The checkpoint Saccharomyces cerevisiae Rad9 protein contains a tandem tudor domain that recognizes DNA.
International audienceDNA damage checkpoints are signal transduction pathways that are activated after genotoxic insults to protect genomic integrity. At the site of DNA damage, 'mediator' proteins are in charge of recruiting 'signal transducers' to molecules 'sensing' the damage. Budding yeast Rad9, fission yeast Crb2 and metazoan 53BP1 are presented as mediators involved in the activation of checkpoint kinases. Here we show that, despite low sequence conservation, Rad9 exhibits a tandem tudor domain structurally close to those found in human/mouse 53BP1 and fission yeast Crb2. Moreover, this region is important for the resistance of Saccharomyces cerevisiae to different genotoxic stresses. It does not mediate direct binding to a histone H3 peptide dimethylated on K79, nor to a histone H4 peptide dimethylated on lysine 20, as was demonstrated for 53BP1. However, the tandem tudor region of Rad9 directly interacts with single-stranded DNA and double-stranded DNAs of various lengths and sequences through a positively charged region absent from 53BP1 and Crb2 but present in several yeast Rad9 homologs. Our results argue that the tandem tudor domains of Rad9, Crb2 and 53BP1 mediate chromatin binding next to double-strand breaks. However, their modes of chromatin recognition are different, suggesting that the corresponding interactions are differently regulated
The effectiveness of scoliosis screening programs: methods for systematic review and expert panel recommendations formulation
Background: Literature on scoliosis screening is vast, however because of the observational nature of available data and methodological flaws, data interpretation is often complex, leading to incomplete and sometimes, somewhat misleading conclusions. The need to propose a set of methods for critical appraisal of the literature about scoliosis screening, a comprehensive summary and rating of the available evidence appeared essential.
METHODS:
To address these gaps, the study aims were: i) To propose a framework for the assessment of published studies on scoliosis screening effectiveness; ii) To suggest specific questions to be answered on screening effectiveness instead of trying to reach a global position for or against the programs; iii) To contextualize the knowledge through expert panel consultation and meaningful recommendations. The general methodological approach proceeds through the following steps: Elaboration of the conceptual framework; Formulation of the review questions; Identification of the criteria for the review; Selection of the studies; Critical assessment of the studies; Results synthesis; Formulation and grading of recommendations in response to the questions. This plan follows at best GRADE Group (Grades of Recommendation, Assessment, Development and Evaluation) requirements for systematic reviews, assessing quality of evidence and grading the strength of recommendations.
CONCLUSIONS:

De la lecture à la résolution de problèmes: des habiletés spécifiques à développer
La réussite des élèves en mathématiques, particulièrement la réussite en résolution de problèmes écrits, a souvent été associée aux compétences en lecture. Si plusieurs recherches ont permis d’établir clairement un lien entre le rendement en lecture des élèves et leur rendement en mathématiques, nous ne savons pas encore précisément quelles habiletés spécifiques en lecture constituent les meilleurs indicateurs du rendement en résolution de problèmes écrits de mathématiques. Notre objectif consiste à préciser les habiletés liées à la lecture que les bons solutionneurs mettent à profit en contexte de résolution de problèmes écrits de mathématiques. Pour atteindre les objectifs de la recherche, nous avons utilisé un devis de recherche quantitatif. L’échantillon de l’étude est composé de 73 élèves de 4e année du primaire. Les participants ont complété deux épreuves de compréhension en lecture, soit un texte narratif et un texte informatif, ainsi qu’un ensemble de problèmes écrits de mathématiques. Les analyses de corrélation nous ont permis d’établir des liens entre les différentes variables à l’étude. Les résultats soutiennent que certaines habiletés spécifiques en lecture constituent des indicateurs à privilégier afin de juger du rendement en résolution de problèmes écrits de mathématiques des élèves.Mots-clés : Mathématiques, résolution de problèmes écrits, situation-problème, compréhension en lecture, enseignement primaire
The Online Citizens Project: Effects oftransitional peer support groups facilitated by peer support workers for persons living with severe mental illness in times of the Covid-19 pandemic
From August to November 2020, the Online Citizens Project has been offered as a transitional peer support group to persons living with Serious Mental Illness (SMI) in the province of Quebec, Canada. The Citizens’ Project is a support group where participants share their challenges and accomplishments with each other and receive honest and confidential feedback. These groups had a personal-civic recovery focus and consisted of a series of 10 weekly 90-minute online workshops. To evaluate the impact of the intervention on the participants' sense of citizenship, all study participants completed the 23-item French version of the Citizenship Measure before (T1) and after (T2) the intervention (?14 weeks between T1 and T2). The mean and standard deviation differences between the two measurement times were compared between the experimental group and the control group. The mean score to the Citizenship Measure for the experimental group varied by -0.4%. The mean score for the experimental group varied by -5.5%. For the control group, there was a decrease in the means for each of the 5 sub-scales of the Citizenship Measure. In total, the difference was statistically significant (P=.04). Results suggest that the Online Citizens Project had a protective effect on the sense of citizenship of the participants living with SMI in the experimental group compared to those in the control group. The Citizenship Measure can be used to gauge the effects of such an online intervention, intentionally designed to promote the exercise of citizenship, namely the Citizens Project.
De agosto a novembro de 2020, o Projeto Cidadãos Online foi oferecido como um grupo de apoio de pares de transição para pessoas que vivem com Doenças Mentais Graves (SMI) na província de Quebec, Canadá. O Projeto dos Cidadãos é um grupo de apoio onde os participantes compartilham seus desafios e realizações uns com os outros e recebem feedback honesto e confidencial. Esses grupos tinham como foco o recovery e cidadania individual e consistiam em uma série de 10 grupos semanais de trabalho online com 90 minutos de duração. Todos os participantes do estudo completaram a versão francesa de 23 itens da Medida de Cidadania antes (T1) e depois (T2) da intervenção (?14 semanas entre T1 e T2) para avaliar o sentido de cidadania e o impacto da intervenção nos participantes. As diferenças de média e desvio padrão entre os dois tempos de medição foram comparadas entre o grupo experimental e o grupo controle. A pontuação média da Medida de Cidadania para o grupo experimental variou em -0,4%. A pontuação média para o grupo experimental variou em -5,5%. Para o grupo controle, houve diminuição das médias para cada uma das 5 subescalas da Medida de Cidadania. No total, a diferença foi estatisticamente significativa (P = 0,04). Os resultados sugerem que o Projeto Cidadãos Online teve um efeito protetor no senso de cidadania dos participantes que vivem com SMI no grupo experimental em comparação com aqueles no grupo de controle. A Medida de Cidadania pode ser utilizada para medir os efeitos dessa intervenção online, intencionalmente concebida para promover o exercício da cidadania, nomeadamente o Projecto Cidadão
Emotion processing in late adulthood: the effect of emotional valence and face age on behavior and scanning patterns
Age-related differences in emotion recognition are well-documented in older adults aged 65 and above, with stimulus valence and the age of the model being key influencing factors. This study examined these variables across three experiments using a novel set of images depicting younger and older models expressing positive and negative emotions (e.g., happy vs. sad; interested vs. bored). Experiment 1 focused on valence- arousal dimensions, Experiment 2 on emotion recognition accuracy, and Experiment 3 on visual fixation patterns. Age-related differences were found in emotion recognition. No significant age-related differences in gaze behavior were found; both age groups looked more at the eye region. The positivity effect—older adults’ tendency to prioritize positive over negative information—did not consistently manifest in recognition performance or scanning patterns. However, older adults evaluated positive emotions differently than negative emotions, rating negative facial expressions as less negative and positive emotions as more arousing compared to younger adults. Finally, emotions portrayed by younger models were rated as more intense and more positive than those portrayed by older adults by both older and younger adults. We conclude that the positivity effect and own-age bias may be more complex and nuanced than previously thought
