    Numerical investigation of the conditioning for plane wave discontinuous Galerkin methods

    We present a numerical study to investigate the conditioning of the plane wave discontinuous Galerkin discretization of the Helmholtz problem. We provide empirical evidence that the spectral condition number of the plane wave basis on a single element depends algebraically on the mesh size and the wave number, and exponentially on the number of plane wave directions; we also test its dependence on the element shape. We show that the conditioning of the global system can be improved by orthogonalization of the local basis functions with the modified Gram-Schmidt algorithm, which results in significantly fewer GMRES iterations for solving the discrete problem iteratively.Comment: Submitted as a conference proceeding; minor revisio

    Erosion-oxidation of pressure vessel steel P265GH

    The behaviour of pressure vessel steel P265GH was studied in a centrifugal high-temperature erosion apparatus under impacts by silica (SiO2) particles moving at velocities ranging from 20 to 60 m s-1 and contacting the surface at the angles of 30° and 90°. Besides particle impacts, the steel was simultaneously exposed to air and elevated temperatures of 350 and 450°C. For comparison, the tests were also performed in the absence of erodent particles. After the tests, the material behaviour was evaluated in terms of occurred weight changes and surface characteristics, the latter ones of which were investigated by using, for example, scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS). In the analysis of the test results, special attention was paid to the composition and microstructure of the used silica particles and how these influence the elemental distribution on the exposed surfaces. The results show that the particle impacts introduce weight losses that follow a ductile angle-dependency, i.e., relatively greater weight losses at the shallow than at the steep impact angle. Although evident oxide scales developed on the surfaces at the test temperatures, they did not provide the steel with protection against particle impacts. Particle debris was detected embedded in the surfaces particularly under impacts at 90°, with softer particle constituents being preferentially deposited. These results are discussed in terms of the erosion-oxidation behaviour of the steel and the consequences of the heterogeneous erodent particle quality

    On stability of discretizations of the Helmholtz equation (extended version)

    We review the stability properties of several discretizations of the Helmholtz equation at large wavenumbers. For a model problem in a polygon, a complete kk-explicit stability (including kk-explicit stability of the continuous problem) and convergence theory for high order finite element methods is developed. In particular, quasi-optimality is shown for a fixed number of degrees of freedom per wavelength if the mesh size hh and the approximation order pp are selected such that kh/pkh/p is sufficiently small and p=O(logk)p = O(\log k), and, additionally, appropriate mesh refinement is used near the vertices. We also review the stability properties of two classes of numerical schemes that use piecewise solutions of the homogeneous Helmholtz equation, namely, Least Squares methods and Discontinuous Galerkin (DG) methods.     Spectral Vector Beams for High-Speed Spectroscopic Measurements

    Structured light harnessing multiple degrees of freedom has become a powerful approach to use complex states of light in fundamental studies and applications. Here, we investigate the light field of an ultrafast laser beam with a wavelength-depended polarization state, a beam we term spectral vector beam. We demonstrate a simple technique to generate and tune such structured beams and demonstrate their spectroscopic capabilities. By only measuring the polarization state using fast photodetectors, it is possible to track pulse-to-pulse changes in the frequency spectrum caused by, e.g. narrowband transmission or absorption. In our experiments, we reach read-out rates of around 6 MHz, which is limited by our technical ability to modulate the spectrum and can in principle reach GHz read-out rates. In simulations we extend the spectral range to more than 1000 nm by using a supercontinuum light source, thereby paving the way to various applications requiring high-speed spectroscopic measurements.Comment: 11 pages, 12 figure

    Long-term clinical and economic outcomes in previously untreated paediatric patients with severe haemophilia A : A nationwide real-world study with 700 person-years

    AimFor previously untreated patients (PUPs) with severe haemophilia A in Finland for the past 2 decades, the standard practice has been to start early primary prophylaxis. We evaluated the long-term clinical outcomes and costs of treatment with high-dose prophylaxis in PUPs from birth to adolescence, including immune tolerance induction (ITI). MethodsFrom the medical records of all PUPs born between June 1994 and May 2013 in Finland, we retrospectively extracted data on clinical outcomes and healthcare use. Using linear mixed models, we analysed longitudinal clinical outcome data. To analyse skewed cost data, including zero costs, we applied hurdle regression. ResultsAll 62 patients received early regular prophylaxis; totally, they have had treatment for nearly 700 patient-years. The median age of starting home treatment was 1.1years. The mean (SD) annual treatment costs (Europerkg) were 4391Euro (3852). For ages 1-3, ITI comprised over half of the costs; in other groups, prophylactic FVIII treatment dominated. With these high costs, however, clinical outcomes were desirable; median (IQR) ABR was low at 0.19 (0.07-0.46) and so was AJBR at 0.06 (0-0.24). Thirteen (21%) patients developed a clinically significant inhibitor, 10 (16%) with a high titre. All ITIs were successful. The mean costs for ITI were 383448Euro (259085). The expected ITI payback period was 1.81 (95% CI 0.62-12.12) years. ConclusionsEarly high-dose prophylaxis leads to excellent long-term clinical outcomes, and early childhood ITI therapy seems to turn cost-neutral generally already in 2years.Peer reviewe

    Hilbert-Huang versus Morlet wavelet transformation on mismatch negativity of children in uninterrupted sound paradigm

    Background. Compared to the waveform or spectrum analysis of event-related potentials (ERPs), time-frequency representation (TFR) has the advantage of revealing the ERPs time and frequency domain information simultaneously. As the human brain could be modeled as a complicated nonlinear system, it is interesting from the view of psychological knowledge to study the performance of the nonlinear and linear time-frequency representation methods for ERP research. In this study Hilbert-Huang transformation (HHT) and Morlet wavelet transformation (MWT) were performed on mismatch negativity (MMN) of children. Participants were 102 children aged 8–16 years. MMN was elicited in a passive oddball paradigm with duration deviants. The stimuli consisted of an uninterrupted sound including two alternating 100 ms tones (600 and 800 Hz) with infrequent 50 ms or 30 ms 600 Hz deviant tones. In theory larger deviant should elicit larger MMN. This theoretical expectation is used as a criterion to test two TFR methods in this study. For statistical analysis MMN support to absence ratio (SAR) could be utilized to qualify TFR of MMN. Results. Compared to MWT, the TFR of MMN with HHT was much sharper, sparser, and clearer. Statistically, SAR showed significant difference between the MMNs elicited by two deviants with HHT but not with MWT, and the larger deviant elicited MMN with larger SAR. Conclusion. Support to absence ratio of Hilbert-Huang Transformation on mismatch negativity meets the theoretical expectations, i.e., the more deviant stimulus elicits larger MMN. However, Morlet wavelet transformation does not reveal that. Thus, HHT seems more appropriate in analyzing event-related potentials in the time-frequency domain. HHT appears to evaluate ERPs more accurately and provide theoretically valid information of the brain responses.peerReviewe

    Fatal Outcome in Bacteremia is Characterized by High Plasma Cell Free DNA Concentration and Apoptotic DNA Fragmentation: A Prospective Cohort Study

    INTRODUCTION: Recent studies have shown that apoptosis plays a critical role in the pathogenesis of sepsis. High plasma cell free DNA (cf-DNA) concentrations have been shown to be associated with sepsis outcome. The origin of cf-DNA is unclear. METHODS: Total plasma cf-DNA was quantified directly in plasma and the amplifiable cf-DNA assessed using quantitative PCR in 132 patients with bacteremia caused by Staphylococcus aureus, Streptococcus pneumoniae, ß-hemolytic streptococcae or Escherichia coli. The quality of cf-DNA was analyzed with a DNA Chip assay performed on 8 survivors and 8 nonsurvivors. Values were measured on days 1-4 after positive blood culture, on day 5-17 and on recovery. RESULTS: The maximum cf-DNA values on days 1-4 (n = 132) were markedly higher in nonsurvivors compared to survivors (2.03 vs 1.26 ug/ml, p<0.001) and the AUCROC in the prediction of case fatality was 0.81 (95% CI 0.69-0.94). cf-DNA at a cut-off level of 1.52 ug/ml showed 83% sensitivity and 79% specificity for fatal disease. High cf-DNA (>1.52 ug/ml) remained an independent risk factor for case fatality in a logistic regression model. Qualitative analysis of cf-DNA showed that cf-DNA displayed a predominating low-molecular-weight cf-DNA band (150-200 bp) in nonsurvivors, corresponding to the size of the apoptotic nucleosomal DNA. cf-DNA concentration showed a significant positive correlation with visually graded apoptotic band intensity (R = 0.822, p<0.001). CONCLUSIONS: Plasma cf-DNA concentration proved to be a specific independent prognostic biomarker in bacteremia. cf-DNA displayed a predominating low-molecular-weight cf-DNA band in nonsurvivors corresponding to the size of apoptotic nucleosomal DNA

    Evaluating the Assumptions of Surface Reflectance and Aerosol Type Selection Within the MODIS Aerosol Retrieval Over Land: The Problem of Dust Type Selection

    Aerosol Optical Depth (AOD) and Angstrom exponent (AE) values derived with the MODIS retrieval algorithm over land (Collection 5) are compared with ground based sun photometer measurements at eleven sites spanning the globe. Although, in general, total AOD compares well at these sites (R2 values generally over 0.8), there are cases (from 2 to 67% of the measurements depending on the site) where MODIS clearly retrieves the wrong spectral dependence, and hence, an unrealistic AE value. Some of these poor AE retrievals are due to the aerosol signal being too small (total AOD<0.3) but in other cases the AOD should have been high enough to derive accurate AE. However, in these cases, MODIS indicates AE values close to 0.6 and zero fine model weighting (FMW), i.e. dust model provides the best fitting to the MODIS observed reflectance. Yet, according to evidence from the collocated sun photometer measurements and back-trajectory analyses, there should be no dust present. This indicates that the assumptions about aerosol model and surface properties made by the MODIS algorithm may have been incorrect. Here we focus on problems related to parameterization of the land-surface optical properties in the algorithm, in particular the relationship between the surface reflectance at 660 and 2130 nm

    Peptic Fluorescent "Signal-On" and "Signal-Off" Sensors Utilized for the Detection Protein Post-Translational Modifications

    Protein post-translational modifications (PTMs) are typically enzyme-catalyzed events generating functional diversification of proteome; thus, multiple PTM enzymes have been validated as potential drug targets. We have previously introduced energy-transfer-based signal-modulation method called quenching resonance energy transfer (QRET), and utilize it to monitor PTM addition or removal using the developed peptide-break technology. Now we have reinvented the QRET technology, and as a model, we introduced the tunable fluorescent "signal-on" and "signal-off" detection scheme in the peptide-break PTM detection. Taking the advantage of time-resolved fluorescence-based single-label detection technology, we were able to select the signal direction upon PTM addition or removal by simply introducing different soluble Eu3+-signal-modulating molecule. This enables the selection of positive signal change upon measurable event, without any additional labeling steps, changes in assay condition or Eu3+-reporter. The concept functionality was demonstrated with four Eu3+-signal modulators in a high-throughput compatible kinase and phosphatase assays using signal-on and signal-off readout at 615 nm or time-resolved Forster resonance energy transfer at 665 nm. Our data suggest that the introduced signal modulation methodology provides a transitional fluorescence-based single-label detection concept not limited only to PTM detection
