    Semisynthetic flavonoid 7-O-galloylquercetin activates Nrf2 and induces Nrf2-dependent gene expression in RAW264.7 and Hepa1c1c7 cells

    The natural flavonoid quercetin is known to activate the transcription factor Nrf2, which regulates the expression of cytoprotective enzymes such as heme oxygenase-1 (HO-1) and NAD(P)H:quinone oxidoreductase 1 (NQO1). In this study, a novel semisynthetic flavonoid 7-O-galloylquercetin (or quercetin-7-gallate, 3) was prepared by direct galloylation of quercetin, and its effect on the Nrf2 pathway was examined. A luciferase reporter assay showed that 7-O-galloylquercetin, like quercetin, significantly activated transcription via the antioxidant response element in a stably transfected human AREc32 reporter cell line. In addition, 7-O-galloylquercetin caused the accumulation of Nrf2 and induced the expression of HO-1 at both the mRNA and protein levels in murine macrophage RAW264.7 cells. The induction of HO-1 by 7-O-galloylquercetin was significantly suppressed by N-acetyl-l-cysteine and SB203580, indicating the involvement of reactive oxygen species and p38 mitogen-activated protein kinase activity, respectively. HPLC/MS analyses also showed that 7-O-galloylquercetin was not degalloylated to quercetin, but it was conjugated with glucuronic acid and/or methylated in RAW264.7 cells. Furthermore, 7-O-galloylquercetin was found to increase the protein levels of Nrf2 and HO-1, and also the activity of NQO1 in murine hepatoma Hepa1c1c7 cells. Taken together, we conclude that 7-O-galloylquercetin increases Nrf2 activity and induces Nrf2-dependent gene expression in RAW264.7 and Hepa1c1c7 cells

    This paper provides a basic insight into the contemporary options when choosing the right Linux distribution for multimedia center or home theater. There is an emphasis on support for single board computers. In the first part, a few important audio and video related aspects of multimedia distributions are mentioned, such as usage of drivers, surround sound support, decoding CDs, DVDs and Blu-ray discs, peripherals requirement and TV broadcast implementation. Also, there are mentioned the most used frontend applications and pictures of their graphical user interfaces are included. The second part is about experimental testing of 9 different distributions on an laptop with x86\_64 architecture. The user interface, installation process, functionality and overall image were evaluated for every distribution. The third part describes the process of installation of the Kodi app on four different platforms including problems that occurred during the process

Method In this work were used observation analyses of moving activity by the technique of indirect observation ofplayer's game achievement in soccer with the help ofinvolved quantitative and qualitative method. Findings The most effective field places were in the middle of the field and in front of the penalty area which in 9 and 20 sec. of combined offense. The most incident offensive plays are when the player participates in offensive play just one time.     Diplomová práce je zaměřena na návrh a vypracování projektové dokumentace polyfunkčního domu. Objekt je navržen na parcele na ulic Sportovců a Waldhauserova, v Kutné Hoře. Návrh domu respektuje územní plán obce a zároveň dotváří stávající zástavbu. Objekt má jedno podzemní a čtyři nadzemní podlaží. V objektu jsou sklepy, kotelna, dva obchody a čtrnáct bytových jednotek. Objekt je navržen z konstrukčního systému Vapis.The master´s thesis is focused on the preparation and design of project documentation of the multi-functional building. The building is designed on the corner of Sportovců and Waldhauserova streets in Kutná Hora. The concept of the building respects the territorial plan of the city and complements the existing buildings. The building has one underground and four floors. There is basement, two shops and fourteen flats. The building is designed from construction system Vapis.

    Browser extension adding functionality to the information system InSIS

    The subject of this bachelor thesis is to create an extension for web browsers that adds new functionalities to the school information system InSIS. The purpose of these added functionalities is to improve the productivity of system users and reduce deficiencies when working with the system, which are often mentioned by students. The goal of the thesis is to design and subsequently implement an extension for web browsers together with a supporting server for storing user data. Another goal is to compare available technologies for both parts of the software solution. The last goal is to evaluate feedback from users of the web extension obtained through a questionnaire survey that answers the defined research questions. The result of the thesis is a functional, coherent software solution with easy installation that is distributed via the Google Web Store and Firefox Addons web stores. At the time of writing, this extension has more than 300 installations and 120 active users.Předmětem této bakalářské práce je vytvoření rozšíření do webových prohlížečů, které přidává nové funkcionality do školního informačního systému InSIS. Účelem těchto přidaných funkcionalit je zlepšení produktivity uživatelů systému a redukce nedostatků při práci se systémem, které jsou často zmiňovány studenty. Cílem práce je návrh a následná implementace rozšíření do webových prohlížečů společně s podpůrným serverem pro ukládání dat uživatelů. Dalším cílem je srovnání dostupných technologií pro obě části softwarového řešení. Posledním cílem je vyhodnocení zpětné vazby od uživatelů webového rozšíření získané metodou dotazníkového šetření, které odpovídá na stanovené výzkumné otázky. Výsledkem práce je funkční, koherentní softwarové řešení se snadnou instalací distribuované na platformách Google Web Store a Firefox Addons. Toto rozšíření má v době psaní více než 300 instalací a 120 aktivních uživatelů
