    GSK-3 protein and the heart: friend or foe?

    Metabolic syndrome manifesting as obesity, insulin resistance and type 2 diabetes mellitus is currently pandemic. Each of these, in its own right, is strongly related to the development of cardiovascular disease. The cardiomyopathy associated with these disorders is characterised by curtailed glucose uptake and utilisation, elevated risk of damage after ischaemia and contractile dysfunction. Current research have indicated that the serine/threonine kinase, glycogen synthase kinase 3 (GSK-3), may play a central role in the development of all these dysfunctions. The development of new generations of inhibitors of this kinase, has renewed interest in its utilisation as therapeutic target. This review has therefore focused on the role of GSK-3 in the development of the obesity-related cardiomyopathy and has highlighted and discussed the detrimental as well as beneficial effects of the GSK-3 inhibitors that are currently available. We have discussed the different roleplayers such as the insulin signalling pathway, modulation of apoptosis and mitochondrial function, SERCA2 expression and regulation of the development of hypertrophy in the context of GSK-3 activity

There is also strong evidence that thefinancial problems that stem from mismanagement are one of the causal factors of the highlevels of pilfering, bribery, corruption and theft in the country’s public and private sectors(ACRC, 2009). In spite of this, very little is being done to address this deficiency. There isespecially a dire need that employers should empower their workforce with appropriatefinancial management skills. South African practitioners cannot, however, yet prove beyond a doubt that these services meet the needs of client systems. Especially one gap still remains on the accountability continuum. This is the extent to which clients themselves are satisfied with the services they receive. Part of this gap has now been filled with the development and testing of client satisfaction scales for an occupational setting.     Signalling pathways activated by glucagon-like peptide-1 (7-36) amide in the rat heart and their role in protection against ischaemia

    The original publication is available at http://www.cvja.co.za/Glucagon-like peptide-1 is an incretin hormone proposed to have insulinomimetic effects on peripheral insulin-sensitive tissue. We examined these effects on the heart by using isolated, perfused rat hearts and adult ventricular myocytes. During normoxic perfusion, no effects of escalating concentrations of GLP-1 on either heart rate or left ventricular developed pressure were found. With functional performance as readout, we found that GLP-1 directly protected the heart against damage incurred by global low-flow ischaemia. This protection was sensitive to the presence of iodo-acetate, implicating activation of glycolysis, and was abolished by wortmannin, indicative of PI-3-kinase as mediator of protection. I n addition, GLP-1 had an infarct-sparing effect when supported by the presence of the dipeptidyl peptidase-IV inhibitor valine pyrrolidide. GLP-1 could not directly activate protein kinase B (also called Akt) or the extracellular regulated kinases Erk1/2 in hearts or cardiocytes under normoxic conditions, but phosphorylation of the AMP-activated kinase (AMPK) on Thr172 was enhanced. I n addition, the glycolytic enzyme phosphofructokinase- 2 was activated dose dependently. During reperfusion after ischaemia, modulation of the phosphorylation of PKB/Akt as well as AMPK was evident. GLP-1 therefore directly protected the heart against low-flow ischaemia by enhancing glycolysis, probably via activation of AMP kinase and by modulating the profile of activation of the survival kinase PKB/Akt.Publisher's versio

    Contaminant mobilisation by fluid-rock interaction and related transport mechanisms in platinum tailings

    Contaminant release and transport in platinum tailings are poorly studied in literature. This study serves to characterise these processes. The tailings facility is located in Steelpoort, Mpumalanga, South Africa on Critical Zone rocks of the Rustenburg Layered Suite in The Bushveld Igneous Complex. Tailings material samples were collected by hand auger- and direct push probe drilling at specific locations to represent the different materials present in the tailings facility. Water samples were collected from monitoring boreholes as well as the Steelpoort River. The samples were analysed using XRD, XRF, ABA, NAG, Reflected Light Microscopy, Acid Leaching Tests and ICP scans. Using the collected data, a geochemical model was constructed for the interpretation of mineral phase dissolution and to trace the mineral phases releasing contaminants. Analysis-, test- and modelling results showed that alteration mineral phases formed within ten years in the tailings material and that the existing alteration phases viz. talc and chlorite, as well as sulphides, are the major contributors of contaminants. Elevated pH values as well as major cation and anion concentrations were found in the fluid discharging from the tailings as well as the in groundwater, with little to no heavy metals, which were traced directly to the chromite phase. This suggests that platinum tailings do not contribute to heavy metal contamination or acid rock drainage but may increase aquifer salinity and alkalinity. The flow through the tailings, underlying vadose zone and fractured rock aquifer was characterised using permeameter- and pumping tests. From the data collected, an unsaturated flow model was developed to characterise the flow through the tailings. From the model, discharge from the tailings was calculated to take place at 0.7m per decade into the underlying vadose zone with fracture flow in the aquifer ranging from 0.46-0.026m/d, as calculated from pumping test results. Contaminant migration into the Steelpoort River is possibly inhibited by the Dwarsriver Fault, based on the chemical data and hydraulic conductivities calculated.     Cardioprotective and anti-hypertensive effects of Prosopis glandulosa in rat models of pre-diabetes

    AIM: Obesity and type 2 diabetes present with two debilitating complications, namely, hypertension and heart disease. The dried and ground pods of Prosopis glandulosa (commonly known as the Honey mesquite tree) which is part of the Fabaceae (or legume) family are currently marketed in South Africa as a food supplement with blood glucose-stabilising and anti-hypertensive properties. We previously determined its hypoglycaemic effects, and in the current study we determined the efficacy of P glandulosa as anti-hypertensive agent and its myocardial protective ability. METHODS: Male Wistar rats were rendered either pre-diabetic (diet-induced obesity: DIO) or hypertensive (high-fat diet: HFD). DIO animals were treated with P glandulosa (100 mg/kg/day for the last eight weeks of a 16-week period) and compared to age-matched controls. Hearts were perfused ex vivo to determine infarct size. Biometric parameters were determined at the time of sacrifice. Cardiac-specific insulin receptor knock-out (CIRKO) mice were similarly treated with P glandulosa and infarct size was determined. HFD animals were treated with P glandulosa from the onset of the diet or from weeks 12–16, using captopril (50 mg/kg/day) as the positive control. Blood pressure was monitored weekly. RESULTS: DIO rats and CIRKO mice: P glandulosa ingestion significantly reduced infarct size after ischaemia–reperfusion. Proteins of the PI-3-kinase/PKB/Akt survival pathway were affected in a manner supporting cardioprotection. HFD model: P glandulosa treatment both prevented and corrected the development of hypertension, which was also reflected in alleviation of water retention. CONCLUSION: P glandulosa was cardioprotective and infarct sparing as well as anti-hypertensive without affecting the body weight or the intra-peritoneal fat depots of the animals. Changes in the PI-3-kinase/PKB/Akt pathway may be causal to protection. Results indicated water retention, possibly coupled to vasoconstriction in the HFD animals, while ingestion of P glandulosa alleviated both. We concluded that treatment of pre-diabetes, type 2 diabetes or hypertension with P glandulosa poses possible beneficial health effects.Department of HE and Training approved lis

    Recolonisation of the Robberg Peninsula by the Cape Fur Seal Arctocephalus pusillus pusillus and its prey preferences

    The Cape fur seal Arctocephalus pusillus pusillus colony at the Robberg Peninsula, Plettenberg Bay, on the south-east coast of South Africa, was driven to extirpation by indiscriminate harvesting by the late 1800s and seals only began to recolonise this site in the 1990s. This study describes the recolonisation process from 2000 to 2009, exploring within- and between-year variation in the number of seals using the site. Numbers increased over the study period from less than 300 animals to over 3 100. Year and month were important in explaining variability in seal counts, whereas sea condition, time of day and lunar phase had minimal explanatory power. Within-year variation in seal counts decreased during the study period, which may indicate an increasing proportion of resident (as opposed to transient) seals in the colony. However, the colony is currently still in a transition phase with a low ratio of breeding to non-breeding animals and low numbers of pups born on the colony (currently still < 100 per year). The influx of seals to the Robberg area may be associated with an increase in prey availability in the area. The relative protection afforded by the Nature Reserve status of the Robberg Peninsula and the existence of a Marine Protected Area adjacent to it are likely to contribute to the growth of this colony. However, human interference associated with fishing and/or ecotourism on the Peninsula may prevent the colony from developing into a breeding colony. Faecal (scat) sampling was employed to study the diet of this increasing seal population at Robberg. Species composition and size of prey were determined, temporal variation in the diet was explored, and the potential for competition between seals and the fisheries around Plettenberg Bay was investigated. Of the 445 scats collected, 90 % contained hard prey remains. These comprised of 3 127 identified otoliths representing 15 teleost prey species, 25 cephalopod beaks representing three 6 species and three feathers representing two bird species. The seals' most important prey species in terms of numerical abundance and frequency of occurrence in the diet were anchovy, sardine, horse mackerel, sand tongue-fish and shallow-water hake (in decreasing order of importance). The proportion of anchovy in the diet increased during the study period, while sardine decreased. Sardine was the only species that increased significantly in the diet during the upwelling season. Little evidence was found of direct competition between seals and linefisheries in Plettenberg Bay, both in terms of prey species composition and quantities consumed. Scat sampling in seals holds promise as a method to track long-term changes in prey species availability. The conservation and management of this colony are discussed in light of the research findings

Aangesien die geskiedenis en die f undering van die kerklied deur ander navorsers reeds volledig uitgewerk is, is hoofstuk 1 bloot 'n inleidende oorsig wat die agtergrond van die studie verskaf. In hoofstuk 2 is 'n kategorisering van die Gesange in extenso gedoen en in hoofstuk 3 is die inhoud van die kategoriee kortliks beskryf. Die doel hiermee is om vas te stel oor watter onderwerpe die meeste gesing word en waar die lakunes voorkom. Aan die hand van hierdie kategorisering is daar uiteindelik vyf onderwerpe afgebaken wat ooglopend Bybelsteologiese leemtes of selfs afwykings vertoon wat in die toekoms aangespreek behoort te word. Die onderwerpe is onderskeidelik: die Godsbeskouing, die skepping, die sonde, die doop en die toekomsverwagting. In hoof stuk 4 het hierdie onderwerpe af sonderlik ender die soeklig gekom. Daar is o.a. gevind dat die inhoud van die Gesange 'n duidelike reflektering is van die tradisionele kerkleer en dus nie volkome daartoe in staat is om die moderne mens (die jeug veral!) aan te spreek nie. Die gevolgtrekking waartoe die navorser geraak het, is dat liedere wat die eietyds geloofsproblematiek aanspreek 'n sine qua non geword het. Die navorsing oor die inhoud van die Gesange het voorts ook vreemde teologiese aksente uitgewys. So bv. is daar 'n duidelike invloed van die Grieks platonistiese dualisme te bespeur waar die Jenseitige teenoor die Diesseitige af gespeel word. Di t het veral aan die lig gekom met die bespreking oor die skepping. Eodem modo is daar spore van die Pietistiese individualisme in die Gesange aangetref waar die vertikale belewing met God (bekering, heiliging) die horisontale belewing van verantwoordelikheid, oorskadu! Hierdie sending, aanbevelings die godsdiens omgewingsbewaring) is in hoof stuk krities-waarderende opmerkings saamgevat.Research into the theological content of the Evangelical Hymns was initiated after it was established that such a study had not been attempted in South Africa in Reformed circles. Since the history and grounding of the church hymn have already been discussed in full by other researchers, chapter 1 merely offers an introductory review which provides some background to the study. In chapter 2 a categorisation of the Hymns was done in extenso and chapter 3, in turn, deals with the contents of each specific category. The aim is to determine which subjects are prevalent in Hymns and which ones are not addressed at all. On the basis of this categorisation, five subjects were finally defined which revealed obvious biblical-theological lacunae, if not deviations, all of which need to be addressed in the future. The subjects are: the ways of thinking about God, the creation, sin, baptism and hopes for the future. In chapter 4 these subjects were examined in detail. It was proved, inter alia, that the contents of the Hymns clearly reflect the traditional church doctrine and consequently fail to a large extent to address modern man (especially the young people of our time). The conclusion reached by the researcher is that Hymns addressing the contemporary problematic nature of faith have become a sine qua non. Furthermore, research on the contents of the Hymns also revealed foreign theological accents such as clear influences of the Greek Platonistic dualism where the Jenseitige is compared to the Diesseitige. This became particularly evident in the discussion on the creation. Eodem modo traces of the Pietistic indiviudalism were detected in the Hymns, where the vertical experience of God (conversion, sanctification) completely overshadows the horizontal experience of religion (social responsibility, mission work, nature conservation)! Chapter 5 comprises these recommendations in the form of remarks of a criticalappreciation nature.Eine Untersuchung in Bezug auf den theologischen Inhalt der Evangelischen Kirchenlieder wurde in die Wege geleitet nachdem festgestellt wurde, daB ein solches Studium noch nie vorhin in Ref ormierten Kreisen in Slidaf rika vorgenommen worden war. Da die Geschichte und die Fundierung des Kirchenliedes von andern Forschern schon in all en Einzelheiten ausgearbeitet worden sind, bietet Kapitel 1 bloB eine einleitende Ubersicht als Hintergrund flir das Studium. In Kapitel 2 wurden die Kirchenlieder in extenso nach Kategorien geordnet und in Kapitel 3 findet sich der Inhalt jeder einzelnen Kategorie vor. Es sollte festgestellt werden, welche Themen sich am meisten in den Kirchenliedern erkennen lassen und welche liberhaupt nicht berlihrt werden. An Hand dieser Anordnung in Kategorien wurden zum Schluss flinf Themen definiert, die augenfiillig biblisch-theologische Lucken, wenn nicht sogar Abweichungen, offenbaren; es sind Themen, denen in der Zukunft zweifellos einem gerechten Platz Gottesauffassung, die zukommen sollen. Die themen sind: Schopfung, Siinde, die Taufe und Zukunftserwartungen. In Kapitel 4 wurden diese Themen in allen Einzelheiten untersucht. Es stellte sich unter anderern heraus, das der Inhalt der Kirchenlieder eine klare Widerspiegelung der traditionellen Kirchenlehre ist und deshalb den Ansprilchen des modernen Menschen (vor allern der Jugend) nicht vollig gerecht werden konnen. Die schluflfolgerung des Forschers ist deshalb, das Lieder, welche den zeitgenossischen Glaubensproblernatik berilhren, eine sine qua non geworden sind. Die Forschung in Bezug auf den Inhalt der Kirchenlieder offenbarte weiterhin auch fremde theologische Akzente. Es laflt sich zurn Beispiel einen deutlichen Einfluss des griechisch-platonistischen Dualismus aufweisen wo das Jenseitige dern Diesseitigen gegenilbergestellt wird. Es wurde besonders deutlich in der Diskussion Uber die Schopfung.
