    Light-induced Fourier transform infrared (FTIR) spectroscopic investigations of the primary donor oxidation in bacterial photosynthesis

    AbstractFourier transform infrared (FTIR) difference spectroscopy of the primary electron donor (P) photo-oxidation has been performed for reaction centers (RCs) and chromatophores of purple photosynthetic bacteria. In the 1800–650 cm−1 spectral region highly reproducible absorbance changes were obtained that can be related to specific changes of individual bond absorption. Several bands in the difference spectra are tentatively assigned to changes of intensity and position of the keto and ester CO vibrations of the P bacteriochlorophylls, and a possible interpretation in terms of changes of their environment or type of bonding to the protein is given. Small difference bands in the amide I and II region allow only minor protein conformational changes

    Roadside verges and cemeteries: Comparative analysis of anthropogenic orchid habitats in the Eastern Mediterranean

    Several important habitats have become threatened in the last few centuries in the Mediterranean Basin due to major changes adopted in land-use practices. The consequent loss of natural and seminatural orchid habitats leads to the appreciation of small anthropogenic habitats, such as cemeteries and roadside verges. Colonization of cemeteries and roadside verges by orchids has long been known, but no study to date compared the suitability of these two anthropogenic habitats for orchids. Therefore, in this paper our aim was to survey cemeteries and roadside verges and to compare these two habitats regarding their role in conserving Mediterranean terrestrial orchids. We conducted field surveys in three Mediterranean islands, Cyprus, Crete, and Lesbos, where both cemeteries and roadside verges were sampled on a geographically representative scale. We found a total of almost 7,000 orchid individuals, belonging to 77 species in the two anthropogenic habitat types. Roadside verges hosted significantly more individuals than cemeteries in Crete and Lesbos, and significantly more species across all three islands. Our results suggest that although cemeteries have a great potential conservation value in other parts of the world, intensive maintenance practices that characterized cemeteries in these three islands renders them unable to sustain valuable plant communities. On the other hand, roadside verges play a prominent role in the conservation of Mediterranean orchids in Cyprus and Greece. The pioneer status of roadside verges facilitates their fast colonization, while roads serve as ecological corridors in fragmented landscapes

    Finite size effects and the order of a phase transition in fragmenting nuclear systems

    We discuss the implications of finite size effects on the determination of the order of a phase transition which may occur in infinite systems. We introduce a specific model to which we apply different tests. They are aimed to characterise the smoothed transition observed in a finite system.     Concerted EP2 and EP4 receptor signaling stimulates autocrine prostaglandin E(2) activation in human podocytes

    Glomerular hyperfiltration is an important mechanism in the development of albuminuria. During hyperfiltration, podocytes are exposed to increased fluid flow shear stress (FFSS) in Bowman's space. Elevated Prostaglandin E2 (PGE(2)) synthesis and upregulated cyclooxygenase 2 (Cox2) are associated with podocyte injury by FFSS. We aimed to elucidate a PGE2 autocrine/paracrine pathway in human podocytes (hPC). We developed a modified liquid chromatography tandem mass spectrometry (LC/ESI-MS/MS) protocol to quantify cellular PGE(2), 15-keto-PGE(2), and 13,14-dihydro-15-keto-PGE(2) levels. hPC were treated with PGE(2) with or without separate or combined blockade of prostaglandin E receptors (EP), EP2, and EP4. Furthermore, the effect of FFSS on COX2, PTGER2, and PTGER4 expression in hPC was quantified. In hPC, stimulation with PGE(2) led to an EP2- and EP4-dependent increase in cyclic adenosine monophosphate (cAMP) and COX2, and induced cellular PGE(2). PTGER4 was downregulated after PGE(2) stimulation in hPC. In the corresponding LC/ESI-MS/MS in vivo analysis at the tissue level, increased PGE(2) and 15-keto-PGE(2) levels were observed in isolated glomeruli obtained from a well-established rat model with glomerular hyperfiltration, the Munich Wistar Frömter rat. COX2 and PTGER2 were upregulated by FFSS. Our data thus support an autocrine/paracrine COX2/PGE(2) pathway in hPC linked to concerted EP2 and EP4 signaling

    Sulfur Isotopic Measurements from a West Antarctic Ice Core: Implications for Sulfate Source and Transport

    Measurements of delta(34)S covering the years 1935-76 and including the 1963 Agung (Indonesia) eruption were made on a West Antarctic firn core, RIDSA (78.73 degrees S, 116.33 degrees W; 1740m a.s.l.), and results are used to unravel potential source functions in the sulfur cycle over West Antarctica. The delta(34)S values Of SO42- range from 3.1 parts per thousand to 9.9 parts per thousand. These values are lower than those reported for central Antarctica, from near South Pole station, of 9.3-18.1 parts per thousand (Patris and others, 2000). While the Agung period is isotopically distinct at South Pole, it is not in the RIDSA dataset, suggesting differences in the source associations for the sulfur cycle between these two regions. Given the relatively large input of marine aerosols at RIDSA (determined from Na+ data and the seasonal SO42- cycle), there is likely a large marine biogenic SO42- influence. The delta(34)S values indicate, however, that this marine biogenic SO42-, with a well-established delta(34)S of 18 parts per thousand, is mixing with SO42- that has extremely negative delta(34)S values to produce the measured isotope values in the RIDSA core. We suggest that the transport and deposition of stratospheric SO42- in West Antarctica, combined with local volcanic input, accounts for the observed variance in delta(34)S values

    A 700 year record of Southern Hemisphere extratropical climate variability

    Annually dated ice cores from West and East Antarctica provide proxies for past changes in atmospheric circulation over Antarctica and portions of the Southern Ocean, temperature in coastal West and East Antarctica, and the frequency of South Polar penetration of El Niño events. During the period AD 1700–1850, atmospheric circulation over the Antarctic and at least portions of the Southern Hemisphere underwent a mode switch departing from the out-of-phase alternation of multi-decadal long phases of EOF1 and EOF2 modes of the 850 hPa field over the Southern Hemisphere (as defined in the recent record by Thompson and Wallace, 2000; Thompson and Solomon, 2002) that characterizes the remainder of the 700 year long record. From AD 1700 to 1850, lower-tropospheric circulation was replaced by in-phase behavior of the Amundsen Sea Low component of EOF2 and the East Antarctic High component of EOF1. During the first phase of the mode switch, both West and East Antarctic temperatures declined, potentially in response to the increased extent of sea ice surrounding both regions. At the end of the mode switch, West Antarctic coastal temperatures rose and East Antarctic coastal temperatures fell, respectively, to their second highest and lowest of the record. Polar penetration of El Niño events increased during the mode switch. The onset of the AD 1700–1850 mode switch coincides with the extreme state of the Maunder Minimum in solar variability. Late 20th-century West Antarctic coastal temperatures are the highest in the record period, and East Antarctic coastal temperatures close to the lowest. Since AD 1700, extratropical regions of the Southern Hemisphere have experienced significant climate variability coincident with changes in both solar variability and greenhouse gase

    The ice-breaker effect: Singing mediates fast social bonding

    It has been proposed that singing evolved to facilitate social cohesion. However, it remains unclear whether bonding arises out of properties intrinsic to singing or whether any social engagement can have a similar effect. Furthermore, previous research has used one-off singing sessions without exploring the emergence of social bonding over time. In this semi-naturalistic study, we followed newly formed singing and non-singing (crafts or creative writing) adult education classes over seven months. Participants rated their closeness to their group and their affect, and were given a proxy measure of endorphin release, before and after their class, at three timepoints (months 1, 3 and 7). We show that although singers and non-singers felt equally connected by timepoint 3, singers experienced much faster bonding: singers demonstrated a significantly greater increase in closeness at timepoint 1, but the more gradual increase shown by non-singers caught up over time. This represents the first evidence for an ‘ice-breaker effect’ of singing in promoting fast cohesion between unfamiliar individuals, which bypasses the need for personal knowledge of group members gained through prolonged interaction. We argue that singing may have evolved to quickly bond large human groups of relative strangers, potentially through encouraging willingness to coordinate by enhancing positive affect

    Centennial-scale variability of the Southern Hemisphere westerly wind belt in the eastern Pacific over the past two millennia

    We present the first high-resolution (sub-annual) dust particle data set from West Antarctica, developed from the West Antarctic Ice Sheet (WAIS) Divide deep ice core (79.468° S, 112.086° W), and use it to reconstruct changes in atmospheric circulation over the past 2400 years. We find a background dust flux of ~4 mg m−2 year−1 and a mode particle size of 5–8 μm diameter. Through comparing the WAIS Divide record with other Antarctic ice core particle records, we observe that coastal and lower-elevation sites have higher dust fluxes and coarser particle size distributions (PSDs) than sites on the East Antarctic plateau, suggesting input from local dust sources at these lower-elevation sites. In order to explore the use of the WAIS Divide dust PSD as a proxy for past atmospheric circulation, we make quantitative comparisons between both mid-latitude zonal wind speed and West Antarctic meridional wind speed and the dust size record, finding significant positive interannual relationships. We find that the dust PSD is related to mid-latitude zonal wind speed via cyclonic activity in the Amundsen Sea region. Using our PSD record, and through comparison with spatially distributed climate reconstructions from the Southern Hemisphere (SH) middle and high latitudes, we infer that the SH westerlies occupied a more southerly position from circa 1050 to 1400 CE (Common Era), coinciding with the Medieval Climate Anomaly (MCA). Subsequently, at ca. 1430 CE, the wind belt shifted equatorward, where it remained until the mid-to-late twentieth century. We find covariability between reconstructions of El Niño–Southern Oscillation (ENSO) and the mid-latitude westerly winds in the eastern Pacific, suggesting that centennial-scale circulation changes in this region are strongly influenced by the tropical Pacific. Further, we observe increased coarse particle deposition over the past 50 years, consistent with observations that the SH westerlies have been shifting southward and intensifying in recent decades
