    The legalized marijuana industry is lucrative but surrounded with uncertainties. The divergence between state and federal law has pushed this industry into a state of limbo. Furthermore, at the federal level, the lack of enforcing the prohibition has only exacerbated the uncertainty. Historically, the federal government has taken a very relaxed approach and allowed marijuana businesses to operate with minimal interference. As a result, there is a thriving legalized marijuana industry operating throughout the majority of the United States. However, there are many obstacles which plague and threaten the future of this relatively young industry.     Construction and Calibration of a Low-Cost 3D Laser Scanner with 360â—¦ Field of View for Mobile Robots

    Navigation of many mobile robots relies on environmental information obtained from three-dimensional (3D) laser scanners. This paper presents a new 360◦ field-of-view 3D laser scanner for mobile robots that avoids the high cost of commercial devices. The 3D scanner is based on spinning a Hokuyo UTM- 30LX-EX two-dimensional (2D) rangefinder around its optical center. The proposed design profits from lessons learned with the development of a previous 3D scanner with pitching motion. Intrinsic calibration of the new device has been performed to obtain both temporal and geometric parameters.     Wave-front phase-modulation control and focusing of second-harmonic light generated in transparent nonlinear random structures

    We theoretically investigate how phase-only spatial light modulation can enable controlling and focusing the second-harmonic light generated in transparent nonlinear random structures. The studied structures are composed of domains with random sizes and antiparallel polarization, which accurately model widely used ferroelectric crystals such as strontium barium niobate. Using a first-principles Green-function formalism, we account for the effect that spatial light modulation of the fundamental beam introduces into the second-order nonlinear frequency conversion occurring in the considered class of structures. This approach provides a complete description of the physical origin of the second-harmonic light generation in the system, as well as the optimization of the light intensity in any arbitrary direction. Our numerical results show how the second-harmonic light is influenced by both the disorder in the structure and the boundaries of the crystal. Particularly, we find that the net result from the interplay between disorder and boundary effects is strongly dependent on the dimensions of the crystal and the observation direction. Remarkably, our calculations also show that although in general the maximum possible enhancement of the second-order light is the same as the one corresponding to linear light scattering in turbid media, in the Cerenkov phase matching direction the enhancement can exceed the linear limit. The theoretical analysis presented in this work expands the current understanding of light control in complex media and could contribute to the development of a new class of imaging and focusing techniques based on nonlinear frequency mixing in random optical materials.Peer ReviewedPostprint (published version

    Improved linkage analysis of Quantitative Trait Loci using bulk segregants unveils a novel determinant of high ethanol tolerance in yeast

    Background: Bulk segregant analysis (BSA) coupled to high throughput sequencing is a powerful method to map genomic regions related with phenotypes of interest. It relies on crossing two parents, one inferior and one superior for a trait of interest. Segregants displaying the trait of the superior parent are pooled, the DNA extracted and sequenced. Genomic regions linked to the trait of interest are identified by searching the pool for overrepresented alleles that normally originate from the superior parent. BSA data analysis is non-trivial due to sequencing, alignment and screening errors. Results: To increase the power of the BSA technology and obtain a better distinction between spuriously and truly linked regions, we developed EXPLoRA (EXtraction of over-rePresented aLleles in BSA), an algorithm for BSA data analysis that explicitly models the dependency between neighboring marker sites by exploiting the properties of linkage disequilibrium through a Hidden Markov Model (HMM). Reanalyzing a BSA dataset for high ethanol tolerance in yeast allowed reliably identifying QTLs linked to this phenotype that could not be identified with statistical significance in the original study. Experimental validation of one of the least pronounced linked regions, by identifying its causative gene VPS70, confirmed the potential of our method. Conclusions: EXPLoRA has a performance at least as good as the state-of-the-art and it is robust even at low signal to noise ratio's i.e. when the true linkage signal is diluted by sampling, screening errors or when few segregants are available

    Expression and signal transduction of the glucagon receptor in βTC3 cells

    AbstractThe expression and signal transduction of the glucagon receptor (GR) have been studied in βTC3 cells. Northern blot and RT-PCR analysis indicated the expression of the GR gene in βTC3 cells. One-5 nM glucagon stimulated a 2.5-fold increase in the IPs production. At glucagon concentrations higher than 5 nM, the production of IPs was blunted but not abolished. The accumulation of intracellular cAMP was observed following the stimulation with 5 nM of glucagon. A maximal 4.5-fold increase in cAMP was observed using 250 nM glucagon and higher. Comparative studies using a glucagon anatogonist, des-His1[Glu]9glucagon, showed no effect on intracellular cAMP and IPs in βTC3 cells. Our data shows that the GR gene is expressed in βTC3 cells. The GR in βTC3 cells transmits its intracellular signal by causing the accumulation of both IPs and cAMP
