Combined Acidification and Solar Drying of Pig Slurries for Nutrient Recovery and Controlled Atmospheric Emissions
The solar drying of pig slurries was tested in a pilot-scale greenhouse (10 m2 footprint), operated with forced ventilation under low and high solar irradiation in Mediterranean conditions. Gaseous emissions were prevented through slurry acidification and by the biofiltration of exhaust gases. Air relative humidity and temperature in and out the greenhouse, as well as the weight of a slurry sample, were monitored online to command the ventilation regime. Daily average drying rate values ranged from 0.3 to 2.8 kg m−2 d−1 and displayed a direct dependency with solar radiation until the pig slurry lost a 60% of its initial weight, with a solar energy efficiency of about 26%. Upon further drying, the water content from pig slurries stabilized at around 10%. Mass balances between the initial slurry and dried product were closed for total solids and organic matter, but the recovery of nutrients ranged from 69% to 81%, apparently because of precipitation and incrustation phenomena. The NPK composition of the final product was 4.3–2.5–3.8 and fulfilled current regulations for solid organic fertilizers. Operational costs of the drying process and fertilizing quality parameters were also discussed.info:eu-repo/semantics/publishedVersio
External Load Demands and Positional Differences in Elite Futsal Using UWB Technology
A statistical mechanical description of metastable states and hysteresis in the 3D soft-spin random-field model at T=0
We present a formalism for computing the complexity of metastable states and
the zero-temperature magnetic hysteresis loop in the soft-spin random-field
model in finite dimensions. The complexity is obtained as the Legendre
transform of the free-energy associated to a certain action in replica space
and the hysteresis loop above the critical disorder is defined as the curve in
the field-magnetization plane where the complexity vanishes; the nonequilibrium
magnetization is therefore obtained without having to follow the dynamical
evolution. We use approximations borrowed from condensed-matter theory and
based on assumptions on the structure of the direct correlation functions (or
proper vertices), such as a local approximation for the self-energies, to
calculate the hysteresis loop in three dimensions, the correlation functions
along the loop, and the second moment of the avalanche-size distribution.Comment: 28 pages, 12 figure
Isolation of human fibroadipogenic progenitors and satellite cells from frozen muscle biopsies
Altres ajuts: Association Française contre les Myopathies (22525)Altres ajuts: Fundación Isabel GemioSkeletal muscle contains multiple cell types that work together to maintain tissue homeostasis. Among these, satellite cells (SC) and fibroadipogenic progenitors cells (FAPs) are the two main stem cell pools. Studies of these cells using animal models have shown the importance of interactions between these cells in repair of healthy muscle, and degeneration of dystrophic muscle. Due to the unavailability of fresh patient muscle biopsies, similar analysis of interactions between human FAPs and SCs is limited especially among the muscular dystrophy patients. To address this issue here we describe a method that allows the use of frozen human skeletal muscle biopsies to simultaneously isolate and grow SCs and FAPs from healthy or dystrophic patients. We show that while the purified SCs differentiate into mature myotubes, purified FAPs can differentiate into adipocytes or fibroblasts demonstrating their multipotency. We find that these FAPs can be immortalized and the immortalized FAPs (iFAPs) retain their multipotency. These approaches open the door for carrying out personalized analysis of patient FAPs and interactions with the SCs that lead to muscle loss
Full-bandwidth electrophysiology of seizures and epileptiform activity enabled by flexible graphene microtransistor depth neural probes
Mapping the entire frequency bandwidth of brain electrophysiological signals is of paramount importance for understanding physiological and pathological states. The ability to record simultaneously DC-shifts, infraslow oscillations (<0.1 Hz), typical local field potentials (0.1-80 Hz) and higher frequencies (80-600 Hz) using the same recording site would particularly benefit preclinical epilepsy research and could provide clinical biomarkers for improved seizure onset zone delineation. However, commonly used metal microelectrode technology suffers from instabilities that hamper the high fidelity of DC-coupled recordings, which are needed to access signals of very low frequency. In this study we used flexible graphene depth neural probes (gDNPs), consisting of a linear array of graphene microtransistors, to concurrently record DC-shifts and high-frequency neuronal activity in awake rodents. We show here that gDNPs can reliably record and map with high spatial resolution seizures, pre-ictal DC-shifts and seizure-associated spreading depolarizations together with higher frequencies through the cortical laminae to the hippocampus in a mouse model of chemically induced seizures. Moreover, we demonstrate the functionality of chronically implanted devices over 10 weeks by recording with high fidelity spontaneous spike-wave discharges and associated infraslow oscillations in a rat model of absence epilepsy. Altogether, our work highlights the suitability of this technology for in vivo electrophysiology research, and in particular epilepsy research, by allowing stable and chronic DC-coupled recordings
The major upgrade of the MAGIC telescopes, Part II: A performance study using observations of the Crab Nebula
MAGIC is a system of two Imaging Atmospheric Cherenkov Telescopes located in
the Canary island of La Palma, Spain. During summer 2011 and 2012 it underwent
a series of upgrades, involving the exchange of the MAGIC-I camera and its
trigger system, as well as the upgrade of the readout system of both
telescopes. We use observations of the Crab Nebula taken at low and medium
zenith angles to assess the key performance parameters of the MAGIC stereo
system. For low zenith angle observations, the standard trigger threshold of
the MAGIC telescopes is ~50GeV. The integral sensitivity for point-like sources
with Crab Nebula-like spectrum above 220GeV is (0.66+/-0.03)% of Crab Nebula
flux in 50 h of observations. The angular resolution, defined as the sigma of a
2-dimensional Gaussian distribution, at those energies is < 0.07 degree, while
the energy resolution is 16%. We also re-evaluate the effect of the systematic
uncertainty on the data taken with the MAGIC telescopes after the upgrade. We
estimate that the systematic uncertainties can be divided in the following
components: < 15% in energy scale, 11-18% in flux normalization and +/-0.15 for
the energy spectrum power-law slope.Comment: 21 pages, 25 figures, accepted for publication in Astroparticle

Reversible Integration of Microfluidic Devices with Microelectrode Arrays for Neurobiological Applications
The majority of current state-of-the-art microfluidic devices are fabricated via replica molding of the fluidic channels into PDMS elastomer and then permanently bonding it to a Pyrex surface using plasma oxidation. This method presents a number of problems associated with the bond strengths, versatility, applicability to alternative substrates, and practicality. Thus, the aim of this study was to investigate a more practical method of integrating microfluidics which is superior in terms of bond strengths, reversible, and applicable to a larger variety of substrates, including microfabricated devices. To achieve the above aims, a modular microfluidic system, capable of reversible microfluidic device integration, simultaneous surface patterning and multichannel fluidic perfusion, was built. To demonstrate the system’s potential, the ability to control the distribution of A549 cells inside a microfluidic channel was tested. Then, the system was integrated with a chemically patterned microelectrode array, and used it to culture primary, rat embryo spinal cord neurons in a dynamic fluidic environment. The results of this study showed that this system has the potential to be a cost effective and importantly, a practical means of integrating microfluidics. The system’s robustness and the ability to withstand extensive manual handling have the additional benefit of reducing the workload. It also has the potential to be easily integrated with alternative substrates such as stainless steel or gold without extensive chemical modifications. The results of this study are of significant relevance to research involving neurobiological applications, where primary cell cultures on microelectrode arrays require this type of flexible integrated solution
Dysferlin Forms a Dimer Mediated by the C2 Domains and the Transmembrane Domain In Vitro and in Living Cells
Dysferlin was previously identified as a key player in muscle membrane repair and its deficiency leads to the development of muscular dystrophy and cardiomyopathy. However, little is known about the oligomerization of this protein in the plasma membrane. Here we report for the first time that dysferlin forms a dimer in vitro and in living adult skeletal muscle fibers isolated from mice. Endogenous dysferlin from rabbit skeletal muscle exists primarily as a ∼460 kDa species in detergent-solubilized muscle homogenate, as shown by sucrose gradient fractionation, gel filtration and cross-linking assays. Fluorescent protein (YFP) labeled human dysferlin forms a dimer in vitro, as demonstrated by fluorescence correlation spectroscopy (FCS) and photon counting histogram (PCH) analyses. Dysferlin also dimerizes in living cells, as probed by fluorescence resonance energy transfer (FRET). Domain mapping FRET experiments showed that dysferlin dimerization is mediated by its transmembrane domain and by multiple C2 domains. However, C2A did not significantly contribute to dimerization; notably, this is the only C2 domain in dysferlin known to engage in a Ca-dependent interaction with cell membranes. Taken together, the data suggest that Ca-insensitive C2 domains mediate high affinity self-association of dysferlin in a parallel homodimer, leaving the Ca-sensitive C2A domain free to interact with membranes
Gene Expression Profiling in Limb-Girdle Muscular Dystrophy 2A
Limb-girdle muscular dystrophy type 2A (LGMD2A) is a recessive genetic disorder caused by mutations in calpain 3 (CAPN3). Calpain 3 plays different roles in muscular cells, but little is known about its functions or in vivo substrates. The aim of this study was to identify the genes showing an altered expression in LGMD2A patients and the possible pathways they are implicated in. Ten muscle samples from LGMD2A patients with in which molecular diagnosis was ascertained were investigated using array technology to analyze gene expression profiling as compared to ten normal muscle samples. Upregulated genes were mostly those related to extracellular matrix (different collagens), cell adhesion (fibronectin), muscle development (myosins and melusin) and signal transduction. It is therefore suggested that different proteins located or participating in the costameric region are implicated in processes regulated by calpain 3 during skeletal muscle development. Genes participating in the ubiquitin proteasome degradation pathway were found to be deregulated in LGMD2A patients, suggesting that regulation of this pathway may be under the control of calpain 3 activity. As frizzled-related protein (FRZB) is upregulated in LGMD2A muscle samples, it could be hypothesized that β-catenin regulation is also altered at the Wnt signaling pathway, leading to an incorrect myogenesis. Conversely, expression of most transcription factor genes was downregulated (MYC, FOS and EGR1). Finally, the upregulation of IL-32 and immunoglobulin genes may induce the eosinophil chemoattraction explaining the inflammatory findings observed in presymptomatic stages. The obtained results try to shed some light on identification of novel therapeutic targets for limb-girdle muscular dystrophies
