    The Bacillus subtilis signaling protein SpoIVB defines a new family of serine peptidases

    The protein SpoIVB plays a key role in signaling in the sigma (K) checkpoint of Bacillus subtilis. This regulatory mechanism coordinates late gene expression during development in this organism and we have recently shown SpoIVB to be a serine peptidase. SpoIVB signals by transiting a membrane, undergoing self-cleavage, and then by an unknown mechanism activating a zinc metalloprotease, SpoIVFB, which cleaves pro-sigma (K) to its active form, sigma (K), in the outer mother cell chamber of the developing cell. In this work we have characterized the serine peptidase domain of SpoIVB. Alignment of SpoIVB with homologues from other spore formers has allowed site-specific mutagenesis of all potential active site residues within the peptidase domain. We have defined the putative catalytic domain of the SpoIVB serine peptidase as a 160-amino-acid residue segment at the carboxyl terminus of the protein. His236 and Ser378 are the most important residues for proteolysis, with Asp363 being the most probable third member of the catalytic triad. In addition, we have shown that mutations at residues Asn290 and His394 lead to delayed signaling in the sigma (K) checkpoint. The active site residues suggest that SpoIVB and its homologues from other spore formers are members of a new family of serine peptidases of the trypsin superfamily

    Drosophila RecQ4 Is Directly Involved in Both DNA Replication and the Response to UV Damage in S2 Cells.

    The RecQ4 protein shows homology to both the S.cerevisiae DNA replication protein Sld2 and the DNA repair related RecQ helicases. Experimental data also suggest replication and repair functions for RecQ4, but the precise details of its involvement remain to be clarified.Here we show that depletion of DmRecQ4 by dsRNA interference in S2 cells causes defects consistent with a replication function for the protein. The cells show reduced proliferation associated with an S phase block, reduced BrdU incorporation, and an increase in cells with a subG1 DNA content. At the molecular level we observe reduced chromatin association of DNA polymerase-alpha and PCNA. We also observe increased chromatin association of phosphorylated H2AvD--consistent with the presence of DNA damage and increased apoptosis.Analysis of DmRecQ4 repair function suggests a direct role in NER, as the protein shows rapid but transient nuclear localisation after UV treatment. Re-localisation is not observed after etoposide or H₂O₂ treatment, indicating that the involvement of DmRecQ4 in repair is likely to be pathway specific.Deletion analysis of DmRecQ4 suggests that the SLD2 domain was essential, but not sufficient, for replication function. In addition a DmRecQ4 N-terminal deletion could efficiently re-localise on UV treatment, suggesting that the determinants for this response are contained in the C terminus of the protein. Finally several deletions show differential rescue of dsRNA generated replication and proliferation phenotypes. These will be useful for a molecular analysis of the specific role of DmRecQ4 in different cellular pathways

    Critical Crossover Between Yosida-Kondo Dominant Regime and Magnetic Frustration Dominant Regime in the System of a Magnetic Trimer on a Metal Surface

    Quantum Monte Carlo simulations were carried out for the system of a magnetic trimer on a metal surface. The magnetic trimer is arranged in two geometric configurations, viz., isosceles and equilateral triangles. The calculated spectral density and magnetic susceptibility show the existence of two phases: Yosida-Kondo dominant phase and magnetic frustration dominant phase. Furthermore, a critical transition between these two phases can be induced by changing the configuration of the magnetic trimers from isosceles to equilateral triangle.Comment: 8 pages, 4 figures; accepted for publication in J.     Sero-epidemiological survey and risk factors for Hepatitis B Virus (HBV) infection among pregnant women in Logo LGA, Benue State, Nigeria

    Hepatitis B virus (HBV) infection is associated with chronic liver diseases, cirrhosis and hepatocellular carcinoma, liver failure and death. The prevalence of Hepatitis B virus carrier and infectivity status and social characteristics among three hundred and ten pregnant women in Logo LGA, Benue State, Nigeria, was determined through random anonymous testing of volunteers attending antenatal clinics of different hospitals within the community. Thirty of three hundred and ten blood samples tested positive for HBV infection. 11.9% were in the 3rd trimester of their pregnancy and 58.1% were within the age bracket of 21-30 years. Illiterates women constituted 14.4% of those sampled while civil servants were 6.7%. HBV carrier status was determined by the presence of Hepatitis B surface antigen (HBsAg). Repeated reactive samples were confirmed by Enzyme linked immounosorbent assay (ELISA) Kit (Diagnostic Automation, Inc., USA). Maternal HBV infectivity status was determined by testing all HBsAg positive samples for the presence of hepatitis B e antigen (HBeAg). A total of thirty (9.7%) pregnant women identified as carriers of HBV and eleven of the thirty tested positive for HBeAg. Hence, 3.6% (11/310) of the entire study population was found to have high viral replication as well as high risk of transmitting HBV to their neonates. The frequency of HBV carrier did not vary with age, however, it varies significantly with the previous use of contraceptives and the anaemic status of the subjects (P< 0.05) .This study demonstrates the endemicity of HBV infection in Logo and high infectivity rate, suggest that HBV is likely to be acquired by both vertical and horizontal means of transmission. Testing for HBsAg is recommended for all pregnant women at first prenatal visit so that positive mothers receive prompt intervention.Keywords: Hepatitis B e antigen, Hepatitis B surface antigen, seropositivity, pregnant women, neonate

    To the Editor: In many countries, the private sector (practitioners not employed by government and nongovernment institutions, e.g., hospitals, pharmacies) is a major source of care, even for poor persons, and the area where services for the public are widely available (1,2). However, little information is available from high-incidence countries about the role of the private sector in tuberculosis (TB) detection and treatment (3). In Vietnam, ≤40% of all TB cases in Ho Chi Minh City (the largest city in Vietnam and with the highest rate of economic growth in the country) were estimated to be treated in the private sector (4), and half of all patients with a diagnosis of TB in the public sector (National Tuberculosis Program [NTP]) in Ho Chi Minh City initially sought help in the private sector (5).     Miniaturized multisensor system with a thermal gradient: Performance beyond the calibration range

    Two microchips, each with four identical microstructured sensors using SnO2 nanowires as sensing material (one chip decorated with Ag nanoparticles, the other with Pt nanoparticles), were used as a nano-electronic nose to distinguish five different gases and estimate their concentrations. This innovative approach uses identical sensors working at different operating temperatures thanks to the thermal gradient created by an integrated microheater. A system with in-house developed hardware and software was used to collect signals from the eight sensors and combine them into eight-dimensional data vectors. These vectors were processed with a support vector machine allowing for qualitative and quantitative discrimination of all gases after calibration. The system worked perfectly within the calibrated range (100% correct classification, 6.9% average error on concentration value). This work focuses on minimizing the number of points needed for calibration while maintaining good sensor performance, both for classification and error in estimating concentration. Therefore, the calibration range (in terms of gas concentration) was gradually reduced and further tests were performed with concentrations outside these new reduced limits. Although with only a few training points, down to just two per gas, the system performed well with 96% correct classifications and 31.7% average error for the gases at concentrations up to 25 times higher than its calibration range. At very low concentrations, down to 20 times lower than the calibration range, the system worked less well, with 93% correct classifications and 38.6% average error, probably due to proximity to the limit of detection of the sensors

    It is difficult to track the philosophy foundation and epistemology of systemic functional grammar (SFG) formulated by Halliday in the 1980s as this kind of grammar views language as a systemic resource for meaning. Besides, it has had global impacts on linguistics and flourished in contemporary linguistic theory. Anyone who is familiar with Halliday\u27s work realizes that his SFG is an approach designed to analyze English texts. Halliday (1994: xv) explicitly states that “to construct a grammar for purposes of text analysis: one that would make it possible to say sensible and useful things about any text, spoken or written, in modern English.” The aim of this study is not about the applicability of SFG to text analysis as many researchers and scholars do.     Telehealth technology: Potentials, challenges and research directions for developing countries

    Telehealth has been developed and successfully applied in clinical practices, gained a strong interest and demonstrated its usefulness for medical diagnosis, treatments and rehabilitation worldwide. The advent of high speed communication technology and complex signal processing techniques, and recent advancements in cloud and cognitive computing, has created a new wave of opportunities for delivering remote healthcare applications and services, where the cost-effective diagnosis and treatment solutions as well as healthcare services are important and need to be deployed widely. Nevertheless, there is still a significant challenge in fully adopting this technology due to asymmetry among the healthcare centers, hospitals and the user-ends, especially in developing countries. This paper provides an overview of the telehealth, then to addresses the possible telehealth technologies and applications that could be applied to improve the healthcare service performance, with the focus on the developing countries. The incorporation of different technologies in telehealth including, Internet of Things (IoT), cloud and cognitive computing, medical image processing and effective encoding is introduced and discussed. Finally, the possible research directions, challenges for the efficient telehealth, and potential research and technology collaborations are outlined
