    Daily and Seasonal Foraging Activity of the Oriental Non-army Ant Doryline Cerapachys sulcinodis Species Complex (Hymenoptera: Formicidae)

    The ant subfamily Dorylinae contains the true army ant genera, the species of which are dominant predators, as well as non-army ants. Some of the non-army ant genera are closely related to true army ants, so they could be an important key in understanding the evolution of the army ant adaptive syndrome. Nevertheless, there has been little field research on the non-army ant doryline species because many of them are very rare. Cerapachys is one of the genera closely related to the Old-World army ants. We found some species of Cerapachys are commonly distributed in northern Thailand, so we investigated the daily and seasonal foraging activities of a non-army doryline ant of Cerapachys sulcinodis species complex in the seasonal tropical area of northern Thailand. Workers of the C. sulcinodis complex were diurnal and actively foraged during the rainy season, and their reproduction and foraging activity paused during the dry season. This report is the first on the foraging activity of non-army doryline ants under field conditions.

    Transcription factor hepatocyte nuclear factor 1-beta (HNF-1β) enhances checkpoint kinase 1 (Chk1) activation and promotes G2/M cell cycle progression in ovarian clear cell carcinoma (CCC) following exposure to diverse genotoxic agents including bleomycin. However, the underlying mechanism leading to checkpoint activation of HNF-1β still remains largely unknown. To clarify the effects of HNF-1β on cell cycle checkpoints, human CCC cell lines were transfected with siRNAs targeting HNF-1β, Claspin, USP28, or a control vector. Ubiquitination and stabilization of Claspin protein by HNF-1β was assessed by immunoprecipitation. Loss-of-function studies using RNAi-mediated gene silencing indicated that HNF-1β facilitated the Claspin expression after treatment with a genotoxic agent bleomycin, resulting in accumulation of phosphorylated Chk1 (p-Chk1) and promotion of survival in CCC cell lines. This study showed for the first time that USP28, a de-ubiquitinase crucial for Claspin expression, is one target gene of HNF-1β. Knockdown of endogenous USP28 suppressed the Claspin expression and p-Chk1 activation and cell viability. Our findings identify a novel pathway of the HNF-1β-USP28-Claspin-Chk1 axis in checkpoint signal amplification in response to DNA damage.     Ants are of high density on the host plant of golden tortoise beetle, Ipomoea carnea, and are being attracted by extrafloral nectaries on petiols.They are important predators of immature stages (eggs, larvae and pupae) of the beetles.The experiment was aimed to know how the golden tortoise beetles defense themselves against ants on I. carnea.The results indicated that golden tortoise beetle,both larvae and adults showed odd habits.They have a specific external organ, a protective camouflage with excreta of the larvae and oviposition strategy.The ant-beetle interaction on the host plants are discussed

    Malignant transformation of endometriosis is a rare and still poorly understood event, but is associated with the distortion of the pro-oxidant and anti-oxidant balance. The aim of the present study was to quantify the numbers of macrophages polarized as M1 or M2 phenotypes and the expression of heme oxygenase (HO)-1 in tissue sections from patients with benign ovarian endometrioma (OE) and its malignant transformation (endometriosis-associated ovarian cancer, EAOC). We performed a retrospective study at the Department of Gynecology, Nara Medical University hospital from December 2012 to March 2015. This study included 53 patients with OE (n = 33) and EAOC (n = 20), and we evaluated polarized functional status of macrophages by immunohistochemical staining of CD68, CD11c, CD163 and HO-1. The number of the M1 phenotype (CD11c+, p = 0.001) and the M2 phenotype (CD163+, p = 0.009) was significantly lower in EAOC patients than in OE patients. Analyzing the correlations between the studied markers, the expression of CD68, CD11c, and CD163 proteins significantly correlated with each other (p < 0.001). The number of M2 phenotypes expressing HO-1 was significantly decreased in the EAOC group, compared with the OE group (P < 0.001), demonstrating sustained downregulation of an antioxidant marker, HO-1, in EAOC. In conclusion, reduced number of M2 macrophages expressing HO-1 may have an important role in promoting malignant transformation of OE.博士(医学)・乙第1434号・令和元年9月27日Copyright © 2018.     Novel Heterogenous CHS1 Mutations Identified in Five Japanese Patients with Chediak-Higashi Syndrome

    Chediak-Higashi syndrome (CHS) is a rare, autosomal recessive disorder characterized by oculocutaneous albinism, recurrent bacterial infections and progressive neurological dysfunction. We demonstrate novel heterogenous mutations of CHS1, the responsive gene of CHS, identified in five Japanese patients with CHS. Patients 1, 2, and 3 were siblings, and they had albinism of the skin and hair. They all had a heterogenous two-base deletion (c.5541-5542 del AA, p.Q1847fsX1850) in exon 18. Patient 4 had a heterogenous single-base insertion (c.3944-3945 ins C, p.T1315fsX1331) in exon 10. The patient exhibited severe early-onset phenotype and suffered from hemophagocytic lymphohistiocytosis. Patient 5 had two heterogenous nonsense mutations; c.7982C>G, p.S2661X in exon 30 and c.8281A>T, p.R2761X in exon 31. The patient suffered from infections in childhood and had visual disturbance and albinism of the skin and hair. The CHS1 mutations described here have not been reported previously

    Collaboration between kindergarten teachers and researchers in practice research : Difference in recognition of practice research <Collaboration with the local community>

    In practice research on early childhood care and education, collaboration between practitioners and researchers has been actively implemented. However, such partnerships have not always been successful. In such cases, it is necessary to consider the possibility of a difference in the recognition of practice research between the two parties. The purpose of this study was to clarify such recognition differences between kindergarten teachers and researchers. Interviews with kindergarten teachers and researchers who collaborated in performing practice research were qualitatively analyzed. Results showed that recognition of five items of research - research themes, scope, duration, results, and application possibilities - differed between researchers and kindergarten teachers. Teachers, being more practice oriented, prefer more concrete research themes and scope, whereas researchers lean more toward abstract themes and scope. In addition, teachers give little consideration to research duration, whereas researchers focus on the need to gather and analyze the data within the determined period. Moreover, neither of the parties is fully aware of these differences. Consequently, interaction in the collaboration may come to resemble a “tug-of-war” with both partners asserting their own viewpoints

    Transcriptome profiling of the spermatheca identifies genes potentially involved in the long-term sperm storage of ant queens

    Females of social Hymenoptera only mate at the beginning of their adult lives and produce offspring until their death. In most ant species, queens live for over a decade, indicating that ant queens can store large numbers of spermatozoa throughout their long lives. To reveal the prolonged sperm storage mechanisms, we identified enriched genes in the sperm-storage organ (spermatheca) relative to those in body samples in Crematogaster osakensis queens using the RNA-sequencing method. The genes encoding antioxidant enzymes, proteases, and extracellular matrix-related genes, and novel genes that have no similar sequences in the public databases were identified. We also performed differential expression analyses between the virgin and mated spermathecae or between the spermathecae at 1-week and 1-year after mating, to identify genes altered by the mating status or by the sperm storage period, respectively. Gene Ontology enrichment analyses suggested that antioxidant function is enhanced in the spermatheca at 1-week after mating compared with the virgin spermatheca and the spermatheca at 1-year after mating. In situ hybridization analyses of 128 selected contigs revealed that 12 contigs were particular to the spermatheca. These genes have never been reported in the reproductive organs of insect females, suggesting specialized roles in ant spermatheca

Vytvářený systém se bude týkat problematiky 3D vizualizace úlohy na hledání modelu množiny formulí v predikátové logice prvního řádu a modelování důležitých vlastností relací. Nástroj bude také obsahovat nápovědu pro vytvoření modelu množiny. V první části práce, je čtenář seznámen s teoretickými základy z dané oblasti, na které plynule navazuje specifikace požadavků na nový systém. Dále je text práce zaměřen především na analýzu a návrh jednotlivých částí systému, která je doplněna o třídní a sekvenční diagramy UML.The goal of this thesis is to Provide design and analysis of a new system, which could then be used as a teaching tool in subject called Mathematical Logic. The new system will consist of two main parts. The first part will be dealing with the problem of 3D visualization of formula set models. The second part will include an environment for modeling important properties of relations. Our system will also include an aid for creating formula set models. In the first part of the thesis we focus on theoretical background and requirement specification for the new system.
