    Complexity of diatom response to Lateglacial and Holocene climate and environmental change in ancient, deep, and oligotrophic Lake Ohrid (Macedonia/Albania)

    Lake Ohrid (Macedonia/Albania) is a rare example of a deep, ancient Mediterranean lake and is a key site for palaeoclimate research in the northeastern Mediterranean region. This study conducts the first high-resolution diatom analysis during the Lateglacial and Holocene in Lake Ohrid. It demonstrates a complex diatom response to temperature change, with a direct response to temperature-induced productivity and an indirect response to temperature-related stratification/mixing regime and epilimnetic nutrient availability. During the Lateglacial (ca. 12 300–11 800 cal yr BP), the low-diversity dominance of hypolimnetic Cyclotella fottii indicates low temperature-dependent lake productivity. During the earliest Holocene (ca. 11 800–10 600 cal yr BP), although the slight increase in small, epilimnetic C. minuscula suggests climate warming and enhanced thermal stratification, diatom concentration remains very low as during the Lateglacial, indicating that temperature increase was muted. The early Holocene (ca. 10 600–8200 cal yr BP) marked a sustained increase in epilimnetic taxa, with mesotrophic C. ocellata indicating high temperature-induced lake productivity between ca. 10 600–10 200 cal yr BP and between ca. 9500–8200 cal yr BP, and with C. minuscula in response to low nutrient availability in the epilimnion between ca. 10 200–9500 cal yr BP. During the mid Holocene (ca. 8200–2600 cal yr BP), when sedimentological and geochemical proxies provide evidence for high temperature, anomalously low C. ocellata abundance is probably a response to epilimnetic nutrient limitation, almost mimicking the Lateglacial flora apart from mesotrophic Stephanodiscus transylvanicus indicative of high temperature-induced productivity in the hypolimnion. During the late Holocene (ca. 2600–0 cal yr BP), high abundance and fluctuating composition of epilimnetic taxa is largely a response to enhanced anthropogenic nutrient input. In this deep, oligotrophic lake, this study demonstrates the strong influence of lake physical and chemical processes in mediating the complex response of diatoms to climate change with particular respect to temperature

    Lake Ohrid (Macedonia and Albania) is a rare example of a deep, ancient Mediterranean lake and is a key site for palaeoclimate research in the northeastern Mediterranean region. This study conducts the analysis of diatoms as a proxy for Lateglacial and Holocene climate and environmental change in Lake Ohrid at a higher resolution than in previous studies. While Lake Ohrid has the potential to be sensitive to water temperature change, the data demonstrate a highly complex diatom response, probably comprising a direct response to temperature-induced lake productivity in some phases and an indirect response to temperature-related lake stratification or mixing and epilimnetic nutrient availability in others. The data also demonstrate the possible influence of physical limnological (e.g. the influence of wind stress on stratification or mixing) and chemical processes (e.g. the influence of catchment dynamics on nutrient input) in mediating the complex response of diatoms. During the Lateglacial (ca. 12 300–11 800 cal yr BP), the low-diversity dominance of hypolimnetic Cyclotella fottii indicates low lake productivity, linked to low water temperature. Although the subsequent slight increase in small, epilimnetic C. minuscula during the earliest Holocene (ca. 11 800–10 600 cal yr BP) suggests climate warming and enhanced stratification, diatom concentration remains as low as during the Lateglacial, suggesting that water temperature increase was muted across this major transition. The early Holocene (ca. 10 600–8200 cal yr BP) is characterised by a sustained increase in epilimnetic taxa, with mesotrophic C. ocellata indicating high water-temperature-induced productivity between ca. 10 600–10 200 cal yr BP and between ca. 9500–8200 cal yr BP and with C. minuscula in response to low nutrient availability in the epilimnion between ca. 10 200–9500 cal yr BP. During the middle Holocene (ca. 8200–2600 cal yr BP), when sedimentological and geochemical proxies provide evidence for maximum Holocene water temperature, anomalously low C. ocellata abundance is probably a response to epilimnetic nutrient limitation, almost mimicking the Lateglacial flora apart from the occurrence of mesotrophic Stephanodiscus transylvanicus in the hypolimnion. During the late Holocene (ca. 2600 cal yr BP–present), high abundance and fluctuating composition of epilimnetic taxa are probably a response more to enhanced anthropogenic nutrient input, particularly nitrogen enrichment, than to climate. Overall, the data indicate that previous assumptions concerning the linearity of diatom response in this deep, ancient lake are invalid, and multi-proxy analysis is essential to improve understanding of palaeolimnological dynamics in future research on the long, Quaternary sequence

    Intra-ER sorting of the peroxisomal membrane protein Pex3 relies on its luminal domain.

    Pex3 is an evolutionarily conserved type III peroxisomal membrane protein required for peroxisome formation. It is inserted into the ER membrane and sorted via an ER subdomain (the peroxisomal ER, or pER) to peroxisomes. By constructing chimeras between Pex3 and the type III ER membrane protein Sec66, we have been able to separate the signals that mediate insertion of Pex3 into the ER from those that mediate sorting within the ER to the pER subdomain. The N-terminal 17-amino acid segment of Pex3 contains two signals that are each sufficient for sorting to the pER: a chimeric protein containing the N-terminal domain of Pex3 fused to the transmembrane and cytoplasmic segments of Sec66 sorts to the pER in wild type cells, and does not colocalise with peroxisomes. Subsequent transport to existing peroxisomes requires the Pex3 transmembrane segment. When expressed in Drosophila S2R+ cells, ScPex3 targeting to peroxisomes is dependent on the intra-ER sorting signals in the N-terminal segment. The N-terminal segments of both human and Drosophila Pex3 contain intra-ER sorting information and can replace that of ScPex3. Our analysis has uncovered the signals within Pex3 required for the various steps of its transport to peroxisomes. Our generation of versions of Pex3 that are blocked at each stage along its transport pathway provides a tool to dissect the mechanism, as well as the molecular machinery required at each step of the pathway

    Risks, alternative knowledge strategies and democratic legitimacy: the conflict over co-incineration of hazardous industrial waste in Portugal.

    The decision to incinerate hazardous industrial waste in cement plants (the socalled ‘co-incineration’ process) gave rise to one of the most heated environmental conflicts ever to take place in Portugal. The bitterest period was between 1997 and 2002, after the government had made a decision. Strong protests by residents, environmental organizations, opposition parties, and some members of the scientific community forced the government to backtrack and to seek scientific legitimacy for the process through scientific expertise. The experts ratified the government’s decision, stating that the risks involved were socially acceptable. The conflict persisted over a decade and ended up clearing the way for a more sustainable method over which there was broad social consensus – a multifunctional method which makes it possible to treat, recover and regenerate most wastes. Focusing the analysis on this conflict, this paper has three aims: (1) to discuss the implications of the fact that expertise was ‘confiscated’ after the government had committed itself to the decision to implement co-incineration and by way of a reaction to the atmosphere of tension and protest; (2) to analyse the uses of the notions of ‘risk’ and ‘uncertainty’ in scientific reports from both experts and counter-experts’ committees, and their different assumptions about controllability and criteria for considering certain practices to be sufficiently safe for the public; and (3) to show how the existence of different technical scientific and political attitudes (one more closely tied to government and the corporate interests of the cement plants, the other closer to the environmental values of reuse and recycling and respect for the risk perception of residents who challenged the facilities) is closely bound up with problems of democratic legitimacy. This conflict showed how adopting more sustainable and lower-risk policies implies a broader view of democratic legitimacy, one which involves both civic movements and citizens themselves

    Innovations in air sampling to detect plant pathogens

    Many innovations in the development and use of air sampling devices have occurred in plant pathology since the first description of the Hirst spore trap. These include improvements in capture efficiency at relatively high air-volume collection rates, methods to enhance the ease of sample processing with downstream diagnostic methods and even full automation of sampling, diagnosis and wireless reporting of results. Other innovations have been to mount air samplers on mobile platforms such as UAVs and ground vehicles to allow sampling at different altitudes and locations in a short space of time to identify potential sources and population structure. Geographical Information Systems and the application to a network of samplers can allow a greater prediction of airborne inoculum and dispersal dynamics. This field of technology is now developing quickly as novel diagnostic methods allow increasingly rapid and accurate quantifications of airborne species and genetic traits. Sampling and interpretation of results, particularly action-thresholds, is improved by understanding components of air dispersal and dilution processes and can add greater precision in the application of crop protection products as part of integrated pest and disease management decisions. The applications of air samplers are likely to increase, with much greater adoption by growers or industry support workers to aid in crop protection decisions. The same devices are likely to improve information available for detection of allergens causing hay fever and asthma or provide valuable metadata for regional plant disease dynamics

    Bounds from Primordial Black Holes with a Near Critical Collapse Initial Mass Function

    Recent numerical evidence suggests that a mass spectrum of primordial black holes (PBHs) is produced as a consequence of near critical gravitational collapse. Assuming that these holes formed from the initial density perturbations seeded by inflation, we calculate model independent upper bounds on the mass variance at the reheating temperature by requiring the mass density not exceed the critical density and the photon emission not exceed current diffuse gamma-ray measurements. We then translate these results into bounds on the spectral index n by utilizing the COBE data to normalize the mass variance at large scales, assuming a constant power law, then scaling this result to the reheating temperature. We find that our bounds on n differ substantially (\delta n > 0.05) from those calculated using initial mass functions derived under the assumption that the black hole mass is proportional to the horizon mass at the collapse epoch. We also find a change in the shape of the diffuse gamma-ray spectrum which results from the Hawking radiation. Finally, we study the impact of a nonzero cosmological constant and find that the bounds on n are strengthened considerably if the universe is indeed vacuum-energy dominated today.Comment: 24 pages, REVTeX, 5 figures; minor typos fixed, two refs added, version to be published in PR

    Translation to practice: a randomised controlled study of an evidenced based booklet targeted at breast care nurses in the United Kingdom

    BACKGROUND: In the United Kingdom (UK), it was documented that a problem of knowledge transfer existed within the speciality of breast-cancer care, thus depriving patients of receiving optimal care. Despite increasingly robust research evidence indicating recommendation of whole body exercise for people affected by breast cancer, commensurate changes to practice were not noted amongst breast-care nurses (BCNs). AIM: To evaluate the effect of a targeted booklet, Exercise and Breast Cancer: A Booklet for Breast-Care Nurses, on changes in knowledge, reported practice, and attitudes of BCNs in the UK. METHOD: A prospective, experimental approach was used for designing a pre- and post-test randomised controlled study. Comparisons of knowledge, reported practice, and attitudes based on responses to a questionnaire were made at two time-points in two groups of BCNs (control and experimental). The unit of randomisation and analysis was hospital clusters of BCNs. The sample comprised 92 nurses from 62 hospitals. Analysis consisted of descriptive statistics and clustered regression techniques: clustered logistic regression for knowledge items, clustered linear regression for knowledge scores, ologit for attitude and reported practice items, and clustered multiple regression for paired and multiple variable analysis. RESULTS: A statistically significant increase in knowledge and changes in reported practice and attitudes were found. Robust variables affecting knowledge acquisition were: promotion of health, promotion of exercise, and understanding how exercise can reduce cancer-related fatigue. DISCUSSION: The study has shown that evidence-based printed material, such as an information booklet, can be used as an effective research dissemination method when developed for needs, values, and context of a target audience. CONCLUSIONS: This practical approach to research dissemination could be replicated and applied to other groups of nurses.</p

    The Formation of Cosmic Structures in a Light Gravitino Dominated Universe

    We analyse the formation of cosmic structures in models where the dark matter is dominated by light gravitinos with mass of 100 100 eV -- 1 keV, as predicted by gauge-mediated supersymmetry (SUSY) breaking models. After evaluating the number of degrees of freedom at the gravitinos decoupling (g∗g_*), we compute the transfer function for matter fluctuations and show that gravitinos behave like warm dark matter (WDM) with free-streaming scale comparable to the galaxy mass scale. We consider different low-density variants of the WDM model, both with and without cosmological constant, and compare the predictions on the abundances of neutral hydrogen within high-redshift damped Ly--α\alpha systems and on the number density of local galaxy clusters with the corresponding observational constraints. We find that none of the models satisfies both constraints at the same time, unless a rather small Ω0\Omega_0 value (\mincir 0.4) and a rather large Hubble parameter (\magcir 0.9) is assumed. Furthermore, in a model with warm + hot dark matter, with hot component provided by massive neutrinos, the strong suppression of fluctuation on scales of \sim 1\hm precludes the formation of high-redshift objects, when the low--zz cluster abundance is required. We conclude that all different variants of a light gravitino DM dominated model show strong difficulties for what concerns cosmic structure formation. This gives a severe cosmological constraint on the gauge-mediated SUSY breaking scheme.Comment: 28 pages,Latex, submitted for publication to Phys.Rev.

    The Physics of Cluster Mergers

    Clusters of galaxies generally form by the gravitational merger of smaller clusters and groups. Major cluster mergers are the most energetic events in the Universe since the Big Bang. Some of the basic physical properties of mergers will be discussed, with an emphasis on simple analytic arguments rather than numerical simulations. Semi-analytic estimates of merger rates are reviewed, and a simple treatment of the kinematics of binary mergers is given. Mergers drive shocks into the intracluster medium, and these shocks heat the gas and should also accelerate nonthermal relativistic particles. X-ray observations of shocks can be used to determine the geometry and kinematics of the merger. Many clusters contain cooling flow cores; the hydrodynamical interactions of these cores with the hotter, less dense gas during mergers are discussed. As a result of particle acceleration in shocks, clusters of galaxies should contain very large populations of relativistic electrons and ions. Electrons with Lorentz factors gamma~300 (energies E = gamma m_e c^2 ~ 150 MeV) are expected to be particularly common. Observations and models for the radio, extreme ultraviolet, hard X-ray, and gamma-ray emission from nonthermal particles accelerated in these mergers are described.Comment: 38 pages with 9 embedded Postscript figures.
