CTL are held to be an important host defense mechanism in persistent herpes-virus infections. We have therefore studied the nature and specificity of human cytomegalovirus (HCMV)-specific CTL in normal persistently infected individuals. This was achieved by using vaccinia recombinants encoding viral genes expressed at different stages of the virus replicative cycle, a structural glycoprotein gB (vac.gB) and the major 72-kD immediate early nonstructural protein (vac.IE) of HCMV, combined with limiting dilution analysis of the CTL response. In two subjects, 43 and 58% of HCMV CTL precursors (CTLp) lysed vac.IE-infected cells, in contrast to less than 6% lysing gB-infected cells. HCMV-specific CTL could also be generated by secondary in vitro stimulation with vac.gB- but not vac.IE-infected autologous fibroblasts. The high frequency of 72-kD IE protein-specific CTL suggests that this is at least a major recognition element for the HCMV-specific CTL response in asymptomatic persistently infected individuals, and CTL with this specificity may be important in maintaining the normal virus/host equilibrium

Host Inflammatory Response to Mosquito Bites Enhances the Severity of Arbovirus Infection
Aedes aegypti mosquitoes are responsible for trans- mitting many medically important viruses such as those that cause Zika and dengue. The inoculation of viruses into mosquito bite sites is an important and common stage of all mosquito-borne virus infec- tions. We show, using Semliki Forest virus and Bunyamwera virus, that these viruses use this inflam- matory niche to aid their replication and dissemina- tion in vivo. Mosquito bites were characterized by an edema that retained virus at the inoculation site and an inflammatory influx of neutrophils that coordi- nated a localized innate immune program that inad- vertently facilitated virus infection by encouraging the entry and infection of virus-permissive myeloid cells. Neutrophil depletion and therapeutic blockade of inflammasome activity suppressed inflammation and abrogated the ability of the bite to promote infec- tion. This study identifies facets of mosquito bite inflammation that are important determinants of the subsequent systemic course and clinical outcome of virus infection
Foot health education for people with rheumatoid arthritis : the practitioner's perspective
Background: Patient education is considered to be a key role for podiatrists in the management of patients with rheumatoid arthritis (RA). Patient education has undoubtedly led to improved clinical outcomes, however no attempts have been made to optimise its content or delivery to maximise benefits within the context of the foot affected by rheumatoid arthritis. The aim of this study was to identify the nature and content of podiatrists' foot health education for people with RA. Any potential barriers to its provision were also explored.
Methods: A focus group was conducted. The audio dialogue was recorded digitally, transcribed verbatim and analysed using a structured, thematic approach. The full transcription was verified by the focus group as an accurate account of what was said. The thematic analysis framework was verified by members of the research team to ensure validity of the data.
Results: Twelve members (all female) of the north west Podiatry Clinical Effectiveness Group for Rheumatology participated. Six overarching themes emerged: (i) the essence of patient education; (ii) the content; (iii) patient-centred approach to content and timing; (iv) barriers to provision; (v) the therapeutic relationship; and (vi) tools of the trade.
Conclusion: The study identified aspects of patient education that this group of podiatrists consider most important in relation to its: content, timing, delivery and barriers to its provision. General disease and foot health information in relation to RA together with a potential prognosis for foot health, the role of the podiatrist in management of foot health, and appropriate self-management strategies were considered to be key aspects of content, delivered according to the needs of the individual. Barriers to foot health education provision, including financial constraints and difficulties in establishing effective therapeutic relationships, were viewed as factors that strongly influenced foot health education provision. These data will contribute to the development of a patient-centred, negotiated approach to the provision of foot health education for people with RA
Antagonism, accommodation and agonism in critical management studies: alternative organizations as allies
Critical Management Studies has long been engaged in discussions about the purpose of critique and the possibilities of engagement. A recent expression calls for Critical Management Studies to moderate its ânegativeâ critique of management and instead use words like care, engagement and affirmation in order to enable âprogressiveâ engagement with managers. This âperformative turnâ has been poorly received by some who see it as a dilution of radical intent. We argue for a middle ground between the antagonistic versions of Critical Management Studies that appear to want to oppose management, and âperformativeâ scholars who appear to accommodate with managerialism. We do this by planting the debate firmly within an empirical setting and a crisis that the first author experienced as a âcritical scholarâ when conducting an ethnography at a sustainable financial services firm. In order to do this, we explore Chantal Mouffeâs concept of agonism to establish a particular mode of political engagement that acknowledges a space between being âforâ and being âagainstâ. We conclude by suggesting that the exploration of alternative forms of organization and management, themselves already involved in struggle against a hegemonic present, should be the proper task of a discipline that wishes to engage with the present and remain âcritical
Photocurrent measurements of supercollision cooling in graphene
The cooling of hot electrons in graphene is the critical process underlying
the operation of exciting new graphene-based optoelectronic and plasmonic
devices, but the nature of this cooling is controversial. We extract the hot
electron cooling rate near the Fermi level by using graphene as novel
photothermal thermometer that measures the electron temperature () as it
cools dynamically. We find the photocurrent generated from graphene
junctions is well described by the energy dissipation rate , where the heat capacity is and is the
base lattice temperature. These results are in disagreement with predictions of
electron-phonon emission in a disorder-free graphene system, but in excellent
quantitative agreement with recent predictions of a disorder-enhanced
supercollision (SC) cooling mechanism. We find that the SC model provides a
complete and unified picture of energy loss near the Fermi level over the wide
range of electronic (15 to 3000 K) and lattice (10 to 295 K) temperatures
