    Nuevas estrategias terapéuticas en diabetes mellitus tipo 1

    El principal determinante del riesgo de complicaciones derivadas de la diabetes mellitus tipo 1 se debe a los altos niveles de glucosa en sangre mantenidos durante largo tiempo. Para conseguir un beneficio terapéutico en pacientes con diabetes mellitus es necesario desarrollar tratamientos que permitan de manera segura, efectiva y estable mantener la normoglucemia. Lamentablemente, el tratamiento de la diabetes mellitus tipo 1 mediante el aporte exógeno de insulina no es capaz de conseguir niveles estables de glucosa en sangre, de manera que con frecuencia se producen casos de severa hipoglucemia o hiperglucemia. Hasta la fecha la única solución para reestablecer de manera permanente la normoglucemia se consigue mediante el trasplante de páncreas o de islotes pancreáticos. Sin embargo, a medida que se incrementa el número de centros especializados en el trasplante de islotes, mayor es la necesidad de islotes para su trasplante. Así pues, el estudio de nuevas fuentes de células productoras de insulina así como de nuevos tratamientos que permitan preservar o incluso aumentar la masa de células beta en los pacientes con diabetes mellitus representa un objetivo de primera necesidad en este campo. En este sentido, en la última década ha habido un avance significativo en el campo de la biología de las células madre. Sin embargo, la identificación de células apropiadas para la generación de nuevas células beta, además del desarrollo de técnicas para la caracterización de estas células, así como de ensayos y modelos animales apropiados para probar su capacidad de diferenciación tanto in vitro como in vivo son de vital importancia para la puesta en marcha de nuevas estrategias terapéuticas basadas en la aplicación de las células madre para el tratamiento de la diabetes mellitus tipo 1

    OpenAIRE is the European infrastructure enabling researchers to comply with the European Union requirements for Open Access to research results. OpenAIRE collects metadata from data sources across Europe and beyond and defines interoperability guidelines to assist providers in exposing their information in a way that is compatible with OpenAIRE. This contribution focuses on a specific type of data source, CRIS systems, and the respective OpenAIRE guidelines, based on CERIF XML. A range of issues, spanning different aspects of information representation and exchange, needed to be addressed by the guidelines in order to define a complete solution for interoperability

    Genetic polymorphisms in key hypoxia-regulated downstream molecules and phenotypic correlation in prostate cancer

    Background In this study we sought if, in their quest to handle hypoxia, prostate tumors express target hypoxia-associated molecules and their correlation with putative functional genetic polymorphisms. Methods Representative areas of prostate carcinoma (n = 51) and of nodular prostate hyperplasia (n = 20) were analysed for hypoxia-inducible factor 1 alpha (HIF-1α), carbonic anhydrase IX (CAIX), lysyl oxidase (LOX) and vascular endothelial growth factor (VEGFR2) immunohistochemistry expression using a tissue microarray. DNA was isolated from peripheral blood and used to genotype functional polymorphisms at the corresponding genes (HIF1A +1772 C > T, rs11549465; CA9 + 201 A > G; rs2071676; LOX +473 G > A, rs1800449; KDR – 604 T > C, rs2071559). Results Immunohistochemistry analyses disclosed predominance of positive CAIX and VEGFR2 expression in epithelial cells of prostate carcinomas compared to nodular prostate hyperplasia (P = 0.043 and P = 0.035, respectively). In addition, the VEGFR2 expression score in prostate epithelial cells was higher in organ-confined and extra prostatic carcinoma compared to nodular prostate hyperplasia (P = 0.031 and P = 0.004, respectively). Notably, for LOX protein the immunoreactivity score was significantly higher in organ-confined carcinomas compared to nodular prostate hyperplasia (P = 0.015). The genotype-phenotype analyses showed higher LOX staining intensity for carriers of the homozygous LOX +473 G-allele (P = 0.011). Still, carriers of the KDR−604 T-allele were more prone to have higher VEGFR2 expression in prostate epithelial cells (P < 0.006). Conclusions Protein expression of hypoxia markers (VEGFR2, CAIX and LOX) on prostate epithelial cells was different between malignant and benign prostate disease. Two genetic polymorphisms (LOX +473 G > A and KDR−604 T > C) were correlated with protein level, accounting for a potential gene-environment effect in the activation of hypoxia-driven pathways in prostate carcinoma.     The HIF1A Functional Genetic Polymorphism at Locus +1772 Associates with Progression to Metastatic Prostate Cancer and Refractoriness to Hormonal Castration

    The hypoxia inducible factor 1 alpha (HIF1a) is a key regulator of tumour cell response to hypoxia, orchestrating mechanisms known to be involved in cancer aggressiveness and metastatic behaviour. In this study we sought to evaluate the association of a functional genetic polymorphism in HIF1A with overall and metastatic prostate cancer (PCa) risk and with response to androgen deprivation therapy (ADT). The HIF1A +1772 C>T (rs11549465) polymorphism was genotyped, using DNA isolated from peripheral blood, in 1490 male subjects (754 with prostate cancer and 736 controls cancer-free) through Real-Time PCR. A nested group of cancer patients who were eligible for androgen deprivation therapy was followed up. Univariate and multivariate models were used to analyse the response to hormonal treatment and the risk for developing distant metastasis. Age-adjusted odds ratios were calculated to evaluate prostate cancer risk. Our results showed that patients under ADT carrying the HIF1A +1772 T-allele have increased risk for developing distant metastasis (OR, 2.0; 95%CI, 1.1-3.9) and an independent 6-fold increased risk for resistance to ADT after multivariate analysis (OR, 6.0; 95%CI, 2.2-16.8). This polymorphism was not associated with increased risk for being diagnosed with prostate cancer (OR, 0.9; 95%CI, 0.7-1.2). The HIF1A +1772 genetic polymorphism predicts a more aggressive prostate cancer behaviour, supporting the involvement of HIF1a in prostate cancer biological progression and ADT resistance. Molecular profiles using hypoxia markers may help predict clinically relevant prostate cancer and response to ADT

    "Objectives: We aimed to describe the sociodemographic, clinical, and therapeutic characteristics of Peruvian military personnel diagnosed with HIV. Furthermore, we determined the frequency of highly active antiretroviral therapy (HAART) adherence, the complications and mortality. Methods: We retrospectively reviewed the medical records of patients diagnosed with HIV, confirmed by Western blot at a single institution from 1989-2020. Descriptive analyses were performed for all the variables using mean and standard deviation (SD) in the case of quantitative variables, and frequency and percentage for qualitative variables. Results:Ofthe 161 patients included, 95.7% were males and the mean age was 39.59 years (SD= 16.45 years). Most had college or higher education (67.7%) and were on active duty at diagnosis (77%). 35.4% had AIDS at diagnosis; 28.6% opportunistic infections; and 8.1% tuberculosis. The median follow-up was five years. 94.4% of the patients received HAART as the principal treatment; of these, 88.8% was adherent to treatment, and death was reported in 6.8%. There were no factors associated with adherence. Conclusions: Although the sociodemographic characteristics of Peruvian military personnel are similar to those of other countries, our findings suggest that Peruvian military personnel present higher adherence to HAART compared to previous studies in other military populations from South American countries. Further studies are recommended to assess specific factors attributed to these successful outcomes in the military personnelthat can be applied in other hospitals.

    Does the exotic equal pollution? Landscape methods for solving the dilemma of using native versus non‐native plant species in drylands

    There is a need to resolve methods to determine the merits of native versus nonnative plant use in drylands and indeed in more temperate areas around the world. This is because whilst plant introductions may have positive objectives, they can have significant negative landscape and environmental impacts. A key discussion on this issue focuses on whether the use of non-native plant species can be considered to be pollution and pollutive based on the concept that pollution can be regarded as ‘matter out of place’. The consequences of putting the wrong plant species in the wrong place can be extremely detrimental to the landscape character, quality and value of the land, let alone the effects on ecosystem structure and functioning as well as on biodiversity. These effects can also affect human communities who may rely on the landscape, for example, for tourism. It is thus necessary that the discussion on how decisions are made in determining plant choice evolves so that the right decisions are made when planting is necessary, for the land, for nature and for the people. This discussion has been initiated through COST Action ES1104, which focused on the restoration of degraded dry and arid lands.     Henri Poincaré: The Status of Mechanical Explanations and the Foundations of Statistical Mechanics

    The first goal of this paper is to show the evolution of Poincaré’s opinion on the mechanistic reduction of the principles of thermodynamics, placing it in the context of the science of his time. The second is to present some of his work in 1890 on the foundations of statistical mechanics. He became interested first in thermodynamics and its relation with mechanics, drawing on the work of Helm-holtz on monocyclic systems. After a period of skepticism concerning the kinetic theory, he read some of Maxwell’s memories and contributed to the foundations of statistical mechanics. I also show that Poincaré's contributions to the founda-tions of statistical mechanics are closely linked to his work in celestial mechanics and its interest in probability theory and its role in physics

    Search for single production of a vectorlike T quark decaying into a Higgs boson and top quark with fully hadronic final states using the ATLAS detector

The Higgs-boson and top-quark candidates are identified in the all-hadronic decay mode, where H→bb¯ and t→bW→bqq¯′ are reconstructed as large-radius jets. The candidate Higgs boson, top quark, and associated B hadrons are identified using tagging algorithms. No significant excess is observed above the background, so limits are set on the production cross section of a singlet T quark at 95% confidence level, depending on the mass mT and coupling κT of the vectorlike T quark to Standard Model particles. In the considered mass range between 1.0 and 2.3 TeV, the upper limit on the allowed coupling values increases with mT from a minimum value of 0.35 for 1.07 < mT < 1.4 TeV to 1.6 for mT = 2.3 Te

    Prevalencia de ansiedad y depresión utilizando la escala de ansiedad y depresión (HADS) en pacientes con Síndrome de Intestino Irritable

    Introducción: en ausencia de una anormalidad estructural. Es una de las enfermedades que se ha asociado frecuentemente con enfermedades psiquiátricas como ansiedad y depresión. El objetivo del estudio fue determinar la prevalencia de depresión y ansiedad en pacientes con SII empleando la encuesta Hospital Anxiety and Depression Scale (HADS) en pacientes que acuden a consultorio de gastroenterología en la Clínica Internacional Sede Lima (CILM). Materiales y Métodos: Estudio descriptivo no comparativo en el cual se utilizaron encuestas autoaplicadas en consultorio externo de la CILM. Se utilizó Microsoft Excel para tabular y STATA versión 17 para calcular las frecuencias absolutas y relativas. Resultados: Se incluyó a 67 pacientes: sexo: femenino (77.61%), rango de edad: 36-45 años (56.72%) y variedad de sintomas en SII: estreñimiento (55.22%). La prevalencia de ansiedad fue de 71.64% mientras que la de depresión fue de 26.87%. Conclusiones: Los p cientes con SII tuvieron mayor prevalencia de síntomas compatibles con ansiedad en comparación con depresión.Introduction: Irritable bowel syndrome (IBS) is characterized by the presence of intestinal disturbances in the absence of structural abnormalities. IBS is one of the diseases that has been frequently associated with psychiatric diseases such as anxiety and depression. The aim of this study was to determine the prevalence of depression and anxiety in patients with IBS using the Hospital Anxiety and Depression Scale (HADS) survey in patients attending the gastroenterology clinic of the International Clinic of Lima (CILM). Materials and Methods: Descriptive non- comparative study where self-administered surveys were used in the outpatient clinic of the CILM. Microsoft Excel was used for tabulation and STATA version 17 for calculation of absolute and relative frequencies. Results: Sixty-seven patients were included. Sex: female (77.61%), age range 36-45 (56.72%) and variety of IBS symptoms: constipation (55.22%). The prevalence of anxiety was 71.64% while that of depression was 26.87%. Conclusions: Patients with IBS presented a higher prevalence of symptoms compatible with anxiety compared to depression
