    The particle carriers of field-aligned currents in the Earth's magnetotail during a substorm

    Although the particle carriers of field-aligned currents (FACs) in the Earth's magnetotail play an important role in the transfer of momentum and energy between the solar wind, magnetosphere, and ionosphere, the characteristics of the FAC carriers have been poorly understood. Taking advantage of multiinstrument magnetic field and plasma data collected by the four spacecraft of the Cluster constellation as they traversed the northern plasma sheet boundary layer in the magnetotail on 14 September 2004, we identified the species type and energy range of the FAC carriers for the first time. The results indicate that part of tailward FACs is carried by energetic keV ions, which are probably originated from the ionosphere through outflow, and they are not too small (~2 nA/m2) to be ignored. The earthward (tailward) FACs are mainly carried by the dominant tailward (earthward) motion of electrons, and higher-energy electrons (from ~0.5 to 26 keV) are the main carriers

    Host Inflammatory Response to Mosquito Bites Enhances the Severity of Arbovirus Infection

    Aedes aegypti mosquitoes are responsible for trans- mitting many medically important viruses such as those that cause Zika and dengue. The inoculation of viruses into mosquito bite sites is an important and common stage of all mosquito-borne virus infec- tions. We show, using Semliki Forest virus and Bunyamwera virus, that these viruses use this inflam- matory niche to aid their replication and dissemina- tion in vivo. Mosquito bites were characterized by an edema that retained virus at the inoculation site and an inflammatory influx of neutrophils that coordi- nated a localized innate immune program that inad- vertently facilitated virus infection by encouraging the entry and infection of virus-permissive myeloid cells. Neutrophil depletion and therapeutic blockade of inflammasome activity suppressed inflammation and abrogated the ability of the bite to promote infec- tion. This study identifies facets of mosquito bite inflammation that are important determinants of the subsequent systemic course and clinical outcome of virus infection

    Impact of the solar wind dynamic pressure on the field‐aligned currents in the magnetotail: Cluster observation

    We statistically investigate the influence of the solar wind dynamic pressure (SW Pdyn) on the field-aligned currents (FACs) in the magnetotail with 1492 FAC cases from July to October in 2001 and 2004, which covers 74 Cluster crossings of the plasma sheet boundary layer (PSBL) in both storm time and non-storm time. The FAC density in the magnetotail is derived from the magnetic field data with the four-point measurement of Cluster, and the SW Pdyn is taken from ACE data. The results indicate the FAC density becomes stronger with increasing SW Pdyn. The statistics show that the FAC occurrence increased monotonically with SW Pdyn in the three levels (Weak: SW Pdyn5 nPa). The FAC density increased with increasing SW Pdyn, while its footprint (invariant latitude, ILAT) in the polar region decreased with increasing SW Pdyn. The response of the FAC to SW Pdyn in the magnetotail had a north-south hemispheric asymmetry. The FAC density had a better correlation with SW Pdyn in the Northern hemisphere, while the footprint had a better correlation with SW Pdyn in the Southern hemisphere. Possible underlying mechanisms for our results are analyzed and discussed. However, it requires more observations and simulation studies to find out the mechanism of north-south asymmetry

    Phenoloxidase activity acts as a mosquito innate immune response against infection with semliki forest virus

    Several components of the mosquito immune system including the RNA interference (RNAi), JAK/STAT, Toll and IMD pathways have previously been implicated in controlling arbovirus infections. In contrast, the role of the phenoloxidase (PO) cascade in mosquito antiviral immunity is unknown. Here we show that conditioned medium from the Aedes albopictus-derived U4.4 cell line contains a functional PO cascade, which is activated by the bacterium Escherichia coli and the arbovirus Semliki Forest virus (SFV) (Togaviridae; Alphavirus). Production of recombinant SFV expressing the PO cascade inhibitor Egf1.0 blocked PO activity in U4.4 cell- conditioned medium, which resulted in enhanced spread of SFV. Infection of adult female Aedes aegypti by feeding mosquitoes a bloodmeal containing Egf1.0-expressing SFV increased virus replication and mosquito mortality. Collectively, these results suggest the PO cascade of mosquitoes plays an important role in immune defence against arboviruses

    Boundary layer plasma flows from high-latitude reconnection in the summer hemisphere for northward IMF: THEMIS multi-point observations

    On 2008-07-11, the THEMIS spacecraft, separated both longitudinally and radially, traversed the dayside low-latitude boundary layer (LLBL) under extended northward IMF. They detected southward flows of magnetosheath plasma from magnetopause reconnection poleward of the northern cusp, which were cold-dense, and had southward velocity similar to 100 km/s and longitudinal extent >3 R-E. These features all agree with a global MHD simulation of the magnetosphere for similar conditions, in which under large geomagnetic dipole tilt, an LLBL forms via poleward-of-the-cusp reconnection first in the summer hemisphere and later in the other. Contrary to the simulation, however, the observed LLBL was mostly magnetically closed, characterized by balanced field-aligned and anti-field-aligned electron fluxes, and was less thick (<= 0.5 R-E). The former suggests comparable reconnection rate in both hemispheres, while the latter suggests the actual reconnection rate being lower, and/or the plasma transport toward the magnetotail being faster, than in the simulation.     Early and Late Pathogenic Events of Newborn Mice Encephalitis Experimentally Induced by Itacaiunas and Curionópolis Bracorhabdoviruses Infection

    In previous reports we proposed a new genus for Rhabdoviridae and described neurotropic preference and gross neuropathology in newborn albino Swiss mice after Curionopolis and Itacaiunas infections. In the present report a time-course study of experimental encephalitis induced by Itacaiunas and Curionopolis virus was conducted both in vivo and in vitro to investigate cellular targets and the sequence of neuroinvasion. We also investigate, after intranasal inoculation, clinical signs, histopathology and apoptosis in correlation with viral immunolabeling at different time points. Curionopolis and Itacaiunas viral antigens were first detected in the parenchyma of olfactory pathways at 2 and 3 days post-inoculation (dpi) and the first clinical signs were observed at 4 and 8 dpi, respectively. After Curionopolis infection, the mortality rate was 100% between 5 and 6 dpi, and 35% between 8 and 15 dpi after Itacaiunas infection. We identified CNS mice cell types both in vivo and in vitro and the temporal sequence of neuroanatomical olfactory areas infected by Itacaiunas and Curionopolis virus. Distinct virulences were reflected in the neuropathological changes including TUNEL immunolabeling and cytopathic effects, more intense and precocious after intracerebral or in vitro inoculations of Curionopolis than after Itacaiunas virus. In vitro studies revealed neuronal but not astrocyte or microglial cytopathic effects at 2 dpi, with monolayer destruction occurring at 5 and 7 dpi with Curionopolis and Itacaiunas virus, respectively. Ultrastructural changes included virus budding associated with interstitial and perivascular edema, endothelial hypertrophy, a reduced and/or collapsed small vessel luminal area, thickening of the capillary basement membrane, and presence of phagocytosed apoptotic bodies. Glial cells with viral budding similar to oligodendrocytes were infected with Itacaiunas virus but not with Curionopolis virus. Thus, Curionopolis and Itacaiunas viruses share many pathological and clinical features present in other rhabdoviruses but distinct virulence and glial targets in newborn albino Swiss mice brain

    Intravenously Administered Alphavirus Vector VA7 Eradicates Orthotopic Human Glioma Xenografts in Nude Mice

    VA7 is a neurotropic alphavirus vector based on an attenuated strain of Semliki Forest virus. We have previously shown that VA7 exhibits oncolytic activity against human melanoma xenografts in immunodeficient mice. The purpose of this study was to determine if intravenously administered VA7 would be effective against human glioma.In vitro, U87, U251, and A172 human glioma cells were infected and killed by VA7-EGFP. In vivo, antiglioma activity of VA7 was tested in Balb/c nude mice using U87 cells stably expressing firefly luciferase in subcutaneous and orthotopic tumor models. Intravenously administered VA7-EGFP completely eradicated 100% of small and 50% of large subcutaneous U87Fluc tumors. A single intravenous injection of either VA7-EGFP or VA7 expressing Renilla luciferase (VA7-Rluc) into mice bearing orthotopic U87Fluc tumors caused a complete quenching of intracranial firefly bioluminescence and long-term survival in total 16 of 17 animals. In tumor-bearing mice injected with VA7-Rluc, transient intracranial and peripheral Renilla bioluminescence was observed. Virus was well tolerated and no damage to heart, liver, spleen, or brain was observed upon pathological assessment at three and ninety days post injection, despite detectable virus titers in these organs during the earlier time point.VA7 vector is apathogenic and can enter and destroy brain tumors in nude mice when administered systemically. This study warrants further elucidation of the mechanism of tumor destruction and attenuation of the VA7 virus

    MHC class II-alpha chain knockout mice support increased viral replication that is independent of their lack of MHC class II cell surface expression and associated immune function deficiencies

    MHCII molecules are heterodimeric cell surface proteins composed of an α and β chain. These molecules are almost exclusively expressed on thymic epithelium and antigen presenting cells (APCs) and play a central role in the development and function of CD4 T cells. Various MHC-II knockout mice have been generated including MHC-IIAα-/- (I-Aα-/-), MHC-IIAβ-/- (I-β-/-) and the double knockout (I-Aαxβ-/-). Here we report a very striking observation, namely that alphaviruses including the avirulent strain of Semliki Forest virus (aSFV), which causes asymptomatic infection in wild-type C57BL6/J (B6) mice, causes a very acute and lethal infection in I-Aα-/-, but not in I-β-/- or I-Aαxβ-/-, mice. This susceptibility to aSFV is associated with high virus titres in muscle, spleen, liver, and brain compared to B6 mice. In addition, I-Aα-/- mice show intact IFN-I responses in terms of IFN-I serum levels and IFN-I receptor expression and function. Radiation bone marrow chimeras of B6 mice reconstituted with I-Aα-/- bone marrow expressed B6 phenotype, whereas radiation chimeras of I-Aα-/- mice reconstituted with B6 bone marrow expressed the phenotype of high viral susceptibility. Virus replication experiments both in vivo and in vitro showed enhanced virus growth in tissues and cell cultures derived form I-Aα-/- compared to B6 mice. This enhanced virus replication is evident for other alpha-, flavi- and poxviruses and may be of great benefit to producers of viral vaccines. In conclusion, I-Aα-/- mice exhibit a striking susceptibility to virus infections independent of their defective MHC-II expression.
