    Incorporating usability evaluation into iterative development of an online platform to support research participation in Parkinson\u27s disease: A mixed methods protocol

Introduction Many people with Parkinson\u27s (PwP) are not given the opportunity or do not have adequate access to participate in clinical research. To address this, we have codeveloped with users an online platform that connects PwP to clinical studies in their local area. It enables site staff to communicate with potential participants and aims to increase the participation of the Parkinson\u27s community in research. This protocol outlines the mixed methods study protocol for the usability testing of the platform. Methods and analysis We will seek user input to finalise the platform\u27s design, which will then be deployed in a limited launch for beta testing. The beta version will be used as a recruitment tool for up to three studies with multiple UK sites. Usability data will be collected from the three intended user groups: PwP, care partners acting on their behalf and site study coordinators. Usability questionnaires and website analytics will be used to capture user experience quantitatively, and a purposive sample of users will be invited to provide further feedback via semistructured interviews. Quantitative data will be analysed using descriptive statistics, and a thematic analysis undertaken for interview data. Data from this study will inform future platform iterations. Ethics and dissemination Ethical approval was obtained from the University of Plymouth (3291; 3 May 2022).     Assessing the needs of older people in urban settings : integration of emotive, physiological and built environment data

    Design of the built environment for navigability and walkability is an increasingly important aspect of urban planning. This focus derives in part from increasing interest in lifestyles and behaviours including level of physical activity and health outcomes. Geographical Information Systems and virtual realities are playing a significant role in advancing this agenda: examples exist of integrating qualitative data (words about or visual images of places) and quantitative data (numerical descriptions of places). However there remain opportunities for exploring alternative ways of linking different types of data (physiological measurements, emotional response, street walkability and urban design quality) to address issues of urban planning and renewal. Using a case study approach this paper explores the application of geographic information science and systems to participatory approaches in built environment planning with the aim of exploring older people’s response to an unfamiliar urban environment. It examines different ways of combining temporally and spatial referenced qualitative and quantitative data. The participants in the study were a group of 44 older people (60+) from Swansea, Wales, who viewed a filmed walking route around Colchester, England. Whilst viewing the film they gave an oral commentary and physiological readings were made, which have been integrated with primary data collected on the built environment along the walking route. Proximity and inverse distance weighting approaches for combining these datasets produce complementary results in respect of older people’s physiological and emotive response to variation in the walkability and design quality of a walking route through an unfamiliar town centre. The results reveal participants experienced an elevated average heart close to Colchester Town railway station and expressed a comparatively negative emotional response to this location. Conversely participants experienced lower average heart rate, indicating reduced stress, in Brook Street where the overall Urban Design Quality score was relatively low

    Understanding pathogen transmission dynamics in waterbird communities: At what scale should interactions be studied?

    Pathogen transmission in animal populations is contingent on interactions between and within species. Often standard ornithological data (e.g. total counts at a wetland) are the only data available for assessing the risks of avian pathogen transmission. In this paper we ask whether these data can be used to infer fine-scale transmission patterns. We tested for non-randomness in waterbird assemblages and explored waterbird interactions using social network analysis. Certain network parameter values were then compared to a data set on avian influenza prevalence in southern Africa. Our results showed that species associations were strongly non-random, implying that most standard ornithological data sets would not provide adequate information on which to base models of pathogen spread. In both aquatic and terrestrial networks, all species regularly associated closely with other network members. The spread of pathogens through the community could thus be rapid. Network analysis together with detailed, fine-scale observations offers a promising avenue for further research and management-oriented applications

    An Endosomal NAADP-Sensitive Two-Pore Ca(2+) Channel Regulates ER-Endosome Membrane Contact Sites to Control Growth Factor Signaling.

    Membrane contact sites are regions of close apposition between organelles that facilitate information transfer. Here, we reveal an essential role for Ca(2+) derived from the endo-lysosomal system in maintaining contact between endosomes and the endoplasmic reticulum (ER). Antagonizing action of the Ca(2+)-mobilizing messenger NAADP, inhibiting its target endo-lysosomal ion channel, TPC1, and buffering local Ca(2+) fluxes all clustered and enlarged late endosomes/lysosomes. We show that TPC1 localizes to ER-endosome contact sites and is required for their formation. Reducing NAADP-dependent contacts delayed EGF receptor de-phosphorylation consistent with close apposition of endocytosed receptors with the ER-localized phosphatase PTP1B. In accord, downstream MAP kinase activation and mobilization of ER Ca(2+) stores by EGF were exaggerated upon NAADP blockade. Membrane contact sites between endosomes and the ER thus emerge as Ca(2+)-dependent hubs for signaling

    Host specificity and co-speciation in avian haemosporidia in the Western Cape, South Africa

    Host and pathogen ecology are often closely linked, with evolutionary processes often leading to the development of host specificity traits in some pathogens. Host specificity may range from ‘generalist’, where pathogens infect any available competent host; to ‘specialist’, where pathogens repeatedly infect specific host species or families. Avian malaria ecology in the region remains largely unexplored, despite the presence of vulnerable endemic avian species. We analysed the expression of host specificity in avian haemosporidia, by applying a previously developed host specificity index to lineages isolated from wetland passerines in the Western Cape, South Africa. Parasite lineages were isolated using PCR and identified when possible using matching lineages deposited in GenBank and in MalAvi. Parasitic clades were constructed from phylogenetic trees consisting of Plasmodium and Haemoproteus lineages. Isolated lineages matched some strains of Plasmodium relictum , P. elongatum , Haemoproteus sylvae and H. lanii . Plasmodium lineages infected a wide range of hosts from several avian families in a generalist pattern of infection. Plasmodium spp. also exhibited an infection trend according to host abundance rather than host species. By contrast, Haemoproteus lineages were typically restricted to one or two host species or families, and displayed higher host fidelity than Plasmodium spp. The findings confirm that a range of host specificity traits are exhibited by avian haemosporidia in the region. The traits show the potential to not only impact infection prevalence within specific host species, but also to affect patterns of infection at the community level

    Breeding latitude predicts timing but not rate of spring migration in a widespread migratory bird in South America

    Identifying the processes that determine avian migratory strategies in different environmental contexts is imperative to understanding the constraints to survival and reproduction faced by migratory birds across the planet. We compared the spring migration strategies of Fork‐tailed Flycatchers (Tyrannus s. savana) that breed at south‐temperate latitudes (i.e., austral migrants) vs. tropical latitudes (i.e., intratropical migrants) in South America. We hypothesized that austral migrant flycatchers are more time‐selected than intratropical migrants during spring migration. As such, we predicted that austral migrants, which migrate further than intratropical migrants, will migrate at a faster rate and that the rate of migration for austral migrants will be positively correlated with the onset of spring migration. We attached light‐level geolocators to Fork‐tailed Flycatchers at two tropical breeding sites in Brazil and at two south‐temperate breeding sites in Argentina and tracked their movements until the following breeding season. Of 286 geolocators that were deployed, 37 were recovered ~1 year later, of which 28 provided useable data. Rate of spring migration did not differ significantly between the two groups, and only at one site was there a significantly positive relationship between date of initiation of spring migration and arrival date. This represents the first comparison of individual migratory strategies among conspecific passerines breeding at tropical vs. temperate latitudes and suggests that austral migrant Fork‐tailed Flycatchers in South America are not more time‐selected on spring migration than intratropical migrant conspecifics. Low sample sizes could have diminished our power to detect differences (e.g., between sexes), such that further research into the mechanisms underpinning migratory strategies in this poorly understood system is necessary.Fil: Jahn, Alex.     What’s sex got to do with it? A family-based investigation of growing up heterosexual during the twentieth century

    This paper explores findings from a cross-generational study of the making of heterosexual relationships in East Yorkshire, which has interviewed women and men within extended families. Using a feminist perspective, it examines the relationship between heterosexuality and adulthood, focussing on sexual attraction, courtship, first kisses, first love and first sex, as mediated within family relationships, and at different historical moments. In this way, the contemporary experiences of young people growing up are compared and contrasted with those of mid-lifers and older adults who formed heterosexual relationships within the context of the changing social and sexual mores of the 1960s/1970s, and the upheavals of World War Two
