    Effects of microwave vs. convection oven heating on the formation of oxidation products in canola (Brassica rapa subsp. oleifera) oil

    Research on the effects of microwave vs. "conventional" heating of dietary oils on lipid oxidation has been very limited. In this study, canola oil (Brassica rapa subsp. oleifera) was heated in either convection or microwave oven to compare the effects of heating methods on triacylglycerol (TAG) oxidation. Peroxide and p-anisidine values (PV and p-AV, respectively) were determined and liquid chromatography-mass spectrometric (LC-MS) analysis of non-oxidized and oxidized TAG molecular species was performed. Neither of the heat treatments caused any considerable changes in PV of the oil samples. However, increase in p-AV was observed. The change was higher in the oil heated in microwave oven, demonstrating a higher increase in the amount of secondary oxidation products. The changes were accompanied by a decrease in the polyunsaturated TAG molecular species ACN:DB (acyl carbon number: number of double bonds) 54: 7 and 54: 6, this change also being higher in the oil heated in microwave oven

    Fatty Acid Composition of Developing Sea Buckthorn (Hippophae rhamnoides L.) Berry and the Transcriptome of the Mature Seed

    Background: Sea buckthorn (Hippophae rhamnoides L.) is a hardy, fruit-producing plant known historically for its medicinal and nutraceutical properties. The most recognized product of sea buckthorn is its fruit oil, composed of seed oil that is rich in essential fatty acids, linoleic (18:2\u3c9-6) and \u3b1-linolenic (18:3\u3c9-3) acids, and pulp oil that contains high levels of monounsaturated palmitoleic acid (16:1\u3c9-7). Sea buckthorn is fast gaining popularity as a source of functional food and nutraceuticals, but currently has few genomic resources; therefore, we explored the fatty acid composition of Canadian-grown cultivars (ssp. mongolica) and the sea buckthorn seed transcriptome using the 454 GS FLX sequencing technology. Results: GC-MS profiling of fatty acids in seeds and pulp of berries indicated that the seed oil contained linoleic and \u3b1-linolenic acids at 33-36% and 30-36%, respectively, while the pulp oil contained palmitoleic acid at 32-42%. 454 sequencing of sea buckthorn cDNA collections from mature seeds yielded 500,392 sequence reads, which identified 89,141 putative unigenes represented by 37,482 contigs and 51,659 singletons. Functional annotation by Gene Ontology and computational prediction of metabolic pathways indicated that primary metabolism (protein>nucleic acid>carbohydrate>lipid) and fatty acid and lipid biosynthesis pathways were highly represented categories. Sea buckthorn sequences related to fatty acid biosynthesis genes in Arabidopsis were identified, and a subset of these was examined for transcript expression at four developing stages of the berry. Conclusion: This study provides the first comprehensive genomic resources represented by expressed sequences for sea buckthorn, and demonstrates that the seed oil of Canadian-grown sea buckthorn cultivars contains high levels of linoleic acid and \u3b1-linolenic acid in a close to 1:1 ratio, which is beneficial for human health. These data provide the foundation for further studies on sea buckthorn oil, the enzymes involved in its biosynthesis, and the genes involved in the general hardiness of sea buckthorn against environmental conditions.Peer reviewed: YesNRC publication: Ye

    Seduced by the dark side: integrating molecular and ecological perspectives on the influence of light on plant defence against pests and pathogens.

    Plants frequently suffer attack from herbivores and microbial pathogens, and have evolved a complex array of defence mechanisms to resist defoliation and disease. These include both preformed defences, ranging from structural features to stores of toxic secondary metabolites, and inducible defences, which are activated only after an attack is detected. It is well known that plant defences against pests and pathogens are commonly affected by environmental conditions, but the mechanisms by which responses to the biotic and abiotic environments interact are only poorly understood. In this review, we consider the impact of light on plant defence, in terms of both plant life histories and rapid scale molecular responses to biotic attack. We bring together evidence that illustrates that light not only modulates defence responses via its influence on biochemistry and plant development but, in some cases, is essential for the development of resistance. We suggest that the interaction between the light environment and plant defence is multifaceted, and extends across different temporal and biological scales

    Fish Oil Finishing Diet Maintains Optimal n-3 Long-Chain Fatty Acid Content in European Whitefish (Coregonus lavaretus)

    This study examined the effect of substituting vegetable oil for fish oil in feed, with subsequent re-introduction of fish oil-rich feed (finishing feeding) in late stages of growth, on the fatty acids of cultivated European whitefish (Coregonus lavaretus). Restorative finishing feeding with fish oil-rich feed for 15 and 25 weeks was sufficient to change the total content of nutritionally valuable long-chain n-3 fatty acids, eicosapentaenoic acid (20:5n-3) and docosahexaenoic acid (22:6n-3), to correspond to that of fish fed the fish oil-rich feed throughout their lifespan. Under natural conditions, 15 and 25 weeks correspond to weight gains of 75% and 100% (i.e. doubling), respectively. Also, the fatty acid profile of the fish was restored after finishing periods of 15 and 25 weeks. Limiting the use of fish oil by lowering the overall fat content of the feed (no vegetable oil added) resulted in a decrease in the long-chain n-3 fatty acids. Based on the results, after receiving a vegetable oil-rich diet, restorative fish oil-rich feeding in the last stages of growth in European whitefish is nutritionally justified in order to balance nutritional gain for consumers with sustainable use of finite marine oils. The results encourage commercial efforts to further utilize and optimize finishing feeding practices.201
