X-ray analog pixel array detector for single synchrotron bunch time-resolved imaging
Dynamic x-ray studies may reach temporal resolutions limited by only the
x-ray pulse duration if the detector is fast enough to segregate synchrotron
pulses. An analog integrating pixel array detector with in-pixel storage and
temporal resolution of around 150 ns, sufficient to isolate pulses, is
presented. Analog integration minimizes count-rate limitations and in-pixel
storage captures successive pulses. Fundamental tests of noise and linearity as
well as high-speed laser measurements are shown. The detector resolved
individual bunch trains at the Cornell High Energy Synchrotron Source (CHESS)
at levels of up to 3.7x10^3 x-rays/pixel/train. When applied to turn-by-turn
x-ray beam characterization single-shot intensity measurements were made with a
repeatability of 0.4% and horizontal oscillations of the positron cloud were
detected. This device is appropriate for time-resolved Bragg spot single
crystal experiments.Comment: 9 pages, 11 figure
Time-resolved investigation of nanometer scale deformations induced by a high flux x-ray beam
We present results of a time-resolved pump-probe experiment where a Si sample was exposed to an intense 15 keV beam and its surface monitored by measuring the wavefront deformation of a reflected optical laser probe beam. By reconstructing and back propagating the wavefront, the deformed surface can be retrieved for each time step. The dynamics of the heat bump, build-up and relaxation, is followed with a spatial resolution in the nanometer range. The results are interpreted taking into account results of finite element method simulations. Due to its robustness and simplicity this method should find further developments at new x-ray light sources (FEL) or be used to gain understanding on thermo-dynamical behavior of highly excited materials. A Functionally Different Immune Phenotype in Cattle Is Associated With Higher Mastitis Incidence
A novel vaccine against bovine viral diarrhea (BVD) induced pathogenic antibody production in 5–10% of BVD-vaccinated cows. Transfer of these antibodies via colostrum caused Bovine neonatal pancytopenia (BNP) in calves, with a lethality rate of 90%. The exact immunological mechanisms behind the onset of BNP are not fully understood to date. To gain further insight into these mechanisms, we analyzed the immune proteome from alloreactive antibody producers (BNP cows) and non-responders. After in vitro stimulation of peripheral blood derived lymphocytes (PBL), we detected distinctly deviant expression levels of several master regulators of immune responses in BNP cells, pointing to a changed immune phenotype with severe dysregulation of immune response in BNP cows. Interestingly, we also found this response pattern in 22% of non-BVD-vaccinated cows, indicating a genetic predisposition of this immune deviant (ID) phenotype in cattle. We additionally analyzed the functional correlation of the ID phenotype with 10 health parameters and 6 diseases in a retrospective study over 38 months. The significantly increased prevalence of mastitis among ID cows emphasizes the clinical relevance of this deviant immune response and its potential impact on the ability to fight infections
Oxalic acid, versatile peroxidase secretion and chelating ability of Bjerkandera fumosa in rich and limited culture conditions
Efficient ligninolytic systems of wood-degrading fungi include not only oxidizing enzymes, but also low-molecular-weight effectors. The ability of Bjerkandera fumosa to secrete oxalic acid and versatile peroxidase (VP) in nitrogen-rich and nitrogen-limited media was studied. Higher activity of VP was determined in the nitrogen-limited media but greater concentration of oxalic acid was observed in the cultures of B. fumosa without nitrogen limitation. Ferric ions chelating ability of Bjerkandera fumosa studied in ferric ions limited media was correlated with the increased level of oxalic acid. The presence of hydroxamate-type siderophores in B. fumosa media were also detected. Oxalate decarboxylase was found to be responsible for regulation of oxalic acid concentration in the tested B. fumosa cultures
Variational Approach to Molecular Kinetics
The eigenvalues and eigenvectors of the molecular dynamics propagator (or transfer operator) contain the essential information about the molecular thermodynamics and kinetics. This includes the stationary distribution, the metastable states, and state-to-state transition rates. Here, we present a variational approach for computing these dominant eigenvalues and eigenvectors. This approach is analogous the variational approach used for computing stationary states in quantum mechanics. A corresponding method of linear variation is formulated. It is shown that the matrices needed for the linear variation method are correlation matrices that can be estimated from simple MD simulations for a given basis set. The method proposed here is thus to first define a basis set able to capture the relevant conformational transitions, then compute the respective correlation matrices, and then to compute their dominant eigenvalues and eigenvectors, thus obtaining the key ingredients of the slow kinetics

Jasmonic acid and coronatin induce odor production in plants
Negative effects like defoliation and senescence (aging) are not the only phenomena triggered in plants by the phytohormone jasmonic acid (1); it also stimulates tuber growth, tendril coiling, and the release of odorous substances that can function as stress signals in plant defense. In the latter case only minute concentrations of jasmonic acid are required, for example, 100 nmol mL−1 for the tobacco plant. Even lower levels of the structurally related phytotoxin coronatin (ca. 1 nmol mL−1) induce the production and release of volatiles
