    Granulometric characterisation of nanoparticles by the PCS method.

    In the theoretical section of presented diploma thesis are discussed various nanoparticles such as micelles and liposomes, solid lipid nanoparticles and polymeric nanoparticles. Special attention was focused on copolymer PLGA. There are characterized different procedures in the preparation of nanoparticles. From the approches for nanoparticles evaluation are chosen measuring size of nanoparticles and their zeta potential. The experiment was almost unambiguously focused on study of effect of diffrent factors on nanopartlicles size and distribution from linear PLGA and PLGA used for branching of tripentaerythritol and polyacrylic acid. Z přístupů k hodnocení nanočástic je vybráno měření jejich velikosti metodou PCS a měření zeta potenciálu.     The paper presents results of the preliminary structural analysis of model of the endoprosthesis of the hip joint. Basics of anatomy and biomecha-nical analysis of the hip joint were introduced. Based on data from atlas of human anatomy and medical imaging, the prototype of endoprosthesis was modelled using Solid Edge ST8 software. After determining physical properties of structural materials, the Finite Elements Analysis of the model was conducted using in SolidWorks software under various load conditions.     The diagnostic difficulties of eosinophilia in clinical practice - case series

    Introduction: Eosinophilia, defined as elevated level of eosinophils in peripheral blood above 5 x 109/L, is the hematological disorder, which may occur in multiple conditions, such as allergies, gastrointestinal, autoimmune diseases, parasite, fungal infections as well as drug related eosinophilia. Although hematological causes of eosinophilia (idiopathic, myeloproliferative and lymphocytic variant) should be taken into consideration. Aim of the study: The aim of our study was to performed the difficulties related to the differential diagnosis of eosinophilia, especially associated with the diverse symptoms. Materials and methods: The study included 5 patients hospitalized in the Department of Hematooncology and Bone Marrow Transplantation, due to eosinophilia associated with diverse symptoms. Medical history, physical examination, the peripheral blood as well as bone marrow samples analysis and the genetic tests for the presence of mutations or rearrangements that detect leukemia or lymphoma were analyzed. Moreover, patients were evaluated due to the presence of parasites. Results: The case series revealed that eosinophilia may have a various etiological background. Three patient demonstrated the reactive eosinophilia, caused by bacterial, parasite infections and eosinophilic granulomatosis with polyangitis, however in two other cases the chronic eosinophilic leukemia with abnormalities of PDGFRA were diagnosed. Conclusions: Eosinophilia is an important diagnostic and prognostic feature in a varied range of pathological conditions from infections, allergies to malignancies. For this reason, it is an enormous diagnostic as well as therapeutic challenge and requires an interdisciplinary clinical approach, especially in cases with unclear manifestations

    BACKGROUND: Plant-based diets (PBDs) are increasingly recommended for human and planetary health. However, comprehensive evidence on the health effects of PBDs in children remains incomplete, particularly in vegans. OBJECTIVES: To quantify differences in body composition, cardiovascular risk, and micronutrient status of vegetarian and vegan children relative to omnivores and to estimate prevalence of abnormal micronutrient and cholesterol status in each group. METHODS: In a cross-sectional study, Polish children aged 5-10 y (63 vegetarian, 52 vegan, 72 matched omnivores) were assessed using anthropometry, deuterium dilution, DXA, and carotid ultrasound. Fasting blood samples, dietary intake, and accelerometry data were collected. RESULTS: All results are reported relative to omnivores. Vegetarians had lower gluteofemoral adiposity but similar total fat and lean mass. Vegans had lower fat indices in all regions but similar lean mass. Both groups had lower bone mineral content (BMC). The difference for vegetarians attenuated after accounting for body size but remained in vegans (total body minus the head: -3.7%; 95% CI: -7.0, -0.4; lumbar spine: -5.6%; 95% CI: -10.6, -0.5). Vegetarians had lower total cholesterol, HDL, and serum B-12 and 25-hydroxyvitamin D [25(OH)D] without supplementation but higher glucose, VLDL, and triglycerides. Vegans were shorter and had lower total LDL (-24 mg/dL; 95% CI: -35.2, -12.9) and HDL (-12.2 mg/dL; 95% CI: -17.3, -7.1), high-sensitivity C-reactive protein, iron status, and serum B-12 (-217.6 pmol/L; 95% CI: -305.7, -129.5) and 25(OH)D without supplementation but higher homocysteine and mean corpuscular volume. Vitamin B-12 deficiency, iron-deficiency anemia, low ferritin, and low HDL were more prevalent in vegans, who also had the lowest prevalence of high LDL. Supplementation resolved low B-12 and 25(OH)D concentrations. CONCLUSIONS: Vegan diets were associated with a healthier cardiovascular risk profile but also with increased risk of nutritional deficiencies and lower BMC and height. Vegetarians showed less pronounced nutritional deficiencies but, unexpectedly, a less favorable cardiometabolic risk profile. Further research may help maximize the benefits of PBDs in children

    Efficacy, pharmacokinetics and safety of subcutaneous versus intravenous CT-P13 in rheumatoid arthritis: a randomized phase I/III trial

    Objective. To assess non-inferiority of s.c. to i.v. CT-P13 in RA. Methods. Patients with active RA and inadequate response to MTX participated in this phase I/III double-blind study at 76 sites. Patients received CT-P13 i.v. 3 mg/kg [week (W) 0 and W2] before randomization (1:1) at W6 to CT-P13 s.c. via pre-filled syringe (PFS) 120 mg biweekly until W28, or CT-P13 i.v. 3 mg/kg every 8 weeks until W22. Randomization was stratified by country, W2 serum CRP and W6 body weight. From W30, all patients received CT-P13 s.c. In a usability sub- study, patients received CT-P13 s.c. via auto-injector (W46–54) then PFS (W56–64). The primary endpoint was change (decrease) from baseline in disease activity score in 28 joints (DAS28)-CRP at W22 (non-inferiority margin: -0.6). Results. Of 357 patients enrolled, 343 were randomized to CT-P13 s.c. (n ¼ 167) or CT-P13 i.v. (n ¼ 176) at W6. The least-squares mean change (decrease) from baseline (standard error) in DAS28-CRP at W22 was 2.21 (0.22) for CT-P13 s.c. (n ¼ 162) and 1.94 (0.21) for CT-P13 i.v. [n ¼ 168; difference 0.27 (95% CI: 0.02, 0.52)], establishing non-inferiority. Efficacy findings were similar between arms at W54. Safety was similar between arms throughout: 92 (54.8%; CT-P13 s.c.) and 117 (66.9%; CT-P13 i.v.) patients experienced treatment-emergent adverse events (from W6). There were no treatment-related deaths or new safety findings. Usability was similar for CT-P13 s.c. via auto-injector or PFS. Conclusion. CT-P13 s.c. was non-inferior to CT-P13 i.v. in active RA. The convenience of s.c. administration could benefit patients

    Potential range of impact of an ecological trap network: the case of timber stacks and the Rosalia longicorn

    Although the negative impact of timber stacks on populations of saproxylic beetles is a well-known phenomenon, there is relatively little data concerning the scale of this impact and its spatial aspect. Beech timber stored in the vicinity of the forest can act as an ecological trap for the Rosalia longicorn (Rosalia alpina), so in this study we have attempted to determine the spatial range of the impact of a network of timber stacks. Timber stacks in the species’ range in the study area were listed and monitored during the adult emergence period in 2014–2016. Based on published data relating to the species’ dispersal capabilities, buffers of four radii (500, 1000, 1600, 3000 m) were delineated around the stacks and the calculated ranges of potential impact. The results show that the percentage of currently known localities of the Rosalia longicorn impacted by stacks varies from 19.7 to 81.6%, depending on the assumed impact radius. The percentage of forest influenced by timber stacks was 77% for the largest-radius buffer. The overall impact of the ecological trap network is accelerated by fragmentation of the impact-free area. It was also found that forests situated close to the timber stacks where the Rosalia longicorn was recorded were older and more homogeneous in age and species composition than those around stacks where the species was absent. Such results suggest that timber stacks act as an ecological trap in the source area of the local population
