    The historical background of coal conversion is reviewed and the programmatic status (operational, construction, design, proposed) of coal gasification processes is tabulated for both commercial and demonstration projects as well as for large and small pilot plants. Both second and third generation processes typically operate at higher temperatures and pressures than first generation methods. Much of the equipment that has been tested has failed. The most difficult problems are in process control. The mechanics of three-phase flow are not fully understood. Companies participating in coal conversion projects are ordering duplicates of failure prone units. No real solutions to any of the significant problems in technology development have been developed in recent years

    Detection of equine atypical myopathy-associated hypoglycin A in plant material: Optimisation and validation of a novel LC-MS based method without derivatisation

    Hypoglycin A (HGA) toxicity, following ingestion of material from certain plants, is linked to an acquired multiple acyl-CoA dehydrogenase deficiency known as atypical myopathy, a commonly fatal form of equine rhabdomyolysis seen worldwide. Whilst some plants are known to contain this toxin, little is known about its function or the mechanisms that lead to varied HGA concentrations between plants. Consequently, reliable tools to detect this amino acid in plant samples are needed. Analytical methods for HGA detection have previously been validated for the food industry, however, these techniques rely on chemical derivatisation to obtain accurate results at low HGA concentrations. In this work, we describe and validate a novel method, without need for chemical derivatisation (accuracy = 84–94%; precision = 3–16%; reproducibility = 3–6%; mean linear range R2 = 0.999). The current limit of quantitation for HGA in plant material was halved (from 1μg/g in previous studies) to 0.5μg/g. The method was tested in Acer pseudoplatanus material and other tree and plant species. We confirm that A. pseudoplatanus is most likely the only source of HGA in trees found within European pastures

    Phylogeny of Rosellinia capetribulensis sp. nov. and its allies (Xylariaceae)

    A new Rosellinia species, R. capetribulensis isolated from Calamus sp. in Australia is described. R. capetribulensis is characterized by perithecia immersed within a carbonaceous stroma surrounded by subiculum-like hyphae, asci with large, barrel-shaped amyloid apical apparatus and large dark brown spores. Morphologically, R. capetribulensis appears to be similar to R. bunodes, R. markhamiae and R. megalospora. To gain further insights into the phylogeny of this new taxon we analyzed the ITS-5.8S rDNA using maximum parsimony and likelihood methods. In addition, a morphological dataset also was analyzed phylogenetically to investigate possible affinities. ITS rDNA based phylogenies reveal that R. capetribulensis is closely related to other Rosellinia species showing closest affinity to R. arcuata, R. necatrix and R. pepo. However, analysis of R. capetribulensis forms an unsupported branch sister to these taxa. Results from the morphological matrix indicate a close morphological affinity to members of Rosellinia subgenus Rosellinia. Despite that ITS rDNA and morphological analyses present difficulties in constructing a proper phylogenetic framework among Rosellinia and allied genera, there is sufficient evidence to support the establishment of the new taxon in the genus Rosellinia. The morphological similarities and differences between R. capetribulensis and allied genera such as Astrocystis and Entoleuca are also briefly discussed.published_or_final_versio

    STS-114 Micrometeoroid/Orbital Debris (MMOD) Post-Flight Assessment

    NASA Johnson Space Center (JSC) personnel assisted Kennedy Space Center (KSC) inspection teams in the identification of 41 micrometeoroid/orbital debris (MMOD) impact sites on the OV-103 vehicle (Discovery) during STS-114 postflight inspections. There were 14 MMOD impacts reported on the crew module windows (Figure 1). The largest impact feature, a 6.6 mm x 5.8 mm crater on window #4, was caused by a particle with an estimated diameter of 0.22 mm (Figure 2). This impact was among the largest ever recorded on a crew module window. The window was removed and replaced. Scanning Electron Microscope/Energy Dispersive X-ray (SEM/EDX) analysis of dental mold samples from the impact site to determine particle origin was inconclusive, possibly due to contamination picked up on the ferry flight from Edwards Air Force Base to KSC. The radiators on the inside of the payload bay doors sustained 19 impacts (Figure 3) with one of the impacts causing a face sheet perforation. The 0.61 mm diameter hole was produced by a particle with an estimated diameter of 0.4 mm, which approaches the 0.5-mm critical particle diameter of the wing leading edge reinforced carbon-carbon (RCC) panel high-temperature regions (Zone 3, Figure 4) that was established during Return to Flight testing of the RCC panels. An inspection of the payload bay door exterior insulation (FRSI) revealed a 5.8 mm x 4.5 mm defect that was caused by an MMOD particle with unknown composition, as the sample obtained was contaminated. Figure 5 provides a summary of the exterior surface survey that was conducted following the STS-114 mission. Two windows were removed and replaced due to hypervelocity impact. Nineteen impacts were recorded on the payload bay door radiators, with one face sheet penetration. Three impact sites were identified on the FRSI. There were four hypervelocity impact sites detected on the wing leading edge RCC panels. One impact was detected on the top cover of the TPS sample box (TSB) payload that was mounted on a carrier in the aft portion of the payload bay

    In vivo electroporation of morpholinos into the adult zebrafish retina.

    Many devastating inherited eye diseases result in progressive and irreversible blindness because humans cannot regenerate dying or diseased retinal neurons. In contrast, the adult zebrafish retina possesses the robust ability to spontaneously regenerate any neuronal class that is lost in a variety of different retinal damage models, including retinal puncture, chemical ablation, concentrated high temperature, and intense light treatment. Our lab extensively characterized regeneration of photoreceptors following constant intense light treatment and inner retinal neurons after intravitreal ouabain injection. In all cases, resident Müller glia re-enter the cell cycle to produce neuronal progenitors, which continue to proliferate and migrate to the proper retinal layer, where they differentiate into the deficient neurons. We characterized five different stages during regeneration of the light-damaged retina that were highlighted by specific cellular responses. We identified several differentially expressed genes at each stage of retinal regeneration by mRNA microarray analysis. Many of these genes are also critical for ocular development. To test the role of each candidate gene/protein during retinal regeneration, we needed to develop a method to conditionally limit the expression of a candidate protein only at times during regeneration of the adult retina. Morpholino oligos are widely used to study loss of function of specific proteins during the development of zebrafish, Xenopus, chick, mouse, and tumors in human xenografts. These modified oligos basepair with complementary RNA sequence to either block the splicing or translation of the target RNA. Morpholinos are stable in the cell and can eliminate or knockdown protein expression for three to five days. Here, we describe a method to efficiently knockdown target protein expression in the adult zebrafish retina. This method employs lissamine-tagged antisense morpholinos that are injected into the vitreous of the adult zebrafish eye. Using electrode forceps, the morpholino is then electroporated into all the cell types of the dorsal and central retina. Lissamine provides the charge on the morpholino for electroporation and can be visualized to assess the presence of the morpholino in the retinal cells. Conditional knockdown in the retina can be used to examine the role of specific proteins at different times during regeneration. Additionally, this approach can be used to study the role of specific proteins in the undamaged retina, in such processes as visual transduction and visual processing in second order neurons

This is because it is only possible to obtain small samples of material in some situations, for example, the scoop samples that are removed from in-service components, the heat-affected zones that are created when welds are used to join components and the desire to produce only small amounts of material in alloy development programmes. It is therefore important to review and compare the theoretical basis and practical aspects of each of the small specimen creep testing methods, in order to clearly understand which of the methods is the best for any specific application.     Southern high-latitude Digisonde observations of ionosphere E-region Bragg scatter during intense lacuna conditions

    International audienceDuring summer months at solar cycle minimum, F-region lacuna and slant-Es conditions (SEC) are common features of daytime ionograms recorded around local magnetic noon at Casey, Antarctica. Digisonde measurements of drift velocity height profiles show that the occurrence of lacuna prevents the determination of F-region drift velocities and also affects E-region drift velocity measurements. Unique E-region spectral features revealed as intervals of Bragg scatter superimposed on typical background E-region reflection were observed in Digisonde Doppler spectra during intense lacuna conditions. Daytime E-region Doppler spectra recorded at carrier frequencies from 1.5 to 2.7MHz, below the E-region critical frequency foE, have two side-peaks corresponding to Bragg scatter at approximately ±1-2Hz symmetrically located on each side of a central-peak corresponding to near-zenith total reflections. Angle-of-arrival information and ray-tracing simulations show that echo returns are coming from oblique directions most likely resulting from direct backscatter from just below the total reflection height for each sounding frequency. The Bragg backscatter events are shown to manifest during polar lacuna conditions, and to affect the determination of E-region background drift velocities, and as such must be considered when using standard Doppler-sorted interferometry (DSI) techniques to estimate ionospheric drift velocities. Given the Doppler and spatial separation of the echoes determined from high-resolution Doppler measurements, we are able to estimate the Bragg scatter phase velocity independently from the bulk E-region motion. The phase velocity coincides with the ExB direction derived from in situ fluxgate magnetometer records. When ionospheric refraction is considered, the phase velocity amplitudes deduced from DSI are comparable to the ion-acoustic speed expected in the E-region. We briefly consider the plausibility that these previously unreported polar cap E-region Bragg scatter Doppler spectral signatures, observed at Casey in December 1996 during SEC/lacuna conditions may be linked to ionosphere irregularities. These irregularities may possibly be generated by primary plasma waves triggered by current-driven instabilities, that is to say, a hybrid of the "modified two-stream" and "gradient drift" instability mechanisms
