Alternatives to in-feed antibiotics and their effects on development and stability of intestinal microbial community in weaning piglets
Der Magen-Darm-Trakt von Schweinen ist durch ein vielfältiges und dynamisches Mikrobiota bewohnt, das eine wichtige Rolle für die Gesundheit des Wirtes spielt. Die Thesis liefert Daten zu der Zusammensetzung und zu Veränderungen der intestinalen porcinen Mikrobiota vor und nach dem Absetzen. Im Hinblick auf die Suche nach

SelbständigkeitserklärungGreen micro-algae have been used as nutritional supplement or food source in
Asiatic countries for hundreds of years. Use of the green micro-algae in human
and animal nutrition has been studied intensively since the mid-50s of last
century. Since then many groups have investigated different micro-algae
cultivated under different conditions. Beneficial immunomodulatory properties
of the green micro-algae (mainly Chlorella vulgaris) were discovered and its
potential for use in human medicine was reported. In my dissertation five
experiments are described, in which we investigated the nutritional value of
micro-algal protein as well as assessing the safety of feeding the micro-algae
on animal health. We also investigated if feeding micro-algae can influence
the reproductive efficiency and growth parameters of laboratory animals. In
order to make the constituents of micro-algae accessible to mammals the cell
wall must be destroyed. Several processes for making micro-algae more
digestible have been developed but their efficacy and economic utility were
poor. New efficient and cheap cell-opening methods are needed if the micro-
algae are to fulfill their potential as novel, widely-available, natural,
modern feed supplements. Treatment of algal cells with electrical field
(electroporation) or ultrasound (ultrasonication) prior to spray-drying have
shown promise as ways of releasing algal nutrients. We aimed to investigate if
these new methods are indeed efficient at enhancing the bioavailability of
algal protein, using rodents as models. This experiment was part of project
Entwicklung und Produktionseinführung von speziellem Kraftfutter mit Zusatz
von aufgewerteten Mikroalgen ( Development and introduction into production
of special concentrate with supplement of valuated micro-algae ) of the
Frankerförder Forschungsgeselschaft Ltd. Three processed products derived from
the green micro-algae Chlorella vulgaris (C1) were therefore
investigated:spray dried only,; electroporated and spray dried and, finally,
ultrasonicated and spray dried. A nitrogen-balance study was undertaken. Male
Wistar rats with a mean weight of 135-145 grams were housed separately in
metabolism cages and were fed the three algal products as the sole protein
source at 150 mg N per 100 g of body weight. A control group of rats was fed
with casein at the same amount as mentioned above. Apparent crude protein
digestibility for spray dried C. vulgaris was 46.9 12.7% (mean SD), for
electroporated Ch. vulgaris 44.3 7.5%, for ultrasonicated micro-algae 56.7
13.7%. The protein efficiency ratio was 1.4 0.3; 1.0 0.5; 2.1 0.3,
respectively. N-balance was 41.86 32.8 mg; 31.3 17.3 mg; and 66.7 30.1
mg, respectively. The biological value was 93 9.5 %; 93.6 10 %; and 101
5%, respectively. The differences between nutritional parameters (except for
biological value) for ultrasonized and electroporated C. vulgaris were
statistically significant (p<0.05 in HSD-Tukey test). The digestibility and
biological value of C. vulgaris was enhanced by ultrasonic treatment and
reduced by electroporation, thus ultrasonication may be a helpful
technological process in processing of the green micro-algae in food industry.
Micro-algae contain high amounts of nucleic acids, which are source of purine
bases that after absorption are transformed to uric acid (in humans) and
allantoin (most animals). Thus feeding microalgae could lead to an increase of
uric acid formation with subsequent development or enhancement of pathological
conditions associated with this metabolite(i.e. gout, kideny stones). To
investigate this and general algal influence on animal, we fed adult and
growing rats with a diet containing 20% of micro-algae (supplemental or as
sole protein source) for one month. Uric acid and allantoin were measured
periodically in serum and urine. Urine production was also recorded. Multiple
serum biochemistry and haematological parameters were measured. Internal
organs were weighed and histological examinations were performed for
intestines, liver and kidneys. No negative influence of micro-algal feeding
could be observed. Uric acid level did not change after algal feeding.
Allantoin excretion increased in the first two weeks of algal feeding and
thereafter returned to the initial level. Serum biochemistry and haematology
showed a decrease of full name (ALP) activity in algal group, no other changes
were observed. Intestines were heavier and longer in rats fed micro-algae. No
histological abnormalities were observed in organ slices. Feeding rats with
micro-algae at 20% in feed led to no adverse effects and this amount can be
considered as safe. The effect of feed supplementation with 1.0 % spray-dried
green alga Chlorella vulgaris powder on mouse reproduction and growth was
investigated. The study was continued over three generations of mice, strain
Fzt:DU. Females (F0) were fed the control diet (commercial chow) or the algae-
supplemented diet starting from 21st day of life (weaning). They were mated
randomly on the 63rd day of life and all gave birth to pups. Litters were
weighed, counted and then standardized (4 males and 5 females per litter).
Pups were weighed and counted on day 10 and 21. After weaning 2 females and 2
males (F1) from each litter were kept further. Females were mated on 63rd day
of life. On the 18th day of pregnancy 57 and 59 (control and algae group) were
sacrificed, 51 and 53 (respectively) gave birth to pups. Live and dead pups,
absorbed fetuses and corpora lutei were counted, live fetuses were weighed.
Live-born pups were counted and weighed and kept by dams without
standardization, their number and weight were recorded on days 10 and 21. 2
females and 2 males (F2) were weaned and kept further, the procedure as above
was repeated. No difference in number of fetuses, corpora lutei or live-born
pups was noted between groups or between generations. Litters from the algae
group were slightly heavier at the weaning, the females and males from algae
group also developed slightly better than the ones from control group, and
this tendency was seen in all generations. Technological processing of
microalgae by ultrasonication can be efficiently used in practice, but still
the industry must find a way to reduce the processing costs. There are still
more experiments needed to establish the amount of micro-algae in feed that
will be efficient and beneficial for farm animals without increasing animal
production costs.Grüne Mikroalgen werden in Asien seit Jahrhunderten als Nahrungsmittel oder
Nahrungsergänzung genutzt. Das Interesse am Einsatz grüner Mikroalgen in der
Human- und Tierernährung wurde in den 50er Jahren des vergangenen Jahrhunderts
geweckt. Seitdem untersuchten Forscher diverse Mikroalgen, die unter den
verschiedensten Bedingungen kultiviert wurden. Es wurden positive immun-
modulatorische Eigenschaften der grünen Mikroalgen (vor allem von Chlorella
vulgaris) entdeckt und ein möglicher Einsatz in der Humanmedizin abgeleitet.
In dieser Dissertation werden fünf Experimente beschrieben, in denen sowohl
der Nährwert des Mikroalgenproteins als auch die Gewährleistung der
Tiergesundheit bei der Verfütterung von Mikroalgen untersucht wurden. Des
Weiteren wurde untersucht, ob die Reproduktion und das Wachstum von
Labortieren durch die Verfütterung von Mikroalgen beeinflusst werden. Um die
Inhaltstoffe der Mikroalgen im Verdauungstrakt zugänglich zu machen, muss
deren Zellwand zerstört werden. Verschiedene Verfahren, die zu diesem Zweck
entwickelt wurden, waren bisher nicht effizient und unökonomisch. Neue,
effiziente und billige Zellaufschlussverfahren werden jedoch benötigt, um die
Mikroalgen zu einem neuen, leicht verfügbaren, natürlichen und modernen
Futtermittel zu machen. Daher wurden Behandlungsverfahren im elektrischen Feld
(Elektroporation) oder mittels Ultraschall entwickelt. Ziel unserer
Untersuchungen war, zu prüfen, ob diese neuen Methoden positiv zu bewerten
sind. Der Versuch war Teil des Projektes Entwicklung und
Produktionseinführung von speziellem Kraftfutter mit Zusatz von aufgewerteten
Mikroalgen der Frankerförder Forschungsgesellschaft mbH. Es wurden drei
behandelte Produkte der Grünalge Chlorella vulgaris (C1) untersucht nur
sprühgetrocknet; elektroporiert und sprühgetrocknet; Ultraschall-behandelt und
sprühgetrocknet. Dazu wurde ein N-Bilanz-Versuch mit männlichen Wistar-Ratten
mit einem mittleren Gewicht von 135-145 g durchgeführt. Diese wurden einzeln
in Stoffwechselkäfigen gehalten und erhielten die drei Algenprodukte als
alleinige Proteinquelle in Höhe von 150 mg N je 100 g Körpergewicht. Eine
interne Kontrollgruppe erhielt Kasein in gleicher Dosierung. Die scheinbare
Verdaulichkeit der sprühgetrockneten Chlorella vulgaris betrug 46.9 ± 12.7 %
(Mittelwert ± SD), die der elektroporierten Chlorella vulgaris 44.3 ± 7.5 %
und die der Ultraschall-behandelten Mikroalge 56.7 ± 13.7 %. Die Protein
efficiency ratio (PER) betrug jeweils 1.4 ± 0.3; 1.0 ± 0.5 und 2.1 ± 0.3. Die
N-Bilanz betrug 41.86 ± 32.8 mg; 31.3 ± 17.3 mg und 66.7 ± 30.1 mg und die
biologische Wertigkeit jeweils 93 ± 9.5 %; 93.6 ± 10 % und 101 ± 5 %. Die
Unterschiede zwischen den Parametern waren mit Ausnahme der biologischen
Wertigkeit für elektroporierte und Ultraschall-behandelte Chlorella vulgaris
statistisch signifikant (p<0.05; HSD-Tukey-Test). Die Verdaulichkeit und die
biologische Wertigkeit von Chlorella vulgaris wurden durch die
Ultraschallbehandlung erhöht und durch die Elektroporation verringert, was
zeigt, dass die Ultraschallbehandlung ein hilfreicher technologischer Prozess
in der praktischen Verarbeitung von grünen Mikroalgen in der
Nahrungsmittelindustrie sein könnte. Mikroalgen haben einen hohen Gehalt an
Nukleinsäuren, deren Purinbasen nach ihrer Absorption in Harnsäure (beim
Menschen) und Allantoin (bei den meisten Tieren) umgewandelt werden. Dies kann
zu einer erhöhten Harnsäurebildung und somit zu einem erhöhten Risiko von
Krankheiten führen, die mit diesem Stoffwechselprodukt assoziiert sind
(beispielsweise Gicht, Nierensteine). Um diese und die allgemeine Wirkung von
Algen auf Tiere zu prüfen, wurden wachsende und ausgewachsene Ratten über
einen Zeitraum von drei Monaten mit Rationen gefüttert, die 20 % Mikroalgen
als alleinige oder zusätzliche Proteinquelle enthielten. Im Serum und Harn
wurden regelmäßig Harnsäure und Allantoin bestimmt. Auch die Harnmenge wurde
erfasst. Es wurde das biochemisches Profil vom Serum und das Blutbild
erstellt. Die inneren Organe wurden gewogen und histologische Untersuchungen
des Darmes, der Leber und der Nieren vorgenommen. Es konnte kein negativer
Effekt der Verfütterung von Mikroalgen festgestellt werden. Der
Harnsäuregehalt änderte sich durch die Verfütterung der Algen nicht. Die
Allantoinausscheidung erhöhte sich in den ersten zwei Wochen nach Beginn der
Algenfütterung und erreichte danach wieder den Ausgangswert. Das biochemische
Profil und das Blutbild zeigten lediglich eine Senkung der ALP-Aktivität in
der Algengruppe, während die anderen erfassten Parameter unbeeinflusst
blieben. Der Darm war schwerer und länger bei Ratten, die die Mikroalgen
erhielten. In den Zellen der untersuchten Organe wurden keine histologischen
Abnormalitäten festgestellt. Die Fütterung von Ratten mit 20 % Mikroalgen in
der Ration führte zu keinen nachteiligen Effekten und kann daher auch in
dieser Höhe - als unbedenklich angesehen werden. Es wurde der Einfluss einer
Gabe von 1 % sprühgetrocknetem Pulver der Grünalge Chlorella vulgaris auf die
Reproduktion und das Wachstum von Mäusen untersucht. Die Studie wurde an
Mäusen der Linie Fzt:DU über drei Generationen vorgenommen. Die weiblichen
Tiere der F0-Generation erhielten ab dem 21. Lebenstag (Absetztag) entweder
eine Kontrollration oder eine Algenration. Sie wurden am 63. Lebenstag
angepaart und alle Tiere wurden tragend. Die Würfe wurden gewogen, gezählt und
standardisiert (4 männliche und 5 weibliche je Wurf). Die Jungen wurden am 10.
und 21. Lebenstag gezählt und gewogen. Nach dem Absetzen verblieben je Wurf 2
männliche und 2 weibliche Tiere (F1) für weitere Untersuchungen. Die
weiblichen Nachkommen wurden erneut am 63. Tag angepaart. Am 18. Tag der
Trächtigkeit wurden 57 und 59 Tiere (Kontroll- und Algengruppe) getötet, wobei
die lebenden, toten und absorbierten Föten sowie die corpora lutei gezählt
wurden. Auch wurden die lebenden Föten gewogen. Die von den verbliebenen
angepaarten weiblichen Nachkommen (51 bzw. 53 Muttertiere) geborenen Jungen
wurden gezählt und gewogen und blieben bei der Mutter, ohne dass eine
Standardisierung der Würfe vorgenommen wurde. Sie wurden erneut am 10. und 21
Lebenstag gewogen und gezählt. Je 2 männliche und 2 weiblich Tiere (F2) wurden
abgesetzt und verblieben für weitere Untersuchungen, die nach dem bereits
beschriebenen Schema wiederholt wurden. Es konnten keine Unterschiede in der
Zahl der Föten, corpora lutei und geborenen Jungtieren zwischen den Gruppen
oder den Generationen festgestellt werden. Die Würfe der Algengruppe waren am
Absetztag etwas schwerer und entwickelten sich nach dem Absetzen etwas besser
als die der Kontrollgruppe. Diese Effekt wurde über alle Generationen
beobachtet. Der technologische Prozess der Ultraschallbehandlung kann effektiv
in der Praxis eingesetzt werden, wobei Möglichkeiten gefunden werden müssen,
um die Kosten der Herstellung von Mikroalgen zu verringern. Es sind weitere
experimentelle Untersuchungen nötig, um die Algenmenge in Rationen zu
ermitteln, die positive Effekte bei landwirtschaftlichen Nutztieren bewirkt,
ohne die Produktionskosten in den Betrieben zusätzlich zu erhöhen

in der praktischen Verarbeitung von grünen Mikroalgen in der
Nahrungsmittelindustrie sein könnte. Mikroalgen haben einen hohen Gehalt an
Nukleinsäuren, deren Purinbasen nach ihrer Absorption in Harnsäure (beim
Menschen) und Allantoin (bei den meisten Tieren) umgewandelt werden. Dies kann
zu einer erhöhten Harnsäurebildung und somit zu einem erhöhten Risiko von
Krankheiten führen, die mit diesem Stoffwechselprodukt assoziiert sind
(beispielsweise Gicht, Nierensteine). Um diese und die allgemeine Wirkung von
Algen auf Tiere zu prüfen, wurden wachsende und ausgewachsene Ratten über
einen Zeitraum von drei Monaten mit Rationen gefüttert, die 20 % Mikroalgen
als alleinige oder zusätzliche Proteinquelle enthielten. Im Serum und Harn
wurden regelmäßig Harnsäure und Allantoin bestimmt. Auch die Harnmenge wurde
erfasst. Es wurde das biochemisches Profil vom Serum und das Blutbild
erstellt. Die inneren Organe wurden gewogen und histologische Untersuchungen
des Darmes, der Leber und der Nieren vorgenommen. Es konnte kein negativer
Effekt der Verfütterung von Mikroalgen festgestellt werden. Der
Harnsäuregehalt änderte sich durch die Verfütterung der Algen nicht. Die
Allantoinausscheidung erhöhte sich in den ersten zwei Wochen nach Beginn der
Algenfütterung und erreichte danach wieder den Ausgangswert. Das biochemische
Profil und das Blutbild zeigten lediglich eine Senkung der ALP-Aktivität in
der Algengruppe, während die anderen erfassten Parameter unbeeinflusst
blieben. Der Darm war schwerer und länger bei Ratten, die die Mikroalgen
erhielten. In den Zellen der untersuchten Organe wurden keine histologischen
Abnormalitäten festgestellt. Die Fütterung von Ratten mit 20 % Mikroalgen in
der Ration führte zu keinen nachteiligen Effekten und kann daher auch in
dieser Höhe - als unbedenklich angesehen werden. Es wurde der Einfluss einer
Gabe von 1 % sprühgetrocknetem Pulver der Grünalge Chlorella vulgaris auf die
Reproduktion und das Wachstum von Mäusen untersucht. Die Studie wurde an
Mäusen der Linie Fzt:DU über drei Generationen vorgenommen. Die weiblichen
Tiere der F0-Generation erhielten ab dem 21. Lebenstag (Absetztag) entweder
eine Kontrollration oder eine Algenration. Sie wurden am 63. Lebenstag
angepaart und alle Tiere wurden tragend. Die Würfe wurden gewogen, gezählt und
standardisiert (4 männliche und 5 weibliche je Wurf). Die Jungen wurden am 10.
und 21. Lebenstag gezählt und gewogen. Nach dem Absetzen verblieben je Wurf 2
männliche und 2 weibliche Tiere (F1) für weitere Untersuchungen. Die
weiblichen Nachkommen wurden erneut am 63. Tag angepaart. Am 18. Tag der
Trächtigkeit wurden 57 und 59 Tiere (Kontroll- und Algengruppe) getötet, wobei
die lebenden, toten und absorbierten Föten sowie die corpora lutei gezählt
wurden. Auch wurden die lebenden Föten gewogen. Die von den verbliebenen
angepaarten weiblichen Nachkommen (51 bzw. 53 Muttertiere) geborenen Jungen
wurden gezählt und gewogen und blieben bei der Mutter, ohne dass eine
Standardisierung der Würfe vorgenommen wurde. Sie wurden erneut am 10. und 21
Lebenstag gewogen und gezählt. Je 2 männliche und 2 weiblich Tiere (F2) wurden
abgesetzt und verblieben für weitere Untersuchungen, die nach dem bereits
beschriebenen Schema wiederholt wurden. Es konnten keine Unterschiede in der
Zahl der Föten, corpora lutei und geborenen Jungtieren zwischen den Gruppen
oder den Generationen festgestellt werden. Die Würfe der Algengruppe waren am
Absetztag etwas schwerer und entwickelten sich nach dem Absetzen etwas besser
als die der Kontrollgruppe. Diese Effekt wurde über alle Generationen
beobachtet. Der technologische Prozess der Ultraschallbehandlung kann effektiv
in der Praxis eingesetzt werden, wobei Möglichkeiten gefunden werden müssen,
um die Kosten der Herstellung von Mikroalgen zu verringern. Es sind weitere
experimentelle Untersuchungen nötig, um die Algenmenge in Rationen zu
ermitteln, die positive Effekte bei landwirtschaftlichen Nutztieren bewirkt,
ohne die Produktionskosten in den Betrieben zusätzlich zu erhöhen
Morphology of the transverse ligament of the atlas and the alar ligaments in the silver fox (Vulpes vulpes var)
Background
Recent new anatomical and histological features of craniocervical junction in dogs and cats were described providing evidence of differences between the carnivore species. No information on these structures in foxes exists.
Results
Two parts of the alar ligaments were found. A longer one aroused from dens of axis to the internal (medial) surface of the occipital condyles and was called apical part. A shorter part originated from the entire length of the lateral edge of the dens of axis and terminated on the internal wall of the vertebral foramen of atlas and thus was called the lateral part. The transverse ligament of the atlas was widened in the mid region, above the dens of axis, and thickened at enthesis. Periosteal fibrocartilage was detected in the transverse ligament of the atlas at the enthesis, and sesamoid fibrocartilage was present on periphery in the middle of the ligament.
Conclusions
The craniocervical junction in foxes differs in part from other carnivores such as dogs and cats but resembles that of mesaticephalic dogs. The sesamoid and periosteal fibrocartilage supports the transverse ligament of the atlas whereas the alar ligaments have no cartilage
Quantitation of angiogenesis in vitro induced by VEGF-A and FGF-2 in two different human endothelial cultures : an all-in-one assay
Angiogenic therapy is considered to be a promising tool for treatment of ischemic diseases. Many in vivo and in vitro
assays have been developed to identify potential proangiogenic drugs and to investigate their mode of action. However, until now no validated system exists that would allow quantitation of angiogenesis in vitro in only one assay. Here, a previously established all-in-one in vitro assay based on staging of the angiogenic cascade was validated by quantitation of the effects of the known proangiogenic factors VEGF-A and FGF-2. Both growth factors were applied separately or in combination to human endothelial cell cultures derived from the heart and the foreskin, and angiogenesis was quantitated over 30 days of culture. Additionally, gene expression of VEGFR-1, VEGFR-2 and FGFR-1 at 3, 10, 20 or 40 days of cultivation was quantitated by RT-qPCR. In both cultures, VEGF-A as well as FGF-2 induced a run through all defined stages of angiogenesis in vitro. Application of VEGF-A only led to formation of irregular globular endothelial structures, while FGF-2 resulted in development of regular capillary-like structures. Quantitation of the angiogenic effects of VEGF-A and transcripts of VEGFR-1 and VEGFR-2 showed that a high VEGFR-1/VEGFR-2 ratio evoked deceleration of angiogenesis
Elevated dietary zinc oxide levels do not have a substantial effect on porcine reproductive and respiratory syndrome virus (PPRSV) vaccination and infection
Background Porcine reproductive and respiratory syndrome virus (PRRSV) is one
of the most important infectious agents for the swine industry worldwide. Zinc
(Zn) salts, which are widely used as a dietary supplement in swine nutrition,
have shown antiviral effects in vitro as well as in vivo. The purpose of this
study was to determine the influence of dietary zinc oxide supplementation on
vaccination and challenge infection with PRRSV. Findings The clinical course
of PRRS and the success of vaccination with an experimental inactivated
vaccine were compared between animals receiving a conventional diet (50 ppm
Zn, control group) and diets supplemented with Zn oxide (ZnO) at final Zn
concentrations of 150 or 2,500 ppm. Pigs receiving higher dietary Zn levels
showed a tendency towards higher neutralizing antibody levels after infection,
while dietary Zn levels did not substantially influence the number of
antiviral IFN-gamma secreting cells (IFN-gamma-SC) or percentages of blood
immune cell subsets after infection. Finally, feeding higher dietary Zn levels
reduced neither clinical symptoms nor viral loads. Conclusions Our results
suggest that higher levels of dietary ZnO do not have the potential to
stimulate or modulate systemic immune responses after vaccination and
heterologous PRRSV infection to an extent that could improve the clinical and
virological outcome
High-dose dietary zinc oxide mitigates infection with transmissible gastroenteritis virus in piglets
Zinc (Zn) supplementation has been shown to reduce the incidence of diarrhea
and to protect animals from intestinal diseases, but the mechanisms of this
protective effect against virus infection in vivo have not yet been
elucidated. Transmissible gastroenteritis virus (TGEV) causes diarrhea in
piglets with an age-dependent decrease of severity. RESULTS: We used 60 weaned
piglets that were divided into three groups to evaluate the effect of
different Zn levels added to a conventional diet (50 mg Zn/kg diet, Znlow,
control group). The other groups received the diet supplemented with ZnO at
final concentrations of 150 mg Zn/kg diet (Znmed), or 2,500 mg/kg diet
(Znhigh). Oral challenge infection with TGEV was performed when the pigs had
been fed for 1 week with the respective diet. Half of the piglets of each
group were sacrificed at day 1 and 18 after challenge infection. Fecal
consistency was improved and body weights increased in the Znhigh group when
compared to the other groups, but no direct effect of Zn concentrations in the
diet on fecal TGEV shedding and mucosal immune responses was detectable.
However, in the Znhigh group, we found a prevention of villus atrophy and
decreased caspase-3-mediated apoptosis of jejunal epithelium. Furthermore,
pigs receiving high Zn diet showed a down-regulation of interferon (IFN)-α,
oligoadenylate synthetase (OAS), Zn transporter SLC39A4 (ZIP4), but up-
regulation of metallothionein-1 (MT1), as well as the Zn transporters SLC30A1
(ZnT1) and SLC30A5 (ZnT5). In addition, forskolin-induced chloride secretion
and epithelial resistance were controlled at a physiological level in the
Znhigh but not the other groups. Finally, in the Znhigh group, we documented
an earlier and higher systemic TGEV-specific serum antibody response.
CONCLUSIONS: These results suggest that high dietary Zn could provide enhanced
protection in the intestinal tract and stimulate the systemic humoral immune
response against TGEV infection

Dietary Enterococcus faecium NCIMB 10415 and Zinc Oxide Stimulate Immune Reactions to Trivalent Influenza Vaccination in Pigs but Do Not Affect Virological Response upon Challenge Infection
Swine influenza viruses (SIV) regularly cause significant disease in pigs
worldwide. Since there is no causative treatment of SIV, we tested if
probiotic Enterococcus (E.) faecium NCIMB 10415 or zinc (Zn) oxide as feed
supplements provide beneficial effects upon SIV infection in piglets. Seventy-
two weaned piglets were fed three different diets containing either E. faecium
or different levels of Zn (2500 ppm, Zn(high); 50 ppm, Zn(low)). Half of the
piglets were vaccinated intramuscularly (VAC) twice with an inactivated
trivalent SIV vaccine, while all piglets were then infected intranasally with
H3N2 SIV. Significantly higher weekly weight gains were observed in the E.
faecium group before virus infection, and piglets in Zn(high) and E. faecium
groups gained weight after infection while those in the control group
(Zn(low)) lost weight. Using ELISA, we found significantly higher
H3N2-specific antibody levels in the E. faecium+VAC group 2 days before and at
the day of challenge infection as well as at 4 and 6 days after challenge
infection. Higher hemagglutination inhibition (HI) titers were also observed
in the Zn(high)+VAC and E. faecium+VAC groups at 0, 1 and 4 days after
infection. However, there were no significant differences in virus shedding
and lung lesions between the dietary groups. Using flow cytometry analysis
significantly higher activated T helper cells and cytotoxic T lymphocyte
percentages in the PBMCs were detected in the Zn(high) and E. faecium groups
at single time points after infection compared to the Zn(low) control group,
but no prolonged effect was found. In the BAL cells no influence of dietary
supplementation on immune cell percentages could be detected. Our results
suggest that feeding high doses of zinc oxide and particularly E. faecium
could beneficially influence humoral immune responses after vaccination and
recovery from SIV infection, but not affect virus shedding and lung pathology
