    The Development of Xenopus tropicalis Transgenic Lines and their Use in Studying Lens Developmental Timing in Living Embryos

    The generation of reporter lines for observing lens differentiation in vivo demonstrates a new strategy for embryological manipulation and allows us to address a long-standing question concerning the timing of the onset of differentiation. Xenopus tropicalis was used to make GFP reporter lines with &#;1-crystallin promoter elements directing GFP expression within the early lens. X. tropicalis is a close relative of X. laevis that shares the same ease of tissue manipulation with the added benefits of a diploid genome and faster life cycle. The efficiency of the Xenopus transgenic technique was improved in order to generate greater numbers of normal, adult transgenic animals and to facilitate in vivo analysis of the crystallin promoter. This transgene is transmitted through the germline, providing an accurate and consistent way to monitor lens differentiation. This line permitted us to distinguish models for how the onset of differentiation is controlled: by a process intrinsic to differentiating tissue or one dependent on external cues. This experiment would not have been feasible without the sensitivity and accuracy provided by the in vivo reporter. We find that, in specified lens ectoderm transplanted from neural tube stage donors to younger neural-plate-stage hosts, the onset of differentiation, as measured by expression of the crystallin/GFP transgene, is delayed by an average of 4.4 hours. When specified lens ectoderm is explanted into culture, the delay was an average of 16.3 hours relative to control embryos. These data suggest that the onset of differentiation in specified ectoderm can be altered by the environment and imply that this onset is normally controlled by external cues rather than by an intrinsic mechanism

    Let S be a subset of the signature of relation algebra. Let R(S) be the closure under isomorphism of the class of proper S-structures and let F(S) be the closure under isomorphism of the class of proper S-structures over finite bases. Based on previous work, we prove that membership of R(S) is undecidable when (Formula presented), and for any of these signatures S if converse is excluded from S then membership of F(S) is also undecidable, for finite S-structures. We prove that the equational theories of F(S) and R(S) are undecidable when S includes composition and the signature of boolean algebra. If all operators in S are positive and it does not include negation, or if it can define neither domain, range nor composition, then the equational theory of either class is decidable. Open cases for decidability of the equational theory of R(S) are when S can define negation but not meet (or join) and either domain, range or composition. Open cases for the decidability of the equational theory of F(S) are (i) when S can define negation, converse and either domain, range or composition, and (ii) when S contains negation but not meet (or join) and either domain, range or composition

    Structured benefit-risk assessment: a review of key publications and initiatives on frameworks and methodologies.

    Introduction The conduct of structured benefit-risk assessment (BRA) of pharmaceutical products is a key area of interest for regulatory agencies and the pharmaceutical industry. However, the acceptance of a standardized approach and implementation are slow. Statisticians play major roles in these organizations, and have a great opportunity to be involved and drive the shaping of future BRA. Method We performed a literature search of recent reviews and initiatives assessing BRA methodologies, and grouped them to assist those new to BRA in learning, understanding, and choosing methodologies. We summarized the key points and discussed the impact of this emerging field on various stakeholders, particularly statisticians in the pharmaceutical industry. Results We provide introductory, essential, special interest, and further information and initiatives materials that direct readers to the most relevant materials, which were published between 2000 and 2013.  Based on recommendations in these materials we supply a toolkit of advocated BRA methodologies. Discussion Despite initiatives promoting these methodologies, there are still barriers, one of which being the lack of a consensus on the most appropriate methodologies among stakeholders. However, this opens up opportunities, for statisticians in the pharmaceutical industry especially, to champion appropriate BRA methodology use throughout the pharmaceutical product lifecycle. Conclusions This article may serve as a starting point for discussions and to reach a mutual consensus for methodology selection in a particular situation. Regulators and pharmaceutical industry should continue to collaborate to develop and take forward BRA methodologies, and by clear communication develop a mutual understanding of the key issues.     A Mixed-Methods Analysis of the Educational Needs of Employers and Non-English Speaking Workers in Arboriculture

    Arboricultural employers and non-English speaking workers were surveyed to acquire information about their outreach educational needs. Results suggest that language barriers sometimes reduce job performance and threaten worker safety and relationships. Respondents stated that employee performance would improve if workers received English language and cultural training. Method of education mattered, with face-to-face learner and educator contact as the preferred method. It is recommended that arboriculture employers and outreach educators provide English language and cultural outreach education opportunities to arboriculture workers whose first language is not English, then follow this with arboriculture training and information (such as pesticide safety)

    River chloride trends in snow-affected urban watersheds: increasing concentrations outpace urban growth rate and are common among all seasons

    AbstractChloride concentrations in northern U.S. included in this study have increased substantially over time with average concentrations approximately doubling from 1990 to 2011, outpacing the rate of urbanization in the northern U.S. Historical data were examined for 30 monitoring sites on 19 streams that had chloride concentration and flow records of 18 to 49years. Chloride concentrations in most studied streams increased in all seasons (13 of 19 in all seasons; 16 of 19 during winter); maximum concentrations occurred during winter. Increasing concentrations during non-deicing periods suggest that chloride was stored in hydrologic reservoirs, such as the shallow groundwater system, during the winter and slowly released in baseflow throughout the year. Streamflow dependency was also observed with chloride concentrations increasing as streamflow decreased, a result of dilution during rainfall- and snowmelt-induced high-flow periods. The influence of chloride on aquatic life increased with time; 29% of sites studied exceeded the concentration for the USEPA chronic water quality criteria of 230mg/L by an average of more than 100 individual days per year during 2006–2011. The rapid rate of chloride concentration increase in these streams is likely due to a combination of possible increased road salt application rates, increased baseline concentrations, and greater snowfall in the Midwestern U.S. during the latter portion of the study period

    Global Stability of a Premixed Reaction Zone (Time-Dependent Liñan’s Problem)

    Global stability properties of a premixed, three-dimensional reaction zone are considered. In the nonadiabatic case (i.e., when there is a heat exchange between the reaction zone and the burned gases) there is a unique, spatially one-dimensional steady state that is shown to be unstable (respectively, asymptotically stable) if the reaction zone is cooled (respectively, heated) by the burned mixture. In the adiabatic case, there is a unique (up to spatial translations) steady state that is shown to be stable. In addition, the large-time asymptotic behavior of the solution is analyzed to obtain sufficient conditions on the initial data for stabilization. Previous partial numerical results on linear stability of one-dimensional reaction zones are thereby confirmed and extended
