of 3-methyl-N-(substituted phenyl)-5-oxo-4,5-dihydro-1H-pyrazole-1-carbothioamide (3a-g) were synthesized by the reaction of thiosemicarbazides (1) and ethylacetoacetate (2) in DMF. The synthesized compounds were characterized on the basis of IR, 1H-NMR, 13C-NMR and elemental analysis. All synthesized compounds were screened for antimicrobial activities using disc diffusion technique against four bacterial pathogens viz Staphylococcus aureus, Escherichia coli, Bacillus anthracis and Pseudomonas aeruginosa & two fungal pathogens namely Candida albicanes, Aspergillus niger. Chloramphenicol and Fluconazole were used as standard drug respectively. The compounds exhibit moderate activity

    Development of stability indicating assay method for the simultaneous estimation of Metformin hydrochloride and Glipizide by RP-HPLC method

    A simple, sensitive an isocratic RP-HPLC method for the estimation of Metformin Hydrochloride (MET) and Glipizide (GPZ) in combined dosage form using Inertsil C8 column (250.6 mm, 5 ) in an isocratic mode with mobile phase comprising Acetonitrile: Water (70:30) and one drop of Triethylamine. The flow rate was 1.0 ml/min and effluent was monitored at 222 nm. The retention times were found to be 3.40 min for MET and 4.44 min for GPZ. The assay exhibited a linear dynamic range of 100-500 g/ml of MET and 1-5 g/mL for GPZ .The calibration curves were linear (r = 0.9985 for MET and r = 0.9955 for GPZ) over the entire linear range. Recovery was found to be 99.94 % + .17 for MET and 99.61 % + 0.89 for GPZ. % RSD of system precision were observed 0.0429 for MET and 0.7212 for GPZ

    A uracil nitroso amine based colorimetric sensor for the detection of Cu²⁺ ions from aqueous environment and its practical applications

    A simple uracil nitroso amine based colorimetric chemosensor (UNA-1) has been synthesized and screened for its cation recognition ability. Sensor UNA-1 exhibited a high sensitivity and selectivity towards Cu²⁺ ions in aqueous medium in the presence of a wide range of other competing cations (Ag⁺, Al³⁺, Ba²⁺+, Ca²⁺, Cd²⁺, Co²⁺, Cr³⁺, Cs⁺, Fe²⁺, Fe³⁺, Li⁺, Mg²⁺, Mn²⁺, Na⁺, Ni²⁺, Pb²⁺, Zn²⁺, Hg²⁺ and Sr²⁺). With Cu²⁺, the sensor UNA-1 gave a distinct color change from colorless to dark yellow by forming a complex of 1:1 stoichiometry. Furthermore, sensor UNA-1 was successfully utilized in the preparation of test strips and supported silica for the detection of Cu²⁺ ions from aqueous environment

    Prevalence of hypertension and its association with anthropometric parameters in adult population of Raipur city, Chhattisgarh, India

    Background: Obesity is now an important emerging public health problem in India. It is one of the major risk factor for hypertension. Overweight persons have two to six fold increase in the risk of developing hypertension. This study was carried out with objective to find out prevalence of Hypertension and assess its association with four obesity-related indices- body mass index (BMI), waist circumference (WC), waist-to-hip ratio (WHR), and waist-to-height ratio (WHtR).Methods: A cross-sectional community-based study was done among 640 study subjects of age group 25-59 years during July 2015 to June 2016. Multi stage random sampling method was used. Data collection was done using WHO STEPS instrument V 3.1. Privacy and confidentiality of data was maintained. Data was entered in the Microsoft excel, collected data was checked for its completeness and correctness before data analysis with the help of PSPP software.Results: Prevalence of hypertension among study subjects was found to be 22.19% (142 out of 640).With respect to body mass index, waist circumference, waist hip ratio and waist height ratio, 4.38%, 64.35%, 85.33%, 76.2% were found to be obese respectively. Hypertension was strongly associated with BMI (p<0.001), waist circumference (p<0.001), waist hip ratio (p<0.05), waist height ratio (p<0.05). Out of the four anthropometric variables waist hip ratio (WHR) was most strongly associated with hypertension (β=420.236, p=0.007).Conclusions: The study shows a rising burden of hypertension among study population. As WHR is the best indicator for measuring obesity, so measurement of WHR should be made compulsory in healthy life style clinics along with other parameters

    RF Based Remote Control for Electrical Appliances

    This work presented here is to control electrical appliances through RF based remote system. From anywhere without any line of sight, RF based wireless remote control system can change the state of the electrical appliances either in off state or in on state. The controlling circuit is built around RF transmitter and RF Receiver modules which are operating at certain frequency along with a encoder and a decoder with few passive components. The four different channels at the encoder IC are used as input switches and the four channels at the decoder output are connected to the electrical devices through a relay. Here the transmission technique is amplitude shift keying (ASK) and the circuit is powered with 9 Volt. The main objective of this work is to control electrical appliances without line of sight requirement using the RF technology. It has many applications like we can control any independent electrical appliance such as T.V, room light, fan just from a remote. Operating them manually is a tedious job and become hectic sometimes. If one can control devices like fan, TV, lights and music system with a remote from a distance place just by pressing the button, life will become simpler. This will make our life more comfortable and easier

    Chemosensor for micro to nano-molar detection of Ag+ and Hg2+ ions in pure aqueous media and its applications in cell imaging

    The pyridine substituted thiourea derivative PTB-1 was synthesized and characterized by spectroscopic techniques as well as by single crystal X-ray crystallography. The metal ion sensing ability of PTB-1 was explored by various experimental (naked-eye, UV-Vis, fluorescence, mass spectrometry and 1H NMR spectroscopy) and theoretical (B3LYP/6-31G**/LANL2DZ) methods. PTB-1 exhibited a highly selective naked-eye detectable color change from colorless to dark brown and UV-Vis spectral changes for the detection of Ag+ with a detection limit of 3.67 µM in aqueous medium. The detection of Ag+ ions was achieved by test paper strip and supported silica methods. In contrast, PTB-1 exhibited a 23-fold enhanced emission at 420 nm in the presence of Hg2+ ions with a nano-molar detection limit of 0.69 nM. Finally, the sensor PTB-1 was applied successfully for the intracellular detection of Hg2+ ions in a HepG2 liver cell line, which was monitored by use of confocal imaging techniques

    The malleable brain: plasticity of neural circuits and behavior: A review from students to students

    One of the most intriguing features of the brain is its ability to be malleable, allowing it to adapt continually to changes in the environment. Specific neuronal activity patterns drive long-lasting increases or decreases in the strength of synaptic connections, referred to as long-term potentiation (LTP) and long-term depression (LTD) respectively. Such phenomena have been described in a variety of model organisms, which are used to study molecular, structural, and functional aspects of synaptic plasticity. This review originated from the first International Society for Neurochemistry (ISN) and Journal of Neurochemistry (JNC) Flagship School held in Alpbach, Austria (Sep 2016), and will use its curriculum and discussions as a framework to review some of the current knowledge in the field of synaptic plasticity. First, we describe the role of plasticity during development and the persistent changes of neural circuitry occurring when sensory input is altered during critical developmental stages. We then outline the signaling cascades resulting in the synthesis of new plasticity-related proteins, which ultimately enable sustained changes in synaptic strength. Going beyond the traditional understanding of synaptic plasticity conceptualized by LTP and LTD, we discuss system-wide modifications and recently unveiled homeostatic mechanisms, such as synaptic scaling. Finally, we describe the neural circuits and synaptic plasticity mechanisms driving associative memory and motor learning. Evidence summarized in this review provides a current view of synaptic plasticity in its various forms, offers new insights into the underlying mechanisms and behavioral relevance, and provides directions for future research in the field of synaptic plasticity.Fil: Schaefer, Natascha.     Moving with the Times: The Health Science Alliance (HSA) Biobank, Pathway to Sustainability

    Human biobanks are recognised as vital components of translational research infrastructure. With the growth in personalised and precision medicine, and the associated expansion of biomarkers and novel therapeutics under development, it is critical that researchers can access a strong collection of patient biospecimens, annotated with clinical data. Biobanks globally are undertaking transformation of their operating models in response to changing research needs; transition from a ‘classic’ model representing a largely retrospective collection of pre-defined specimens to a more targeted, prospective collection model, although there remains a research need for both models to co-exist. Here we introduce the Health Science Alliance (HSA) Biobank, established in 2012 as a classic biobank, now transitioning to a hybrid operational model. Some of the past and current challenges encountered are discussed including clinical annotation, specimen utilisation and biobank sustainability, along with the measures the HSA Biobank is taking to address these challenges. We describe new directions being explored, going beyond traditional specimen collection into areas involving bioimages, microbiota and live cell culture. The HSA Biobank is working in collaboration with clinicians, pathologists and researchers, piloting a sustainable, robust platform with the potential to integrate future needs
