Regulatory mechanism of the cyclin-dependent kinase 16 through the cyclin Y/14-3-3 complex

CDK16 is a cell-cycle-related cyclin-dependent kinase that functions primarily in neurite growth processes, axonal transport regulation and sperm development. It also plays a role in the progression of X-linked intellectual disability and, together with its activating partner cyclin Y, in the development and progression of various types of cancer. Consequently, CDK16 and cyclin Y could represent potential new therapeutic targets. Both CDK16 and cyclin Y are structurally atypical among similar proteins, and the mechanism of activation of CDK16 by cyclin Y is unclear. However, it is clear from recent studies that the interaction between CDK16 and cyclin Y requires the prior interaction of cyclin Y with 14-3-3 proteins. In addition, for cyclin Y to interact with 14-3-3 proteins, its phosphorylation on the S100 and S326 binding sites of 14-3- 3 is required. To investigate the mechanism of activation of CDK16 by cyclin Y, we decided to characterize the interaction between CDK16, cyclin Y and 14-3-3 proteins in detail. The interaction between CDK16, cyclin Y and 14-3-3 proteins was characterized using biophysical methods including fluorescence anisotropy measurements, native electrophoresis, size exclusion chromatography and protein crystallography. Podílí se především na procesech růstu neuritů, regulaci axonálního transportu a vývoji spermií. Hraje také roli v progresi XLID (z angl. X-linked intellectual dis- ability) a spolu se svým aktivačním partnerem cyklinem Y ve vývoji a progresi různých typů rakoviny. CDK16 a cyklin Y by proto mohly představovat potenciální nové terapeutické cíle. CDK16 i cyklin Y jsou strukturně atypické mezi podobnými proteiny a mecha- nismus aktivace CDK16 cyklinem Y zatím není známý. Z předchozích studií však vyplývá, že interakce mezi CDK16 a cyklinem Y vyžaduje předchozí interakci cyk- linu Y s 14-3-3 proteiny. Pro interakci cyklinu Y s 14-3-3 proteiny je nutná fosforylace jeho 14-3-3 vazebných míst, S100 a S326. S cílem zjistit přesný mechanismus aktivace CDK16 jsme se rozhodli blíže charakterizovat interakci mezi CDK16, cyklinem Y a 14-3-3 proteiny. Pro charakterizaci interakcí mezi CDK16, cyklinem Y a 14-3-3 proteiny byla využita řada biofyzikálních metod, včetně fluorescenční anisotropie, nativní gelové elektroforézy, gelové permeační chromatografie a proteinové krystalografie. English texts in English teaching

Diplomová práce se zabývá autentickými materiály při výuce anglického jazyka. Práce je rozdělena do dvou hlavních částí. Teoretická část poskytuje důležité informace o autentických materiálech v obecné rovině, jejich klasifikaci a výhody a nevýhody jejich použití. Práce také přináší bližší pohled na "reading skills" a "reading strategies", tato část je následována částí popisující kritéria pro výběr vhodného písemného autentického materiálu. Praktická část se zabývá náročností vybraných písemných autentických materiálů. Tato kapitola se skládá ze tří hlavních částí. Zaprvé byly texty analyzovány online programem zvaným "The Lexile Analyzer". Poté byly tyto texty zanalyzovány zkušenými učiteli vyučujícími na základních školách. Na závěr byly porovnány výsledky zpracované online analyzátorem a výsledky zpracované učiteli. Dále byli učitelé požádáni, aby zanalyzovali texty také z pohledu jejich vhodnosti ve výuce anglického jazyka. Jejich komentáře jsou uvedeny. Tyto výsledky a komentáře jsou doplněny o vyjádření učitelů týkající se používání autentických materiálů ve výuce anglického jazyka. Byli také požádáni, aby uvedli kritéria, které berou v úvahu při výběru těcho materiálů. Na závěr je obecně rozebrán dopad na jazykové dovednosti studentů.Katedra anglického jazykaObhájenoThis thesis deals with written authentic materials and their use in English teaching. It is divided into two main parts. The theoretical part provides essential information about authentic materials in general, their classification and both advantages and disadvantages of their use. The thesis presents a closer look at reading skills and reading strategies followed by a section desribing the criteria for selecting an appropriate written authentic material. The research part deals with the difficulty, ie. readability, of chosen written authentic materials. This chapter consists of three main parts. First of all, the texts were analyzed by an online program for evaluating text readability called the Lexile Analyzer. Then, these texts were analyzed by professional teachers teaching at primary schools. Finally, the results generated by the Lexile Analyzer and those carried out by the teachers were compared. Futhermore, the teachers were also asked to assess the texts in terms of theirs suitablity for English teaching. Their comments are stated. These results and comments are accompanied by teachers´ statements on using authentic materials for English classes and explaining what criteria they take into considerations when choosing them. Finally, the impact on students language performance is discussed

Cultural and historical factors influencing China's behaviour in international relations

Cílem této bakalářské práce je analýza vlivu kulturních hodnot a historie na postavení a vystupování Číny v mezinárodních vztazích. Práce je obecně zaměřena na období novověku čínských dějin, zejména poté na období druhé poloviny 20. století. V rámci kulturních vlivů jsou nejdůležitější vybrané charakteristiky vyplývající z čínské filozofie a kultury aplikovány na zahraniční politiku, tato část zahrnuje i rozbor kulturních paradigmat Číny z pohledu Geerta Hofstedeho. Stejně tak v rámci historie je proveden rozbor nejdůležitějších vybraných momentů čínských dějin a jejich odrazu jak v daném období, tak v současnosti. V závěru je nejen zhodnoceno, zda mají dané faktory na čínskou zahraniční politiku nějaký vliv, ale také zda je tento vliv významný. Součástí práce je i zhodnocení, nakolik uvedený vliv historie a kultury odpovídá realitě, neboli nakolik se shoduje teorie a praxe zahraniční politiky Číny.The aim of this bachelor's thesis is to make an analysis of a cultural and historical influence on a China's position and behaviour in international relations. The work examines the period of Chinese modern times, particularly the second half of the twentieth century. The main selected characteristics of Chinese philosophy and culture are applied on the Chinese foreign policy; the analysis and application of Geert Hofstede's cultural paradigms for Chinese culture is included as well. The main selected aspects of Chinese history and their influence are analysed after that, both in the given period and in present. The conclusion summarises the influence of these factors on Chinese foreign policy and assesses whether this influence is significant. It also contains the evaluation whether the observed influence complies with the reality, it means whether the theory of Chinese foreign policy corresponds with its practice

Cultural and historical factors influencing China's behaviour in international relations

The aim of this bachelor's thesis is to make an analysis of a cultural and historical influence on a China's position and behaviour in international relations. The work examines the period of Chinese modern times, particularly the second half of the twentieth century. The main selected characteristics of Chinese philosophy and culture are applied on the Chinese foreign policy; the analysis and application of Geert Hofstede's cultural paradigms for Chinese culture is included as well. The main selected aspects of Chinese history and their influence are analysed after that, both in the given period and in present. The conclusion summarises the influence of these factors on Chinese foreign policy and assesses whether this influence is significant. Current aspects of cross-strait relations

The aim of this master thesis is to assess present situation and current aspects of cross-strait relations in the area of mutual negotiation, achieved progress as well as possible future development. First, a brief summary of existing historic development of mutual relations of China and Taiwan is presented with the emphasis laid on historic moments influencing attitude and relations of both states. The main factors that have a significant influence on views and behaviour of both China and Taiwan and on their relationship as well are identified in the next chapter. The last chapter deals with the future of cross-strait relations; the main possible alternatives of prospective development of mutual relations of both sides of the Taiwan Strait with reference to the above mentioned factors are presented. The conclusion summarizes the main problems of current cross-strait relations and it also evaluates which variant of the future development of mutual relations of China and Taiwan is the most probable one
