    Free vibration of a three-layered sandwich beam using the dynamic stiffness method and experiment

    In this paper, an accurate dynamic stiffness model for a three-layered sandwich beam of unequal thicknesses is developed and subsequently used to investigate its free vibration characteristics. Each layer of the beam is idealised by the Timoshenko beam theory and the combined system is reduced to a tenth-order system using symbolic computation. An exact dynamic stiffness matrix is then developed by relating amplitudes of harmonically varying loads to those of the responses. The resulting dynamic stiffness matrix is used with particular reference to the Wittrick-Williams algorithm to carry out the free vibration analysis of a few illustrative examples. The accuracy of the theory is confirmed both by published literature and by experiment. The paper closes with some concluding remarks. (c) 2007 Elsevier Ltd. All rights reserved

    Stochastic orbital migration of small bodies in Saturn's rings

    Many small moonlets, creating propeller structures, have been found in Saturn's rings by the Cassini spacecraft. We study the dynamical evolution of such 20-50m sized bodies which are embedded in Saturn's rings. We estimate the importance of various interaction processes with the ring particles on the moonlet's eccentricity and semi-major axis analytically. For low ring surface densities, the main effects on the evolution of the eccentricity and the semi-major axis are found to be due to collisions and the gravitational interaction with particles in the vicinity of the moonlet. For large surface densities, the gravitational interaction with self-gravitating wakes becomes important. We also perform realistic three dimensional, collisional N-body simulations with up to a quarter of a million particles. A new set of pseudo shear periodic boundary conditions is used which reduces the computational costs by an order of magnitude compared to previous studies. Our analytic estimates are confirmed to within a factor of two. On short timescales the evolution is always dominated by stochastic effects caused by collisions and gravitational interaction with self-gravitating ring particles. These result in a random walk of the moonlet's semi-major axis. The eccentricity of the moonlet quickly reaches an equilibrium value due to collisional damping. The average change in semi-major axis of the moonlet after 100 orbital periods is 10-100m. This translates to an offset in the azimuthal direction of several hundred kilometres.     Aggregation number distributions and mesoglobules in dilute solutions of diblock and triblock copolymers

    We investigate the aggregation number and size distributions for inter-molecular clusters of amphiphilic diblock and triblock copolymers in poor solvent at very low concentrations. Diblocks and triblocks with hydrophilic ends are shown to possess narrow distributions corresponding to formation of monodispersed mesoglobules. Diblocks with hydrophobic ends are found to produce inter-cluster multimers due to bridging by the hydrophilic middle blocks, resulting in polydisperse distributions. Implications of these observations for preparation of monodispersed nanoparticles and, potentially, understanding of the quaternary structure of proteins are discussed.Comment: 4 pages, 4 PS figures.     Analysis of composite wing structures with a morphing leading edge

    One of the main challenges for the civil aviation industry is the reduction of its environmental impact. Over the past years, improvements in performance efficiency have been achieved by simplifying the design of the structural components and using composite materials to reduce the overall weight. These approaches however, are not sufficient to meet the current demanding requirements set for a „greener‟ aircraft. Significant changes in drag reduction and fuel consumption can be obtained by using new technologies, such as smart morphing structures. These concepts will in fact help flow laminarisation, which will increase the lift to drag ratio. Furthermore, the capability to adapt the wing shape will enable to optimise the aerodynamic performance not only for a single flight condition but during the entire mission. This will significantly improve the aircraft efficiency. The current research work has been carried out as part of the European Commission founded Seventh Framework Program called „Smart High Lift Device for the Next Generation Wing‟ (SADE), which main aim is to develop and study morphing high lift devices. The author‟s investigation focused on developing a design concept for the actuation mechanism of a morphing leading edge device. A detailed structural analysis has been carried out in order to demonstrate its feasibility.In the first phase of the research the attention was directed on the preliminary design and analysis of the composite wing box. The parameters of the key structural components, such as skin, spars, ribs and stringers were set to satisfy the static stress and buckling requirements. Moreover, numerical and experimental studies were conducted to analyse the static failure and buckling behaviour of two typical composite wing structural components: a spar section and a web and base joint assembly. In the second stage of the research, a design for the morphing leading edge actuation mechanism was developed. The actuation system was designed in such a way that the target shape was reached with minimum actuation force demand. A geometrical nonlinear FE analysis was conducted to simulate the leading edge morphing deflection and ensure that structural strength requirements were satisfied. Furthermore, the behaviour of the skin integrated with the internal actuation mechanism was modelled under the aerodynamic pressure, at different flight conditions and gust loads, in order to prove that the proposed actuation system can compete with the conventional rigid rib. This study demonstrated that a feasible morphing leading edge design for a next generation large aircraft wing can be achieved.     A local prescription for the softening length in self-gravitating gaseous discs

    In 2D-simulations of self-gravitating gaseous discs, the potential is often computed in the framework of "softened gravity" initially designed for N-body codes. In this special context, the role of the softening length LAMBDA is twofold: i) to avoid numerical singularities in the integral representation of the potential (i.e., arising when the relative separation vanishes), and ii) to acount for stratification of matter in the direction perpendicular to the disc mid-plane. So far, most studies have considered LAMBDA as a free parameter and various values or formulae have been proposed without much mathematical justification. In this paper, we demonstrate by means of a rigorous calculus that it is possible to define LAMBDA such that the gravitational potential of a flat disc coincides at order zero with that of a geometically thin disc of the same surface density. Our prescription for LAMBDA, valid in the local, axisymmetric limit, has the required properties i) and ii). It is mainly an analytical function of the radius and disc thickness, and is sensitive to the vertical stratification. For mass density profiles considered (namely, profiles expandable over even powers of the altitude), we find that LAMBDA : i) is independant of the numerical mesh, ii) is always a fraction of the local thickness H, iii) goes through a minimum at the singularity (i.e., at null separation), and iv) is such that 0.13 < LAMBDA/H < 0.29 typically (depending on the separation and on density profile).     Pifithrin-alpha inhibits p53 signaling after interaction of the tumor suppressor protein with hsp90 and its nuclear translocation

    Pifithrin-alpha (PFTalpha) was originally thought to be a specific inhibitor of signaling by the tumor suppressor protein p53. However, the laboratory that discovered pifithrin recently reported that the compound also inhibits heat shock and glucocorticoid receptor (GR) signaling, and they suggested that PFTalpha targets a factor common to all three signal transduction pathways, such as the hsp90/hsp70-based chaperone machinery (Komarova, E. A., Neznanov, N., Komarov, P. G., Chernov, M. V., Wang, K., and Gudkov, A. V. (2003) J. Biol. Chem. 278, 15465-15468). Because it is important for the mechanistic study of this machinery to identify unique inhibitors of chaperone action, we have examined the effect of PFTalpha on transcriptional activation, the hsp90 heterocomplex assembly, and hsp90-dependent nuclear translocation for both p53 and the GR. At concentrations where PFTalpha blocks p53-mediated induction of p21/Waf-1 in human embryonic kidney cells, we observed no inhibition of GR-mediated induction of a chloramphenicol acetyl transferase reporter in LMCAT cells. PFTalpha did, however, cause a left shift in the dexamethasone dose response curve by increasing intracellular dexamethasone concentration, apparently by competing for dexamethasone efflux from the cell. The assembly of p53 or GR heterocomplexes with hsp90 and immunophilins was not affected by PFTalpha either in vivo or in vitro and did not affect the nuclear translocation of either transcription factor. Thus, we conclude that PFTalpha does not inhibit GR-mediated induction or the function of the chaperone machinery, and, as originally thought, it may specifically inhibit p53 signaling by acting at a stage after p53 translocation to the nucleus.Fil: Murphy, Patrick J..     During embryonic life, the initially paired pharyngeal arch arteries (PAAs) follow a precisely orchestrated program of persistence and regression that leads to the formation of the mature aortic arch and great vessels. When this program fails, specific cardiovascular defects arise that may be life threatening or mild, according to the identity of the affected artery. Fourth PAA-derived cardiovascular defects occur commonly in DiGeorge syndrome and velocardiofacial syndrome (22q11DS), and in Tbx1(+/-) mice that model the 22q11DS cardiovascular phenotype. Tbx1 is expressed in pharyngeal mesoderm, endoderm and ectoderm, and, in addition, we show that it is expressed in precursors of the endothelial cells that line the PAAs, thus expanding the number of tissues in which Tbx1 is potentially required for fourth PAA development. In this study, we have used cell fate mapping and tissue-specific gene deletion, driven by six different Cre lines, to explore Tbx1 gene-dosage requirements in the embryonic pharynx for fourth PAA development. Through this approach, we have resolved the spatial requirements for Tbx1 in this process, and we show pharyngeal epithelia to be a critical tissue. We also thereby demonstrate conclusively that the role of Tbx1 in fourth PAA development is cell non-autonomous

    A Genetic Variant in the IL-17 Promoter Is Functionally Associated with Acute Graft-Versus-Host Disease after Unrelated Bone Marrow Transplantation

    Interleukin IL-17 is a proinflammatory cytokine that has been implicated in the pathogenesis of various autoimmune diseases. The single nucleotide polymorphism (SNP), rs2275913, in the promoter region of the IL-17 gene is associated with susceptibility to ulcerative colitis. When we examined the impact of rs2275913 in a cohort consisting of 438 pairs of patients and their unrelated donors transplanted through the Japan Marrow Donor Program, the donor IL-17 197A allele was found to be associated with a higher risk of acute graft-versus-host disease (GVHD; hazard ratio [HR], 1.46; 95% confidence interval [CI], 1.00 to 2.13; P = 0.05). Next, we investigated the functional relevance of the rs2275913 SNP. In vitro stimulated T cells from healthy individuals possessing the 197A allele produced significantly more IL-17 than those without the 197A allele. In a gene reporter assay, the 197A allele construct induced higher luciferase activity than the 197G allele, and the difference was higher in the presence of T cell receptor activation and was abrogated by cyclosporine treatment. Moreover, the 197A allele displayed a higher affinity for the nuclear factor activated T cells (NFAT), a critical transcription factor involved in IL-17 regulation. These findings substantiate the functional relevance of the rs2275913 polymorphism and indicate that the higher IL-17 secretion by individuals with the 197A allele likely accounts for their increased risk for acute GVHD and certain autoimmune diseases
