    Cooperative interactions of protein kinase C and cAMP-dependent protein kinase systems in human promyelocytic leukemia HL60 cells

    AbstractInteractions of protein kinase C (PKC) and cAMP-dependent protein kinase (PKA) systems were investigated in HL60 cells. It was found that the differentiating effects of 12-O-tetradecanoylphorbol-13-acetate (TPA) were potentiated by dibutyryl cAMP (dbcAMP) or prostaglandin E2 (PGE2). In addition, dbcAMP or PGE2 inhibited TPA-induced binding of PKC to plasma membrane, leading to decreased protein phosphorylation, and promoted subsequent redistribution of enzyme to the nuclear membrane region. The findings are consistent with the hypothesis that PKC and PKA systems regulate cooperatively the phenotypical differentiation of leukemic cells

    Bilateral Waveform Similarity Overlap-and-Add Based Packet Loss Concealment for Voice over IP

    This paper invested a bilateral waveform similarity overlap-and-add algorithm for voice packet lost. Since Packet lost will cause the semantic misunderstanding, it has become one of the most essential problems in speech communication. This investment is based on waveform similarity measure using overlap-and-Add algorithm and provides the bilateral information to enhance the speech signal reconstruction. Traditionally, it has been improved that waveform similarity overlap-and-add (WSOLA) technique is an effective algorithm to deal with packet loss concealment (PLC) for real-time time communication. WSOLA algorithm is widely applied to deal with the length adaptation and packet loss concealment of speech signal. Time scale modification of audio signal is one of the most essential research topics in data communication, especially in voice of IP (VoIP). Herein, the proposed the bilateral WSOLA (BWSOLA) that is derived from WSOLA. Instead of only exploitation one direction speech data, the proposed method will reconstruct the lost voice data according to the preceding and cascading data. The related algorithms have been developed to achieve the optimal reconstructing estimation. The experimental results show that the quality of the reconstructed speech signal of the bilateral WSOLA is much better compared to the standard WSOLA and GWSOLA on different packet loss rate and length using the metrics PESQ and MOS. The significant improvement is obtained by bilateral information and proposed method. The proposed bilateral waveform similarity overlap-and-add (BWSOLA) outperforms the traditional approaches especially in the long duration data loss

    Immunocytochemical localization of protein kinase C in resting and activated human neutrophils

    AbstractAn immunocytochemical method was used to determine possible changes in the subcellular distribution of protein kinase C (PKC) in human neutrophils in response to opsonized latex beads and zymosan. While in resting cells most of the PKC immunoreactivity was localized in the cytoplasm, a redistribution of PKC to the plasma and phagosomal membranes was observed in cells treated with latex beads or zymosan for 5–20 min, suggesting a participation of PKC in endocytosis

    Geochemical characterization of the mining district of Linares (Jaen, Spain) by means of XRF and ICP-AES

    Linares (Jaen, Spain) has been subjected to an intense mining and metallurgic activity during 2500 years. Basically, lead and silver have been extracted from galena; as well as copper and zinc. Different studies have been conducted to evaluate trace element concentrations to obtain relationships with anthropogenic and geologic factors. The analytical results of these studies have been obtained with ICP-AES. This analytical technique is relatively expensive and inaccessible in zones with scarce economic means. In this study, XRF analyses have been conducted and the obtained results are presented, as well as a comparison with the previously obtained ICP results. In both cases, a cluster analysis has been made to try to identify the same relations in the target area. 5 groups have been identified, mainly related with lithology. Only 10 of the 122 grid squares (each one of 1 km2) are classified in different groups if the cluster analysis is conducted with XRF results or ICP results. ICP classifies better where these grid squares are located, mostly, in contact zones of different lithologies

    A guanosine 5′-triphosphate-dependent protein kinase is localized in the outer envelope membrane of pea chloroplasts

    A guanosine 5-triphosphate (GTP)-dependent protein kinase was detected in preparations of outer chloroplast envelope membranes of pea (Pisum sativum L.) chloroplasts. The protein-kinase activity was capable of phosphorylating several envelope-membrane proteins. The major phosphorylated products were 23- and 32.5-kilo-dalton proteins of the outer envelope membrane. Several other envelope proteins were labeled to a lesser extent. Following acid hydrolysis of the labeled proteins, most of the label was detected as phosphoserine with only minor amounts detected as phosphothreonine. Several criteria were used to distinguish the GTP-dependent protein kinase from an ATP-dependent kinase also present in the outer envelope membrane. The ATP-dependent kinase phosphorylated a very different set of envelope-membrane proteins. Heparin inhibited the GTP-dependent kinase but had little effect upon the ATP-dependent enzyme. The GTP-dependent enzyme accepted phosvitin as an external protein substrate whereas the ATP-dependent enzyme did not. The outer membrane of the chloroplast envelope also contained a phosphotransferase capable of transferring labeled phosphate from [-32P]GTP to ADP to yield (-32P]ATP. Consequently, addition of ADP to a GTP-dependent protein-kinase assay resulted in a switch in the pattern of labeled products from that seen with GTP to that typically seen with ATP

    The use of synchrotron edge topography to study polytype nearest neighbour relationships in SiC

    A brief review of the phenomenon of polytypism is presented and its prolific abundance in Silicon Carbide discussed. An attempt has been made to emphasise modern developments in understanding this unique behaviour. The properties of Synchrotron Radiation are shown to be ideally suited to studies of polytypes in various materials and in particular the coalescence of polytypes in SiC. It is shown that with complex multipolytypic crystals the technique of edge topography allows the spatial extent of disorder to be determined and, from the superposition of Laue type reflections, neighbourhood relationships between polytypes can be deduced. Finer features have now been observed with the advent of second generation synchrotrons, the resolution available enabling the regions between adjoining polytypes to be examined more closely. It is shown that Long Period Polytypes and One Dimensionally Disordered layers often found in association with regions of high defect density are common features at polytype boundaries. An idealised configuration termed a "polytype sandwich" is presented as a model for the structure of SiC grown by the modified Lely technique. The frequency of common sandwich edge profiles are classified and some general trends of polytype neighbourism are summarised

    Three-generation flavor transitions and decays of supernova relic neutrinos

    If neutrinos have mass, they can also decay. Decay lifetimes of cosmological interest can be probed, in principle, through the detection of the redshifted, diffuse neutrino flux produced by all past supernovae--the so-called supernova relic neutrino (SRN) flux. In this work, we solve the SRN kinetic equations in the general case of three-generation flavor transitions followed by invisible (nonradiative) two-body decays. We then use the general solution to calculate observable SRN spectra in some representative decay scenarios. It is shown that, in the presence of decay, the SRN event rate can basically span the whole range below the current experimental upper bound--a range accessible to future experimental projects.     Matter effects and CP violating neutrino oscillations with non-decoupling heavy neutrinos

    The evolution equation for active and sterile neutrinos propagating in general anisotropic or polarized background environment is found and solved for a special case when heavy neutrinos do not decouple, resulting in non-unitary mixing among light neutrino states. Then new CP violating neutrino oscillation effects appear. In contrast to the standard unitary neutrino oscillations these effects can be visible even for two flavour neutrino transitions and even if one of the elements of the neutrino mixing matrix is equal to zero. They do not necessarily vanish with δm2→0\delta m^{2} \to 0 and they are different for various pairs of flavour neutrino transitions (νe→νμ\nu_e \to \nu_\mu), (νμ→ντ\nu_\mu \to \nu_\tau), (ντ→νe\nu_\tau \to \nu_e). Neutrino oscillations in vacuum and Earth's matter are calculated for some fixed baseline experiments and a comparison between unitary and non-unitary oscillations are presented. It is shown, taking into account the present experimental constraints, that heavy neutrino states can affect CP and T asymmetries. This is especially true in the case of νμ→ντ\nu_\mu \to \nu_\tau oscillations.Comment: 18 pages, 6 fig

    Model Independent Information On Solar Neutrino Oscillations

    We present the results of a Bayesian analysis of solar neutrino data in terms of nu_e->nu_{mu,tau} oscillations, independent from the Standard Solar Model predictions for the solar neutrino fluxes. We show that such a model independent analysis allows to constraint the values of the neutrino mixing parameters in limited regions around the usual SMA, LMA, LOW and VO regions. Furthermore, there is a strong indication in favor of large neutrino mixing and large values of Delta m^2 (LMA region). We calculate also the allowed ranges of the neutrino fluxes and we show that they are in good agreement with the Standard Solar Model prediction. In particular, the ratio of the 8B flux with its Standard Solar Model prediction is constrained in the interval [0.45,1.42] with 99.73% probability. Finally, we show that the hypothesis of no neutrino oscillations is strongly disfavored in a model independent way with respect to the hypothesis of neutrino oscillations.Comment: 40 pages, 20 figures.     Linear Response, Validity of Semi-Classical Gravity, and the Stability of Flat Space

    A quantitative test for the validity of the semi-classical approximation in gravity is given. The criterion proposed is that solutions to the semi-classical Einstein equations should be stable to linearized perturbations, in the sense that no gauge invariant perturbation should become unbounded in time. A self-consistent linear response analysis of these perturbations, based upon an invariant effective action principle, necessarily involves metric fluctuations about the mean semi-classical geometry, and brings in the two-point correlation function of the quantum energy-momentum tensor in a natural way. This linear response equation contains no state dependent divergences and requires no new renormalization counterterms beyond those required in the leading order semi-classical approximation. The general linear response criterion is applied to the specific example of a scalar field with arbitrary mass and curvature coupling in the vacuum state of Minkowski spacetime.
