Analysis of Marketing Activities and Suggestion of a New Marketing Strategy of Sailtime Travel Agency

Tahle práce se zabývá sestavením návrhu marketingové strategie pro cestovní kancelář. Konkrétními doporučeními forem propagace se má stát marketingová strategie úspěšnější, má přinést majiteli firmy vyšší zisky a také získání nových zákazníků. Návrhy týkající se marketingové strategie vychází z dotazníkového šetření, následného vyhodnocení výsledků a nalezení nejvhodnějších návrhů umožňující zefektivnění marketingové komunikace a které mohou vést ke zvýšení počtu klientů společnosti.This thesis deals with formulation of a marketing strategy’s proposal for a travel agency. Due to specific recommendations of the forms of promotion the marketing strategy is expected to become more successful, brings higher profits for the owner and gain new customers. Suggestions regarding the marketing strategy are based on a questionnaire survey, the subsequent evaluation of the results and findings the most suitable proposals to allow more efficient marketing communication that may lead to an increase in the number of clients.

Development of Personal Income Tax in the Czech Republic

Import 30/10/2012Autorka bakalářské práce se zaměřuje na vývoj zdaňování příjmů fyzických osob v České republice. V první části autorka vymezuje nejdůležitější pojmy týkající se daně z příjmů, kdy také vysvětluje princip zdaňování příjmů ze závislé a podnikatelské činnosti spolu s problematikou odvodu daně. Tyto skutečnosti vymezuje autorka s platnou legislativou z roku 2012. Druhá část práce obsahuje změny v zákoně o daních z příjmů od roku 1993, které mají vliv na výši odvedené daně poplatníkem. V neposlední řadě se autorka věnuje aplikaci těchto změn na konkrétní fyzickou osobu, která je zaměstnaná a zároveň podniká. Autorka sleduje vývoj odvedené výše daně a vývoj složitosti při sestavování daňových přiznání v letech 1995, 2005, 2008 a 2011, kdy následně nastiňuje i budoucí vývoj problematiky těchto daní v souvislosti připravované reformy.The author of the Thesis focuses on the development of personal income taxation in the Czech Republic. In the first part the author defines the most important concepts relating to income tax, also the author explains the principle of taxation of income from employment and business activities and the author deals with the issue of tax too. The author defines all these problems in connection with laws of 2012. The second part of the Thesis includes changes in the law on income taxes since 1993. These changes affect the amount of tax paid. The author examines the application of these changes to a specific person who is employee and businessman too. The author follows the evolution of the amount of tax paid and the author follows the evolution of complicacy in the preparation of tax returns in 1995, 2005, 2008 and 2011. Regulatory mechanism of the cyclin-dependent kinase 16 through the cyclin Y/14-3-3 complex

CDK16 is a cell-cycle-related cyclin-dependent kinase that functions primarily in neurite growth processes, axonal transport regulation and sperm development. It also plays a role in the progression of X-linked intellectual disability and, together with its activating partner cyclin Y, in the development and progression of various types of cancer. Consequently, CDK16 and cyclin Y could represent potential new therapeutic targets. Both CDK16 and cyclin Y are structurally atypical among similar proteins, and the mechanism of activation of CDK16 by cyclin Y is unclear. However, it is clear from recent studies that the interaction between CDK16 and cyclin Y requires the prior interaction of cyclin Y with 14-3-3 proteins. In addition, for cyclin Y to interact with 14-3-3 proteins, its phosphorylation on the S100 and S326 binding sites of 14-3- 3 is required. To investigate the mechanism of activation of CDK16 by cyclin Y, we decided to characterize the interaction between CDK16, cyclin Y and 14-3-3 proteins in detail. The interaction between CDK16, cyclin Y and 14-3-3 proteins was characterized using biophysical methods including fluorescence anisotropy measurements, native electrophoresis, size exclusion chromatography and protein crystallography. Podílí se především na procesech růstu neuritů, regulaci axonálního transportu a vývoji spermií. Hraje také roli v progresi XLID (z angl. X-linked intellectual dis- ability) a spolu se svým aktivačním partnerem cyklinem Y ve vývoji a progresi různých typů rakoviny. CDK16 a cyklin Y by proto mohly představovat potenciální nové terapeutické cíle. CDK16 i cyklin Y jsou strukturně atypické mezi podobnými proteiny a mecha- nismus aktivace CDK16 cyklinem Y zatím není známý. Z předchozích studií však vyplývá, že interakce mezi CDK16 a cyklinem Y vyžaduje předchozí interakci cyk- linu Y s 14-3-3 proteiny. Pro interakci cyklinu Y s 14-3-3 proteiny je nutná fosforylace jeho 14-3-3 vazebných míst, S100 a S326. S cílem zjistit přesný mechanismus aktivace CDK16 jsme se rozhodli blíže charakterizovat interakci mezi CDK16, cyklinem Y a 14-3-3 proteiny. Pro charakterizaci interakcí mezi CDK16, cyklinem Y a 14-3-3 proteiny byla využita řada biofyzikálních metod, včetně fluorescenční anisotropie, nativní gelové elektroforézy, gelové permeační chromatografie a proteinové krystalografie. Vizualization and optical measurements of two-phase flows for pressure-swirl atomizer

Tato práce se zabývá studií Simplex a SR verze původní trysky používané ve spalovací komoře malého proudového motoru. Pro lepší porozumění procesu rozprašování byl vyroben transparentní model obou trysek v měřítku 10:1. Tvorba spreje u tlakové vířivé trysky je komplexní proces dvoufázového proudění. Vnitřní a vnější proudění bylo zkoumáno pomocí Laserového Dopplerovského Anemometru a vysokorychlostní kamery při vstupních tlacích p = 0.5, 1.0 a 1.5 MPa s použitím kerosinu a p-Cymenu. Z hlediska vnitřního proudění byla provedena charakteristika vzdušného jádra a zkoumán vliv různých SFR hodnot na délku a průměr jádra. Dále byly objasněny rychlostní profily společně s jejich fluktuačními složkami vzhledem k Reynoldsovu číslu a použité kapalině. Také jednoduchý numerický model byl vyvinut k odhadu vnitřního proudění pomocí trackování částic. V případě vnějšího proudění byl zkoumán vliv vstupního tlaku na rozpadovou vzdálenost spreje a úhel kužele spreje.The present thesis deals with a study of both Simplex and SR versions of the original atomizer used in a combustion chamber of the small turbojet engine. For a better understanding of the spray formation process, a transparent model of both atomizers was manufactured in the scale of 10:1. The spray formation of a pressure swirl atomizer is a complex process of the two-phase flow. The internal and external flow was examined using Laser Doppler Anemometry and a high-speed imaging at inlet pressures of p = 0.5, 1.0 and 1.5 MPa with the use of kerosene and p-Cymene as working liquids. In terms of internal flow, the air core characteristic was performed, and the influence of different SFR values on the air core length and diameter was investigated. The velocity profiles of the internal flow together with their fluctuation components have been further clarified, considering Reynolds number and used liquid. Also, the simple numerical model was developed to estimate the internal flow field by particle tracking. In the case of external flow, the dependence of the inlet pressure on the liquid sheet breakup length and the spray cone angle was investigated.

Vizualization of liquid break-up from atomization nozzles

V oblasti mechaniky tekutin hraje vizualizace významnou roli. Fotografie poskytují vhled do zásad chování tekutin a obecně pomáhají k lepšímu pochopení fyzikálních procesů proudění. Tato práce se zabývá vizualizací rozpadu kapalin u rozprašovacích trysek. Práce popisuje základy rozpadu kapalin, stejně jako metody pro vizualizaci. Dále je v práci probrána a experimentálně testována analýza prvků ve vizualizačních metodách. Laboratorní testy byly provedeny na malé tlakové vířivé trysce určené pro spalování kerosinu v proudovém leteckém motoru a část experimentů byla provedena na tlakové spirálové trysce. Práce dokumentuje vliv zdroje světla na kvalitu fotografie. Chování spreje je navíc analyzováno pomocí makrofotografie se zaměřením na primární rozpad, který hraje hlavní roli v celém procesu tvorby spreje a sprejových charakteristik.In the field of experimental fluid mechanics, visualizations play an important role. The images give an insight into the principal of fluid behavior and help with understanding of physical processes in flows in general. This thesis deals with visualization of liquid breakup from atomization nozzles. The thesis describes fundamentals of liquid atomization, as techniques for spray visualization. Further in this work an analysis of elements in the visualization techniques are discussed and tested experimentally. Laboratory tests were performed using small pressure-swirl atomizer originally designed for the combustion of kerosene in turbojet aircraft engine and a part of the experiments was carried out at a pressure spiral atomizer. The thesis documents the influence of the light source to image quality. Moreover, spray behavior is analysed using macro-photography with focus on primary breakup which plays major role in the whole process of the spray formation and spray characteristics.

Kukučín’s Unawakened in Gabriela Preissovás digest (The Slovak story-teller in the first Czech reception)

The study deals with the prose writer Martin Kukučín´s „Prague period“, namely the troubles with the adaptation of his representative psychological prose Neprebudený (Unawakened) in the Czech enviroment. The prose writer G. Preissová – later the author of the works such as Gazdina roba (Woman housekeeper) a Její pastorkyně (Her stepdaughter) – wishing to introduce Kukučín to Czech literature in her retold version titled Nemoudrý (Unwise) transformed his prose with many levels of meaning into a one-level text of popular light fiction. The study presents this adaptation mechanism and draws attention to the meaning shifts and reductions in comparison with the original. At the same time it describes the relationship between the Slovak prose writer and the Czech cultural enviroment. The author of the study builds on the methodological stimuli of W.Schmidt´s narrative transformation, which presents the mechanism of dealing with the text in the process of adaptation – digest, which is characterised as „accelerated narative“. The study is concluded with the finding that inspirations by Kukučín´s type of prose could later be seen in the Czech literary enviroment

Analysis and Assessment of the Financial Situation of the Company

Teoretická část obsahuje vymezení základních pojmů pro pochopení zkoumané problematiky. Praktická část obsahuje finanční analýzu společnosti, provedenou pomocí horiozntální a vertikální analýzy, rozdílových ukazatelů, poměrových ukazatelů, rozboru hospodaření Du – Pontova pyramidálního rozkladu a zhodnocení společnosti na základě bankrotních a bonitních modelů. V závěru práce je zařazeno doporučení budoucích kroků vedoucích k lepšímu hospodaření analyzované společnosti.The goal of this master’s work is evaluation of the financial situation of the company Aqua saning, Ltd. within the years 2008 to 2012 at the basis of elementary methods of the financial analysis. The theoretical part contains the definition of basic concepts for understanding the issues examined. The practical part contains a financial analysis to be performed by using of horizontal and vertical analysis, differential ratios, financial ratios, analysis of economic Du-Pont pyramidal decomposition and evaluation of the company under bankruptcy and credible models.
