    Modificaciones funcionales, personales y sociales de las fracturas de la extremidad proximal del fémur en pacientes mayores

    Se ha realizado un estudio retrospectivo revisando las historias clínicas de 550 pacientes mayores de 69 años que sufrieron una fractura de cadera durante 1995 y 1996 en Salamanca. Cerca de la mitad de los pacientes estudiados tenían independencia casi total y el 40.22% cambió su tipo de residencia. La mayoría de los pacientes perdieron actividad y se encontraron diferencias estadísticamente significativas en la existencia o no de complicaciones. Casi todas las fracturas ocurrieron tras una caída casual por tropiezos o resbalones. El 57.67% de los pacientes tenían una intensa osteopenia. Alrededor del 25% de los pacientes realizaron rehabilitación, sobre todo los tratados con enclavados endomedulares. La mortalidad fue de 31.37% y se encontraron diferencias estadísticamente significativas al relacionarla con la actividad física previa y postratamiento y el grado de dependencia previa a la fractura. Estos cambios en la función y dependencia, tienen poca influencia en la rehabilitación y mortalidad de los pacientes.Case-history reviews and at-home interviews we used to study retrospectively the 550 patients older than 69 years who suffered a hip fracture between 1995 and 1996 in Salamanca (Spain). The average hospitalization time was 15 days. Almost half of patients were not dependent at all and 40.22% of them moved to another residence. Most patients lost activity, and it was statistically correlated to the adverse events reported. Almost every fracture occurred after a casual fall due to stumbling or sliding. 57.67 % of patients had a severe osteopenia. The most frequent fracture reported was pertrochanteric (44.67%) followed by subcapital, especially those treated with intramedullary nailing. Mortality was 31.37% and statistical differences were found between degree of activity before and after the fracture. These changes in function and type of residence have a scarce influence on rehabilitation and mortality of patients

    Conscious mobility for urban spaces: case studies review and indicator framework design

    A lack of data collection on conscious mobility behaviors has been identified in current sustainable and smart mobility planning, development and implementation strategies. This leads to technocentric solutions that do not place people and their behavior at the center of new mobility solutions in urban centers around the globe. This paper introduces the concept of conscious mobility to link techno-economic analyses with user awareness on the impact of their travel decisions on other people, local urban infrastructure and the environment through systematic big data collection. A preliminary conscious mobility indicator framework is presented to leverage behavioral considerations to enhance urban-community mobility systems. Key factors for conscious mobility analysis have been derived from five case studies. The sample offers regional diversity (i.e., local, regional and the global urban contexts), as well as different goals in the transformation of conventional urban transport systems, from improving public transport efficiency and equipment electrification to mitigate pollution and climate risks, to focusing on equity, access and people safety. The case studies selected provide useful metrics on the adoption of cleaner, smarter, safer and more autonomous mobility technologies, along with novel people-centric program designs to build an initial set of conscious mobility indicators frameworks. The parameters were applied to the city of Monterrey, Nuevo Leon in Mexico focusing on the needs of the communities that work, study and live around the local urban campus of the Tecnologico de Monterrey’s Distrito Tec. This case study, served as an example of how conscious mobility indicators could be applied and customized to a community and region of interest. This paper introduces the first application of the conscious mobility framework for urban communities’ mobility system analysis. This more holistic assessment approach includes dimensions such as society and culture, infrastructure and urban spaces, technology, government, normativity, economy and politics, and the environment. The expectation is that the conscious mobility framework of analysis will become a useful tool for smarter and sustainable urban and mobility problem solving and decision making to enhance the quality of life all living in urban communities

    Alkynyl N-BODIPYs as Reactive Intermediates for the Development of Dyes for Biophotonics

    A new approach for the rapid multi-functionalization of BODIPY dyes towards biophotonics is reported. It is based on novel N-BODIPYs, through reactive intermediates with alkynyl groups to be further derivatized by click chemistry. This approach has been exemplified by the development of new dyes for cell bio-imaging, which have proven to successfully internalize into pancreatic cancer cells and accumulate in the mitochondria. The in vitro suitability for photodynamic therapy (PDT) was also analyzed and confirmed our compounds to be promising PDT candidates for the treatment of pancreatic cancer

    Topo-Iberia Project: CGPS crustal velocity field in the Iberian Peninsula and Morocco

    A new continuous GPS network was installed under the umbrella of a research project called 'Geociencias en Iberia: Estudios integrados de topografı´a y evolución 4D (Topo-Iberia)', to improve understanding of kinematic behavior of the Iberian Peninsula region. Here we present a velocity field based on the analysis of the 4 years of data from 25 stations constituting the network, which were analyzed by three different analysis groups contributing to the project. Different geodetic software packages (GIPSY-OASIS, Bernese and GAMIT) as well as different approaches were used to estimate rates of present day crustal deformation in the Iberian Peninsula and Morocco. In order to ensure the consistency of the velocity fields determined by the three groups, the velocities obtained by each analysis center were transformed into a common Eurasia Reference Frame. After that, the strain rate field was calculated. The results put in evidence more prominent residual motions in Morocco and southernmost part of the Iberian Peninsula. In particular, the dilatation and shear strain rates reach their maximum values in the Central Betics and northern Alboran Sea. A small region of high shear strain rate is observed in the east-central part of the peninsula and another deformation focus is located around the Strait of Gibraltar and the Gulf of Cadiz

    Cranberry-derived phenolic metabolites and urinary tract infections

    Resumen del póster presentado a la 7th International Conference on Polyphenols and Health, celebrada en Tours (Francia) del 27 al 30 de octubre de 2015.The beneficial effects of cranbeny products against urinary tract infections (UTIs) have been attributed, at least partly, to their A-type proanthocyanidin (PAC) content. A-type PACs have shown uropathogenic Escherichia coli (UPEC)-anti-adhesive activity, although they are unlikely to appear in urine at relevant concentrations as they are poorly absorbed. One leading hypothesis is that PAC-derived metabolites present in urine would operate in the phase of UPEC adyherence to uroepithelial cells, preventing bacterial colonization. In addition to this, and as it is becoming evident that the intestine is a reservoir for uropathogenic bacteria, other hypothesis is that. A-type proanthocyanidins specifically decrease the transient intestinal colonisation b UPEC, consequently reducing the risk of UTI incidence. In any case, gut microbiota (and its inter-individual variability) seems to be an important factor to be considered. In this communication, we summarize our results from different approaches aimed to look into the mechanisms that are behind the protedive action of cranberry polyphenols against ITUs: 1)in vitro fermentations of cranbeny polyphenols with colonic microbiota, that were performed to access the microbial-derived metabolic profile of cranbeny polyphenols as well as their effect on gut microbiota survival, 2) an in vivo trial with model mouse intraurethral-inoculated wilh UPEC, that evaluated the effectiveness of cranbeny supplementation in bacterial infection as well as its impact on faecal phenolic metabolism and faecal microbiota, 3) testing the UPEC-antiadhesive capacity of cranbeny phenolic compounds and their metabolites in bladder epithelial cell culíures, and 4) ex vivo studies of UPEC-antiadhesive capacity of mice mines collected after cranbeny supplementation.Ministry of Economy and Competitiveness (MiNECO) (Projects AGL-2010-17499 and AGL2012-40172-C02-01) and the Comunidad Autónoma de Madrid (Project ALIBIRD S2013/ABI-2728), Spain.Peer Reviewe

    Analysis of gene network bifurcation during optic cup morphogenesis in zebrafish

    Sight depends on the tight cooperation between photoreceptors and pigmented cells, which derive from common progenitors through the bifurcation of a single gene regulatory network into the neural retina (NR) and retinal-pigmented epithelium (RPE) programs. Although genetic studies have identified upstream nodes controlling these networks, their regulatory logic remains poorly investigated. Here, we characterize transcriptome dynamics and chromatin accessibility in segregating NR/RPE populations in zebrafish. We analyze cis-regulatory modules and enriched transcription factor motives to show extensive network redundancy and context-dependent activity. We identify downstream targets, highlighting an early recruitment of desmosomal genes in the flattening RPE and revealing Tead factors as upstream regulators. We investigate the RPE specification network dynamics to uncover an unexpected sequence of transcription factors recruitment, which is conserved in humans.     Generating new fanca-deficient hnscc cell lines by genomic editing recapitulates the cellular phenotypes of fanconi anemia

    Fanconi anemia (FA) patients have an exacerbated risk of head and neck squamous cell carcinoma (HNSCC). Treatment is challenging as FA patients display enhanced toxicity to standard treatments, including radio/chemotherapy. Therefore, better therapies as well as new disease models are urgently needed. We have used CRISPR/Cas9 editing tools in order to interrupt the human FANCA gene by the generation of insertions/deletions (indels) in exon 4 in two cancer cell lines from sporadic HNSCC having no mutation in FA-genes: CAL27 and CAL33 cells. Our approach allowed efficient editing, subsequent purification of single-cell clones, and Sanger sequencing validation at the edited locus. Clones having frameshift indels in homozygosis did not express FANCA protein and were selected for further analysis. When compared with parental CAL27 and CAL33, FANCA-mutant cell clones displayed a FA-phenotype as they (i) are highly sensitive to DNA interstrand crosslink (ICL) agents such as mitomycin C (MMC) or cisplatin(ii) do not monoubiquitinate FANCD2 upon MMC treatment and therefore (iii) do not form FANCD2 nuclear foci, and (iv) they display increased chromosome fragility and G2 arrest after diepoxybutane (DEB) treatment. These FANCA-mutant clones display similar growth rates as their parental cells. Interestingly, mutant cells acquire phenotypes associated with more aggressive disease, such as increased migration in wound healing assays. Therefore, CAL27 and CAL33 cells with FANCA mutations are phenocopies of FA-HNSCC cells
