    A Contingent Valuation Study Of Buriti (mauritia Flexuosa L.f.) In The Main Region Of Production In Brazil: Is Environmental Conservation A Collective Responsibility?

    Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)The immature leaves of the buriti palm (Mauritia flexuosa) are widely harvested in the municipality of Barreirinhas, Maranhão, for the production of handicrafts, which are sold to locals and tourists. The increasing demand for these artisanal goods is stimulating the emergence of an informal market for immature buriti leaves, leading to an intensification of their extraction and resulting in negative effects on local buriti palm populations and the ecosystem. Thus, the objective of the present study was to assess the environmental value of the buriti palm tree based on the maximum willingness to pay (WTP) for its conservation, using the contingent valuation method. Among the respondents, 99.74% reported that the palm species should be protected and the majority of them (65.75%) agreed to pay for its conservation (annual WTP R$ 179.49 ± 222.05). Multivariate analysis revealed that the WTP was not influenced by the socio-economic profile of the respondents. The main reasons for non-WTP are related to dissatisfaction with the government and the belief that financial contributions would not solve problems of environmental damage. Overall, the evaluated population believes that environmental conservation is not an obligation of the government or that of the population, but is a collective responsibility. © 2016, Sociedade Botanica do Brasil.     Development of structural layers PVC incorporating phase change materials for thermal energy storage

    The use of poly(vinyl chloride) (PVC) structural layers incorporating phase change materials (PCM) for latent heat thermal energy storage (LHTES) has become more attractive in the recent years compared to other supporting materials. In this study, PVC layers with different types of PCM were prepared using blending and compression moulding methods. Two types of synthesized PCM, one based on paraffin and calcium carbonate (PCM@CaCO3) and the other on paraffin, silica and graphene oxide (PCM@SiGO) have been developed to enhance the thermal conductivity of the PVC matrix and thus achieve a more effective charging and discharging process. PVC layers prepared using a commercial PCM (PCM@BASF) were also prepared for comparison. SEM images and DSC results reveal homogeneous distribution of the PCM in PVC layers and that most PCM particles are undamaged. The shell material (in the case of PCM@BASF) and the shape stability (in the case of synthesized PCM@CaCO3 and PCM@SiGO) prevent leakage of molten paraffin during the PVC layer production. The thermal conductivity profile of the PVC layer without PCM have a decreasing tendency with the temperature increase when determined using different measurement approaches, the transient plane heat source method (HotDisk Analyser, TPS 2500 S) and thermal flux meter method (steady-state method). However, for PVC layers with PCM the thermal conductivity profile shows a different behaviour when the mean surface temperature of the specimen is below the phase change transition temperature range (increasing tendency). During phase change transition (18–26 °C), the thermal conductivity presents two distinct tendencies. Firstly, the thermal conductivity reveals a decreasing tendency as the mean temperature of the specimen rises and afterwards an increasing tendency. Secondly, when the mean surface temperature is above the phase change transition temperature range, the thermal conductivity profile shows a decreasing tendency, independent of the PCM. The mechanical properties of PVC layers were also assessed and the results obtained revealed that the incorporation of PCM into the PVC matrix reduces the mechanical performance of the composites, however for LHETS applications not subjected to high tensile stress levels (over 1 kPa), this is not a significant drawback.publishe

    Nutritional composition and bioactivity of Umbilicus rupestris (Salisb.)Dandy: An underexploited edible wild plant

    The inclusion of edible wild plants in human diet has been receiving an increasing attention, as they represent an easily accessible source of nutrients, vitamins and antioxidants. In this work, the leaves of Umbelicus rupestris (Salisb.) Dandy, an edible species for which only scarce data is available in literature, were thoroughly evaluated for its nutritional profile, chemical characterization and bioactive properties. Being considered a succulent plant, the leaves revealed a very high content of moisture, with several beneficial compounds, including omega-3 polyunsaturated fatty acids, tocopherols and different polyphenols. A total of twelve flavonoids, three phenolic acids and one phenylpropanoid glucoside were identified in the decoction and/or hydroethanolic extracts, with most of them being described for the first time in this plant.     Carcass characteristics and meat quality of sheep fed buffelgrass silage to replace corn silage

    The aim of the study was to evaluate the carcass characteristics, proximate composition, and sensorial attributes of meat from sheep fed diets in which buffelgrass silage replaced corn silage. Thirty-two intact male crossbred Santa Inês sheep with an average live weight of 20.09 ± 2.0 kg were housed in individual stalls and allotted at random to four treatments in which corn silage was replaced by buffelgrass silage at the levels of 0 (control), 33.3%, 66.6%, and 100%. After an adaption period of 10 days, the sheep were fed for an additional 61 days. Feed was offered ad libitum and corn silage comprised 60% of the diet for the control group. Carcass characteristics, non-carcass components and meat quality were evaluated. Hot carcass yield, cold carcass yield, true carcass yield, trimmings, fat weight, and mesenteric and omental fat weight were highest for the control group (P <0.05). Loin eye area had a quadratic response (P =0.02), with the largest areas being observed in animals fed the diet containing 66.6% buffelgrass silage. Liver weight (P <0.01), luminosity of the meat (P <0.05), and cooking loss (P <0.05) likewise had nonlinear responses to the concentration of buffelgrass silage in the diet. The treatments did not have significant negative influence on the nutritional and organoleptic characteristics of the meat

    The role of natural regeneration to ecosystem services provision and habitat availability: a case study in the Brazilian Atlantic Forest

    Natural regeneration provides multiple benefits to nature and human societies, and can play a major role in global and national restoration targets. However, these benefits are context specific and impacted by both biophysical and socioeconomic heterogeneity across landscapes. Here we investigate the benefits of natural regeneration for climate change mitigation, sediment retention and biodiversity conservation in a spatially explicit way at very high resolution for a region within the global biodiversity hotspot of the Atlantic Forest. We classified current land-use cover in the region and simulated a natural regeneration scenario in abandoned pasturelands, areas where potential conflicts with agricultural production would be minimized and where some early stage regeneration is already occurring. We then modelled changes in biophysical functions for climate change mitigation and sediment retention, and performed an economic valuation of both ecosystem services. We also modelled how land-use changes affect habitat availability for species. We found that natural regeneration can provide significant ecological and social benefits. Economic values of climate change mitigation and sediment retention alone could completely compensate for the opportunity costs of agricultural production over 20 years. Habitat availability is improved for three species with different dispersal abilities, although by different magnitudes. Improving the understanding of how costs and benefits of natural regeneration are distributed can be useful to design incentive structures that bring farmers’ decision making more in line with societal benefits. This alignment is crucial for natural regeneration to fulfil its potential as a large-scale solution for pressing local and global environmental challenges

    Screening polymeric ionic liquids for chromatography-based purification of bacteriophage M13

    M13 bacteriophage is a key instrument in phage display applications, as well as a possible antibacterial therapeutic agent due to its highly restrictive bacterial pathogenesis, and other applications. The traditional phage purification process is usually achieved by gradient ultracentrifugation or a combination of precipitation, centrifugation and microfiltration. These approaches easily lead to long process times, high operational costs, phage aggregation and consequent product loss (approximately 60%). This work is thus focused on an alternative potential large-scale process to achieve high yield and purity while minimizing the operational costs. Electrostatic-based separation processes are also common biomolecules purification techniques. Although anion exchange chromatography has been used before to purify several viral particles, this technique has been poorly reported for the purification of M13 phage. In a recent work, our group has demonstrated the use of a predominant anion exchange process, where a polymeric ionic liquid (PIL) was used as an alternative separation matrix for M13 bacteriophage. In this work, a variety of system parameters was studied, including chemical structure of the cation and the anion, the crosslinker nature and its concentration, either in batch adsorption/elution or chromatographic operation mode. The PIL-based chromatographic operation mode revealed to be a suitable separation process for M13 from directly filtered E. coli supernatant, reaching over 70% M13 recovery and 4.6 purification factor in a single step.     Alteration of PTGS2 Promoter Methylation in Chronic Periodontitis

    Levels of prostaglandin E2 and the prostaglandin-endoperoxide synthase-2 (PTGS2, or COX-2) increase in actively progressing periodontal lesions, but decrease in chronic disease. We hypothesized that chronic inflammation is associated with altered DNA methylation levels within the PTGS2 promoter, with effects on COX-2 mRNA expression. PTGS2 promoter methylation levels from periodontally inflamed gingival biopsies showed a 5.06-fold increase as compared with non-inflamed samples (p = 0.03), and the odds of methylation in a CpG site in the inflamed gingival group is 4.46 times higher than in the same site in the non-inflamed group (p = 0.016). The level of methylation at −458 bp was inversely associated with transcriptional levels of PTGS2 (RT-PCR) (p = 0.01). Analysis of the data suggests that, in chronically inflamed tissues, there is a hypermethylation pattern of the PTGS2 promoter in association with a lower level of PTGS2 transcription, consistent with a dampening of COX-2 expression in chronic periodontitis. These findings suggest that the chronic persistence of the biofilm and inflammation may be associated with epigenetic changes in local tissues at the biofilm-gingival interface
