Kadar Gula Darah Ditinjau dari Gratitude pada Penderita DM

    Get PDF

Class IIa bacteriocin resistance in Enterococcus faecalis V583: The mannose PTS operon mediates global transcriptional responses

<p>Abstract</p> <p>Background</p> <p>The class IIa bacteriocin, pediocin PA-1, has clear potential as food preservative and in the medical field to be used against Gram negative pathogen species as <it>Enterococcus faecalis </it>and <it>Listeria monocytogenes</it>. Resistance towards class IIa bacteriocins appear in laboratory and characterization of these phenotypes is important for their application. To gain insight into bacteriocin resistance we studied mutants of <it>E. faecalis </it>V583 resistant to pediocin PA-1 by use of transcriptomic analyses.</p> <p>Results</p> <p>Mutants of <it>E. faecalis </it>V583 resistant to pediocin PA-1 were isolated, and their gene expression profiles were analyzed and compared to the wild type using whole-genome microarray. Significantly altered transcription was detected from about 200 genes; most of them encoding proteins involved in energy metabolism and transport. Glycolytic genes were down-regulated in the mutants, but most of the genes showing differential expression were up-regulated. The data indicate that the mutants were relieved from glucose repression and putative catabolic responsive elements (<it>cre</it>) could be identified in the upstream regions of 70% of the differentially expressed genes. Bacteriocin resistance was caused by reduced expression of the <it>mpt </it>operon encoding the mannose-specific phosphoenolpyruvate:carbohydrate phosphotransferase system (PTS), and the same transcriptional changes were seen in a <it>mptD</it>-inactivated mutant. This mutant also had decreased transcription of the whole <it>mpt </it>operon, showing that the PTS is involved in its own transcriptional regulation.</p> <p>Conclusion</p> <p>Our data confirm the important role of mannose PTS in class IIa bacteriocin sensitivity and we demonstrate its importance involving global carbon catabolite control.</p

Utilization Biochar of Bamboo and Compost in Improving Yield of Pakchoy Plant

This study aims to determine the effect of biochar and compost and their interaction on the growth and yield of pakchoy plants. This research uses randomized block design with a factorial pattern. Treatment consists of two factors, namely: the dosage of biochar and compost. The first factor was the dosage of biochar consisting of 4 levels, namely: without biochar, 5 t ha-1, 10 t ha-1, 15 t ha-1. The second factor is the dosage of compost, which consists of 4 levels: without compost, 10 t ha-1, 20 t ha-1, and 30 t ha-1. The results showed that the interaction between the dosage of biochar and compost had no significant effect on all observed variables. The compost application had a very significant effect on all observed variables, while the biochar application had no significant effect on all observed variables. Biochar applications at various dosage were not significantly different, but the dosage of biochar 10 t ha-1 tended to give the highest yield of fresh weight of 18.68 g which increased by 19.32% when compared with the lowest yield on treatment without the dosage biochar that is 15.66 g. The application of 20 t ha-1 compost gave the highest yield of fresh weight of 19.75 g which increased by 44% compared to the lowest value in the composted treatment which was 13.71 g

Enantioselective, potentiometric membrane electrodes for enantioanalysis of amino acids of clinical and pharmaceutical importance

    Get PDF
The enantioanalysis of compounds of clinical and pharmaceutical became increasingly important because the enantiomers of the same substance may be markers for different disease or are having a different pathway in the body. The utilization of enantioselective, potentiometric membrane electrodes made the assay of a single enantiomer easier and faster. Also the reliability of the analytical information is higher than that obtained using chromatographic techniques. The proposed electrodes are made by mixing graphite powder with paraffin oil to give carbon paste, which is modified by the addition of a chiral selector (e.g., cylodextrins, maltodextrins, macrocyclic antibiotics and fullerenes). This design is reliable. The high sensitivity, selectivity, enantioselectivity, accuracy and precision made them suitable to be used for the enantioanalysis of different compounds of clinical and pharmaceutical importance (e.g., L-histidine, L-cysteine and R-clenbuterol) in pharmaceutical tablets, and/or serum and urine samples. Fracture usually caused by trauma or physical exertion. Fracture causing common symptom of pain. Pain is an uncomfortable, subjective feeling where only the sufferer can feel it. Almost all pain felt by patients with fracture will be treated with analgesic, but many patients and doctors are less satisfied with long-term administration. This situation encourages the development of nonpharmacological techniques to treat pain such as relaxation and distraction techniques. The purpose of this study was to determine the effectiveness of relaxation technique and distraction technique to reduce the intensity of pain in patients fracture in the Surgery Ward of RSUD Prof. Dr. W. Z. Johannes Kupang. The research method was Pre Experiment with One Group Pretest-Postest design, the selection of sample using Accidental Sampling. Sample of 18 respondents. This research was conducted on 9 February until 16 March 2017 in the Surgery Ward of RSUD Prof. Dr. W. Z. Johannes Kupang. Data collection was done by giving relaxation technique (deep breath) and distraction technique (listening to music), then the researchers measure the scale of the respondent pain using Hayward pain scale. The results used independent t showed that the relaxation technique was more effective in reducing the pain of fracture patients than the distraction technique with pain reduction mean = 2.00 &gt; 1.67. Conclusion, relaxation techniques and distraction techniques are able to reduce the intensity of pain and there was a comparison of pain intensity of fracture patients after relaxation technique and distraction technique.
