    Inhibition of large conductance calcium-dependent potassium channel by Rho-kinase contributes to agonist-induced vasoconstriction

    We tested the hypothesis that Rho-kinase inhibits the large-conductance, calcium and voltage dependent potassium (BKCa) channels thereby promoting vasoconstriction. Our results show that the Rho-kinase inhibitor, Y-27632, induced concentration-dependent relaxation in rat mesenteric artery. The selective BKCa channel inhibitors, iberiotoxin (0.1 mM) and tetraethylammonium (10 mM) increased the EC50 of Y-27632 more than 2-fold and decreased Y-27632-induced maximum relaxation (P<0.05). In the inside-out patch clamp configuration, constitutively active Rho-kinase (1mg/ml) attenuated BKCa channel activity induced by protein kinase G (PKG) (P<0.05). Y-27632 (10 mM) reversed the inhibitory effect of active Rhokinase (P<0.01). Furthermore, in the presence of Y-27632, addition of active Rho-kinase had no effect on PKG-stimulated BKCa channel activity. Taken together, our data suggest that Rho-kinase negatively regulates BKCa channels, thus providing a novel mechanism though which Rho-kinase increases smooth muscle contraction.Keywords: Rho, smooth muscle, hyperpolarization, vascular reactivity, mesenteric artery, patch clam

    Global oral health inequalities: task group--periodontal disease.

    Periodontal diseases constitute one of the major global oral health burdens, and periodontitis remains a major cause of tooth loss in adults worldwide. The World Health Organization recently reported that severe periodontitis exists in 5-20% of adult populations, and most children and adolescents exhibit signs of gingivitis. Likely reasons to account for these prevalent diseases include genetic, epigenetic, and environmental risk factors, as well as individual and socio-economic determinants. Currently, there are fundamental gaps in knowledge of such fundamental issues as the mechanisms of initiation and progression of periodontal diseases, which are undefined; inability to identify high-risk forms of gingivitis that progress to periodontitis; lack of evidence on how to prevent the diseases effectively; inability to detect disease activity and predict treatment efficacy; and limited information on the effects of integration of periodontal health as a part of the health care program designed to promote general health and prevent chronic diseases. In the present report, 12 basic, translational, and applied research areas have been proposed to address the issue of global periodontal health inequality. We believe that the oral health burden caused by periodontal diseases could be relieved significantly in the near future through an effective global collaboration.published_or_final_versio

    Infection of a yellow baboon with simian immunodeficiency virus from African green monkeys:evidence for cross-species transmission in the wild

    Many African primates are known to be naturally infected with simian immunodeficiency viruses (SIVs), but only a fraction of these viruses has been molecularly characterized. One primate species for which only serological evidence of SIV infection has been reported is the yellow baboon (Papio hamadryas cynocephalus). Two wild-living baboons with strong SIVAGM seroreactivity were previously identified in a Tanzanian national park where baboons and African green monkeys shared the same habitat (T. Kodama, D. P. Silva, M. D. Daniel, J. E. Phillips-Conroy, C. J. Jolly, J. Rogers, and R. C. Desrosiers, AIDS Res. Hum. Retroviruses 5:337-343, 1989). To determine the genetic identity of the viruses infecting these animals, we used PCR to examine SIV sequences directly in uncultured leukocyte DNA. Targeting two different, nonoverlapping genomic regions, we amplified and sequenced a 673-bp gag gene fragment and a 908-bp env gene fragment from one of the two baboons. Phylo-genetic analyses revealed that this baboon was infected with an SIVAGM strain of the vervet subtype. These results provide the first direct evidence for simian-to-simian cross-species transmission of SIV in the wild

    Do online resources give satisfactory answers to questions about meaning and phraseology?

    In this paper we explore some aspects of the differences between printed paper dictionaries and online dictionaries in the ways in which they explain meaning and phraseology. After noting the importance of the lexicon as an inventory of linguistic items and the neglect in both linguistics and lexicography of phraseological aspects of that inventory, we investigate the treatment in online resources of phraseology – in particular, the phrasal verbs wipe out and put down – and we go on to investigate a word, dope, that has undergone some dramatic meaning changes during the 20th century. In the course of discussion, we mention the new availability of corpus evidence and the technique of Corpus Pattern Analysis, which is important for linking phraseology and meaning and distinguishing normal phraseology from rare and unusual phraseology.     Creation of ultracold molecules from a Fermi gas of atoms

    Since the realization of Bose-Einstein condensates (BEC) in atomic gases an experimental challenge has been the production of molecular gases in the quantum regime. A promising approach is to create the molecular gas directly from an ultracold atomic gas; for example, atoms in a BEC have been coupled to electronic ground-state molecules through photoassociation as well as through a magnetic-field Feshbach resonance. The availability of atomic Fermi gases provides the exciting prospect of coupling fermionic atoms to bosonic molecules, and thus altering the quantum statistics of the system. This Fermi-Bose coupling is closely related to the pairing mechanism for a novel fermionic superfluid proposed to occur near a Feshbach resonance. Here we report the creation and quantitative characterization of exotic, ultracold 40^{40}K2_2 molecules. Starting with a quantum degenerate Fermi gas of atoms at T < 150 nanoKelvin we scan over a Feshbach resonance to adiabatically create over a quarter million trapped molecules, which we can convert back to atoms by reversing the scan. The small binding energy of the molecules is controlled by detuning from the Feshbach resonance and can be varied over a wide range. We directly detect these weakly bound molecules through rf photodissociation spectra that probe the molecular wavefunction and yield binding energies that are consistent with theory

    On-the-fly decoding luminescence lifetimes in the microsecond region for lanthanide-encoded suspension arrays

    Significant multiplexing capacity of optical time-domain coding has been recently demonstrated by tuning luminescence lifetimes of the upconversion nanoparticles called 'τ-Dots'. It provides a large dynamic range of lifetimes from microseconds to milliseconds, which allows creating large libraries of nanotags/microcarriers. However, a robust approach is required to rapidly and accurately measure the luminescence lifetimes from the relatively slow-decaying signals. Here we show a fast algorithm suitable for the microsecond region with precision closely approaching the theoretical limit and compatible with the rapid scanning cytometry technique.We exploit this approach to further extend optical time-domain multiplexing to the downconversion luminescence, using luminescence microspheres wherein lifetimes are tuned through luminescence resonance energy transfer.We demonstrate real-time discrimination of these microspheres in the rapid scanning cytometry, and apply them to the multiplexed probing of pathogen DNA strands. Our results indicate that tunable luminescence lifetimes have considerable potential in high-throughput analytical sciences. © 2014 Macmillan Publishers Limited. All rights reserved
