    Associations between daily lifestyle characteristics and latent depressive symptoms in elementary school children: A cross-sectional survey

    This study aimed to identify associations between daily lifestyle characteristics and latent depressive symptoms in young children by analyzing self-reported questionnaire data. In 2011, in Nagasaki Japan, a cross-sectional survey of 1961 children aged between 10 and 12 years was conducted. Children answered questionnaires that collected demographic information, along with daily lifestyle characteristics, and were administered the Birleson Depression Self-Rating Scale for Children (DSRSC-J). The mean age of the participants was 10.98 years (±0.83 standard deviation [SD]) with a mean DSRSC-J raw score of 11.61 (±6.34 SD). In total, 24.7% of participants reported depressive symptoms. A multiple logistic regression analysis revealed that depressive symptoms were associated with female gender (odds ratio [OR]:1.86; 95% CI: 1.48?2.33), experiencing growth (OR: 1.25; 1.07-1.44), a bedtime later than 11 pm (OR: 1.25; 1.01-1.66), skipping breakfast (OR: 1.55; 1.15-2.10), and not having a set dinner time (OR: 1.65; 1.32?2.07). Conversely, absence of depressive symptoms was associated with involvement in school-related athletic clubs (OR: 0.60, 95% Cl: 0.48?0.76) and at home studying for at least one hour per day (OR: 0.67, 95% Cl: 0.54?0.84). Depressive symptoms in young children were associated with social contact and their parent’s lifestyles. Non-experts in the field of mental health who interact with young children should carefully note the presence of factors associated with depressive symptoms as this would help children with latent depressive symptoms receive prompt and timely care

    Since the amino groups in melamine could not be successfully diazotized, the oxidation of phenylhydrazino-s-triazines by oxidizing agents has been studied in order to give phenylazo-s-triazines. The azo-s-triazines, such as 2-amino-4- ani1ino-6-phenylazo-,( mp 233-234℃) (orange),and 2,4-diani1ino-6-phenylazo-s-triazine, (mp 262-2630 C) (reddish orange), were obtained by oxidation of II and III with an aqueous solution of ferric chloride at 50-70℃ respectively.     A novel strategy for the identification of antigens that are recognised by bovine MHC class I restricted cytotoxic T cells in a protozoan infection using reverse vaccinology

    BACKGROUND: Immunity against the bovine protozoan parasite Theileria parva has previously been shown to be mediated through lysis of parasite-infected cells by MHC class I restricted CD8(+ )cytotoxic T lymphocytes. It is hypothesized that identification of CTL target schizont antigens will aid the development of a sub-unit vaccine. We exploited the availability of the complete genome sequence data and bioinformatics tools to identify genes encoding secreted or membrane anchored proteins that may be processed and presented by the MHC class I molecules of infected cells to CTL. RESULTS: Of the 986 predicted open reading frames (ORFs) encoded by chromosome 1 of the T. parva genome, 55 were selected based on the presence of a signal peptide and/or a transmembrane helix domain. Thirty six selected ORFs were successfully cloned into a eukaryotic expression vector, transiently transfected into immortalized bovine skin fibroblasts and screened in vitro using T. parva-specific CTL. Recognition of gene products by CTL was assessed using an IFN-γ ELISpot assay. A 525 base pair ORF encoding a 174 amino acid protein, designated Tp2, was identified by T. parva-specific CTL from 4 animals. These CTL recognized and lysed Tp2 transfected skin fibroblasts and recognized 4 distinct epitopes. Significantly, Tp2 specific CD8(+ )T cell responses were observed during the protective immune response against sporozoite challenge. CONCLUSION: The identification of an antigen containing multiple CTL epitopes and its apparent immunodominance during a protective anti-parasite response makes Tp2 an attractive candidate for evaluation of its vaccine potential

    SPIN INJECTION INTO FERROMAGNETIC METAL FROM HEAVY METAL OWING TO SPIN HALL EFFECT

    When a current flows through a heavy metal (HM), the spin of the current carriers is polarized by the spin Hall effect (SHE). In a junction of a HM and a ferromagnetic metal (FM), the spin current flows into the FM from the HM and gives spin-transfer torque to the magnetic moments of the FM. We calculate the spin current in a layer of HM/FM with the SHE. We use a spin-resolved electrochemical potential including the SHE in the calculation of the spin current.     Pharmaceutical and preclinical evaluation of Advax adjuvant as a dose-sparing strategy for ant venom immunotherapy

Adjuvants are commonly used for antigen sparing in other vaccines, and thereby could potentially have major benefits to extend JJA supplies if they were to similarly enhance JJA VIT immunogenicity. The purpose of this study was to evaluate the physicochemical and microbiological stability and murine immunogenicity of low-dose JJA VIT formulated with a novel polysaccharide adjuvant referred to as delta inulin or Advax™. Jack Jumper ant venom (JJAV) protein stability was assessed by UPLC-UV, SDS-PAGE, SDS-PAGE immunoblot, and ELISA inhibition. Diffraction light scattering was used to assess particle size distribution of Advax; pH and benzyl alcohol quantification by UPLC-UV were used to assess the physicochemical stability of JJAV diluent, and endotoxin content and preservative efficacy test was used to investigate the microbiological properties of the adjuvanted VIT formulation. To assess the effect of adjuvant on JJA venom immunogenicity, mice were immunised four times with JJAV alone or formulated with Advax adjuvant. JJA VIT formulated with Advax was found to be physicochemically and microbiologically stable for at least 2 days when stored at 4 and 25 °C with a trend for an increase in allergenic potency observed beyond 2 days of storage. Low-dose JJAV formulated with Advax adjuvant induced significantly higher JJAV-specific IgG than a 5-fold higher dose of JJAV alone, consistent with a powerful allergen-sparing effect. The pharmaceutical data provides important guidance on the formulation, storage and use of JJA VIT formulated with Advax adjuvant, with the murine immunogenicity studies providing a strong rationale for a planned clinical trial to test the ability of Advax adjuvant to achieve 4-fold JJAV dose sparing in JJA-allergic human patients

    Comparison of Hospitalization Incidence in Influenza Outpatients Treated With Baloxavir Marboxil or Neuraminidase Inhibitors: A Health Insurance Claims Database Study

    Background: Baloxavir marboxil (baloxavir) is a single-dose, oral antiinfluenza drug with a novel mechanism of action. We compared the incidence of hospitalization in patients treated with baloxavir vs neuraminidase inhibitors.Methods: In this retrospective, observational, cohort study, we used real-world patient data extracted from a Japanese health insurance claims database. The enrollment period was 1 October 2018 to 17 April 2019. On day 1, eligible patients (N = 339 007) received baloxavir, oseltamivir, zanamivir, or laninamivir. Baseline characteristics were standardized using the inverse probability of treatment weighting method. The primary end point was the incidence of hospitalization (days 2–14). Secondary end points included antibacterial use, secondary pneumonia, and additional antiinfluenza drug use.Results: Compared with the baloxavir group, the incidence of hospitalization was greater in the oseltamivir group (risk ratio [RR] and 95% confidence interval [CI], 1.41 [1.00–2.00]; risk difference [RD] and 95% CI, 0.06 [.01–.12]) and zanamivir group (RR, 1.85 [1.23–2.78]; RD, 0.11 [.02–.20]). Oseltamivir-treated patients were less likely to require antibacterials than baloxavir-treated patients (RR, 0.87 [.82–.91]). However, oseltamivir-treated patients were more likely to be hospitalized with antibacterials (RR, 1.70 [1.21–2.38]) or antibacterial injection (RR, 1.67 [1.17–2.38]) than baloxavir-treated patients (post hoc analysis). Compared with baloxavir-treated patients, additional antiinfluenza drug use was greater in oseltamivir-, zanamivir-, and laninamivir-treated patients (RR, 1.51 [1.05–2.18], 2.84 [2.04–3.96], and 1.68 [1.35–2.10], respectively).Conclusions: Baloxavir is an efficacious antiinfluenza treatment that may reduce hospitalization compared with oseltamivir and zanamivir.Clinical Trials Registration: University hospital Medical Information Network Clinical Trials Registry (UMIN000038159)

    Recent Progress of Flower Colour Modification by Biotechnology

    Genetically-modified, colour-altered varieties of the important cut-flower crop carnation have now been commercially available for nearly ten years. In this review we describe the manipulation of the anthocyanin biosynthesis pathway that has lead to the development of these varieties and how similar manipulations have been successfully applied to both pot plants and another cut-flower species, the rose. From this experience it is clear that down- and up-regulation of the flavonoid and anthocyanin pathway is both possible and predictable. The major commercial benefit of the application of this technology has so far been the development of novel flower colours through the development of transgenic varieties that produce, uniquely for the target species, anthocyanins derived from delphinidin. These anthocyanins are ubiquitous in nature, and occur in both ornamental plants and common food plants. Through the extensive regulatory approval processes that must occur for the commercialization of genetically modified organisms, we have accumulated considerable experimental and trial data to show the accumulation of delphinidin based anthocyanins in the transgenic plants poses no environmental or health risk

    Sixteen-year phytoplankton biomass trends in the northwestern Pacific Ocean observed by the SeaWiFS and MODIS ocean color sensors

    Using multisensor/platform biophysical data collected from 1997 to 2013, we investigated trends of the concentrations of phytoplankton biomass (Chl) in the northwestern Pacific Ocean (NWPO) and the probable responsible factors. The trend of rising sea surface temperature (SST) was the main factor maintaining phytoplankton positive net growth and resulted in a trend of increasing Chl at high latitudes in all seasons. At latitudes of 36-46°N, east of 160°E, the trend of rising SST was accompanied by a trend of declining Chl, markedly in spring and fall, which could be ascribed to strengthened stratification. The trends of environmental variables in the Oyashio area have modified conditions in a way detrimental to phytoplankton growth, the result being a trend of declining Chl from spring to fall. Chl south of roughly 36°N exhibited different trends in different seasons because of the different trends of vertical stratification. Whereas the observed 16-year Chl trends were not primarily influenced by interannual climate variability, to some degree they were likely modified by decadal variability associated with a weakened Aleutian Low pressure. This work prompts further comprehensive studies to investigate the probable ecological consequences of the observed Chl trend for high-trophic-level marine organisms in the NWPO

    Genome-Wide Association Study Confirming Association of HLA-DP with Protection against Chronic Hepatitis B and Viral Clearance in Japanese and Korean

    Hepatitis B virus (HBV) infection can lead to serious liver diseases, including liver cirrhosis (LC) and hepatocellular carcinoma (HCC); however, about 85–90% of infected individuals become inactive carriers with sustained biochemical remission and very low risk of LC or HCC. To identify host genetic factors contributing to HBV clearance, we conducted genome-wide association studies (GWAS) and replication analysis using samples from HBV carriers and spontaneously HBV-resolved Japanese and Korean individuals. Association analysis in the Japanese and Korean data identified the HLA-DPA1 and HLA-DPB1 genes with Pmeta = 1.89×10−12 for rs3077 and Pmeta = 9.69×10−10 for rs9277542. We also found that the HLA-DPA1 and HLA-DPB1 genes were significantly associated with protective effects against chronic hepatitis B (CHB) in Japanese, Korean and other Asian populations, including Chinese and Thai individuals (Pmeta = 4.40×10−19 for rs3077 and Pmeta = 1.28×10−15 for rs9277542). These results suggest that the associations between the HLA-DP locus and the protective effects against persistent HBV infection and with clearance of HBV were replicated widely in East Asian populations; however, there are no reports of GWAS in Caucasian or African populations. Based on the GWAS in this study, there were no significant SNPs associated with HCC development. To clarify the pathogenesis of CHB and the mechanisms of HBV clearance, further studies are necessary, including functional analyses of the HLA-DP molecule
