    Teaching Lab: Training Novice Computer Science Teachers

This poster presents the Teaching Lab – a mature and fully developed training course for novice teachers (mostly undergraduate teaching assistants), its core principles, content and unique features as it evolved over five years. Our experience can be helpful to others intending to create or adjust a training program for novice teachers

Zproduktivnění má dopomoci úprava či změna strojů, nástrojů a také řezných podmínek. Stroje se vybírají výhradně ze strojového parku, který má k dispozici firma Armatury KLAD spol. s r. o. Úvodní část práce je věnována představení podniku, ve kterém byla diplomová práce zpracována. Následuje popis samotného produktu, zpětného ventilu. Dále je popsána současná technologie výroby, která obsahuje jednotlivá pracoviště, stroje, nástroje a výrobní podmínky. Další část práce obsahuje návrh na zproduktivnění výroby, díky změně výrobní technologie. Zproduktivnění se dosáhne díky zkrácení výrobních časů a snížením finančních nákladů na kus. Na závěr je zmíněno technicko – hospodářské zhodnocení a porovnání stávajícího a nově navrženého výrobního postupu.Diploma thesis deals with problematic about manufacturing body of lift check valve. Thesis is focused on more productive manufacturing technologies of this product. It should be carried out by modifying or changing machines, tools and cutting conditions too. Machines are chosen only from machine park, of company Armatury KLAD spol. s r.o. Introductory part of thesis is about company, where the diploma thesis was processed. After that there is product (lift check valve) description and also description of current manufacturing procces. This category include severy single workplace, machine, tool and cutting condition. Next part is about proposal for new technology. This technology is more productive, because time for production of one piece is shorter and input costs are lower too.     Cardiolipin-containing lipid membranes attract the bacterial cell division protein diviva

    DivIVA is a protein initially identified as a spatial regulator of cell division in the model organism Bacillus subtilis, but its homologues are present in many other Gram-positive bacteria, including Clostridia species. Besides its role as topological regulator of the Min system during bacterial cell division, DivIVA is involved in chromosome segregation during sporulation, genetic competence, and cell wall synthesis. DivIVA localizes to regions of high membrane curvature, such as the cell poles and cell division site, where it recruits distinct binding partners. Previously, it was suggested that negative curvature sensing is the main mechanism by which DivIVA binds to these specific regions. Here, we show that Clostridioides difficile DivIVA binds preferably to membranes containing negatively charged phospholipids, especially cardiolipin. Strikingly, we observed that upon binding, DivIVA modifies the lipid distribution and induces changes to lipid bilayers containing cardiolipin. Our observations indicate that DivIVA might play a more complex and so far unknown active role during the formation of the cell division septal membrane

    The analysis of serum mineral profile of cows before and after calving: A case study

These include the drying period, parturition, postpartum period and the first 100 days of lactation. The mineral profile displays the balanced feed ration appropriate for the category. Therefore, investigation of serum mineral profile of dry cows and cows in 12th week after calving was the aim of this study. In this work 12 high producing Holstein Friesian dairy cows from dairy farm were used. The analysed elements Ca2+, P, Na+, K+, Mg2+ and chlorides (Cl-) were determined at 4th week after drying  and during 12th week after calving. The data obtained were compared with the standard for dairy cows. Compared to reference interval, before calving 8 and after calving 5 cows had hypocalcaemia. Hypophosphatemia was detected for 2 cows before calving and 3 after calving. For the other elements Na, K, Mg2+ and chlorides (Cl-) no deviations from the reference intervals were found. In blood serum mineral profile between dry cows and cows in 12th week of lactation significant differences were found for Mg Cl- (increase) and K (decrease). Results shown, that main problem of dry and lactating cows was the hypocalcaemia and partly the hypophosphatemia.Keywords: metabolic test, mineral profile, blood serum, dairy cowReferencesAiello, S.     Analysis of movement mechanics in the training process of sport horses

    Article Details: Received: 2020-10-15 | Accepted: 2020-11-27 | Available online: 2021-01-31https://doi.org/10.15414/afz.2021.24.mi-prap.145-149The aim of the study was to determine the effect of training process on changes of quantitative and qualitative indicators of the movement mechanics of sport horses. A load controller was used to evaluate the motion mechanics. The training load was divided into three stages with different lengths, speeds and degrees of load. Tested animals were 14 horses of Slovak Warmblood breed. Our results have shown that gradual increase of load in the tested horses resulted in better spaciousness of movement, which was reflected in the elongation of the horse´s stride, regular movement and significant energy of the pelvic limbs (p<0.05). From our findings we can state that the effect of gender did not have significant effect on the number and length of steps in each degree of load. We have noticed a significant difference (p<0.05) in the „number of steps“ parameter between the age categories of horses in the age up to 7 years (3.460±619) and the category aged up to 14 years (3.383±572).     Non–T Cell Activation Linker (NTAL): A Transmembrane Adaptor Protein Involved in Immunoreceptor Signaling

    A key molecule necessary for activation of T lymphocytes through their antigen-specific T cell receptor (TCR) is the transmembrane adaptor protein LAT (linker for activation of T cells). Upon TCR engagement, LAT becomes rapidly tyrosine phosphorylated and then serves as a scaffold organizing a multicomponent complex that is indispensable for induction of further downstream steps of the signaling cascade. Here we describe the identification and preliminary characterization of a novel transmembrane adaptor protein that is structurally and evolutionarily related to LAT and is expressed in B lymphocytes, natural killer (NK) cells, monocytes, and mast cells but not in resting T lymphocytes. This novel transmembrane adaptor protein, termed NTAL (non–T cell activation linker) is the product of a previously identified WBSCR5 gene of so far unknown function. NTAL becomes rapidly tyrosine-phosphorylated upon cross-linking of the B cell receptor (BCR) or of high-affinity Fcγ- and Fcɛ-receptors of myeloid cells and then associates with the cytoplasmic signaling molecules Grb2, Sos1, Gab1, and c-Cbl. NTAL expressed in the LAT-deficient T cell line J.CaM2.5 becomes tyrosine phosphorylated and rescues activation of Erk1/2 and minimal transient elevation of cytoplasmic calcium level upon TCR/CD3 cross-linking. Thus, NTAL appears to be a structural and possibly also functional homologue of LAT in non–T cells
