Penelitian ini menggunakan Importance Performance Analysis(IPA). Hasil dari penelitian ini adalah kesenjangan antara harapan dan Kenyataan yang diukur menggunakan atribut DINESERV adalah Kenyataan yang diterima oleh konsumen sangat tidak sesuai dengan harapan konsumen dan konsumen sangat tidak puas terutama dengan atribut Convenience Restoran yaitu Jarak dari Restoran “X” di Surabay

    Source and evolution of the ore-forming fluid of the Cuonadong Sn-W-Be polymetallic deposit (southern Tibet, China): constraints from scheelite trace element and Sr isotope geochemistry

    The Cuonadong Sn-W-Be polymetallic deposit is the first rare-metal deposit with an extraordinary metallogenic potential that has been discovered in the Tethyan Himalayan metallogenic belt. The deposit shows a wide range of different mineralization types, including greisen-, pegmatite-, skarn-, and hydrothermal vein-type mineralization. Of these mineralization types, the skarn-type has the largest inferred resources. In order to constrain the source and evolution of the ore-forming fluid, we conducted a detailed study of the skarn-hosted scheelite, including cathodoluminescence imaging, in-situ trace elemental analysis, and bulk Sr isotope analysis. In addition, bulk Sr isotope analysis of fluorite, phlogopite 40Ar-39Ar dating, and marble whole-rock geochemical analysis were also conducted. Phlogopite 40Ar-39Ar dating indicates that the skarn was formed at ca. 15 Ma, which is close to the formation age of the Cuonadong muscovite granite reported in previous studies. Cathodoluminescence images indicate that skarn-hosted scheelite can be subdivided into an older type 1 scheelite and a younger type 2 scheelite. Type 1 scheelite is characterized by a negative Eu anomaly, whereas type 2 scheelite shows a positive Eu anomaly. Considering the close temporal and spatial relationship between the Cuonadong Miocene leucogranite and the ore-bearing skarn, and their similar lanthanide tetrad effect as observed in the REE patterns, we propose that the negative Eu anomaly was inherited from the Cuonadong Miocene leucogranite, while the positive Eu anomaly resulted from the addition of Eu2+ to the ore-forming fluid during greisenization. Sr isotope data show that the fluorite (87Sr/86Sr)i value of 0.728885 is within the range of the Cuonadong Miocene leucogranite (87Sr/86Sr)i values, while scheelite (87Sr/86Sr)i values (0.709717-0.713480) are similar to those of the Cuonadong marble (0.709525-0.712146). Since scheelite is intergrown with fluorite, we propose that the hydrothermal fluid that exsolved from the Cuonadong Miocene leucogranite is responsible for formation of both fluorite and scheelite. Water-rock interaction between this magma-derived fluid and the marble is responsible for the loss of the scheelite magmatic Sr isotopic signature.publishedVersio

    Cancer-induced bone pain sequentially activates the ERK/MAPK pathway in different cell types in the rat spinal cord

    <p>Abstract</p> <p>Background</p> <p>Previous studies have demonstrates that, after nerve injury, extracellular signal-regulated protein kinase (ERK) activation in the spinal cord-initially in neurons, then microglia, and finally astrocytes. In addition, phosphorylation of ERK (p-ERK) contributes to nociceptive responses following inflammation and/or nerve injury. However, the role of spinal cells and the ERK/MAPK pathway in cancer-induced bone pain (CIBP) remains poorly understood. The present study analyzed activation of spinal cells and the ERK/MAPK pathway in a rat model of bone cancer pain.</p> <p>Results</p> <p>A Sprague Dawley rat model of bone cancer pain was established and the model was evaluated by a series of tests. Moreover, fluorocitrate (reversible glial metabolic inhibitor) and U0126 (a MEK inhibitor) was administered intrathecally. Western blots and double immunofluorescence were used to detect the expression and location of phosphorylation of ERK (p-ERK). Our studies on pain behavior show that the time between day 6 and day 18 is a reasonable period ("time window" as the remaining stages) to investigate bone cancer pain mechanisms and to research analgesic drugs. Double-labeling immunofluorescence revealed that p-ERK was sequentially expressed in neurons, microglia, and astrocytes in the L4-5 superficial spinal cord following inoculation of Walker 256 cells. Phosphorylation of ERK (p-ERK) and the transcription factor cAMP response element-binding protein (p-CREB) increased in the spinal cord of CIBP rats, which was attenuated by intrathecal injection of fluorocitrate or U0126.</p> <p>Conclusions</p> <p>The ERK inhibitors could have a useful role in CIBP management, because the same target is expressed in various cells at different times.</p

    Promising plasmid DNA vector based on APTES-modified silica nanoparticles

    Nanoparticles have an enormous potential for development in biomedical applications, such as gene or drug delivery. We developed and characterized aminopropyltriethoxysilane-functionalized silicon dioxide nanoparticles (APTES-SiNPs) for gene therapy. LipofectamineÂź 2000, a commonly used agent, served as a contrast. We showed that APTES-SiNPs had a gene transfection efficiency almost equal to that of Lipofectamine 2000, but with lower cytotoxicity. Thus, these novel APTES-SiNPs can achieve highly efficient transfection of plasmid DNA, and to some extent reduce cytotoxicity, which might overcome the critical drawbacks in vivo of conventional carriers, such as viral vectors, organic polymers, and liposomes, and seem to be a promising nonviral gene therapy vector

    Complete sequence and organization of Antheraea pernyi nucleopolyhedrovirus, a dr-rich baculovirus

    <p>Abstract</p> <p>Background</p> <p>The completion and reporting of baculovirus genomes is extremely important as it advances our understanding of gene function and evolution. Due to the large number of viral genomes now sequenced it is very important that authors present significantly detailed analyses to advance the understanding of the viral genomes. However, there is no report of the <it>Antheraea pernyi </it>nucleopolyhedrovirus (AnpeNPV) genome.</p> <p>Results</p> <p>The genome of AnpeNPV, which infects Chinese tussah silkworm (<it>Antheraea pernyi</it>), was sequenced and analyzed. The genome was 126,629 bp in size. The G+C content of the genome, 53.4%, was higher than that of most of the sequenced baculoviruses. 147 open reading frames (ORFs) that putatively encode proteins of 50 or more amino acid residues with minimal overlap were determined. Of the 147 ORFs, 143 appeared to be homologous to other baculovirus genes, and 4 were unique to AnpeNPV. Furthermore, there are still 29 and 33 conserved genes present in all baculoviruses and all lepidopteran baculoviruses respectively. In addition, the total number of genes common to all lepidopteran NPVs is sill 74, however the 74 genes are somewhat different from the 74 genes identified before because of some new sequenced NPVs. Only 6 genes were found exclusively in all lepidopteran NPVs and 12 genes were found exclusively in all Group I NPVs. AnpeNPV encodes <it>v-trex</it>(Anpe115, a 3' to 5' repair exonuclease), which was observed only in CfMNPV and CfDEFNPV in Group I NPVs. This gene potentially originated by horizontal gene transfer from an ancestral host. In addition, AnpeNPV encodes two <it>conotoxin</it>-like gene homologues (<it>ctls</it>), <it>ctl1 </it>and <it>ctl2</it>, which were observed only in HycuNPV, OpMNPV and LdMNPV. Unlike other baculoviruses, only 3 typical homologous regions (<it>hr</it>s) were identified containing 2~9 repeats of a 30 bp-long palindromic core. However, 24 perfect or imperfect direct repeats (<it>dr</it>s) with a high degree of AT content were found within the intergenic spacer regions that may function as non-<it>hr</it>, <it>ori</it>-like regions found in GrleGV, CpGV and AdorGV. 9 <it>dr</it>s were also found in intragenic spacer regions of AnpeNPV.</p> <p>Conclusion</p> <p>AnpeNPV belongs to Group I NPVs and is most similar to HycuNPV, EppoNPV, OpMNPV and CfMNPV based on gene content, genome arrangement, and amino acid identity. In addition, analysis of genes that flank <it>hr</it>s supported the argument that these regions are involved in the transfer of sequences between the virus and host.</p

    Clearance of Free Silica in Rat Lungs by Spraying with Chinese Herbal Kombucha

    The effects of spraying with kombucha and Chinese herbal kombucha were compared with treatments with tetrandrine in a rat silicosis model. Silica dust (50 mg) was injected into the lungs of rats, which were then treated with one of the experimental treatments for a month. The rats were then killed and the effects of the treatments were evaluated by examining the extent and severity of the histopathological lesions in the animals’ lungs, measuring their organ coefficients and lung collagen contents, determining the dry and wet weights of their lungs, and measuring the free silica content of the dried lungs. In addition, lavage was performed on whole lungs taken from selected rats, and the numbers and types of cells in the lavage fluid were counted. The most effective treatment in terms of the ability to reduce lung collagen content and minimize the formation of pulmonary histopathological lesions was tetrandrine treatment, followed by Chinese herbal kombucha and non-Chinese herbal kombucha. However, the lavage fluid cell counts indicated that tetrandrine treatment had severe adverse effects on macrophage viability. This effect was much less pronounced for the kombucha and Chinese herbal kombucha treatments. Moreover, the free silica levels in the lungs of animals treated with Chinese herbal kombucha were significantly lower than those for any other silica-exposed group. These preliminary results indicate that spraying with Chinese herbal kombucha preparations can effectively promote the discharge of silica dust from lung tissues. Chinese herbal kombucha inhalation may thus be a useful new treatment for silicosis and other pneumoconiosis diseases

    Differential diagnosis of parkinsonism based on deep metabolic imaging indices.

    The clinical presentations of early idiopathic Parkinson's disease (PD) substantially overlap with those of atypical parkinsonian syndromes like multiple system atrophy (MSA) and progressive supranuclear palsy (PSP). This study aimed to develop metabolic imaging indices based on deep learning to support the differential diagnosis of these conditions. Methods: A benchmark Huashan parkinsonian PET imaging (HPPI, China) database including 1275 parkinsonian patients and 863 non-parkinsonian subjects with 18F-FDG PET images was established to support artificial intelligence development. A 3D deep convolutional neural network was developed to extract deep metabolic imaging (DMI) indices, which was blindly evaluated in an independent cohort with longitudinal follow-up from the HPPI, and an external German cohort of 90 parkinsonian patients with different imaging acquisition protocols. Results: The proposed DMI indices had less ambiguity space in the differential diagnosis. They achieved sensitivities of 98.1%, 88.5%, and 84.5%, and specificities of 90.0%, 99.2%, and 97.8% for the diagnosis of PD, MSA, and PSP in the blind test cohort. In the German cohort, They resulted in sensitivities of 94.1%, 82.4%, 82.1%, and specificities of 84.0%, 99.9%, 94.1% respectively. Employing the PET scans independently achieved comparable performance to the integration of demographic and clinical information into the DMI indices. Conclusion: The DMI indices developed on the HPPI database show potential to provide an early and accurate differential diagnosis for parkinsonism and is robust when dealing with discrepancies between populations and imaging acquisitions
