Investigation of the Electric Field and Charge Density Distribution of pristine and defective 2D WSe by Differential Phase Contrast Imaging
Most properties of solid materials are defined by their internal electric
field and charge density distributions which so far have been difficult to
measure with sufficient spatial resolution. For 2D materials, the electric
field at the atomic level in particular influences the optoelectronic
properties. In this study, the atomic-scale electric field and charge density
distribution of 2D WSe are revealed by using an emerging microscopy
technique, differential phase contrast (DPC) imaging in the scanning
transmission electron microscope (STEM). Combined with high-angle annular
dark-field imaging the charge density distribution of bi- and trilayers of
WSe is mapped. A measured higher positive charge density located at the
selenium atomic columns compared to the tungsten atomic columns is reported,
and possible reasons are discussed. Furthermore, the change in the electric
field distribution of a selenium point defect in a trilayer is investigated
exhibiting a characteristic electric field distribution in the vicinity of the
defect: there are characteristic regions with locally enhanced and with locally

Molecular monitoring of minimal residual disease in two patients with MLL-rearranged acute myeloid leukemia and haploidentical transplantation after relapse
This report describes the clinical courses of two acute myeloid leukemia patients. Both had MLL translocations, the first a t(10;11)(p11.2;q23) with MLL-AF10 and the second a t(11;19)(q23;p13.1) with MLL-ELL fusion. They achieved a clinical remission under conventional chemotherapy but relapsed shortly after end of therapy. Both had a history of invasive mycoses (one had possible pulmonary mycosis, one systemic candidiasis). Because no HLA-identical donor was available, a haploidentical transplantation was performed in both cases. Using a specially designed PCR method for the assessment of minimal residual disease (MRD), based on the quantitative detection of the individual chromosomal breakpoint in the MLL gene, all patients achieved complete and persistent molecular remission after transplantation. The immune reconstitution after transplantation is described in terms of total CD3+/CD4+, CD3+/CD8+, CD19+, and CD16+/CD56+ cell numbers over time. The KIR and HLA genotypes of donors and recipients are reported and the possibility of a KIR-mediated alloreactivity is discussed. This report illustrates that haploidentical transplantation may offer a chance of cure without chronic graft-versus-host disease in situations where no suitable HLA-identical donor is available even in a high-risk setting and shows the value of MRD monitoring in the pre- and posttransplant setting
Cryo-EM structures reveal intricate Fe-S cluster arrangement and charging in Rhodobacter capsulatus formate dehydrogenase
Metal-containing formate dehydrogenases (FDH) catalyse the reversible oxidation of formate to carbon dioxide at their molybdenum or tungsten active site. They display a diverse subunit and cofactor composition, but structural information on these enzymes is limited. Here we report the cryo-electron microscopic structures of the soluble Rhodobacter capsulatus FDH (RcFDH) as isolated and in the presence of reduced nicotinamide adenine dinucleotide (NADH). RcFDH assembles into a 360âkDa dimer of heterotetramers revealing a putative interconnection of electron pathway chains. Structural insights into Cullin4-RING ubiquitin ligase remodelling by Vpr from simian immunodeficiency viruses
Viruses have evolved means to manipulate the host's ubiquitin-proteasome system, in order to down-regulate antiviral host factors. The Vpx/Vpr family of lentiviral accessory proteins usurp the substrate receptor DCAF1 of host Cullin4-RING ligases (CRL4), a family of modular ubiquitin ligases involved in DNA replication, DNA repair and cell cycle regulation. CRL4DCAF1 specificity modulation by Vpx and Vpr from certain simian immunodeficiency viruses (SIV) leads to recruitment, poly-ubiquitylation and subsequent proteasomal degradation of the host restriction factor SAMHD1, resulting in enhanced virus replication in differentiated cells. To unravel the mechanism of SIV Vpr-induced SAMHD1 ubiquitylation, we conducted integrative biochemical and structural analyses of the Vpr protein from SIVs infecting Cercopithecus cephus (SIVmus). X-ray crystallography reveals commonalities between SIVmus Vpr and other members of the Vpx/Vpr family with regard to DCAF1 interaction, while cryo-electron microscopy and cross-linking mass spectrometry highlight a divergent molecular mechanism of SAMHD1 recruitment. In addition, these studies demonstrate how SIVmus Vpr exploits the dynamic architecture of the multi-subunit CRL4DCAF1 assembly to optimise SAMHD1 ubiquitylation. Together, the present work provides detailed molecular insight into variability and species-specificity of the evolutionary arms race between host SAMHD1 restriction and lentiviral counteraction through Vpx/Vpr proteins
Structural mechanism of CRL4-instructed STAT2 degradation via a novel cytomegaloviral DCAF receptor
Human cytomegalovirus (CMV) is a ubiquitously distributed pathogen whose rodent counterparts such as mouse and rat CMV serve as common infection models. Here, we conducted global proteome profiling of rat CMV-infected cells and uncovered a pronounced loss of the transcription factor STAT2, which is crucial for antiviral interferon signalling. Via deletion mutagenesis, we found that the viral protein E27 is required for CMV-induced STAT2 depletion. Cellular and in vitro analyses showed that E27 exploits host-cell Cullin4-RING ubiquitin ligase (CRL4) complexes to induce poly-ubiquitylation and proteasomal degradation of STAT2. Cryo-electron microscopy revealed how E27 mimics molecular surface properties of cellular CRL4 substrate receptors called DCAFs (DDB1- and Cullin4-associated factors), thereby displacing them from the catalytic core of CRL4. Moreover, structural analyses showed that E27 recruits STAT2 through a bipartite binding interface, which partially overlaps with the IRF9 binding site. Structure-based mutations in M27, the murine CMV homologue of E27, impair the interferon-suppressing capacity and virus replication in mouse models, supporting the conserved importance of DCAF mimicry for CMV immune evasion

TRY plant trait database â enhanced coverage and open access
Plant traits - the morphological, anatomical, physiological, biochemical and phenological characteristics of plants - determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of traitâbased plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits - almost complete coverage for âplant growth formâ. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and traitâenvironmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives
Chromosome Pairing Does Not Contribute to Nuclear Architecture in Vegetative Yeast Cells
There are several reports of a closer-than-random colocalization of homologous chromosomes in the vegetative nuclei of diploid budding yeast. Here, we studied by fluorescence in situ hybridization (FISH) the nuclear distribution of chromosomes and found a slight tendency toward closer proximity between homologous (allelic) loci than between any nonhomologous chromosomal regions. We show that most of this preferential association is not due to vegetative (also known as somatic) pairing but is caused by the polar orientation of interphase chromosomes (Rabl orientation). We quantified the occasional loss of detectable fluorescence signals that is inherent to the FISH method. Signal loss leads to the occurrence of a single signal that may be misinterpreted as the close association of two homologous chromosomal sites. The nuclear distribution of homologous loci, when corrected for the influence of nuclear architecture and methodological faults, was not different or was only marginally different from a random relative positioning as predicted by computer simulation. We discuss here several possibilities for the residual homologous proximity that do not invoke homology-dependent vegetative pairing, and we conclude that, in diploid budding yeast, constitutive vegetative pairing is a negligible factor for the organization of the interphase nucleus
FCREX: Fuel Cell Range Extender with Cold Start Capability for Cargo Pedelecs
More than 75 % of the Europeans are living in cities and the amount of goods delivered to households is constantly increasing, mainly caused by the increasing market share of online shopping. The associated raise in traffic volume intensifies the urban problems of traffic jams and air pollution. To overcome these issues, new solutions especially for the last mile delivery have to be established providing low exhaust emissions, high transportation flexibility, high operating range and reliable functionality regardless of the weather.
Electrically-operated Cargo Pedelecs (E-Pedelecs) have a high potential to cover the gap of cargo transport solutions in cities. Based on a European research study, 51 % of all motorized trips related to goods transport can be shifted to these bicycles in European cities. These E-Pedelecs provide a (locally) emission-free transport and they can use roads, bikeways, bus lanes and pedestrian areas. Beside this ecofriendly transport and their high flexibility, E-Pedelecs are still struggling to cover the required operating range for multiple-shift operation and to assure a sufficient lifetime under subzero conditions. A German research study identified the battery to be the bottleneck of this problem.
The presented FC REX is a fuel cell range extender system especially designed for E-Pedelecs under all climatic conditions in European cities. A cold start unit is developed to enable freeze start of the fuel cell system and provide full system power in short time and without loss of operating range. All components of the system, including the fuel cell stack, are specially designed with respect to fast and efficient heat transfer, high system efficiency and high power density. The latter is one of the key requirements for a successful integration of the FC REX into E-Pedelecs. The limited installation space was previously the main obstacle for fuel cell powered E-Pedelecs
Experimental investigation of a liquid cooled high temperature proton exchange membrane (HT-PEM) fuel cell coupled to a sodium-alanate tank
In high temperature proton exchange membrane (HT-PEM) fuel cells, waste heat at
approximately 160 °C is produced, which can be used for thermal integration of solid state hydrogen
storage systems. In the present study, a HT-PEM fuel cell stack (400 W) with direct liquid cooling is
characterized and coupled to a separately characterized sodium alanate storage tank (300 g material).
The coupled system is studied in steady-state for 20 min operation and all relevant heat flows are
determined. Even though in lab-scale heat losses at that specific power and temperature level cannot
be completely avoided, it is demonstrated that the amount of heat transferred from the fuel cell stack
to the cooling liquid circuit is sufficient to desorb the necessary amount of hydrogen from the storage
tank. Furthermore, it is shown that the reaction rate of the sodium alanate at 160 °C and 1.7 bar is
adequate to provide the hydrogen to the fuel cell stack. Based on these experimental investigations, a
set of recommendations is given for the future design and layout of similar coupled systems
