Design of University Small-Scale Dairy Processing Facility
This project was assigned to research the feasibility and value of implementing a dairy processing facility on the campus of the University of Nebraska-Lincoln. The facility would process milk produced from cows under the same roof and will serve as an educational experience for Nebraska dairy farmers, UNL students, and K-12 students in the Lincoln-Lancaster County area. If the project is successful and replicated across the state, this facility could have a significant impact on the reduction of milk transportation costs in the Nebraska dairy industry.
The project began with researching milk processing methods and steps from production to consumption. Shortly after this step, information on milk consumption patterns was collected from UNL Dining Services to determine demand on campus. Every unit operation requires certain equipment to effectively ensure the safety and quality of the final product, and mass balances from UNL milk consumption data were used to size equipment and storage capacity. Engineering firms were then consulted to gather information on equipment specifications and prices. Equipment costs and operating costs (estimated with the help of Dr. Howell and other university dairy operations) were entered into a Monte Carlo simulation to analyze return on investment and a breakeven point.
The results from the costs section showed that the fixed costs (equipment and engineering) for the milk processing would be about $1.2 million. The Monte Carlo simulation showed that the project would not turn a profit for 10-12 years, and approximately 2.25 million gallons of milk would need to be processed and sold to recover initial costs. Overall, the project successfully displays data that can be interpreted by the client to decide whether to move forward with the project and the appropriate scale for the project at UNL
The malaria parasite egress protease SUB1 is a calcium-dependent redox switch subtilisin.
Malaria is caused by a protozoan parasite that replicates within an intraerythrocytic parasitophorous vacuole. Release (egress) of malaria merozoites from the host erythrocyte is a highly regulated and calcium-dependent event that is critical for disease progression. Minutes before egress, an essential parasite serine protease called SUB1 is discharged into the parasitophorous vacuole, where it proteolytically processes a subset of parasite proteins that play indispensable roles in egress and invasion. Here we report the first crystallographic structure of Plasmodium falciparum SUB1 at 2.25 Ă…, in complex with its cognate prodomain. The structure highlights the basis of the calcium dependence of SUB1, as well as its unusual requirement for interactions with substrate residues on both prime and non-prime sides of the scissile bond. Importantly, the structure also reveals the presence of a solvent-exposed redox-sensitive disulphide bridge, unique among the subtilisin family, that likely acts as a regulator of protease activity in the parasite
Exercise prevents obesity-induced cognitive decline and white matter damage in mice.
Obesity in the western world has reached epidemic proportions, and yet the long-term effects on brain health are not well understood. To address this, we performed transcriptional profiling of brain regions from a mouse model of western diet (WD)-induced obesity. Both the cortex and hippocampus from C57BL/6J (B6) mice fed either a WD or a control diet from 2 months of age to 12 months of age (equivalent to midlife in a human population) were profiled. Gene set enrichment analyses predicted that genes involved in myelin generation, inflammation, and cerebrovascular health were differentially expressed in brains from WD-fed compared to control diet-fed mice. White matter damage and cerebrovascular decline were evident in brains from WD-fed mice using immunofluorescence and electron microscopy. At the cellular level, the WD caused an increase in the numbers of oligodendrocytes and myeloid cells suggesting that a WD is perturbing myelin turnover. Encouragingly, cerebrovascular damage and white matter damage were prevented by exercising WD-fed mice despite mice still gaining a significant amount of weight. Collectively, these data show that chronic consumption of a WD in B6 mice causes obesity, neuroinflammation, and cerebrovascular and white matter damage, but these potentially damaging effects can be prevented by modifiable risk factors such as exercise
Effect of enhanced medical rehabilitation on functional recovery in older adults receiving skilled nursing care after acute rehabilitation: A randomized clinical trial
Importance: Enhanced medical rehabilitation (EMR) is a systematic and standardized approach for physical and occupational therapists to engage patients. Higher patient engagement in therapy might lead to improved functional recovery in rehabilitation settings, such as skilled nursing facilities (SNFs).
Objective: To determine whether EMR improves older adults\u27 functional recovery.
Design, Setting, and Participants: A double-blind, parallel-group, randomized clinical trial was conducted from July 29, 2014, to July 13, 2018, in 229 adults aged 65 years or older admitted to 2 US SNFs. Participants were randomized to receive EMR (n = 114) vs standard-of-care rehabilitation (n = 115). Intention-to-treat analysis was used.
Interventions: The intervention group received their physical and occupational therapy from therapists trained in EMR. Based on models of motivation and behavior change, EMR is a toolkit of techniques to increase patient engagement and therapy intensity. The control group received standard-of-care rehabilitation from physical and occupational therapists not trained in EMR.
Main Outcomes and Measures: The primary outcome was change in function in activities of daily living and mobility, as assessed with the Barthel Index, which measures 10 basic activities of daily living or mobility items (scale range, 0-100), from SNF admission to discharge; secondary outcomes were gait speed for 10 m, 6-minute walk test, discharge disposition, rehospitalizations, and self-reported functional status at days 30, 60, and 90. To examine the rehabilitation process, therapists\u27 engagement with patients and patient active time during therapy were measured for a sample of the sessions.
Results: Of the 229 participants, 149 (65.1%) were women; 177 (77.3%) were white, and 51 (22.3%) were black; mean (SD) age was 79.3 (8.0) years. Participants assigned to EMR showed greater recovery of function than those assigned to standard of care (mean increase in Barthel Index score, 35 points; 95% CI, 31.6-38.3 vs 28 points; 95% CI, 25.2-31.7 points; P = .007). There was no evidence of a difference in the length of stay (mean [SD], 23.5 [13.1] days). However, there were no group by time differences in secondary outcome measures, including self-reported function after SNF discharge out to 90 days as measured on the Barthel Index (mean [SE] score: EMR, 83.65 [2.20]; standard of care, 84.67 [2.16]; P = .96). The EMR therapists used a median (interquartile range) of 24.4 (21.0-37.3) motivational messages per therapy session vs 2.3 (1.1-2.9) for nontrained therapists (P \u3c .001), and EMR patients were active during a mean (SD) of 52.5 (6.6%) of the therapy session time vs 41.2 (6.8%) for nontrained therapists (P = .001).
Conclusions and Relevance: Enhanced medical rehabilitation modestly improved short-term functional recovery for selected older adults rehabilitating in SNFs. However, there was no evidence that the benefits persisted over the longer term. This study demonstrates the value of engaging and motivating older adults in rehabilitation therapy, but more work is needed to extend these benefits to longer-term outcomes after discharge home.

How can cunent slaughter and dressing procedures in UK pig slaughterhouses be improved to reduce contamination of pig meat with pathogenic bacteria?
In pork slaughterhouses a number of dressing stages have the potential to improve the hygienic condition of the carcass surface. The operations performed at each of these dressing stages can be undertaken using a diverse range of mechanized systems that each have different reductive effects on the levels of microbial contamination. Our results show that pre-washing optimizes the effectiveness of condensation scalding in improving carcass hygiene, and that the same areas of carcasses are consistently not effectively heated by singeing/flaming, independent of the different systems used in pork and bacon slaughterhouses
Validation of Twelve Small Kepler Transiting Planets in the Habitable Zone
We present an investigation of twelve candidate transiting planets from
Kepler with orbital periods ranging from 34 to 207 days, selected from initial
indications that they are small and potentially in the habitable zone (HZ) of
their parent stars. Few of these objects are known. The expected Doppler
signals are too small to confirm them by demonstrating that their masses are in
the planetary regime. Here we verify their planetary nature by validating them
statistically using the BLENDER technique, which simulates large numbers of
false positives and compares the resulting light curves with the Kepler
photometry. This analysis was supplemented with new follow-up observations
(high-resolution optical and near-infrared spectroscopy, adaptive optics
imaging, and speckle interferometry), as well as an analysis of the flux
centroids. For eleven of them (KOI-0571.05, 1422.04, 1422.05, 2529.02, 3255.01,
3284.01, 4005.01, 4087.01, 4622.01, 4742.01, and 4745.01) we show that the
likelihood they are true planets is far greater than that of a false positive,
to a confidence level of 99.73% (3 sigma) or higher. For KOI-4427.01 the
confidence level is about 99.2% (2.6 sigma). With our accurate characterization
of the GKM host stars, the derived planetary radii range from 1.1 to 2.7
R_Earth. All twelve objects are confirmed to be in the HZ, and nine are small
enough to be rocky. Excluding three of them that have been previously validated
by others, our study doubles the number of known rocky planets in the HZ.
KOI-3284.01 (Kepler-438b) and KOI-4742.01 (Kepler-442b) are the planets most
similar to the Earth discovered to date when considering their size and

Life Histories, Demography, and Distribution of a Fluvial Bull Trout Population
To describe the life histories and demography of a fluvial population of Bull Trout Salvelinus confluentus, we PIT-tagged and radio-tagged Bull Trout captured in Mill Creek, a tributary of the Walla Walla River (Washington–Oregon), during 1998–2009. Adult abundance declined 63% during 2006–2010, driven primarily by a 10-fold reduction in subadult-to-adult returns. Larger subadults and fall–winter emigrants survived at higher rates, but they were a small proportion of the subadult migrants. The survival rates of larger, generally older adults were also more than 40% greater than those of smaller adults. Changes in abundance influenced other characteristics of the population. For example, adult upstream movement into spawning areas during 1999–2005 peaked in late July, whereas the smaller runs observed during 2006–2010 peaked in early September, and the relationship between fish size and migration timing shifted. Unlike many adfluvial populations, more than 90% of the adults in Mill Creek spawned annually. Bull Trout that spawned in main-stem Mill Creek were primarily larger migratory adults; however, about 20% of the large adults were strictly or intermittently resident, remaining in the spawning area year-round. The downstream extent of individuals' migratory distributions varied greatly—from just downstream of the spawning area to the mouth of the Walla Walla River and potentially hundreds of kilometers into the Columbia River. Despite a large sample size of radio-tagged fish, radiotelemetry substantially underestimated the distribution and range that were evident from PIT tag detections. Life history terms such as “migratory,” “resident,” and “fluvial” and their associations with body size, movement, and distribution are useful for describing general patterns, but they fail to reflect the diversity and complexity within and among populations. For Bull Trout in Mill Creek, that life history diversity, including small, resident adult forms in the tributaries and a continuum of distribution for large adults, maximizes the use of available habitat and likely contributes to the population's persistence
PTF11eon/SN2011dh: Discovery of a Type IIb Supernova From a Compact Progenitor in the Nearby Galaxy M51
On May 31, 2011 UT a supernova (SN) exploded in the nearby galaxy M51 (the
Whirlpool Galaxy). We discovered this event using small telescopes equipped
with CCD cameras, as well as by the Palomar Transient Factory (PTF) survey, and
rapidly confirmed it to be a Type II supernova. Our early light curve and
spectroscopy indicates that PTF11eon resulted from the explosion of a
relatively compact progenitor star as evidenced by the rapid shock-breakout
cooling seen in the light curve, the relatively low temperature in early-time
spectra and the prompt appearance of low-ionization spectral features. The
spectra of PTF11eon are dominated by H lines out to day 10 after explosion, but
initial signs of He appear to be present. Assuming that He lines continue to
develop in the near future, this SN is likely a member of the cIIb (compact
IIb; Chevalier and Soderberg 2010) class, with progenitor radius larger than
that of SN 2008ax and smaller than the eIIb (extended IIb) SN 1993J progenitor.
Our data imply that the object identified in pre-explosion Hubble Space
Telescope images at the SN location is possibly a companion to the progenitor
or a blended source, and not the progenitor star itself, as its radius (~10^13
cm) would be highly inconsistent with constraints from our post-explosion
photometric and spectroscopic data
