Inferring Population Preferences via Mixtures of Spatial Voting Models
Understanding political phenomena requires measuring the political
preferences of society. We introduce a model based on mixtures of spatial
voting models that infers the underlying distribution of political preferences
of voters with only voting records of the population and political positions of
candidates in an election. Beyond offering a cost-effective alternative to
surveys, this method projects the political preferences of voters and
candidates into a shared latent preference space. This projection allows us to
directly compare the preferences of the two groups, which is desirable for
political science but difficult with traditional survey methods. After
validating the aggregated-level inferences of this model against results of
related work and on simple prediction tasks, we apply the model to better
understand the phenomenon of political polarization in the Texas, New York, and
Ohio electorates. Taken at face value, inferences drawn from our model indicate
that the electorates in these states may be less bimodal than the distribution
of candidates, but that the electorates are comparatively more extreme in their
variance. We conclude with a discussion of limitations of our method and

Cataract research using electronic health records
<p>Abstract</p> <p>Background</p> <p>The eMERGE (electronic MEdical Records and Genomics) network, funded by the National Human Genome Research Institute, is a national consortium formed to develop, disseminate, and apply approaches to research that combine DNA biorepositories with electronic health record (EHR) systems for large-scale, high-throughput genetic research. Marshfield Clinic is one of five sites in the eMERGE network and primarily studied: 1) age-related cataract and 2) HDL-cholesterol levels. The purpose of this paper is to describe the approach to electronic evaluation of the epidemiology of cataract using the EHR for a large biobank and to assess previously identified epidemiologic risk factors in cases identified by electronic algorithms.</p> <p>Methods</p> <p>Electronic algorithms were used to select individuals with cataracts in the Personalized Medicine Research Project database. These were analyzed for cataract prevalence, age at cataract, and previously identified risk factors.</p> <p>Results</p> <p>Cataract diagnoses and surgeries, though not type of cataract, were successfully identified using electronic algorithms. Age specific prevalence of both cataract (22% compared to 17.2%) and cataract surgery (11% compared to 5.1%) were higher when compared to the Eye Diseases Prevalence Research Group. The risk factors of age, gender, diabetes, and steroid use were confirmed.</p> <p>Conclusions</p> <p>Using electronic health records can be a viable and efficient tool to identify cataracts for research. However, using retrospective data from this source can be confounded by historical limits on data availability, differences in the utilization of healthcare, and changes in exposures over time.</p
Updating known distribution models for forecasting climate change impact on endangered species
To plan endangered species conservation and to design adequate management programmes, it is necessary to predict their
distributional response to climate change, especially under the current situation of rapid change. However, these
predictions are customarily done by relating de novo the distribution of the species with climatic conditions with no regard
of previously available knowledge about the factors affecting the species distribution. We propose to take advantage of
known species distribution models, but proceeding to update them with the variables yielded by climatic models before
projecting them to the future. To exemplify our proposal, the availability of suitable habitat across Spain for the endangered
Bonelli’s Eagle (Aquila fasciata) was modelled by updating a pre-existing model based on current climate and topography to
a combination of different general circulation models and Special Report on Emissions Scenarios. Our results suggested that
the main threat for this endangered species would not be climate change, since all forecasting models show that its
distribution will be maintained and increased in mainland Spain for all the XXI century. We remark on the importance of
linking conservation biology with distribution modelling by updating existing models, frequently available for endangered
species, considering all the known factors conditioning the species’ distribution, instead of building new models that are

Driver self-regulation and depressive symptoms in cataract patients awaiting surgery: a cross-sectional study
Background: Cataract is an extremely common visual condition of ageing. Evidence suggests that visual impairment influences driving patterns and self-regulatory behavior among older drivers. However, little is known about the psychological effects of driver self-regulation among older drivers. Therefore, this study aimed to describe driver self-regulation practices among older bilateral cataract patients and to determine the association between self-regulation and depressive symptoms. Methods: Ninety-nine older drivers with bilateral cataract were assessed the week before first eye cataract surgery. Driver self-regulation was measured via the Driving Habits Questionnaire. Depressive symptoms were assessed using the 20-item Center for Epidemiological Studies Depression Scale. Visual, demographic and cognitive data were also collected. Differences between self-regulators and non self-regulators were described and linear regression modeling used to determine the association between driver self-regulation and depressive symptoms score. Results: Among cataract patients, 48% reported self-regulating their driving to avoid at least one challenging situation. The situations most commonly avoided were driving at night (40%), on the freeway (12%), in the rain (9%) and parallel parking (8%). Self-regulators had significantly poorer contrast sensitivity in their worse eye than non self-regulators (p = 0.027). Driver self-regulation was significantly associated with increased depressive symptoms after controlling for potential confounding factors (p = 0.002).Conclusions: Driver self-regulation was associated with increased depressive symptoms among cataract patients. Further research should investigate this association among the general older population. Self-regulation programs aimed at older drivers may need to incorporate mental health elements to counteract unintended psychological effects
Prostacyclin reverses platelet stress fibre formation causing platelet aggregate instability
Prostacyclin (PGI2) modulates platelet activation to regulate haemostasis. Evidence has emerged to suggest that thrombi are dynamic structures with distinct areas of differing platelet activation. It was hypothesised that PGI2 could reverse platelet spreading by actin cytoskeletal modulation, leading to reduced capability of platelet aggregates to withstand a high shear environment. Our data demonstrates that post-flow of PGI2 over activated and spread platelets on fibrinogen, identified a significant reduction in platelet surface area under high shear. Exploration of the molecular mechanisms underpinning this effect revealed that PGI2 reversed stress fibre formation in adherent platelets, reduced platelet spreading, whilst simultaneously promoting actin nodule formation. The effects of PGI2 on stress fibres were mimicked by the adenylyl cyclase activator forskolin and prevented by inhibitors of protein kinase A (PKA). Stress fibre formation is a RhoA dependent process and we found that treatment of adherent platelets with PGI2 caused inhibitory phosphorylation of RhoA, reduced RhoA GTP-loading and reversal of myosin light chain phosphorylation. Phospho-RhoA was localised in actin nodules with PKA type II and a number of other phosphorylated PKA substrates. This study demonstrates that PGI2 can reverse key platelet functions after their initial activation and identifies a novel mechanism for controlling thrombosis
Water quality and planktonic microbial assemblages of isolated wetlands in an agricultural landscape

Wetlands occur in human-altered landscapes, and the ongoing ability of these wetlands to
provide ecosystem services is lacking. Additionally, the apparent lack of connection of some
wetlands, termed geographically isolated, to permanent waters has resulted in little regulatory
recognition. We examined the influence of intensive agriculture on water quality and planktonic
microbial assemblages of intermittently inundated wetlands. We sampled 10 reference and 10
agriculturally altered wetlands in the Gulf Coastal Plain of Georgia. Water quality measures
included pH, alkalinity, dissolved organic carbon, nutrients (nitrate, ammonium, and phosphate),
and filterable solids (dry mass and ash-free dry mass). We measured abundance and relative size
distribution of the planktonic microbial assemblage (< 45 μm) using flow cytometry. Water
quality in agricultural wetlands was characterized by elevated nutrients, pH, and suspended
solids. Autotrophic microbial cells were largely absent from both wetland types. Heterotrophic microbial abundance was influenced by nutrients and suspended matter concentration.
Agriculture caused changes in microbial assemblages forming the base of wetland food webs.
Yet, these wetlands potentially support important ecological services in a highly altered

Endothelial Expression of TGFβ Type II Receptor Is Required to Maintain Vascular Integrity during Postnatal Development of the Central Nervous System
TGFβ signalling in endothelial cells is important for angiogenesis in early embryonic development, but little is known about its role in early postnatal life. To address this we used a tamoxifen inducible Cre-LoxP strategy in neonatal mice to deplete the TypeII TGFβ receptor (Tgfbr2) specifically in endothelial cells. This resulted in multiple micro-haemorrhages, and glomeruloid-like vascular tufts throughout the cerebral cortices and hypothalamus of the brain as well as in retinal tissues. A detailed examination of the retinal defects in these mutants revealed that endothelial adherens and tight junctions were in place, pericytes were recruited and there was no failure of vascular smooth muscle differentiation. However, the deeper retinal plexus failed to form in these mutants and the angiogenic sprouts stalled in their progress towards the inner nuclear layer. Instead the leading endothelial cells formed glomerular tufts with associated smooth muscle cells. This evidence suggests that TGFβ signalling is not required for vessel maturation, but is essential for the organised migration of endothelial cells as they begin to enter the deeper layers of the retina. Thus, TGFβ signalling is essential in vascular endothelial cells for maintaining vascular integrity at the angiogenic front as it migrates into developing neural tissues in early postnatal life
Population based allele frequencies of disease associated polymorphisms in the Personalized Medicine Research Project
<p>Abstract</p> <p>Background</p> <p>There is a lack of knowledge regarding the frequency of disease associated polymorphisms in populations and population attributable risk for many populations remains unknown. Factors that could affect the association of the allele with disease, either positively or negatively, such as race, ethnicity, and gender, may not be possible to determine without population based allele frequencies.</p> <p>Here we used a panel of 51 polymorphisms previously associated with at least one disease and determined the allele frequencies within the entire Personalized Medicine Research Project population based cohort. We compared these allele frequencies to those in dbSNP and other data sources stratified by race. Differences in allele frequencies between self reported race, region of origin, and sex were determined.</p> <p>Results</p> <p>There were 19544 individuals who self reported a single racial category, 19027 or (97.4%) self reported white Caucasian, and 11205 (57.3%) individuals were female. Of the 11,208 (57%) individuals with an identifiable region of origin 8337 or (74.4%) were German.</p> <p>41 polymorphisms were significantly different between self reported race at the 0.05 level. Stratification of our Caucasian population by self reported region of origin revealed 19 polymorphisms that were significantly different (p = 0.05) between individuals of different origins. Further stratification of the population by gender revealed few significant differences in allele frequencies between the genders.</p> <p>Conclusions</p> <p>This represents one of the largest population based allele frequency studies to date. Stratification by self reported race and region of origin revealed wide differences in allele frequencies not only by race but also by region of origin within a single racial group. We report allele frequencies for our Asian/Hmong and American Indian populations; these two minority groups are not typically selected for population allele frequency detection. Population wide allele frequencies are important for the design and implementation of studies and for determining the relevance of a disease associated polymorphism for a given population.</p
