The NAD(P)H oxidase homolog Nox4 modulates insulin-stimulated generation of H\u3csub\u3e2\u3c/sub\u3e0\u3csub\u3e2\u3c/sub\u3e and plays an integral role in insulin signal transduction
Insulin stimulation of target cells elicits a burst of H2O2 that enhances tyrosine phosphorylation of the insulin receptor and its cellular substrate proteins as well as distal signaling events in the insulin action cascade. The molecular mechanism coupling the insulin receptor with the cellular oxidant-generating apparatus has not been elucidated. Using reverse transcription-PCR and Northern blot analyses, we found that Nox4, a homolog of gp91phox, the phagocytic NAD(P)H oxidase catalytic subunit, is prominently expressed in insulin-sensitive adipose cells. Adenovirus-mediated expression of Nox4 deletion constructs lacking NAD(P)H or FAD/NAD(P)H cofactor binding domains acted in a dominant-negative fashion in differentiated 3T3-L1 adipocytes and attenuated insulin-stimulated H2O2 generation, insulin receptor (IR) and IRS-1 tyrosine phosphorylation, activation of downstream serine kinases, and glucose uptake. Transfection of specific small interfering RNA oligonucleotides reduced Nox4 protein abundance and also inhibited the insulin signaling cascade. Overexpression of Nox4 also significantly reversed the inhibition of insulin-stimulated IR tyrosine phosphorylation induced by coexpression of PTP1B by inhibiting PTP1B catalytic activity. These data suggest that Nox4 provides a novel link between the IR and the generation of cellular reactive oxygen species that enhance insulin signal transduction, at least in part via the oxidative inhibition of cellular protein-tyrosine phosphatases (PTPases), including PTP1B, a PTPase that has been previously implicated in the regulation of insulin action
Angola Cameia Development Casing-Settlement Calculations
The amount of axial settlement of casings supported by regions of axial elastic foundations is computed. The differential equation of axial equilibrium, including the foundation stiffnesses, is solved by use of cubic axial finite elements. The analysis is applied to 101 m of a vertical 914-mm (36-in.) casing supporting a 559-mm (22-in.) casing running from 3 m above the mudline to near the bottom of the 1201-m hole. The upper casing is supported by a region of layered clay sediments. The lower casing has a long openhole region, followed by a bottom region encased in cement. A series of loading increments is applied, with the connection of the tree/blowout preventer (BOP) as the last. The settlement at the mudline was calculated to be less than 0.1 m. This study shows that for weak upper foundations, a good cement job is needed to support the tree/BOP without large mudline settlements

Macrozoobenthic Community Structure in Subtidal Soft-bottom Area Along the Coast of Lembeh Island -North Sulawesi
The present paper describes the soft-bottom macrozoobenthic community structure inhabiting Lembeh Island's waters (North Sulawesi). Material for the study was collected from 5 stations in October 2013 using a La Motte grab (600 cm2) and subsequently sieved through a 1 mm square mesh sieve. A total of 1147 individuals belonging to 78 species (taxa) of macrozoobenthos and representative of higher taxonomic groups belonging to 12 phyla were recorded and identified. Univariate analysis showed low abundance of individuals and number of species in the Pintu Kota station which has a black sludge of sediment but Shannon index values at this station is the highest. Instead Motto station relatively far from anthropogenic disturbance showed a high abundance of individuals and number of species but Shannon index values at this station is the lowest. The station is dominated by Tanais sp at a density of 9533 individuals m-2. Shannon index is less sensitive to measure the effect of anthropogenic disturbances compared with the abundance of individuals and number of species. The multivariate analysis (Cluster Analysis and Correspondence Analysis) managed to separate the three groups (essemblage) makrozoobethos: Group A (Posokan), Group B (Motto) and Group C (Pancoran, Mawali and Pintu Kota). with regional economic indicators. It shows that penetration of the website by the provincial government
in Indonesia has yet to promote regional economic development or vice versa.

Pengaruh Dimensi Benda Uji Terhadap Kuat Tekan Beton
Kuat tekan adalah karakteristik mekanik utama dari beton yang dapat diketahui melalui penelitian uji tekan di laboratorium terhadap benda uji. Baik dalam bentuk kubus ataupun silinder dengan ukuran standar: 10cm x 10cm x 10cm dan 15cm x 15cm untuk kubus dan 10cm x 20cm dan 15cm x 30cm untuk silinder. Untuk mendapatkan informasi mengenai kecendrungan harga kuat tekan beton dengan variasi dimensi benda uji, telah dilakukan penelitian-penelitian di laboratoriun untuk mendapatkan komposisi campuran tertentu pada umur beton 28 hari, variasi ukuran benda uji dibuat: 10cm x 10cm x 10cm, 12,5cm x 12,5cm x 12,5cm dan 15cm x 15cm x 15cm untuk kubus dan 10cm x 20cm, 12,5cm x 25cm dan 15cm x 30cm untuk silinder. Dengan jumlah benda uji masing-masing 20 buah untuk setiap ukuran benda uji. Melalui prosedur standar pengujian kuat tekan dan menggunakan formula-formula baku perhitungan tekan rata-rata diperoleh informasi bahwa peningkatan ukuran dimensi benda uji menghasilkan penurunan kuat tekan rata-rata, untuk benda uji kubus dengan ukuran masing-masing: 10cm x 10cm x 10cm, 12,5cm x 12,5cm x 12,5cm dan 15cm x 15cm x 15cm diperoleh kuat tekan rata-rata masing-masing: 32,86MPa, 31,26MPa dan 31,036MPa. Sedangkan untuk silinder dengan kururan 10cm x 20cm, 12,5cm x 25cm dan 15cm x 30cm diperoleh kuat tekan rata-rata masing-masing: 31,47MPa, 30,85MPa dan 30,44MPa
