    Functional Importance of Lipin Phosphorylation

    Highly conserved throughout evolution, lipins are dual functioning proteins found from yeast to humans. Functioning in the cytoplasm as phosphatidate phosphatase enzymes (PAP), lipins produce diacylglycerol that serves as a precursor for neutral fats and membrane phospholipids. Alternatively, nuclear lipins are responsible for the regulation of metabolic genes. Interestingly, both the mammalian lipin 1 paralog and the single Drosophila Lipin ortholog are highly phosphorylated proteins. Target of rapamycin (TOR) has previously been identified as one of the kinases that controls the subcellular localization of both lipin 1 and Drosophila Lipin. However, other serine and threonine kinases are predicted to be important for the phosphorylation of Lipin. Here, I implement both the GAL4/UAS system as well as CRISPR/Cas9 mutagenesis to systematically mutate individual amino acid residues or clusters of phosphorylation sites of Drosophila Lipin to identify their functional importance. Phenotypic characterization of the phosphosite mutants included fat body histology and fat droplet staining, triglyceride and protein content, starvation resistance, and potential developmental delays. Lipin antibody staining was employed to reveal intracellular distribution of the mutant protein. Results support the prediction that these phosphorylation sites are important for both nuclear function and the role of the protein in fat storage. Data reported here will support the understanding of how the activities of these proteins could be specifically targeted

    Products and Kunneth theorems in cyclic homology and cohomology theories

    This thesis is concerned with the construction of products in cyclic homology and cohomology, by use of the methods of acyclic models. A theory HC ˉ͙ (A) which is dual to cyclic cohomology HC *(A) over its natural coefficients is introduced, and products are defined HC ˉ͙ (A) and HC * (A). The product then induced on cyclic homology HC ͙(A) is shown to agree with that defined by Loday and Quillen. By using HC ˉ͙ (A), it is possible to construct a multiplicative chern character ch : K ᵢ (A) → HC ˉᵢ-2α (A). Kunneth theorems in the various theories are proved, and some examples are considered

    Health and Housing Trends of Single Homeless Adults in Chittenden County, Vermont

    Introduction. Homeless individuals are at an increased risk for both physical and mental health comorbidities compared to those with stable housing. Prior studies have demonstrated that many of these individuals do not seek or receive proper medical care. This study provides a landscape analysis of health characteristics in the single adult homeless population in Chittenden County, Vermont. Methods. Interviews were conducted using a modified version of the Vulnerability Index – Service Prioritization Decision Assistance Tool to determine vulnerability indices across a range of different health and social wellness domains. Single home- less adults were sampled from community self-help clinics and transitional living and homeless shelters from September to November 2017. Results. 56 in-person surveys were conducted. Mean age of participants was 45.3 years and over half were male (63%). Extended periods or multiple episodes of homelessness were experienced most by those who reported sleeping outdoors (89%); this group also reported the highest use of emergency services (47%). The highest incidence of abuse/trauma as a contributing factor for homelessness was reported most in the 18-30 age group (71%) and least in the above 51 age group (35%). Nearly all participants owned a cell phone (86%). Conclusions. Single homeless adults self-reported a variety of health and social is- sues, including substance abuse, increased emergency department use, and in- creased abuse, trauma, and deteriorated social relationships. This study highlights different risk factors that contribute to homelessness in this select population.     Who Can I Turn To? Emotional Support Availability in African American Social Networks

    African Americans disproportionately experience psychological distress, such as feelings of sadness, hopelessness, and worthlessness and are disproportionately exposed to risk factors associated with mental illness, such as racial discrimination, violence and poverty. To effectively address African Americans’ mental health needs, it is imperative to identify who African Americans turn to when they experience stressors. The purpose of this study was to assess the extent to which emotional support is provided within African Americans’ social networks and determine the characteristics of social network members who African Americans rely upon for emotional support. Results indicate that African Americans rely on social network members for spiritual and physical health support more so than emotional support. Among both male and female participants, social network members were significantly more likely to be relied upon for emotional support if they were a non-familial network contact, had a close relationship to the participant, and if they also were someone the participant spoke to about his or her physical health. Findings have implications for the development of culturally-sensitive strategies for increasing emotional support provision within African Americans’ social networks

    CanSat Competition: Contributing to the Development of NASA's Vision for Robotic Space Exploration

    CanSat is an international student design-build-launch competition organized by the American Astronautical Society (AAS) and American Institute of Aeronautics and Astronautics (AIAA). The competition is also sponsored by the Naval Research Laboratory (NRL) and the National Aeronautics and Space Administration (NASA). The CanSat competition is designed for college, university and high school students wanting to participate in an applicable space-related competition. The objective of the CanSat competition is to complete space exploration missions by designing a specific system for a small sounding rocket payload which will follow and perform to a specific set of rules and guidelines for each year's competition. The competition encompasses a complete life-cycle of one year which includes all phases of design, integration, testing, judging and competition. The mission guidelines are based from space exploration missions and include bonus requirement options which teams may choose to participate in. The fundamental goal of the competition is to educate future engineers and scientists. This is accomplished by students applying systems engineering practices to a development project that incorporates an end-to-end life cycle, from requirements analysis, through preliminary design, integration and testing, an actual flight of the CanSat, and concluding with a post-mission debrief. This is done specifically with space related missions to bring a unique aspect of engineering and design to the competition. The competition has been progressing since its creation in 2005. The competition was originally meant to purely convey the engineering and design process to its participants, but through many experiences the competition has also undergone a learning experience with respect to systems engineering process and design.     Structural and Contextual Patterns in Family Health History Knowledge among African American Adults: A Mixed-Methods Social Network Analysis Study*

    Background: Family health history is a strong risk factor for many chronic diseases. Ethnic minorities have been found to have a low awareness of their family health history (FHH), which may pose a contributing factor to health disparities. Purpose: The purpose of this mixed-methods social network analysis study was to identify structural and contextual patterns in African American adults’ FHH knowledge based on interpersonal communication exchanges with their family members. Methods: African American adults completed individually administered family network interviews. Participants’ 3-generation family pedigree served as a visual aid to guide their interview. Our primary outcome of interest for this analysis was whether a family member was reported as someone who talks to the participant about their own (i.e., the family member’s) health, which we refer to as a “personal health informant.” To contextualize quantitative findings, participants were asked to describe how they learned about the health history of the relatives they identified during their interview. Results: Participants (n=37) reported an average family network size of 29.4 relatives (SD = 15.5; Range = 10-67). Each participant, on average, named 17% of their familial network as personal health informants. Multivariate regression results showed that participants were more likely to name an alter as a personal health informant if the alter was female (OR = 2.14, p = 0.0519), from the maternal side of the participant’s family (OR = 1.12, p = 0.0006), had one or more chronic health conditions (OR = 2.41, p = 0.0041), was someone who has discussions with the participant about the participant’s health (OR = 16.28, p < 0.0001), was a source of family health information (OR = 3.46, p = 0.0072), and was someone whose health the participant helps to monitor or track (OR = 5.93, p = 0.0002). Complementary qualitative findings indicate that FHH knowledge is facilitated by open, direct communication among relatives. Personal health informants were described as disclosing information for the purposes of informing others for preventive purposes and for gaining social support. Participants also learned about FHH via other methods, including direct observation, during caretaking, and following a relative’s death. Conclusions: Communication and disclosure practices is an important determinant of African Americans’ FHH knowledge. More culturally and contextually meaningful public health efforts are needed to promote family health history sharing, especially regarding paternal family health history, siblings, and extended relatives

    Current practices in clinical neurofeedback with functional MRI—Analysis of a survey using the TIDieR checklist

    Background: A core principle of creating a scientific evidence base is that results can be replicated in independent experiments and in health intervention research. The TIDieR (Template for Intervention Description and Replication) checklist has been developed to aid in summarising key items needed when reporting clinical trials and other well designed evaluations of complex interventions in order that findings can be replicated or built on reliably. Neurofeedback (NF) using functional MRI (fMRI) is a multicomponent intervention that should be considered a complex intervention. The TIDieR checklist (with minor modification to increase applicability in this context) was distributed to NF researchers as a survey of current practice in the design and conduct of clinical studies. The aim was to document practice and convergence between research groups, highlighting areas for discussion and providing a basis for recommendations for harmonisation and standardisation. Methods: The TIDieR checklist was interpreted and expanded (21 questions) to make it applicable to neurofeedback research studies. Using the web-based Bristol Online Survey (BOS) tool, the revised checklist was disseminated to researchers in the BRAINTRAIN European research collaborative network (supported by the European Commission) and others in the fMRI-neurofeedback community. Results: There were 16 responses to the survey. Responses were reported under eight main headings which covered the six domains of the TIDieR checklist: What, Why, When, How, Where and Who. Conclusions: This piece of work provides encouraging insight into the ability to be able to map neuroimaging interventions to a structured framework for reporting purposes. Regardless of the considerable variability of design components, all studies could be described in standard terms of diagnostic groups, dose/duration, targeted areas/signals, and psychological strategies and learning models.
