    Directed Network Laplacians and Random Graph Models

    We consider spectral methods that uncover hidden structures in directed networks. We develop a general framework that allows us to associate methods based on optimization formulations with maximum likelihood problems on random graphs. We focus on two existing spectral approaches that build and analyse Laplacian-style matrices via the minimization of frustration and trophic incoherence. These algorithms aim to reveal directed periodic and linear hierarchies, respectively. We show that reordering nodes using the two algorithms, or mapping them onto a specified lattice, is associated with new classes of directed random graph models. Using this random graph setting, we are able to compare the two algorithms on a given network and quantify which structure is more likely to be present. We illustrate the approach on synthetic and real networks, and discuss practical implementation issues

    Developing pedagogy for responsible leadership: Towards a dialogic theory of democratic education

    This thesis explores the connections between dialogue, education and democracy. It begins by asking: ‘what are the implications of dialogic theory for democratic education’? In doing so it draws on concepts from the work of Arendt, Biesta, Dewey and Wegerif: dialogic space as a productive metaphor for education; an ontology of difference in which meaning emerges through dialogue; and authentic democratic action as ‘coming into being’ in negotiation with others. It then asks, ‘Can we teach for democracy?’ by looking at recent practices of citizenship education in Britain. It argues that genuine democratic education must consider students as already being citizens rather than as citizens-in-training, and must offer them opportunities to express their values in action. A theory of ‘responsible leadership’, based on a ‘pedagogy of challenge’, is proposed as a means to enable students to develop the skills and dispositions needed for democratic participation. Short courses in leadership education for teenagers are identified as sites to test this theory. Two empirical studies are detailed, which use a longitudinal case-study approach primarily based on student interviews. The first was a two-day school-based course for academically able 13-18 year olds; the second was a five-day outdoor residential course for 16-18 year olds. Both studies found significant development in students’ skills and dispositions for learning, including: openness to others’ ideas, confidence, greater self-knowledge and better communication skills. In both cases, students’ personal dispositions and insights endured. However, lack of opportunities for democratic action after the courses meant that learned collaborative skills were not strongly embedded; this also meant that ‘responsible leadership’ was not often demonstrated subsequently.     Mycobacterium avium subspecies paratuberculosis:human exposure through environmental and domestic aerosols

    Mycobacterium avium subspecies paratuberculosis (Map) causes Johne’s disease in animals and is significantly associated with Crohn’s disease (CD) in humans. Our previous studies have shown Map to be present in U.K. rivers due to land deposition from chronic livestock infection and runoff driven by rainfall. The epidemiology of CD in Cardiff showed a significant association with the River Taff, in which Map can be detected on a regular basis. We have previously hypothesized that aerosols from the river might influence the epidemiology of CD. In this preliminary study, we detected Map by quantitative PCR in one of five aerosol samples collected above the River Taff. In addition, we examined domestic showers from different regions in the U.K. and detected Map in three out of 30 independent samples. In detecting Map in river aerosols and those from domestic showers, this is the first study to provide evidence that aerosols are an exposure route for Map to humans and may play a role in the epidemiology of CD

    Respiratory viral infection alters the gut microbiota by inducing inappetence

    ABSTRACT Respiratory viral infections are extremely common, but their impacts on the composition and function of the gut microbiota are poorly understood. We previously observed a significant change in the gut microbiota after viral lung infection. Here, we show that weight loss during respiratory syncytial virus (RSV) or influenza virus infection was due to decreased food consumption, and that the fasting of mice altered gut microbiota composition independently of infection. While the acute phase tumor necrosis factor alpha (TNF-α) response drove early weight loss and inappetence during RSV infection, this was not sufficient to induce changes in the gut microbiota. However, the depletion of CD8+ cells increased food intake and prevented weight loss, resulting in a reversal of the gut microbiota changes normally observed during RSV infection. Viral infection also led to changes in the fecal gut metabolome, with a significant shift in lipid metabolism. Sphingolipids, polyunsaturated fatty acids (PUFAs), and the short-chain fatty acid (SCFA) valerate were all increased in abundance in the fecal metabolome following RSV infection. Whether this and the impact of infection-induced anorexia on the gut microbiota are part of a protective anti-inflammatory response during respiratory viral infections remains to be determined. IMPORTANCE The gut microbiota has an important role in health and disease: gut bacteria can generate metabolites that alter the function of immune cells systemically. Understanding the factors that can lead to changes in the gut microbiome may help to inform therapeutic interventions. This is the first study to systematically dissect the pathway of events from viral lung infection to changes in gut microbiota. We show that the cellular immune response to viral lung infection induces inappetence, which in turn alters the gut microbiome and metabolome. Strikingly, there was an increase in lipids that have been associated with the resolution of disease. This opens up new paths of investigation: first, what is the (presumably secreted) factor made by the T cells that can induce inappetence? Second, is inappetence an adaptation that accelerates recovery from infection, and if so, does the microbiome play a role in this

    Finding the optimal background subtraction algorithm for EuroHockey 2015 video

    Background subtraction is a classic step in a vision-based localization and tracking workflow. Previous studies have compared background subtraction algorithms on publicly available datasets; however comparisons were made only with manually optimized parameters. The aim of this research was to identify the optimal background subtraction algorithm for a set of field hockey videos captured at EuroHockey 2015. Particle Swarm Optimization was applied to find the optimal background subtraction algorithm. The objective function was the F-score, i.e. the harmonic mean of precision and recall. The precision and recall were calculated using the output of the background subtraction algorithm and gold standard labeled images. The training dataset consisted of 15 x 13 second field hockey video segments. The test data consisted of 5 x 13 second field hockey video segments. The video segments were chosen to be representative of the teams present at the tournament, the times of day the matches were played and the weather conditions experienced. Each segment was 960 pixels x 540 pixels and had 10 ground truth labeled frames. Eight commonly used background subtraction algorithms were considered. Results suggest that a background subtraction algorithm must use optimized parameters for a valid comparison of performance. Particle Swarm Optimization is an appropriate method to undertake this optimization. The optimal algorithm, Temporal Median, achieved an F-score of 0.791 on the test dataset, suggesting it generalizes to the rest of the video footage captured at EuroHockey 2015
