    Deformazioni indotte da flussi idrici sotterranei nel Carso Triestino

    Il Carso Classico \ue8 un vasto areale a carso maturo sviluppato in una potente idrostruttura, caratterizzata da intensa e diffusa carsifi cazione anche ben al di sotto del livello del mare. La zona satura \ue8 potente ed estesa con una zona di oscillazione che verso monte raggiunge il centinaio di metri di spessore durante le piene importanti, come risulta dagli strumenti posizionati nell\u2019Abisso di Trebiciano, circa 20 km in linea d\u2019aria dalle sorgenti. Fra l\u2019Abisso di Trebiciano e le foci, a circa 12 km da queste ultime, si apre la Grotta Gigante, all\u2019interno della quale, dal 1960, \ue8 presente una stazione geodetica per la registrazione delle deformazioni crostali. Si analizzano le deformazioni indotte dalle variazioni di livello delle acque di fondo del Carso confrontando i dati geodetici della Grotta Gigante con quelli idrologici dell\u2019Abisso di Trebiciano. Analizzando i dati con campionamento giornaliero, si riconosce una correlazione fra il segnale clinometrico e le piene; i due segnali hanno sfasamento temporale massimo di un giorno. Il segnale clinometrico \ue8 un transiente, per cui il clinometro torna alla posizione iniziale successivamente al passaggio della piena ed ha un\u2019orientazione caratteristica lungo la direttrice N150W-N30E. Il segnale clinometrico \ue8 proporzionale al livello raggiunto nell\u2019Abisso di Trebiciano (100 nrad di inclinazione corrispondono a 9.7 m di incremento di livello). Le piene nell\u2019Abisso di Trebiciano devono superare 23.2 m, per provocare il segnale clinometrico a Grotta Gigante

    The polo-like kinase 1 (PLK1) inhibitor NMS-P937 is effective in a new model of disseminated primary CD56+ acute monoblastic leukaemia

    CD56 is expressed in 15–20% of acute myeloid leukaemias (AML) and is associated with extramedullary diffusion, multidrug resistance and poor prognosis. We describe the establishment and characterisation of a novel disseminated model of AML (AML-NS8), generated by injection into mice of leukaemic blasts freshly isolated from a patient with an aggressive CD56+ monoblastic AML (M5a). The model reproduced typical manifestations of this leukaemia, including presence of extramedullary masses and central nervous system involvement, and the original phenotype, karyotype and genotype of leukaemic cells were retained in vivo. Recently Polo-Like Kinase 1 (PLK1) has emerged as a new candidate drug target in AML. We therefore tested our PLK1 inhibitor NMS-P937 in this model either in the engraftment or in the established disease settings. Both schedules showed good efficacy compared to standard therapies, with a significant increase in median survival time (MST) expecially in the established disease setting (MST = 28, 36, 62 days for vehicle, cytarabine and NMS-P937, respectively). Importantly, we could also demonstrate that NMS-P937 induced specific biomarker modulation in extramedullary tissues. This new in vivo model of CD56+ AML that recapitulates the human tumour lends support for the therapeutic use of PLK1 inhibitors in AML

    Observation of Large Topologically Trivial Fermi-Arcs in the Candidate Type-II Weyl Semimetal WTe2

    We report angle-resolved photoemission experiments resolving the distinct electronic structure of the inequivalent top and bottom (001) surfaces of WTe2. On both surfaces, we identify a surface state that forms a large Fermi-arc emerging out of the bulk electron pocket. Using surface electronic structure calculations, we show that these Fermi arcs are topologically trivial and that their existence is independent of the presence of type-II Weyl points in the bulk band structure. This implies that the observation of surface Fermi arcs alone does not allow the identification of WTe2 as a topological Weyl semimetal. We further use the identification of the two different surfaces to clarify the number of Fermi surface sheets in WTe2.Comment: Accepted in Physical Review B Rapid Communication on 16 Aug 201

    DNA damage in lens epithelial cells exposed to occupationally-relevant X-ray doses and role in cataract formation

    The current framework of radiological protection of occupational exposed medical workers reduced the eye-lens equivalent dose limit from 150 to 20 mSv per year requiring an accurate dosimetric evaluation and an increase understanding of radiation induced effects on Lens cells considering the typical scenario of occupational exposed medical operators. Indeed, it is widely accepted that genomic damage of Lens epithelial cells (LEC) is a key mechanism of cataractogenesis. However, the relationship between apoptosis and cataractogenesis is still controversial. In this study biological and physical data are combined to improve the understanding of radiation induced effects on LEC. To characterize the occupational exposure of medical workers during angiographic procedures an INNOVA 4100 (General Electric Healthcare) equipment was used (scenario A). Additional experiments were conducted using a research tube (scenario B). For both scenarios, the frequencies of binucleated cells, micronuclei, p21-positive cells were assessed with different doses and dose rates. A Monte-Carlo study was conducted using a model for the photon generation with the X-ray tubes and with the Petri dishes considering the two different scenarios (A and B) to reproduce the experimental conditions and validate the irradiation setups to the cells. The simulation results have been tallied using the Monte Carlo code MCNP6. The spectral characteristics of the different X-ray beams have been estimated. All irradiated samples showed frequencies of micronuclei and p21-positive cells higher than the unirradiated controls. Differences in frequencies increased with the delivered dose measured with Gafchromic films XR-RV3. The spectrum incident on eye lens and Petri, as estimated with MCNP6, was in good agreement in the scenario A (confirming the experimental setup), while the mean energy spectrum was higher in the scenario B. Nevertheless, the response of LEC seemed mainly related to the measured absorbed dose. No effects on viability were detected. Our results support the hypothesis that apoptosis is not responsible for cataract induced by low doses of X-ray (i.e. 25 mGy) while the induction of transient p21 may interfere with the disassembly of the nuclear envelop in differentiating LEC, leading to cataract formation. Further studies are needed to better clarify the relationship we suggested between DNA damage, transient p21 induction and the inability of LEC enucleation

    In situ strain tuning of the metal-insulator-transition of Ca2RuO4 in angle-resolved photoemission experiments

    Pressure plays a key role in the study of quantum materials. Its application in angle resolved photoemission (ARPES) studies, however, has so far been limited. Here, we report the evolution of the k-space electronic structure of bulk Ca2RuO4, lightly doped with Pr, under uniaxial strain. Using ultrathin plate-like crystals, we achieve uniaxial strain levels up to -4.1%, sufficient to suppress the insulating Mott phase and access the previously unexplored electronic structure of the metallic state at low temperature. ARPES experiments performed while tuning the uniaxial strain reveal that metallicity emerges from a marked redistribution of charge within the Ru t2g shell, accompanied by a sudden collapse of the spectral weight in the lower Hubbard band and the emergence of a well-defined Fermi surface which is devoid of pseudogaps. Our results highlight the profound roles of lattice energetics and of the multiorbital nature of Ca2RuO4 in this archetypal Mott transition and open new perspectives for spectroscopic measurements

    Quality of life gains in frail and intermediate-fit patients with multiple Myeloma:Findings from the prospective HOVON123 clinical trial

    Background: Frailty in newly-diagnosed multiple myeloma (NDMM) patients is associated with treatment-related toxicity, which negatively affects health-related quality of life (HRQoL). Currently, data on changes in HRQoL of frail and intermediate-fit MM patients during active treatment and post-treatment follow-up are absent. Methods: The HOVON123 study (NTR4244) was a phase II trial in which NDMM patients ≄ 75 years were treated with nine dose-adjusted cycles of Melphalan-Prednisone-Bortezomib (MPV). Two HRQoL instruments (EORTC QLQ-C30 and -MY20) were obtained before start of treatment, after 3 and 9 months of treatment and 6 and 12 months after treatment for patients who did not yet start second-line treatment. HRQoL changes and/or differences in frail and intermediate-fit patients (IMWG frailty score) were reported only when both statistically significant (p &lt; 0.005) and clinically relevant (&gt;MID). Results: 137 frail and 71 intermediate-fit patients were included in the analysis. Compliance was high and comparable in both groups. At baseline, frail patients reported lower global health status, lower physical functioning scores and more fatigue and pain compared to intermediate-fit patients. Both groups improved in global health status and future perspective; polyneuropathy complaints worsened over time. Frail patients improved over time in physical functioning, fatigue and pain. Improvement in global health status occurred earlier than in intermediate-fit patients. Conclusion: HRQoL improved during anti-myeloma treatment in both intermediate-fit and frail MM patients. In frail patients, improvement occurred faster and, in more domains, which was retained during follow-up. This implies that physicians should not withhold safe and effective therapies from frail patients in fear of HRQoL deterioration.</p

    Palaeogenomic analysis of black rat (Rattus rattus) reveals multiple European introductions associated with human economic history

    The distribution of the black rat (Rattus rattus) has been heavily influenced by its association with humans. The dispersal history of this non-native commensal rodent across Europe, however, remains poorly understood, and different introductions may have occurred during the Roman and medieval periods. Here, in order to reconstruct the population history of European black rats, we generated a de novo genome assembly of the black rat, 67 ancient black rat mitogenomes and 36 ancient nuclear genomes from sites spanning the 1st-17th centuries CE in Europe and North Africa. Analyses of mitochondrial DNA confirm that black rats were introduced into the Mediterranean and Europe from Southwest Asia. Genomic analyses of the ancient rats reveal a population turnover in temperate Europe between the 6th and 10th centuries CE, coincident with an archaeologically attested decline in the black rat population.     Joint EUCAR/JRC/CONCAWE Study on: Effects of Gasoline Vapour Pressure and Ethanol Content on Evaporative Emissions from Modern Cars

    A test programme designed to investigate the influence of gasoline vapour pressure and ethanol content on evaporative emissions from modern passenger cars has been carried out by the Joint Research Centre of the European Commission jointly with CONCAWE and EUCAR. Seven gasoline passenger cars representative of current EURO 3/4 emissions technology were tested for evaporative emissions with ten different test fuels. The test fuel matrix comprised 60 and 70 kPa hydrocarbon base fuels with 5 and 10% ethanol splash blends and 5 and 10% ethanol matched volatility blends. The evaporative emission tests were carried out according to a test protocol based on the European homologation test procedure, with no additional vehicle conditioning. Although this test protocol turned out to have a considerable influence on the results, the programme has provided valuable information and several clear conclusions can be drawn. The programme confirmed that vapour pressure (DVPE) is a key fuel variable for evaporative emissions. However the effect of vapour pressure is strongly non-linear; the ethanol blends with final DVPE around 75 kPa gave considerably higher evaporative emissions than the lower volatility fuels in most of the vehicles. Differences between fuels with DVPE in the range 60-70 kPa were small. Additional tests on two vehicles performed after the main programme have raised some questions about possible effects of ethanol on carbon canister working capacity and on the role of permeation in determining evaporative emissions.JRC.H.4-Transport and air qualit
