Inhibition of SERCA and PMCA Ca2+-ATPase activities by polyoxotungstates
Plasma membrane calcium ATPases (PMCA) and sarco(endo) reticulum calcium ATPases (SERCA) are key proteins in the maintenance of calcium homeostasis. Herein, we compare for the first time the inhibition of SERCA and PMCA calcium pumps by several polyoxotungstates (POTs), namely by Wells-Dawson phospho-tungstate anions [P2W18O62]6-(intact, {P2W18}), [P2W17O61]10-(monolacunary, {P2W17}), [P2W15O56]12-(trilacunary, {P2W15}), [H2P2W12O48]12-(hexalacunary, {P2W12}), [H3P2W15V3O62]6- (trivanadium-substituted, {P2W15V3}) and by Preyssler-type anion [NaP5W30O110]14-({P5W30}). The speciation in the solu-tions of tested POTs was investigated by 31P and 51V NMR spectroscopy. The tested POTs inhibited SERCA Ca2+- ATPase activity, whereby the Preyssler POT showed the strongest effect, with an IC50 value of 0.37 mu M. For {P2W17} and {P2W15V3} higher IC50 values were determined: 0.72 and 0.95 mu M, respectively. Phlebologie an deutschen Hautkliniken: eine Bestandsaufnahme im Auftrag der Deutschen Gesellschaft fĂŒr Phlebologie

However, the percentage of dermatologists working in phlebology appears to be decreasing according to the data of the German Society of Phlebology (DGP). To investigate the reasons for this development, we-on behalf of the DGP-sent a questionnaire to 120 German Departments of Dermatology in autumn 2012. In 76 returned questionnaires, the number of physicians with additional fellowship training in phlebology averaged 1.5; the average number of those who fulfill the criteria for training fellows in phlebology was 0.9. In 71.1 % of the departments there was a phlebologist. A special phlebologic outpatient clinic existed in 73.7 % of the departments. Sonography with Doppler (89.5 %) and duplex (86.8 %) was used as the most frequent diagnostic tool. For therapy, compression (94.7 %), sclerotherapy (liquid 78.9 %, foam 63.2 %, catheter 18.4 %), endoluminal thermic procedures (radio wave 28.9 %, laser 17.1 %) and surgery (especially crossectomy and stripping 67.1 %, phlebectomy of tributaries 75 %) were used. The average number of treatments was very heterogenous in the different departments. Phlebology definitely plays an important role in dermatology. Most departments fulfill the formal criteria for the license to conduct advanced training in phlebology. A wide spectrum of phlebological diagnostic and therapeutic procedures is available.Hintergrund
Venenleiden sind eine Volkskrankheit und haben eine enorme sozioökonomische Bedeutung. Dennoch scheint der Anteil der phlebologisch tĂ€tigen Dermatologen entsprechend den Zahlen der Deutschen Gesellschaft fĂŒr Phlebologie (DGP) abzunehmen.
Methoden
Um die Ursachen dieser Entwicklung zu untersuchen, fĂŒhrten wir im Herbst 2012 im Auftrag der DGP eine Befragungsstudie zur Phlebologie an 120 deutschen Hautkliniken durch.
Ergebnisse
In 76 antwortenden Klinien gab es im Mittel 1,5 Phlebologen und 0,9 Ărzte mit Voraussetzungen zur WeiterbildungsermĂ€chtigung Phlebologie. In 71,1â% der Kliniken gab es einen Phlebologen, in 73,7â% eine phlebologische Sprechstunde. Zur Diagnostik wurden besonders die Doppler- (89,5â%) und Duplexsonographie (86,8â%) eingesetzt. In der Therapie fĂŒhrte die Kompressionstherapie (94,7â%), gefolgt von Sklerosierungstherapien (flĂŒssig 78,9â%, Schaum 63,2â%, Katheter 18,4â%), den endoluminalen thermischen Verfahren (Radiowelle 28,9â%, Laser 17,1â%) und den operativen Eingriffen (vor allem Cross- und Saphenektomie 67,1â%, Seitenastexhairese 75â%). Die durchschnittlichen Behandlungszahlen waren sehr heterogen.
Schlussfolgerung
Die Phlebologie spielt in der Dermatologie weiterhin eine wichtige Rolle. Mehrheitlich sind in den Kliniken die Möglichkeiten zur Weiterbildung Phlebologie gegeben. Es wird ein breites Spektrum an phlebologischen Techniken zur Diagnostik und Therapie angeboten
Identification of NAD(P)H Quinone Oxidoreductase Activity in Azoreductases from P. aeruginosa: Azoreductases and NAD(P)H Quinone Oxidoreductases Belong to the Same FMN-Dependent Superfamily of Enzymes
Water soluble quinones are a group of cytotoxic anti-bacterial compounds that are secreted by many species of plants, invertebrates, fungi and bacteria. Studies in a number of species have shown the importance of quinones in response to pathogenic bacteria of the genus Pseudomonas. Two electron reduction is an important mechanism of quinone detoxification as it generates the less toxic quinol. In most organisms this reaction is carried out by a group of flavoenzymes known as NAD(P)H quinone oxidoreductases. Azoreductases have previously been separate from this group, however using azoreductases from Pseudomonas aeruginosa we show that they can rapidly reduce quinones. Azoreductases from the same organism are also shown to have distinct substrate specificity profiles allowing them to reduce a wide range of quinones. The azoreductase family is also shown to be more extensive than originally thought, due to the large sequence divergence amongst its members. As both NAD(P)H quinone oxidoreductases and azoreductases have related reaction mechanisms it is proposed that they form an enzyme superfamily. The ubiquitous and diverse nature of azoreductases alongside their broad substrate specificity, indicates they play a wide role in cellular survival under adverse conditions

Toward artificial musselâglue proteins : differentiating sequence modules for adhesion and switchable cohesion
The synthesis of artificial musselâglue proteins with pH triggered cohesion control mechanism is described by extending the tyrosinase activated polymerization of peptides to sequences having specific modules for cohesion control. The high propensity of those sequence sections to adapt ÎČâsheets is temporarily suppressed by switch defects. This allows enzymatic activation and polymerization to proceed undisturbed. The ÎČâsheet formation is regained after polymerization by changing pH from 5.5 to 6.8, triggering OâN acyl transfer rearrangements that activate the cohesion mechanism. The resulting artificial mussel glue proteins exhibit rapid adsorption on alumina surfaces. The coatings resist harsh hypersaline conditions, and reach remarkable adhesive energies of 2.64 mJ/mâ2 on silica at pH 6.8. In in situ switch experiments, the minor pH change increases the adhesive properties of a coating by 300% and nanoindentation confirms the cohesion mechanism to improve bulkâstiffness by ~200%
