    Technical assessment approach for vendor initiated changes of direct materials

    Hundreds of direct materials are used for each biologic production process. Vendor initiated changes (VIC) to the production process, packaging, raw material sources, and other aspects of these direct materials can and will happen. Each change could have the potential for both process and product impact not only to one product, but to every product that uses it across a manufacturing network. In order to ensure these changes are assessed appropriately and in a timely manner, procedures and processes need to be in place both from the customer and supplier. In addition, if changes are complex, communication between both parties is often necessary. Complex changes may also require a deeper level of collaboration between multiple functional groups from the customer and supplier. This poster will describe Roche/Genentech’s approach for technical assessments of VICs to ensure they are evaluated properly by the necessary subject matter experts (SMEs). It will highlight case studies such as a complex VIC pertaining to Poloxamer 188 (P188) and the effective collaboration between Roche/Genentech and its supplier. Poloxamer 188 is a complex nonionic tri-block co-polymer composed of a central hydrophobic chain of polyoxypropylene (PPO) flanked by two hydrophilic chains of polyoxyethylene (PEO) which is used in biopharmaceutical manufacturing for the cultivation of mammalian cells. The supplier informed Roche/Genentech of a production site change of P188 through the VIC business process. The complexity associated with the site change VIC was further compounded by an ongoing investigation based on observed low cell growth and product yield for some large-scale biomanufacturing processes at Roche/Genentech. Lot-to-lot variability in P188 was identified as the primary source of the observed low growth and yield even though all release specifications were met (Figure below).     Spatiotemporal analysis of air pollution and asthma patient visits in Taipei, Taiwan

    [[abstract]]Background: Buffer analyses have shown that air pollution is associated with an increased incidence of asthma, but little is known about how air pollutants affect health outside a defined buffer. The aim of this study was to better understand how air pollutants affect asthma patient visits in a metropolitan area. The study used an integrated spatial and temporal approach that included the Kriging method and the Generalized Additive Model (GAM). Results: We analyzed daily outpatient and emergency visit data from the Taiwan Bureau of National Health Insurance and air pollution data from the Taiwan Environmental Protection Administration during 2000-2002. In general, children (aged 0-15 years) had the highest number of total asthma visits. Seasonal changes of PM10, NO2, O3 and SO2 were evident. However, SO2 showed a positive correlation with the dew point (r = 0.17, p < 0.01) and temperature (r = 0.22, p < 0.01). Among the four pollutants studied, the elevation of NO2 concentration had the highest impact on asthma outpatient visits on the day that a 10% increase of concentration caused the asthma outpatient visit rate to increase by 0.30% (95% CI: 0.16%??.45%) in the four pollutant model. For emergency visits, the elevation of PM10 concentration, which occurred two days before the visits, had the most significant influence on this type of patient visit with an increase of 0.14% (95% CI: 0.01%??.28%) in the four pollutants model. The impact on the emergency visit rate was non-significant two days following exposure to the other three air pollutants. Conclusion: This preliminary study demonstrates the feasibility of an integrated spatial and temporal approach to assess the impact of air pollution on asthma patient visits. The results of this study provide a better understanding of the correlation of air pollution with asthma patient visits and demonstrate that NO2 and PM10 might have a positive impact on outpatient and emergency settings respectively. Future research is required to validate robust spatiotemporal patterns and trends

    Photoproducts of indomethacin exhibit decreased hydroxyl radical scavenging and xanthine oxidase inhibition activities

    AbstractIndomethacin (IN) is a widely used nonsteroidal anti-inflammatory drug. In this study, four photoproducts of IN (IN1–IN4) were produced and isolated from photoirradiated IN. This study investigated the abilities of IN and its photoproducts to scavenge hydroxyl radicals and inhibit xanthine oxidase (XO). The hydroxyl radical-scavenging activity was measured in vitro by electron spin resonance spectrometry using 5,5-dimethyl-1-pyrroline-N-oxide as a spin trapping agent. Enzyme activity was measured by continuous monitoring of uric acid formation, using xanthine as a substrate. The results showed that, among all the related products, IN has the strongest hydroxyl radical-scavenging (IC50 = 65 μM) and XO inhibitory (IC50 = 86 μM) effects. To further understand the stereochemistry of the reactions between these IN derivatives and XO, we performed computer-aided molecular modeling. IN was the most potent inhibitor with the most favorable interaction in the reactive site.     Gender Difference in Statin Intervention on Blood Lipid Control among Patients with Coronary Heart Disease

    SummaryBackgroundThe aim of this study was to clarify the current status in the effective control of dyslipidemia in Taiwanese women and men with coronary heart disease (CHD).Materials and methodsA total 1584 patients with CHD (1188 men, aged 64.8 ± 11.6 years and 396 women, aged 69.0 ± 9.8 years) from 3486 patients who had atherosclerotic vascular disease and complete lipids measured values [total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), triglycerides (TG), and low-density lipoprotein cholesterol (LDL-C)] were used for analysis.ResultsThe waist, height, weight, and creatinine levels were higher in men than in women. The systolic blood pressure, TC, HDL-C, LDL-C, fasting blood glucose, and platelet were lower in men than in women. Men were more likely to achieve the target goal than women in TC < 160 mg/dL, LDL-C < 100 mg/dL, and TG < 150 mg/dL as well as to achieve HDL-C goal.ConclusionA significant gap was found between the guidelines and clinical practice in statin intervention among these CHD patients, particularly for women.     Associations between Aerosol Types and Chlorophyll-a Concentration over Coastal Area in East Asia from Satellite Observations

    This study improved significantly the relationship between aerosol optical depth (AOD) and sea surface chlorophyll-a concentration (Chl-a), after considering the effects of sea surface temperature (SST), ocean surface current (OSC) and type of aerosols. The decadal satellite-retrieved Orbview-2/SeaWiFS Chl-a and Terra/MODIS AOD550nm data (from March 2000 to December 2010) were used to investigate the impact of atmospheric aerosols on the Chl-a concentration in the coastal water around the region of East Asia (equator to 75°N and from 100°E to 180°E). Two sets of sequential areas (A1 to A10 and B1 to B9) were selected for examining and excluding the influence of SST and OSC. After taking the potential location of aerosol deposition from OSC into account, an obvious correlation between AOD550nm and Chl-a concentration was demonstrated around the site of study area A. For aerosol partition, the Normalized Gradient Aerosol Index (NGAI) was applied to MODIS AOD products for aerosol type identification and mixed status determination. The results indicated that the type of mineral dust (DS) significantly increases the Chl-a while the biomass burning (BB) aerosols may restrain the Chl-a. This seems to be a non-impact of anthropogenic pollutant (AP) on Chl-a within the surface layer. The other area, B (B1 to B9), next to the region of area A, also shows similar results with high consistency; thus, the significant impact of DS aerosols on Chl-a production is suggested over the coastal region of East Asia

    Environmental literacy on ecotourism: a study on student knowledge, attitude, and behavioral intentions in China and Taiwan

    This study aims to gain further insights to Chinese and Taiwanese university students’ environmental literacy on ecotourism. A structural equation model is developed and validated in an effort to explore the differences between Chinese and Taiwanese university students in terms of their environmental knowledge, environmental attitude, and behavioral intentions. The results showed that the ecotourism perception of Chinese and Taiwanese university students affect their behavioral intentions. Chinese university students exhibited a higher correlation between ecotourism knowledge and behavioral intentions than their Taiwanese counterparts. The findings also revealed differences between the Chinese and Taiwanese students in their perception of ecotourism, and this disparity was particularly evident with regards to how ecotourism should be governed. A moderate difference in ecotourism behavioral intentions was also identified, in which Taiwanese university students were less likely to engage in self-empowerment or private empowerment, to be more educated in the field of ecotourism than their Chinese counterparts

    AMiBA Wideband Analog Correlator

    A wideband analog correlator has been constructed for the Yuan-Tseh Lee Array for Microwave Background Anisotropy. Lag correlators using analog multipliers provide large bandwidth and moderate frequency resolution. Broadband IF distribution, backend signal processing and control are described. Operating conditions for optimum sensitivity and linearity are discussed. From observations, a large effective bandwidth of around 10 GHz has been shown to provide sufficient sensitivity for detecting cosmic microwave background variations.Comment: 28 pages, 23 figures, ApJ in press

    The Yuan-Tseh Lee Array for Microwave Background Anisotropy

    The Yuan-Tseh Lee Array for Microwave Background Anisotropy (AMiBA) is the first interferometer dedicated to studying the cosmic microwave background (CMB) radiation at 3mm wavelength. The choice of 3mm was made to minimize the contributions from foreground synchrotron radiation and Galactic dust emission. The initial configuration of seven 0.6m telescopes mounted on a 6-m hexapod platform was dedicated in October 2006 on Mauna Loa, Hawaii. Scientific operations began with the detection of a number of clusters of galaxies via the thermal Sunyaev-Zel'dovich effect. We compare our data with Subaru weak lensing data in order to study the structure of dark matter.     A Taiwanese Propolis Derivative Induces Apoptosis through Inducing Endoplasmic Reticular Stress and Activating Transcription Factor-3 in Human Hepatoma Cells

    Activating transcription factor-(ATF-) 3, a stress-inducible transcription factor, is rapidly upregulated under various stress conditions and plays an important role in inducing cancer cell apoptosis. NBM-TP-007-GS-002 (GS-002) is a Taiwanese propolin G (PPG) derivative. In this study, we examined the antitumor effects of GS-002 in human hepatoma Hep3B and HepG2 cells in vitro. First, we found that GS-002 significantly inhibited cell proliferation and induced cell apoptosis in dose-dependent manners. Several main apoptotic indicators were found in GS-002-treated cells, such as the cleaved forms of caspase-3, caspase-9, and poly(ADP-ribose) polymerase (PARP). GS-002 also induced endoplasmic reticular (ER) stress as evidenced by increases in ER stress-responsive proteins including glucose-regulated protein 78 (GRP78), growth arrest- and DNA damage-inducible gene 153 (GADD153), phosphorylated eukaryotic initiation factor 2α (eIF2α), phosphorylated protein endoplasmic-reticular-resident kinase (PERK), and ATF-3. The induction of ATF-3 expression was mediated by mitogen-activated protein kinase (MAPK) signaling pathways in GS-002-treated cells. Furthermore, we found that GS-002 induced more cell apoptosis in ATF-3-overexpressing cells. These results suggest that the induction of apoptosis by the propolis derivative, GS-002, is partially mediated through ER stress and ATF-3-dependent pathways, and GS-002 has the potential for development as an antitumor drug
