    A falta de padronização e de qualidade das carcaças ofertadas faz com que a carne bovina brasileira receba baixas remunerações, além da limitada expansão para novos mercados. Dessa forma, objetivou-se avaliar as características de peso ao abate, peso da carcaça e seus rendimentos de 68 bovinos machos da raça Nelore Mocho, inteiros, oriundos de uma população segregante para maciez da carne. Os animais foram abatidos ao apresentarem 5 mm de espessura de gordura e/ou 500 kg de peso vivo. Antes do embarque para o frigorifico, os animais foram pesados para obtenção do peso ao abate. Ao final da linha de abate, as meias carcaças foram pesagens individuais para obtenção do peso de carcaça quente e carcaça fria após resfriamento por 24 horas em câmara. O rendimento de carcaça foi obtido em porcentagem do peso da carcaça quente, em relação ao peso vivo ao abate. Cálculo similar foi realizado para rendimento de carcaça fria. Amostras desse músculo foram maturadas por sete dias para mensuração da força de cisalhamento (FC) por meio do aparelho de Warner-Bratzler shear force (WBSF). Foram, então, definidos dois grupos: baixo WBSF (FC≤3,5 kgf) e alto WBSF (FC>3,5 kgf). As médias foram comparadas por meio do teste Duncan a 5% de significância. Não foram observadas diferenças estatisticamente entre os grupos de baixo e alto WBSF, para peso ao abate, peso da carcaça e seus rendimentos. A seleção para a maciez da carne em bovinos Nelore Mocho não afetou as demais características de carcaça. Abstract: Although Brazil exports only 18% of its beef production, the lack of standardization and carcass quality has limited expansion into new markets. Therefore, we aimed to evaluate the characteristics of slaughter weight, carcass weight and dressing percentage of 68 intact male Polled Nelore (OB brand) cattle derived from a population segregating for meat tenderness. The animals were slaughtered with 5 mm of fat thickness and/or 500 kg live weight (LW). After slaughter, half carcasses were weighed and chilled at 0 to 2 °C for 24 hours, then re-weighed and measurements taken of pH, rib eye area (REA) and back fat (BF) at the 12th rib. Samples of the L. dorsi muscle were removed and aged for seven days for measurement of Warner - Bratzler Shear Force (WBSF). Two groups were then defined based on WBSF: low WBSF (≤ 3.5 kg) and high WBSF (> 3.5 kg). Hot (HDP) and cold (CDP) carcass dressing % were obtained as the percentages of hot (HCW, immediately following slaughter) and cold (CCW, after 24 hours at 0 to 2 °C) carcass weights in relation to live weight (LW) at slaughter. No differences were observed between Low and High WBSF groups with regard to LW, HCW, CCW HDP or CDP. Selection for meat tenderness in Polled Nellore should not affect carcass characteristics

    Seascape genomics and phylogeography of the sailfish (Istiophorus platypterus)

    Permeable phylogeographic barriers characterize the vast open ocean, boosting gene flow and counteracting population differentiation and speciation of widely distributed and migratory species. However, many widely distributed species consists of distinct populations throughout their distribution, evidencing that our understanding of how the marine environment triggers population and species divergence are insufficient. The sailfish is a circumtropical and highly migratory billfish that inhabits warm and productive areas. Despite its ecological and socioeconomic importance as a predator and fishery resource, the species is threatened by overfishing, requiring innovative approaches to improve their management and conservation status. Thus, we presented a novel high-quality reference genome for the species and applied a seascape genomics approach to understand how marine environmental features may promote local adaptation and how it affects gene flow between populations. We delimit two populations between the Atlantic and Indo-Western Pacific oceans and detect outlier loci correlated with sea surface temperature, salinity, oxygen, and chlorophyll concentrations. However, the most significant explanatory factor that explains the differences between populations was isolation by distance. Despite recent population drops, the sailfish populations are not inbred. For billfishes in general, genome-wide heterozygosity was found to be relatively low compared to other marine fishes, evidencing the need to counteract overfishing effects. In addition, in a climate change scenario, management agencies must implement state-of-the-art sequencing methods, consider our findings in their management plans, and monitor genome-wide heterozygosity over time to improve sustainable fisheries and the long-term viability of its populations.LA/P/0101/2020info:eu-repo/semantics/publishedVersio

    Volumoso quisto acromioclavicular como apresentação invulgar de rotura massiva da coifa dos rotadores

    Objetivo: Os quistos sinoviais da articulação acromioclavicular (AC), podem ocorrer associadas à artrose AC ou como consequência da rotura massiva da coifa dos rotadores. Com o presente caso, os autores pretendem apresentar e discutir uma situação pouco frequente. Relato de caso: Homem de 81 anos, referenciado à consulta de Ortopedia por tumefação da região superior do ombro direito, indolor e de aumento progressivo nos últimos 2 anos. Avaliação clínica e radiológica compatível com volumoso quisto sinovial da acromioclavicular, associado a rotura massiva da coifa dos rotadores; submetido a tratamento cirúrgico com excisão marginal da lesão; a análise anatomopatológica confirmou o diagnóstico de quisto sinovial. Comentários: Os quistos sinoviais da articulação AC são entidades raras e pouco abordadas na literatura. A sua etiologia permanece por esclarecer ainda que um dos tipos descritos pareça estar associada à omatrose com rotura massiva da coifa dos rotadores (artropatia da coifa); nestes, a rotura da coifa permite a passagem de grandes quantidades de líquido sinovial para o espaço subacromial com formação de um volumoso quisto. Atendendo à sua raridade, o tratamento permanece controverso. O caso vem alertar para a ocorrência dos quistos da articulação AC, como apresentação invulgar de artropatia das coifa dos rotadores, discutindo a sua etiologia e abordagem terapêutica adequada.info:eu-repo/semantics/publishedVersio

Publisher Copyright: © 2022 by the authors.Myotonic dystrophy type 1 (DM1) is an autosomal dominant disease caused by a CTG repeat expansion in the 3′ untranslated region of the dystrophia myotonica protein kinase gene. AKT dephosphorylation and autophagy are associated with DM1. Autophagy has been widely studied in DM1, although the endocytic pathway has not. AKT has a critical role in endocytosis, and its phosphorylation is mediated by the activation of tyrosine kinase receptors, such as epidermal growth factor receptor (EGFR). EGF-activated EGFR triggers the internalization and degradation of ligand–receptor complexes that serve as a PI3K/AKT signaling platform. Here, we used primary fibroblasts from healthy subjects and DM1 patients. DM1-derived fibroblasts showed increased autophagy flux, with enlarged endosomes and lysosomes. Thereafter, cells were stimulated with a high concentration of EGF to promote EGFR internalization and degradation. Interestingly, EGF binding to EGFR was reduced in DM1 cells and EGFR internalization was also slowed during the early steps of endocytosis. However, EGF-activated EGFR enhanced AKT and ERK1/2 phosphorylation levels in the DM1-derived fibroblasts. Therefore, there was a delay in EGF-stimulated EGFR endocytosis in DM1 cells; this alteration might be due to the decrease in the binding of EGF to EGFR, and not to a decrease in AKT phosphorylation.publishersversionpublishe
