    Kν10.1 K⁺ - channel plasma membrane discrete domain partitioning and its functional correlation in neurons

    AbstractKV10.1 potassium channels are implicated in a variety of cellular processes including cell proliferation and tumour progression. Their expression in over 70% of human tumours makes them an attractive diagnostic and therapeutic target. Although their physiological role in the central nervous system is not yet fully understood, advances in their precise cell localization will contribute to the understanding of their interactions and function. We have determined the plasma membrane (PM) distribution of the KV10.1 protein in an enriched mouse brain PM fraction and its association with cholesterol- and sphingolipid-rich domains. We show that the KV10.1 channel has two different populations in a 3:2 ratio, one associated to and another excluded from Detergent Resistant Membranes (DRMs). This distribution of KV10.1 in isolated PM is cholesterol- and cytoskeleton-dependent since alteration of those factors changes the relationship to 1:4. In transfected HEK-293 cells with a mutant unable to bind Ca2+/CaM to KV10.1 protein, Kv10.1 distribution in DRM/non-DRM is 1:4. Mean current density was doubled in the cholesterol-depleted cells, without any noticeable effects on other parameters. These results demonstrate that recruitment of the KV10.1 channel to the DRM fractions involves its functional regulation

    Stability of class II malocclusion treatment with the austro repositioner followed by fixed appliances in brachyfacial patients

    One of the goals of functional-appliance devices is to modify the vertical growth pattern, solving several kinds of malocclusion. This study aimed to evaluate Class II malocclusion treatment’s stability with Austro Repositioner, followed by fixed appliances, and assess its capacity to modify vertical dimensions in brachyfacial patients. A test group of 30 patients (16 boys and 14 girls, mean 11.9 years old) with Class II malocclusion due to mandibular retrognathism and brachyfacial pattern treated with Austro Repositioner and fixed appliance were compared to a matched untreated Class II control group of 30 patients (17 boys and 13 girls, mean age 11.7 years old). Lateral cephalograms were taken at T1 (initial records), T2 (end of treatment), and T3 (one year after treatment). Statistical comparisons were performed with a paired-sample t-test and two-sample t-tests. Significant improvements in the skeletal Class II relationship were observed in the treated group. The ANB angle decreased (4.75°), the SNB angle increased (3.92°), and the total mandibular length (Co-Pg) increased (8.18 mm) (p < 0.001). Vertical dimensions were also significantly modified, the FMA angle increased (3.94°), LAFH-distance increased (3.15 mm), and overbite decreased (3.35 mm). These changes remained stable one year after treatment. The Austro Repositioner was adequate for treating the skeletal Class II malocclusion resulting from the mandible retrusion in brachyfacial patients

    Assessing the trophic ecology of the invasive Atlantic Blue Crab Callinectes sapidus in the coastal waters of the Gulf of Cadiz

    Callinectes sapidus, the invasive blue crab from the west of the Atlantic Ocean, has extended its distribution along the Atlantic coast around the Gulf of Cadiz and increased massively since 2016. Food web studies are useful for understanding changes in ecosystems caused by exotic species. Stable isotope analysis (δ13C and δ15N) were used to assess the potential carbon sources and its trophic relationships among different ecosystems (estuaries and saltmarshes), sexes (male and females) and seasons (summer vs. autumn). Significant differences were found in the δ13C of blue crabs from the estuaries and salt-marshes (-21.2 ±2.6 vs -14.2 ±0.9, respectively). These differences may be explained by an increase in the 13C of the blue crabs from the salt-marshes, probably due to the enrichment of this isotope in the primary producers such as the salt marsh plants [1] and its preys inhabiting this ecosystem. Meanwhile, the more depleted 13C values in the estuary crabs seem to reflect a carbon source from mollusks and fish derived from decomposing detritus. Among the ecosystems analyzed, δ15N was only significantly enriched in the crabs of the Guadalquivir estuary and was higher in males than females. Also, seasonal differences were found in this estuary in both sexes, by a decrease in the 15N values between summer and autumn. Those differences, can be explained by the fact that the Guadalquivir estuary suffers nitrogen hyper-nitrification due to intensive agriculture and is more noticeable in the upper part of the estuary, to which the males are more associated due to their life cycle. Previous studies reported, the diet of C. sapidus seems to be opportunistic, dependent on the food availability in different habitats [2], with a divergence in sexes induced by different spatial distributions. Future studies analyzing the stomach content and trophic behavior should be conducted to clarify our results

    Decerebration induced by surgical transection of cerebral ganglion of crayfish

    Background: Since the neural structures of the crayfish brain closely resemble their equivalent in the mammals. This can be suggested by observing the similarity that exists in the brain divided by the surgical transection of the crayfish brain in which the protocerebrum remains attached to the first two cranial nerves, findings also described by Frederic Bremer in 1935 in cats with cerebral transection.Methods: Total 11 Adult male crayfish were trained to respond with defense reflex, the animals were placed in water at 0°C, remained without any movement, and subsequently through a small incision of 3 mm in diameter in the medial antero region and dorsal cephalothorax region, a surgical section of the cerebral ganglion was performed. Immediately after surgery, metal microelectrodes were implanted to collect the activity of the photoreceptors and visual fibers.Results: Once the defense reflex begins to recover in previously decerebrated crayfish, it means that it shows signs of reconnection. The isolated protocerebrum with the deutocerebrum olfactory lobe remain alive for several days and the neuronal connections were reestablished, as measured throughout the bilateral defense activity. The defense reflex was observed in all animals and then recovered after surgery.Conclusions: The crayfish is an excellent model to work the visual activity, all coding of visual information was suppressed in de-cerebrated crayfish. The recovery of the neural disconnection is observed from 40 days, where the defence reflex appears again before visual stimuli.

    Control de calidad y de almacenamiento de semillas de girasol mediante tecnología de infrarrojo cercano

    This work assesses the application of near infrared spectroscopy technology for the quality control of sunflower seeds direct from farmers and from a storage silo. The results show that the analytical method employing near infrared spectroscopy can be used as a rapid and non-destructive tool for the determination of moisture, fat and high/low oleic acid contents in samples of sunflower seeds. The ranges obtained were comparable to those reported for classic chemical methods, and were between 4.6-21.4% for moisture; 38.4-49.6% for fat, and 60.0-93.1% for oleic acid expressed as percentage of total fatty acids. A stepwise discriminant analysis was performed to determine the most useful wavelengths for classifying sunflower seeds in terms of their (high/low) oleic acid composition. The discriminant model allows the classification of sunflower seeds with high or low oleic acid contents, with a prediction rate of 90.5% for internal validation and of 89.4% for cross-validation.En este trabajo se evalúa la espectroscopía de infrarrojo cercano para su uso en el control de calidad y almacenamiento de semillas de girasol. Los resultados indican que el método analítico empleado puede utilizarse como método de determinación rápida de humedad, grasa y contenidos altos/bajos de ácido oleico. Los rangos de aplicación son comparables con los valores que se han determinado mediante métodos clásicos de análisis, encontrándose entre 4.6-21.4% la humedad, 38.4-49.6% la grasa y 60.0- 93.1% de ácido oleico del total de los ácidos grasos. Además se ha utilizado un análisis discriminarte lineal por pasos determinando las longitudes de onda más adecuadas para la clasificación de semillas de girasol en los grupos alto/bajo oleico. El modelo generado permitió la clasificación de semillas de girasol en los grupos alto y bajo oleico con unos porcentajes de muestras correctamente clasificadas de un 90.5% en validación interna y de un 89.4% en validación cruzada

    Thoracic limb salvage by fibular free flap

    The treatment of most fractures of the ulna and radius is usually performed by anatomical reduction and internal fixation, when damage is extensive and local soft tissue cannot provide a complete wound coverage, locoregional flaps present a suitable reconstructive benefit. A 35-year-old male patient suffered an exposed diaphysio-metaphyseal fracture with multi-fragmented distal radius. The patient was evaluated during a 10-day period at the National Institute of Rehabilitation, where the osteosynthesis material and a severe infectious process with necrosis were identified. Necrosectomy of the posterior compartment and removal of the osteosynthesis material was performed, a skin defect of approximately 22x16 cm was observed with a bone gap of 6 cm of radius and ulna. a fibula-free flap is placed to correct the skin defect and an external fixative used for bone alignment. The fibular free flap presents an excellent therapeutic alternative in the resolution of bone gaps with extensive skin defect. Whenever a trained microsurgery team is available, current scales of limb injury should be considered but not utilized for therapeutic approach, always trying to shift amputation as the first option, to the very last one of them
