Cinètica de la reacció de descomposició tèrmica de l’ascaridol en dissolució
Existen diferencias entre los valores de algunas propiedades fÃsicas de ascaridol natural aislado del vegetal denominado vulgarmente «Paico» (Chenopodium ambrosioides L., Chenopodiaceae) y la sustancia sintética obtenida mediante la reacción de fotooxigenación de α-terpineno. Es más, el uso de decocciones de dicha planta, utilizadas como medicina antihelmÃntica en los humanos, ha despertado un debate cientÃfico debido a las propiedades tóxicas atribuÃdas a su principio activo (ascaridol), el cual serÃa responsable de la muy conocida acción beneficiosa. En este trabajo se ha realizado la preparación de esa sustancia por el método de Schenck, pero trabajando a 0 ºC con alcohol isopropÃlico como solvente y efectuando su separación y purificación a temperatura ambiente por cromatografÃa en columna preparativa, a fin de prevenir su degradación térmica. De esta manera se obtuvieron excelentes rendimientos de ascaridol (ca. 99% GC-FID), de un alto grado de pureza (> 98% GC; RP-HPLC; 1 H y 13C RMN). Además, se informan aquÃ
los parámetros de activación de la reacción de descomposición térmica de ascaridol en solución de n-hexano, alcohol isopropÃlico y metanol, en el ámbito de temperaturas de 120ºC a 170ºC. Es evidente también un efecto de solvente en la homólisis unimolecular de esa sustancia.Discrepancies exist between the values of some physical properties of ascaridole isolated from the vegetal named «Paico» (Chenopodium ambrosioides L., Chenopodiaceae) and the synthetic substance obtained by the photoxygenation reaction of α-terpinene. Moreover, the use of decoctions of the above mentioned plant as anthelmintic medicine in humans arouse a scientific debate because of toxic properties attributed to its main active drug (ascaridole), which would be responsible of the well-known beneficial action. This lead to perform here the preparation of that substance by Schenck’s method, but working at 0 ºC with isopropyl alcohol as solvent and doing its separation and purification at room temperature by preparative column chromatography to prevent its thermal degradation. Thus, excellent yields of ascaridole were obtained (ca. 99 % GC-FID) of high degree of purity (> 98% GC; RP-HPLC; 1 H and 13C NMR). Furthermore, the kinetics of the thermal stability of ascaridole in solution is now advanced, reporting the activation parameters values for their decomposition reactions in n-hexane, isopropyl alcohol and methanol, in the 120º170 ºC temperature range. A solvent effect on the unimolecular

Plateaux vibrants et performance musculaire: quels effets, quels réglages ?
Introduction : Les plateaux vibrants apparaissent comme un nouveau moyen de renforcement musculaire. La littérature sur ce sujet n’en est qu’à ses débuts. Le but de cette revue systématique est de déterminer dans un premier temps si les vibrations peuvent conduire à une amélioration des performances musculaires du membre inférieur dans une population saine. Dans un deuxième temps, l’objectif est de définir les meilleurs réglages (fréquence, amplitude, temps) de ces machines afin d’optimiser les améliorations. Méthode : La recherche dans la littérature a été effectuée sur les bases de données suivantes : Pubmed, PEDro, Cinahl et The Cochrane Library. Onze études randomisées contrôlées ont été incluses. Résultats : L’effet des vibrations sur la hauteur des sauts (CMJ), la force dynamique et la force isométrique semble être généralement positif. L’amélioration atteint souvent 5% de plus que les groupes de contrôle. Quant à l’optimisation des réglages, il apparaît difficile, sur la base de la littérature scientifique, de faire ressortir une combinaison idéale. Cependant, d’après cette revue, une combinaison de haute fréquence (35-45 Hz) et de basse amplitude (2-5 mm), semblerait être la plus efficace. Discussion : Il reste un manque d’évidence statistique sur l’apport des vibrations dans l’amélioration des capacités musculaires. La recherche future se devra de démontrer statistiquement l’intérêt de l’application des vibrations dans l’augmentation des capacités musculaires et de définir les réglages optimaux de ces machines
Utilisation de l’acide poly (propyl acrylique) pour augmenter l’efficacité in vitro des nanoparticules à base de chitosane et ARN messager
L’ARNm a un fort potentiel en thérapie génique pour des applications de vaccination contre le
cancer, le remplacement de protéine défectueuse, l’édition du génome et la reprogrammation de
cellules souches. Néanmoins, la livraison de l’ARNm représente un défi scientifique et
technologique de taille, car elle est limitée par plusieurs barrières telles qu’une dégradation rapide
dans le plasma, la difficulté à pénétrer dans la cellule et une séquestration/dégradation au niveau
des endosomes. Cette barrière endosomale demeure à ce jour l’une des plus difficiles à franchir.
Pour surmonter certains de ces obstacles, plusieurs véhicules de livraison ont été développés Ã
base de virus, lipides, polymères. Le chitosane (CS), un polymère naturel cationique, peut lier et
condenser les acides nucléiques pour former des nanoparticules, ce qui les protège et augmente
leur internalisation cellulaire. Toutefois, la capacité des nanoparticules à base de CS à surmonter
la séquestration endosomale demeure limitée et pourrait être augmentée par l’utilisation d’agents
endosomolytiques.
L’objectif principal de ce projet était d’incorporer l’acide poly(propyle acrylique)(PPAA), un
agent endosomolytique, à des nanoparticules de CS/ARNm dans le but d’accroître leur efficacité.
Le PPAA a été incorporé à différentes concentrations dans des nanoparticules binaires de
CS/ARNm. Les propriétés physico-chimiques (taille, PdI, potentiel-zeta, pH) et l’efficacité de
transfection in vitro des nouvelles formulations ternaires (CS/ARNm/PPAA) ont été
caractérisées. De plus, ces formulations ont été formées avec/sans tampon et en présence/absence
de tréhalose, une molécule utilisée pour la lyophilisation des nanoparticules. En présence de
tréhalose et tampon, le PPAA était principalement incorporé dans les nanoparticules qui avaient
une taille d’environ 170 nm et une charge de surface de 12 mV. Une augmentation de l’efficacité
de transfection (~10 fois) fut observée pour une concentration intermédiaire de PPAA en
présence de tampon et tréhalose. Par la suite, l’efficacité a été augmentée davantage en retirant le
tréhalose et en augmentant la concentration de mélange de l’ARNm. Cette nouvelle formulation a
atteint une efficacité comparable à ~86% du contrôle lipidique LipofectamineTM et ce sans aucun
signe de toxicité. Ces résultats suggèrent que la présence de PPAA dans les nanoparticules
favorise leur libération de l’endosome et/ou diminue leur stabilité. Néanmoins, des études de
trafic intracellulaire et compétition devront être effectuées afin de confirmer ces hypothèses.----------ABSTRACT
Messenger mRNA is a molecule that has a strong potential in gene therapy for a wide variety of
therapeutic applications such as vaccination against infectious disease or cancer, protein
replacement, genome editing and stem cells reprogrammation. However, delivery of mRNA
remains a scientific and technological endeavour as it is limited by many hurdles such as a fast
degradation in plasma, low internalisation kinetics and a sequestration/degradation within
endosomes. This endosomale barrier remains to day one of the hardest to overcome. To
overcome some of these hurdles, delivery vehicles made of virus, lipids or polymers are
developed worldwide. Chitosan (CS), a natural cationic polymer can bind and condense anionic
nucleic acids to spontaneously form nanoparticles. These nanoparticles protect mRNA and help
its cellular internalisation. However, the capacity of these nanoparticles to escape from
endosomes is limited and could be augmented by adding endosomolytique agents into the
nanoparticles.
The main objective of this project was to incorporate an endosomolytique agent in CS/mRNA
nanoparticle in order to increase their transfection efficiency. Poly(propylacrylic acid)(PPAA), an
anionic synthetic polymer was used as endosomolytique agent and added at different
concentrations in the binary (CS/mRNA) nanoparticles. These newly formed ternary
nanoparticles (CS/mRNA/PPAA) had their physicochemical characteristics (size, PdI, zetapotential,
pH) and transfection efficiency analysed. Furthermore, these formulations were made
with/without buffer and with/without trehalose, a disaccharide used for lyophilisation of
nanoparticles. In presence of buffer and trehalose, PPAA was mostly incorporated in the
nanoparticles that had a size of 170 nm and a zeta potential of 12 mV. An augmentation of
transfection efficiency (~10-fold) was observed for an intermediate PPAA concentration. This
bioactivity was further improved by removing threlaose and increasing the mRNA mixing
concentration while forming nanoparticles. This new optimal formulation reached a protein
expression levels comparable to ~86% of the lipid control LipofectamineTM 2000 messenger max,
without any signs of toxicity. These results suggest that PPAA in nanoparticles favorise their
endosomal release and/or decrease their stability. Nevertheless, further studies such as
intracellular trafficking and competition should be performed to confirm these hypotheses
Kondo Effect and Surface-State Electrons
We have used low temperature scanning tunneling spectroscopy and atomic
manipulation to study the role of surface-state electrons in the Kondo effect
of an isolated cobalt atom adsorbed on Ag(111). We show that the observed Kondo
signature remains unchanged in close proximity of a monoatomic step, where the
local density of states of the surface-state electrons is strongly perturbed.
This result indicates a minor role for surface-state electrons in the Kondo
effect of cobalt, compared to bulk electrons. A possible explanation for our

Transferencia de materia hacia la solución en la extracción de productos bioactivos de vegetales y su degradación en mezclas de solventes
A quantitative analytic evaluation of the extraction of the natural antimalarial drug artemisinin, in wild plants of Artemisia annua L., using mixtures of ethanol 70% (v/v)-water has ben carried out. This study has been performed in order to find a justification of the reduced recovery of that drug in their isolation processes previously reported, not corresponding to the values of its dosage in the fresh plant material. The results of this work could be useful to interpret similar situations in the isolation of plant natural products.Se ha realizado una evaluación analÃtica cuantitativa de la extracción de la droga antimalárica natural artemisinina, presente en plantas silvestres de Artemisia annua L., utilizando mezclas de etanol 70% (v/v) - agua. El objetivo ha sido encontrar una justificación de la reducida recuperación de esa droga en los procesos de su aislamiento ya indicados en la literatura y que conducen a la obtención de rendimientos que no corresponden con el dosaje inicial de la droga en el material vegetal fresco. Los resultados de este trabajo podrÃan ser de utilidad para interpretar situaciones análogas en el aislamiento de productos naturales provenientes de vegetales

Interactions Mediated by Surface States: From Pairs and Trios to Adchains and Ordered Overlayers
Since metallic surface states on (111) noble metals are free-electron like,
their propagators can be evaluated analytically. Since they are well-screened,
one can use simple tight-binding formalism to study their effects. The needed
phase shifts can be extracted from experiment. Hence, one can now make
quantitative predictions of these slowly-decaying, oscillatory indirect
interactions. For the (isotropic!) pair interactions (which decay as the
inverse square of adatom-adatom separation), remarkable agreement has been
obtained with experiments by two groups. We have extended the formalism to
consider the full indirect ("triple") interaction of 3 adsorbates, which is the
sum of the 3 constituent pair interactions plus the non-pairwise "trio"
contribution, which tends to decay with the 5/2 power of perimeter. Here, we
concentrate on interactions due to ordered overlayers and to linear defects,
relating the latter to the interactions of (nx1) ordered overlayers and to the
constituent pair interactions. We compare with experimental studies of
interactions of adatoms with adchains and of consequent 1D motion of adatoms
trapped between two such parallel chains. We discuss implications for step-step
interactions (on vicinal surfaces), with attention to the modification of the

Extracción con solventes de artemisinina y otros metabolitos de <i>Artemisia annua</i> L. silvestre
Se han llevado a cabo metodologÃas alternativas para la extracción de artemisinina de hojas e inflorescencias del comunmente denominado ajenjo dulce ("sweetwormwood", Artemisia annua L.), ya sea de ejemplares de crecimiento expontáneo (silvestre) como de aquellos expresamente cultivados en regiones ubicadas en las Provincias de Buenos Aires y de Entre RÃos de la República Argentina. Fueron experimentalmente ensayadas y al mismo tiempo optimizadas, las diferentes condiciones de extracción, algunas si bien ya conocidas, de esa eficaz droga antipalúdica natural. Los extractos correspondientes fueron preparados en distintos solventes y condiciones, ya sea para el análisis de sus contenidos de artemisinina o bien, para el aislamiento de los principales metabolitos presentes en Artemisia annua L. Estos extractos se obtuvieron por operaciones de maceración, percolación o decocción de partes aéreas del vegetal fresco o previamente secado y molido, colocado ya sea en viales de Pyrex o en recipientes cerrados, en presencia de solventes puros o sus mezclas, con distintas caracterÃsticas fÃsicoquÃmicas, ejerciendo vigorosa agitación mecánica y colocados a temperaturas convenientes. Para el dosage de artemisinina en los correspondientes extractos y también poder evaluar su calidad analÃtica se empleó, según su conveniencia experimental para la obtención de resultados confiables, cromatografÃa planar en capa delgada (CCD, TLC), cromatografÃa lÃquida de alta eficiencia en fase reversa (CLAE, RP-HPLC) y cromatografÃa capilar en fase gaseosa (CG). La reproducibilidad y exactitud de los procedimientos analÃticos utilizados se comprobó comparando los resultados obtenidos con los provenientes de aplicar métodos clásicos para la extracción exhaustiva de artemisinina del material vegetal con n-hexano a reflujo (i.e. Soxhlet). La metodologÃa llevada a cabo permitió obtener conclusiones relativas a la eficiencia de los solventes utilizados, lo cual resulta de utilidad para poder adaptar los procedimientos seguidos en la extracción de artemisinina a una mayor escala como también de otras sustancias naturales estructuralmente relacionadas o con similares propiedades fisicoquÃmicas.Departamento de QuÃmic
Probing hot-electron dynamics at surfaces with a cold scanning tunneling microscope
We report on a novel approach to measure the phase relaxation length and femtosecond lifetime of hot quasiparticles on metal surfaces. A 4 K scanning tunneling microscope has been used to study the spatial decay of interference patterns in the local density of states for surface state electrons on Ag(111) and Cu(111). This decay is governed by inelastic; electron-electron scattering. We find a (E - E-F)(-2) energy dependence of the lifetimes for both Ag and Cu, and our values are comparable to the corresponding bulk electron lifetimes. This indicates that electron-electron interaction of hot surface state electrons with the Fermi sea is dominated by the underlying bulk electrons. [S0031-9007(99)09260-1]
