How can we use artificial intelligence (AI) and machine learning (ML) to make video editing as easy as "editing text''? In this thesis, this problem of using AI to support video editing is explored from the human--AI interaction perspective, with the emphasis on using AI to support users. Video is a dual-track medium with audio and visual tracks. Editing videos requires synchronization of these two tracks and precise operations at milliseconds. Making it as easy as editing text might not be currently possible. Then how should we support the users with AI, and what are the current challenges in doing so? There are five key questions that drove the research in this thesis. What is the start of the art in using AI to support video editing? What are the needs and expectations of video professionals from AI? What are the impacts on efficiency and accuracy of subtitles when AI is used to support subtitling? What are the changes in user experience brought on by AI-assisted subtitling? How can multiple AI methods be used to support cropping and panning task? In this thesis, we employed a user experience focused and task-based approach to address the semi-automation in video editing. The first paper of this thesis provided a synthesis and critical review of the existing work on AI-based tools for videos editing and provided some answers to how should and what more AI can be used in supporting users by a survey of 14 video professional. The second paper presented a prototype of AI-assisted subtitling built on a production grade video editing software. It is the first comparative evaluation of both performance and user experience of AI-assisted subtitling with 24 novice users. The third work described an idiom-based tool for converting wide screen videos made for television to narrower aspect ratios for mobile social media platforms. It explores a new method to perform cropping and panning using five AI models, and an evaluation with 5 users and a review with a professional video editor were presented.Doktorgradsavhandlin

The performance of a low temperature solar collector, which acts as an evaporator of a heat pump system, has been investigated. An experimcntal setup has been designed, fabricated and tested under the meteorological conditions of Singapore. Refrigerant 134a has been used in the experiments. Effects of various parameters have been considered. Results show that, with the increase in collector tube length, the dryness fraction of the fluid increases leading to an increase in the heat transfer coefficient between the tube wall and fluid. Results also show that collector is very sensitive to solar radiation. If solar radiation increases, the mass flow rate of fluid inside the collector increases, which increases the collector efficiency as more heat is absorbed. Collector efficiency increases with irradiation, reaches a maximum and then decreases. Collector efficiency, as high as about 0.85 was obtained, which could be attributed to the low temperature operation of the collector

Melioidosis is a frequently fatal infectious disease caused by the Gram negative bacillus Burkholderia pseudomallei. Although it was originally discovered in Myanmar, the disease disappeared from sight for many decades. This study focuses on detection of B. pseudomallei in soil in selected sampling sites in an attempt to start to fill the gaps in the current status of our knowledge of the geographical distribution of B. pseudomallei in soil in Myanmar. This cross-sectional study consists of 400 soil samples from 10 selected study townships from two major paddy growing regions. Bacterial isolation was done using a simplified method for the isolation of Burkholderia pseudomallei from soil. In this study, only 1% (4/400) of soil samples were found to be positive; two of four were found at 90 cm depth and another two positive samples were found at 30 cm and 60 cm. This survey has confirmed the presence of environmental B. pseudomallei in Myanmar indicating that the conditions are in place for melioidosis acquisition

Background: As the prevalence of the malaria has been decreasing in many endemic countries including Myanmar, malaria elimination in Greater Mekong Region was targeted not later than 2030. The relevance of molecular and serological tools to identify residual transmission remains to be established in this setting. Methods: One-year cohort study was conducted and sera samples were collected in every 3 months with active and passive case detection for clinical malaria episodes by RDT, microscopy and molecular method. The sera were used to detect the malaria antibody against PfMSP1-19, PvAMA1, PvDBPII and PvMSP1-19 by protein microarray. Results: Among the recruited 1182 participants, there was no RDT positive case for malaria infection although two vivax infections were detected by microscopy in initial collection. Molecular methods detected the asymptomatic cases of 28/1182 (2.37%) in first, 5/894 (0.42%) in second, 12/944 (1.02%) in third, 6/889 (0.51%) in fourth collection, respectively. Seropositivity rates against the PfMSP1-19, PvMSP1-19, PvAMA1 and PvDBPII were 73/270 (27.0%), 85/270 (31.5%), 65/270 (24.1%) and 160/270 (59.3%), respectively. PfMSP1-19 and PvMSP1-19 showed high and stable antigenicity in acute and subacute samples but declining in 1-year history samples. No cross reactivity of PfMSP1-19 and PvMSP1-19 between the two species and higher seropositivity among the asymptomatic carriers were observed. Mapping data indicated serological surveillance can detect the geographical pattern of malaria infection under low transmission setting. Conclusions: These findings support that PfMSP1-19 and PvMSP1-19 are suggested for serosurveillance of the malaria especially in low transmission setting for further necessary actions have to be carried out to eliminate the malaria.Publisher PDFPeer reviewe

Associations between Micronutrient Status and Post-Hospital Discharge Growth among 2–23-Month-Old Children with Acute Illnesses in Africa and South-Asia

Thesis (Master's)--University of Washington, 2023Introduction: The high prevalence of wasting among children under five in low- and middle-income countries remains a significant public health concern, contributing to elevated morbidity and mortality rates. Recent studies have highlighted that even after hospital discharge, these children continue to face an increased incidence of mortality and nutritional factors may contribute to this elevated risk. Malnutrition has been shown to exert detrimental effects on innate and adaptive immune functions, increasing children's susceptibility to infections. At the same time, diseases can deplete vital micro and macronutrients, resulting in impaired growth, which, in turn, is associated with elevated mortality risk. This study aimed to investigate associations between serum micronutrient levels and changes in growth among children aged 2–23 months following hospital discharge for acute illnesses. Method: This secondary data analysis utilized CHAIN nested case-cohort data. Children aged 2-23 months with acute illnesses who had been admitted to hospitals and discharged as survived across six different countries were included. The associations between micronutrient status at hospital discharge and day 45 follow-up growth outcomes, measured as changes in mid-upper arm circumference Z-score (ΔMUACZ), weight-for-height Z-score (ΔWHZ), length-for-age Z-score (ΔLAZ), and weight-for-age Z-score (ΔWAZ) were examined. Initial univariate regression analyses were conducted to examine the associations between individual micronutrients and early post-discharge growth. Subsequently, multivariate analysis using linear regression was employed to confirm associations between selected micronutrients and growth based on the univariate results and adjusting for potential confounders. Results: Univariate analysis found significant associations between several micronutrients and growth outcomes. High calcium and copper levels were negatively associated with early post-discharge growth, as reflected by changes in ΔMUACZ, ΔWHZ and ΔWAZ scores. Conversely, serum retinol, magnesium, potassium, calcium, B1, B2, B5, B7 and vitamin E each showed positive associations with at least one measure of growth. However, in multivariate analysis, only vitamin B2 maintained a significant association with increased changes in ΔMUACZ and ΔWAZ scores. Conclusion: Micronutrients, including copper, calcium, B vitamins, vitamin A, and vitamin E, demonstrated modest associations with early post-discharge growth. However, these associations were attenuated by adjustment for confounders such as preterm birth, suboptimal breastfeeding practices, stunting, diarrhea, pneumonia, and systemic inflammation. Deficiencies in the aforementioned micronutrients may be negatively associated with early post-discharge growth through various physiological mechanisms. However, micronutrient deficiencies alone are not strong independent determinants of early post-discharge growth. While micronutrient supplementation remains an important intervention regardless of its effect on post-discharge growth, promoting early post-discharge growth through micronutrient supplementation alone is unlikely to be effective without addressing underlying factors, such as preterm birth, feeding practices and infectious morbidities

Serum-Mediated Cleavage of Bacillus anthracis Protective Antigen Is a Two-Step Process That Involves a Serum Carboxypeptidase

    No full text
Our findings identify a serum-mediated modification of PA20 that has not been previously described. These observations further imply that the processing of PA is more complex than currently thought. Additional study is needed to define the contribution of serum processing of PA to the host response and individual susceptibility to anthrax.Much of our understanding of the activity of anthrax toxin is based on in vitro systems, which delineate the interaction between Bacillus anthracis toxins and the cell surface. However, these systems fail to account for the intimate association of B. anthracis with the circulatory system, including the contribution of serum proteins to the host response and processing of anthrax toxins. Using a variety of immunological techniques to inhibit serum processing of B. anthracis protective antigen (PA) along with mass spectrometry analysis, we demonstrate that serum digests PA via 2 distinct reactions. In the first reaction, serum cleaves PA83 into 2 fragments to produce PA63 and PA20 fragments, similarly to that observed following furin digestion. This is followed by carboxypeptidase-mediated removal of the carboxy-terminal arginine and lysines from PA20
