    Conversion of relativistic pair energy into radiation in the jets of active galactic nuclei

    It is generally accepted that relativistic jet outflows power the nonthermal emission from active galactic nuclei (AGN). The composition of these jets -- leptonic versus hadronic -- is still under debate. We investigate the microphysical details of the conversion process of the kinetic energy in collimated relativistic pair outflows into radiation through interactions with the ambient interstellar medium. Viewed from the coordinate system comoving with the pair outflow, the interstellar protons and electrons represent a proton-electron beam propagating with relativistic speed in the pair plasma. We demonstrate that the beam excites both electrostatic and low-frequency magnetohydrodynamic Alfven-type waves via a two-stream instability in the pair background plasma, and we calculate the time evolution of the distribution functions of the beam particles and the generated plasma wave turbulence power spectra. For standard AGN jet outflow and environment parameters we show that the initial beam distributions of interstellar protons and electrons quickly relax to plateau-distributions in parallel momentum, transferring thereby one-half of the initial energy density of the beam particles to electric field fluctuations of the generated electrostatic turbulence. On considerably longer time scales, the plateaued interstellar electrons and protons will isotropise by their self-generated transverse turbulence and thus be picked-up in the outflow pair plasma. These longer time scales are also characteristic for the development of transverse hydromagnetic turbulence from the plateaued electrons and protons.     Evolution and rapid spread of a reassortant A(H3N2) virus that predominated the 2017-2018 influenza season

    The 2017-2018 North American inïŹ‚uenza season caused more hospitalizations and deaths than any year since the 2009 H1N1 pandemic. The majority of recorded inïŹ‚uenza infections were caused by A(H3N2) viruses, with most of the virus's North American diversity falling into the A2 clade. Within A2, we observe a subclade which we call A2/re that rose to comprise almost 70 per cent of A(H3N2) viruses circulating in North America by early 2018. Unlike most fast-growing clades, however, A2/re contains no amino acid substitutions in the hemagglutinin (HA) segment. Moreover, hemagglutination inhibition assays did not suggest substantial antigenic differences between A2/re viruses and viruses sampled during the 2016-2017 season. Rather, we observe that the A2/re clade was the result of a reassortment event that occurred in late 2016 or early 2017 and involved the combination of the HA and PB1 segments of an A2 virus with neuraminidase (NA) and other segmentsa virus from the clade A1b. The success of this clade shows the need for antigenic analysis that targets NA in addition to HA. Our results illustrate the potential for non-HA drivers of viral success and necessitate the need for more thorough tracking of full viral genomes to better understand the dynamics of inïŹ‚uenza epidemics

    On the making and taking of professionalism in the further education workplace

    This paper examines the changing nature of professional practice in English further education. At a time when neo-liberal reform has significantly impacted on this under-researched and over-market-tested sector, little is known about who its practitioners are and how they construct meaning in their work. Sociological interest in the field has tended to focus on further education practitioners as either the subjects of market and managerial reform or as creative agents operating within the contradictions of audit and inspection cultures. In challenging such dualism, which is reflective of wider sociological thinking, the paper examines the ways in which agency and structure combine to produce a more transformative conception of the further education professional. The approach contrasts with a prevailing policy discourse that seeks to re-professionalise and modernise further education practice without interrogating either the terms of its professionalism or the neo-liberal practices in which it resides

": A Qualitative Exploration of Athlete Perceptions of Social Loafing in Training and Competition Contexts - Implications for Team Sport Coaches

    Semi-structured interviews were conducted with twenty team sport athletes representing a range of different sports with the aim of investigating their views relating to social loafing in training and competition. General themes investigated and subsequent content analysis highlighted factors that promote or reduce the extent of social loafing. Determinants of self-loafing were grouped according to three distinct categories: Group processes; Task characteristics and Individual perceptions. Social loafing among others was perceived to be prevalent in both the training and competition scenarios with more examples of social loafing provided in the training situation. The signs of social loafing were grouped according to the following themes: Cognitive & Emotional, Behavioural, Communication and Player Intuition. Consequences of social loafing were found to be detrimental to the effort and performance of both the individual and the team. Athletes were also able to differentiate between perceived social loafing and the perceived use of ‘strategic rest’ in team sports. Implications for the coaching process are far reaching with the need to develop an effective team culture and to provide training sessions that are interesting, engaging and relevant

    Performance over professional learning and the complexity puzzle: lesson observation in England’s further education sector

    Attempts to measure the quality of teaching and learning have resulted in an overreliance on quantitative performance data and the normalisation of a set of reductionist practices in England’s further education sector in recent years. Focusing on lesson observation as an illustrative example and drawing on data from a national study, this article examines the application of observation and its impact on further education teachers’ practice. In viewing lesson observation through a complexity theory lens and contextualising it against the wider neoliberal backdrop of the marketisation of education, we seek to critique the inadequacies of current reductionist approaches to teacher evaluation, whilst simultaneously opening up a debate regarding the consequences of seeing classrooms as complex adaptive systems. In focusing on performative models of lesson observation in particular, the article exposes what we perceive as some of the epistemological and methodological shortcomings of neoliberalism in practice, but also offers an alternative way forward in dealing with the contested practice of evaluating the quality of teaching and learning

    Rapid Identification of Fluorochrome Modification Sites in Proteins by LC ESI-Q-TOF Mass Spectrometry

    Conjugation of either a fluorescent dye or a drug molecule to the Δ-amino groups of lysine residues of proteins has many applications in biology and medicine. However, this type of conjugation produces a heterogeneous population of protein conjugates. Because conjugation of fluorochrome or drug molecule to a protein may have deleterious effects on protein function, the identification of conjugation sites is necessary. Unfortunately, the identification process can be time-consuming and laborious; therefore, there is a need to develop a rapid and reliable way to determine the conjugation sites of the fluorescent label or drug molecule. In this study, the sites of conjugation of fluorescein-5â€Č-isothiocyanate and rhodamine-B-isothiocyanate to free amino groups on the insert-domain (I-domain) protein derived from the α-subunit of lymphocyte function-associated antigen-1 (LFA-1) were determined by electrospray ionization quadrupole time-of-flight mass spectrometry (ESI-Q-TOF MS) along with peptide mapping using trypsin digestion. A reporter fragment of the fluorochrome moiety that is generated in the collision cell of the Q-TOF without explicit MS/MS precursor selection was used to identify the conjugation site. Selected ion plots of the reporter ion readily mark modified peptides in chromatograms of the complex digest. Interrogation of theses spectra reveals a neutral loss/precursor pair that identifies the modified peptide. The results show that one to seven fluorescein molecules or one to four rhodamine molecules were attached to the lysine residue(s) of the I-domain protein. No modifications were found in the metal ion-dependent adhesion site (MIDAS), which is an important binding region of the I-domain
